Silver Nimesulide Complex in Bacterial Cellulose Membranes as an Innovative Therapeutic Method for Topical Treatment of Skin Squamous Cell Carcinoma.

ABSTRACT: Oxidative stress and inflammation act on skin squamous cell carcinoma (SSCC) development and progression. Curative therapy for SSCC patients is mainly based on surgical resection, which can cause various sequelae. Silver ions have in vitro activities over tumor cells, while nimesulide has antioxidant and anti-inflammatory activities. This study aimed to evaluate the effects of a silver(I) complex with nimesulide (AgNMS) incorporated in a sustained release device based on bacterial cellulose membrane, named AgNMS@BCM, on topic SSCC treatment. The antiproliferative effect of AgNMS complex was evaluated in the SCC4, SCC15 and FaDu SCC lines. AgNMS complex activity on exposure of phosphatidylserine (PS) residues and multicaspase activation were evaluated on FaDu cells by flow cytometry. The AgNMS@BCM effects were evaluated in a SSCC model induced by 7,12-dimethylbenzanthracene/12-o-tetradecanoyl-phorbol-13-acetate (DMBA/TPA) in mice. Toxicity and tumor size were evaluated throughout the study. AgNMS complex showed antiproliferative activity in SCC15 and FaDu lines in low to moderate concentrations (67.3 µM and 107.3 µM, respectively), and induced multicaspase activation on FaDu cells. The AgNMS@BCM did not induce toxicity and reduced tumor size up to 100%. Thus, the application of AgNMS@BCM was effective and safe in SSCC treatment in mice, and can be seen as a potential and safe agent for topic treatment of SSCC in humans.

Project description:In recent years, inorganic metal nanoparticle fabrication by extraction of a different part of the plant has been gaining more importance. In this research, cellulose-mediated Ag nanoparticles (cellulose/Ag NPs) with excellent antibacterial and antioxidant properties and photocatalytic activity have been synthesized by the microwave-assisted hydrothermal method. This method is a green, simple, and low-cost method that does not use any other capping or reducing agents. X-ray diffraction (XRD), Fourier transform infrared (FTIR), field emission scanning microscopy (FESEM), transmission electron microscopy (TEM), energy-dispersive X-ray (EDX), and UV-visible spectroscopic techniques were used to investigate the structure, morphology, as well as components of the generated cellulose/Ag NPs. In fact, XRD results confirm the formation of the face-centered cubic phase of Ag nanoparticles, while the FTIR spectra showed that the synergy of carbohydrates and proteins is responsible for the formation of cellulose/Ag NPs by the green method. It was found that the green-synthesized silver nanoparticles showed good crystallinity and a size range of about 20-30 nm. The morphology results showed that cellulose has a cavity-like structure and the green-synthesized Ag NPs were dispersed throughout the cellulose polymer matrix. In comparison to cellulose/Ag NPs and Ag nanoparticles, cellulose/Ag NPs demonstrated excellent antibacterial activity, Proteus mirabilis (MTCC 1771) possessed a maximum inhibition zone of 18.81.5 mm at 2.5 g/mL, and Staphylococcus aureus (MTTC 3615) had a minimum inhibition zone of 11.30.5 mm at 0.5 g/mL. Furthermore, cellulose/Ag NPs also exhibited a significant radical scavenging property against the DDPH free radical, and there was a higher degradation efficiency compared to pure Ag NPs against Rhodamine B as 97.38% removal was achieved. Notably, cellulose/Ag NPs remarkably promoted the transfer and separation of photogenerated electron-hole (e-/h+) pairs, thereby offering prospective application of the photodegradation efficiency for Rhodamine B (RhB) as well as antibacterial applications. With the findings from this study, we could develop efficient and environmentally friendly cellulose/Ag nanoparticles using low-cost, environmentally friendly materials, making them suitable for industrial and technological applications.

Project description:Even though cutaneous atrophy is the major adverse effect of topical glucocorticoids, its molecular mechanisms are poorly understood. We found that glucocorticoids strongly increased the expression of REDD1 (regulated in development and DNA damage response 1), a stress-inducible inhibitor of mTOR, in mouse and human epidermis. We found that REDD1 knockout animals are partially resistant to glucocorticoid-induced epidermal and subcutaneous adipose atrophy which correlated with the protection of CD34+ follicular epithelial stem cells as well as p63+ keratinocyte progenitors in REDD1 knockout epidermis during chronic steroid treatment. At the same time, REDD1 knockout did not affect anti- inflammatory effect of glucocorticoids, as evaluated by ear edema test. Expression profiling revealed that ~ 50% of the glucocorticoid receptor (GR) target genes were not activated in epidermis of REDD1 knockouts, however, the negative effect of glucocorticoids on gene expression was similar to that in w.t. animals. Overall, our findings reveal a novel pathway in GR activation by its target gene/protein REDD1; and indicate an important role of REDD1 in glucocorticoid-induced skin atrophy, and maintenance of the epidermis and subcutaneous adipose. In addition, our findings form the foundation for the development of safer topical glucocorticoid treatment regimens by combining this therapy with REDD1 inhibitors. Seven wk old B6D2 females in the telogen stage of the hair cycle were shaved, and treated 3 days later. Glucocorticoid FA was applied topically (2 ug/animal) in 200 ul acetone to the back skin once or up to four applications every third day as described previously (Chebotaev et al., 2007b). Control animals were treated with acetone only. Skin was harvested 24 h after last FA application or 4-8 h after single FA application as indicated in Figure legends. To evaluate the anti-inflammatory effect of glucocorticoid FA in REDD1 KO and w.t. B6x129 mice, we used ear edema test (Schoepe et al., 2010; Schäcke et al., 2004; Park et al., 2006). Seven wk old female animals were pretreated with FA or vehicle applied to the back of the ear lobe one h before application of nonspecific contact irritant croton oil . Mice were sacrificed and ears were harvested nine hours after CO application, the time point at which we observed maximum ear edema in F1 C57Bl x 129 animals (data not shown). Four mm ear punch biopsies were immediately weighted to assess ear edema. Animals were injected i.p. with bromodeoxyuridine 1 hr before skin was harvested. Total RNA from murine epidermis and whole human skin was isolated with RiboPure kit (Ambion). Total RNA from murine s.c. adipose was isolated with RNeasy Lipid Tissue Kit (Qiagen). The RNA samples were treated with TURBOTM DNase (Ambion). Purity of RNA samples isolated from epidermis and s.c. fat was confirmed by the expression of adipose and keratinocyte cell markers.
