Infrared Laser Ablation Microsampling with a Reflective Objective.

ABSTRACT: A Schwarzschild reflective objective with a numerical aperture of 0.3 and working distance of 10 cm was used for laser ablation sampling of tissue for off-line mass spectrometry. The objective focused the laser to a diameter of 5 μm and produced 10 μm ablation spots on thin ink films and tissue sections. Rat brain tissue sections 50 μm thick were ablated in transmission geometry, and the ablated material was captured in a microcentrifuge tube containing solvent. Proteins from ablated tissue sections were quantified with a Bradford assay, which indicated that approximately 300 ng of protein was captured from a 1 mm² area of ablated tissue. Areas of tissue ranging from 0.01 to 1 mm² were ablated and captured for bottom-up proteomics. Proteins were extracted from the captured tissue and digested for liquid chromatography tandem mass spectrometry (LC-MS/MS) analysis for peptide and protein identification.

Improved spatial resolution of infrared matrix-assisted laser desorption electrospray ionization mass spectrometry imaging using a reflective objective.

Project description:RationaleThe level of visual detail of a mass spectrometry image is dependent on the spatial resolution with which it is acquired, which is largely determined by the focal diameter in infrared laser ablation-based techniques. While the use of mid-IR light for mass spectrometry imaging (MSI) has advantages, it results in a relatively large focal diameter and spatial resolution. The continual advancement of infrared matrix-assisted electrospray ionization (IR-MALDESI) for MSI warranted novel methods to decrease laser ablation areas and thus improve spatial resolution.MethodsIn this work, a Schwarzschild-like reflective objective was incorporated into the novel NextGen IR-MALDESI source and characterized on both burn paper and mammalian tissue using an ice matrix. Ablation areas, mass spectra, and annotations obtained using the objective were compared against the current optical train on the NextGen system without modification.ResultsThe effective resolution was determined to be 55 μm by decreasing the step size until oversampling was observed. Use of the objective improved the spatial resolution by a factor of three as compared against the focus lens.ConclusionsA Schwarzschild-like reflective objective was successfully incorporated into the NextGen source and characterized on mammalian tissue using an ice matrix. The corresponding improvement in spatial resolution facilitates the future expansion of IR-MALDESI applications to include those that require fine structural detail.

Mid-infrared absorption by soft tissue sarcoma and cell ablation utilizing a mid-infrared interband cascade laser.

Project description:Mid-infrared (MIR) light refers to wavelengths ranging from 3 to 30 μm and is the most attractive spectral region for ablation of soft and hard tissues. This is because building blocks of biological tissue, such as water, proteins, and lipids, exhibit molecular vibrational modes in the MIR wavelengths that result in strong MIR light absorption. To date, researchers investigating MIR lasers for surgical applications have used bulky light sources, such as free electron lasers, nonlinear light generators, and carbon dioxide lasers. We demonstrate the use of a tiny (a few microns wide, a few millimeters long) MIR interband cascade laser (ICL) for surgical thermal ablation applications. Our goal is to demonstrate the use of an ICL for surgical thermal ablation and demonstrate its efficacy in ablating normal fibroblasts and primary undifferentiated pleomorphic sarcoma tumor cells (C1619). We conducted Fourier transform infrared spectroscopy analysis of healthy and cancerous tissue samples, which indicated that the absorption of tumor tissue is higher than healthy tissue around 3.3-μm wavelength. These results enabled us to select an ICL emission wavelength, λ, of 3.3 μm to probe normal fibroblast and primary undifferentiated pleomorphic sarcoma cell survival after ICL exposure. We show that the absorption of tumorous tissue is higher than that of healthy tissues around the 3-μm MIR wavelength. We demonstrate that the ICL is able to ablate cancer cells at very low-power levels that can be clinically implemented but that this effect does not appear to be specific to C1619 when compared to normal fibroblasts. Our study demonstrates that ICLs may represent an exciting new avenue toward precise laser-based thermal ablation.

Selective induction of targeted cell death and elimination by near-infrared femtosecond laser ablation.

Project description:The techniques for inducing the death of specific cells in tissue has attracted attention as new methodologies for studying cell function and tissue regeneration. In this study, we show that a sequential process of targeted cell death and removal can be triggered by short-term exposure of near-infrared femtosecond laser pulses. Kinetic analysis of the intracellular accumulation of trypan blue and the assay of caspase activity revealed that femtosecond laser pulses induced immediate disturbance of plasma membrane integrity followed by apoptosis-like cell death. Yet, adjacent cells showed no sign of membrane damage and no increased caspase activity. The laser-exposed cells eventually detached from the substrate after a delay of >54 min while adjacent cells remained intact. On the base of in vitro experiments, we applied the same approach to ablate targeted single cardiac cells of a live zebrafish heart. The ability of inducing targeted cell death with femtosecond laser pulses should find broad applications that benefit from precise cellular manipulation at the level of single cells in vivo and in vitro.

Single-Shot Multi-Stage Damage and Ablation of Silicon by Femtosecond Mid-infrared Laser Pulses.

Project description:Although ultrafast laser materials processing has advanced at a breakneck pace over the last two decades, most applications have been developed with laser pulses at near-IR or visible wavelengths. Recent progress in mid-infrared (MIR) femtosecond laser source development may create novel capabilities for material processing. This is because, at high intensities required for such processing, wavelength tuning to longer wavelengths opens the pathway to a special regime of laser-solid interactions. Under these conditions, due to the λ2 scaling, the ponderomotive energy of laser-driven electrons may significantly exceed photon energy, band gap and electron affinity and can dominantly drive absorption, resulting in a paradigm shift in the traditional concepts of ultrafast laser-solid interactions. Irreversible high-intensity ultrafast MIR laser-solid interactions are of primary interest in this connection, but they have not been systematically studied so far. To address this fundamental gap, we performed a detailed experimental investigation of high-intensity ultrafast modifications of silicon by single femtosecond MIR pulses (λ = 2.7-4.2 μm). Ultrafast melting, interaction with silicon-oxide surface layer, and ablation of the oxide and crystal surfaces were ex-situ characterized by scanning electron, atomic-force, and transmission electron microscopy combined with focused ion-beam milling, electron diffractometry, and μ-Raman spectroscopy. Laser induced damage and ablation thresholds were measured as functions of laser wavelength. The traditional theoretical models did not reproduce the wavelength scaling of the damage thresholds. To address the disagreement, we discuss possible novel pathways of energy deposition driven by the ponderomotive energy and field effects characteristic of the MIR wavelength regime.

New design for highly durable infrared-reflective coatings.

Project description:The fundamental challenge in designing durable infrared-reflective coatings is achieving the ideal combination of both high reflectivity and durability. Satisfying these competing demands is traditionally achieved by deposition of durable layers on highly reflective metals. We overturn the traditional logic of 'first reflectivity and then durability' and propose an alternative of 'first durability and then reflectivity': First, a transition-metal compound is selected as a durable base; then its reflectivity is improved by incorporating silver/gold to form an alloy or by overcoating a multilayer stack. Two validation experiments prove that the new strategy works extremely well: the coatings thus obtained have infrared reflectivities close to that of aluminum, and their hardness and acid and salt corrosion resistances are 27-50, 400-1 500 and 7 500-25 000 times that of aluminum. The traditional mirror coating (e.g., Al/SiO2 films) is more suitable for moderate environments, while our mirror coating that was obtained by the new strategy (e.g., an Ag-doped hafnium nitride film) is more suitable for harsh environments, such as ones with dust, windblown sand, moisture, acid rain or salt fog. This work opens up new opportunities for highly durable infrared-reflective coatings and rejuvenates the study of transition metal compounds in a completely new area of optics.

Easily Processable Temperature-Responsive Infrared-Reflective Polymer Coatings.

Project description:A temperature-responsive near-infrared reflective coating was fabricated based on a side-chain liquid crystal siloxane polymer using a simple wired-bar method. The cholesteric liquid crystalline polymer film showed a blue shift of the reflection band of ∼1000 nm in the IR region upon heating. The temperature-responsive change of the reflection band was reversible. Compared to that of the same mixture system in an alignment cell, the coating showed a significantly faster response. This research demonstrates an easy way to prepare a temperature-responsive IR-reflective coating that shifts its reflection to a shorter wavelength upon heating. As IR radiation of shorter wavelengths is more strongly represented in sunlight than longer wavelengths, this coating could be used to selectively reduce heating of an indoor space when the temperature is high. This is promising for the future application of smart climate control.

Continuous preparation of antimony nanocrystals with near infrared photothermal property by pulsed laser ablation in liquids.

Project description:Antimony nanocrystals (Sb NCs) are of interest in energy storage, catalysis and cancer therapy for its special physical, chemical and biomedical properties. However, methodology challenges still remain in preparation of colloidal Sb NCs, due to the restricted reaction solution systems, high temperature and time costing for common routes. Herein, size controllable colloidal Sb NCs were continuously prepared by pulsed laser ablation of Sb target in different solvents, owning to the metal nanodroplet explosive ejection and thermal evaporation mechanisms. These well dispersed and stable Sb NCs showed excellent photothermal property in the near-infrared-II window.
