Effects of Temephos (Abate®), Spinosad (Natular®), and Diflubenzuron on the Survival of Cyclopoid Copepods.

ABSTRACT: Dracunculus medinensis, also known as the African Guinea worm, is the causative agent of dracunculiasis and the focus of the global Guinea Worm Eradication Program (GWEP). Transmission of D. medinensis to humans occurs primarily by drinking water containing cyclopoid copepods infected with third-stage D. medinensis larvae. A common intervention to interrupt transmission and decrease the number of copepods in infected water bodies is the application of the organophosphate larvicide Abate® (temephos). However, the use of alternative compounds to help decrease copepod populations would be beneficial to the GWEP. We compared the immobilization of copepods by three compounds: Abate, Natular® (spinosad), and diflubenzuron. Our results confirm that neither diflubenzuron nor Natular immobilized copepods as quickly or as effectively as Abate. However, doubling or tripling the suggested concentration of Natular resulted in immobilization rates similar to Abate over 72 hours of continuous exposure. Further research on the possible effects of higher concentrations of Natular on the environment and nontarget organisms is necessary to determine whether this compound can be used safely to control the copepod population.

Project description:BackgroundThe control of the Asian tiger mosquito Aedes albopictus (Diptera: Culicidae) is crucial owing to its high vector competence for more than 20 arboviruses-the most important being dengue, chikungunya and Zika virus. Aedes albopictus has an enormous adaptive potential, and its invasive spreading across urban and suburban environments poses challenges for its control. Therefore, all suitable, cost-effective and eco-friendly control tools should be put into practice. In this context, cyclopoid copepods are already known as effective predators of mosquito larvae. This study reports an essential preliminary step towards the integration of copepods into the vector control strategy in Germany, in order to provide a sustainable tool in an integrated control strategy based on the elimination or sanitation of breeding sites, the use of formulations based on Bacillus thuringiensis israelensis (Bti.) and the sterile insect technique (SIT).MethodsThe predatory potential of native cyclopoid copepods, namely the field-derived species Megacyclops viridis (Crustacea: Cyclopidae), was examined against the larvae of Ae. albopictus, and for comparison, against the larvae of the common house mosquito, Culex pipiens sensu lato (Diptera: Culicidae). The use of different larval instars as prey, and various predator-to-prey ratios, were examined under laboratory and semi-field conditions. The compatibility of Bti. applications along with the use of copepods was assessed in the laboratory.ResultsHigh predation efficiency of M. viridis upon first-instar larvae of Ae. albopictus was observed under laboratory (up to 96%) and semi-field conditions (65.7%). The copepods did not prey upon stages further developed than the first instars, and in comparison with Ae. albopictus, the predation rates on the larvae of Cx. pipiens s.l. were significantly lower.ConclusionsThe results indicate a high predation potential of M. viridis against Ae. albopictus larvae, even though strong larval stage and mosquito species preferences were implicated. The integration of copepods as a promising biocontrol agent to the vector control strategy in Germany is therefore highly recommended, especially because of the excellent compatibility of copepods with the use of Bti. However, further research is required, concerning all the probable parameters that may impact the copepod performance under natural conditions.

Project description:BackgroundThe larvicidal efficacy of the naturally derived insecticide spinosad, for control of immature stages of Anopheles albimanus and associated culicids, was compared to that of synthetic and biological larvicides. Effects on non-target insects were also determined.MethodsA field trial was performed in replicated temporary pools during the rainy season, in southern Mexico. Pools were treated with 10 ppm a.i. spinosad (Tracer 480SC), Bti granules applied at 2 kg/ha (VectoBac WDG, ABG-6511), and 100 ml/ha temephos (50 EC), or an untreated control. Numbers of immature mosquitoes, and aquatic insects in pools were monitored for 20 weeks.ResultsSamples of immature mosquitoes comprised approximately 10% An. albimanus, 70% Culex spp. (mostly Cx. melanoconion and Cx. coronator) and 20% Uranotaenia lowii. The most effective larvicides were spinosad and temephos that eliminated An. albimanus in 16 out of 20 post-treatment samples, or 9 weeks of continuous control of immature stages, respectively. These larvicides resulted in 15 and 5 weeks of elimination of Culex spp., respectively, or 20 and 4 weeks of continuous elimination of U. lowii, respectively. Bti treatment provided little consistent control. Aquatic insects were recorded comprising 3 orders, 20 families, 40 genera and 44 species. Shannon diversity index values (H') for aquatic insects were highest in the control (0.997) and Bti (0.974) treatments, intermediate in the spinosad treatment (0.638) and lowest in the temephos treatment (0.520). Severely affected non-target insects in the spinosad and temephos treated pools were predatory Coleoptera, Hemiptera and Odonata, which in the case of spinosad was likely due to the high concentration applied. Bti had little effect on aquatic insects.ConclusionsThe spinosad treatment retained larvicidal activity for markedly longer than expected. Spinosad is likely to be an effective tool for control of anopheline and other pool-breeding mosquitoes in tropical regions. Non-target effects of spinosad on aquatic insects merit further study, but were likely related to the concentration of the product used.
