Effects of integrated nutrient management on sucrose phosphate synthase enzyme activity and grain quality traits in rice.

ABSTRACT: Cropping systems and management practices can greatly affect rice crop, finally determining their grain yield and quality. In this study, we evaluated the effects of integrated nutrient management practices on sucrose phosphate synthase enzyme (SPS) activity and grain quality traits of rice. Field experiments were conducted at North Bank Plain Agro-climatic Zone of Assam, India from 2013 to 2015 on three rice ecosystems (winter, summer, and pre-monsoon). Selected fertilizer treatments were NPK as recommended inorganic fertilizer, NPK + cow dung, NPK + whole parts of the green manure Sesbania aculeata, NPK + compost of Azolla caroliniana and NPK + rice husk dust. NPK + Azolla compost application resulted in higher SPS activity compared to control. A significant relationship between panicle biomass and SPS enzyme activity was observed in the rice plants grown in different ecosystems. Integrated nutrient fertilizers in rice soil had a significant impact on the grain quality of rice. Grain nitrogen and crude protein content were higher at NPK + green manure Sesbania aculeata applied field irrespective of the ecosystems. NPK and Azolla compost were effective in improving grain productivity and grain quality parameters viz; total carbohydrates, starch, and amylose in rice crop.

Project description:BackgroundSucrose phosphate synthase B (SPSB) gene encoding an important rate-limiting enzyme for sucrose synthesis in sugarcane is mainly expressed on leaves, where its alleles control sucrose synthesis. In this study, genetic variation of SPSB gene represented by different haplotypes in sugarcane was investigated in hybrid clones with high and low sugar content and various accessory species.ResultsA total of 39 haplotypes of SPSB gene with 2, 824 bp in size were identified from 18 sugarcane accessions. These haplotypes mainly distributed on Chr3B, Chr3C, and Chr3D according to the AP85-441 reference genome. Single nucleotide polymorphisms (SNPs) and insertion/deletion (InDels) were very dense (42 bp/sequence variation) including 44 transitional and 23 transversional SNPs among the 39 haplotypes. The sequence diversity related Hd, Eta, and Pi values were all lower in clones of high sucrose content (HS) than those in clones of low sucrose content (LS). The evolutionary network analysis showed that about half SPSB haplotypes (19 out of 39) were clustered into one group, including 6 (HAP4, HAP6, HAP7, HAP9, HAP17 and HAP20) haplotypes under positive selection in comparison to HAP26 identified in Badila (S. officinarum), an ancestry noble cane species and under purification selection (except HAP19 under neutral selection) in comparison to HAP18 identified from India1 (S. spontaneum), an ancestry species with low sugar content but high stress tolerance. The average number of haplotypes under positive selection in HS clones was twice as that in LS. Most of the SNPs and InDels sequence variation sites were positively correlated with sucrose and fiber content and negatively correlated with reducing sugar.ConclusionsA total of 39 haplotypes of SPSB gene were identified in this study. Haplotypes potentially associated with high sucrose synthesis efficiency were identified. The mutations of SPSB haplotypes in HS were favorable and tended to be selected and fixed. The results of this study are informative and beneficial to the molecular assisted breeding of sucrose synthesis in sugarcane in the future.

Project description:Super hybrid rice genotypes have transformed the rate of genetic yield gain primarily due to intersubspecific heterosis, although the physiological basis underpinning this yield transformation has not been well quantified. We assessed the radiation use efficiency (RUE) and nitrogen use efficiency (NUE) of novel hybrid rice genotypes under four management practices representative of rice cropping systems in China. Y-liangyou 900 (YLY900), a new super hybrid rice widely adopted in China, was examined in field experiments conducted in Jingzhou and Suizhou, Hubei Province, China, from 2017 to 2020. Four management practices were conducted: nil fertilizer (CK), conventional farmer practice (FP), optimized cultivation with reduced nitrogen (OPT-N), and optimized cultivation with increased nitrogen (OPT+N). Yield differences across the treatment regimens were significant (p < 0.05). Grain yield of OPT+N in Jingzhou and Suizhou were 11 and 12 t ha-1, which was 14 and 27% greater than yields obtained under OPT-N and FP, respectively. Relative to OPT-N and FP, OPT+N had greater panicle numbers (9 and 18%), spikelets per panicle (7 and 12%), spikelets per unit area (17 and 32%), and total dry weight (9 and 19%). The average RUE of OPT+N was 2.7 g MJ-1, which was 5 and 9% greater than that of OPT-N and FP, respectively, due to higher intercepted photosynthetically active radiation (IPAR). The agronomic efficiency of applied N (AEN) of OPT+N was 17 kg grain kg-1 N, which was 9 and 68% higher than that of OPT-N and FP. These results show that close correlations exist between yield and both the panicles number (R 2 = 0.91) and spikelets per panicle (R 2 = 0.83) in OPT+N. We conclude that grain yields of OPT+N were associated with greater IPAR, RUE, and total dry matter. We suggest that integrated cropping systems management practices are conducive to higher grain yield and resource use efficiency through expansion of sink potential in super hybrid rice production.
