Project description:To reveal the characteristics of climate change and the controlling factors for vegetation dynamics in the Ordos, Inner Mongolia, China, 34 years (1982-2015) of regional climate variables and vegetation dynamics were investigated. The results show that: Annual mean air temperature (TMP) significantly increased with a linear slope of 0.473°C/10yr. Annual precipitation (PRE) had a non-significant positive trend nearly 5 times lower than the trend of potential evapotranspiration (PET). The average Normalized Difference Vegetation Index (NDVI) computed for the region was found to show a significant positive trend (6.131×10-4/yr). However, all climate variables displayed non-significant correlations with NDVI at annual scale. The reduction of desert and the increase of grassland over the past decades were accountable for the increased NDVI. Principal components analysis revealed that the regional climate change can be characterized as changes in temperature, humidity and the availability of radiant energy. Based on principal components regression coefficients, NDVI was mostly sensitive to humidity component, followed by growing season warmth (WMI). Spatially, 93.1% of the pixels displayed positive trend and 61.8% of the pixels displayed significant change over the past decades. Both principal regression analysis and partial correlation analysis revealed that NDVI in eastern part of Ordos was sensitive to TMP, whereas, NDVI in southern and western areas of Ordos displayed the high sensitivity to combined effects of PRE and cloud coverage (CLD). Partial correlation analyses also revealed that TMX was a surrogate for aridity, TMN was a representative of humidity, and temperature variations below the threshold of 5°C (CDI) were less important than WMI. We conclude that regional climate change can be characterized by warming and increased aridity. The significant positive trend of regional NDVI and the non-significant correlations between NDVI and climate variables at annual scale suggests the hidden role of the human activities.

Project description:Soil moisture content (SWC) can change the diversity and composition of soil fungal communities by affecting soil texture and soil nutrients. To explore the response of soil fungal communities to moisture in the grassland ecosystem on the south shore of Hulun Lake, we set up a natural moisture gradient that was subdivided into high (HW), medium (MW), and low (LW) water contents. Vegetation was investigated by quadrat method, and aboveground biomass was collected by the mowing method. Soil physicochemical properties were obtained by internal experiments. The composition of the soil fungal community was determined using high-throughput sequencing technology. The results showed significant differences in soil texture, nutrients, and fungal species diversity under the moisture gradients. Although there was significant clustering of fungal communities in different treatments, the fungal community composition was not significantly different. According to the phylogenetic tree, the Ascomycota and Basidiomycota were the most important branches. The fungal species diversity was smaller when SWC was higher, and in this environment (HW), the fungal-dominant species were significantly related to SWC and soil nutrients. At this time, soil clay formed a protective barrier for the survival of the dominant classes Sordariomycetes and Dothideomycetes and increased their relative abundance. In summary, the fungal community responded significantly to SWC on the southern shore of the Hulun Lake ecosystem in Inner Mongolia, China, and the fungal community composition of the HW group was stable and easier to survive.

Project description:BACKGROUND:Human brucellosis incidence in China was divided into 3 stages, high incidence (1950-1960s), decline (1970-1980s) and re-emergence (1990-2000s). Human brucellosis has been reported in all the 32 provinces, of which Inner Mongolia has the highest prevalence, accounting for over 40% of the cases in China. To investigate the etiology alteration of human brucellosis in Inner Mongolia, the species, biovars and genotypes of 60 Brucella isolates from this province were analyzed. METHODS:Species and biovars of the Brucella strains isolated from outbreaks were determined based on classical identification procedures. Strains were genotyped by multi locus sequence typing (MLST). Sequences of 9 housekeeping genes were obtained and sequence types were defined. The distribution of species, biovars and sequence types (STs) among the three incidence stages were analyzed and compared. RESULTS:The three stages of high incidence, decline and re-emergence were predominated by B. melitensis biovar 2 and 3, B. abortus biovar 3, and B. melitensis biovar 1, respectively, implying changes in the predominant biovars. Genotyping by MLST revealed a total of 14 STs. Nine STs (from ST28 to ST36), accounting for 64.3% of all the STs, were newly defined and different from those observed in other countries. Different STs were distributed among the three stages. ST8 was the most common ST in 1950-1960s and 1990-2000s, while ST2 was the most common in 1970-1980s. CONCLUSIONS:The prevalence of biovars and sequence types of Brucella strains from Inner Mongolia has changed over time in the three stages. Compared with those from other countries, new sequence types of Brucella strains exist in China.

Project description:BACKGROUND: Batai virus (BATV) is a member of the Orthobunyavirus genus of the family Bunyaviridae, and a vector-borne pathogen. Genomic variations of BATV exist in different regions of the world, due to genetic reassortment. Whole-genome sequencing of any isolate is necessary for a phylogenetic analysis. In 1998, a BATV strain was isolated from an Anopheles philippines mosquito in Yunnan Province, China. This strain has not been found to infect any other host. We investigated BATV infection in cattle in Inner Mongolia, China and performed deep sequencing of the genome of the BATV isolate. FINDINGS: Ninety-five blood samples were collected from cattle in Inner Mongolia, China in 2012. The BATV infection rate was 2.1%. Previously, BATV strain NM/12 was isolated from two cattle in Inner Mongolia, China, and the whole genomic sequence of the strain has been available. We determined the complete genomic nucleotide sequences of the small (S), medium (M), and large (L) genome segments using bovine blood obtained in 2012, and the nucleotide homologies of these segments with those from GenBank were 88.7%-97%, 84%-95.4%, and 72.6%-95.8%, respectively. The deduced amino acid identities were 87.2-99.7%, 64.2-96.8%, and 81.1-98.6%. Phylogenetic analyses based on full-length genomic sequences indicated that the M and L segments, and a portion of the S segment, of NM/12 are most closely related to the BATV strains isolated in Asia. The S and M segments of NM/12 were independent of phylogenetic lineages. The L segment was the most closely related to Chittoor/IG-20217 (isolated in India), and distantly related to isolated strains in Italy. Nucleotide substitution rates in the nucleotide sequences that code for the nucleocapsid, envelope glycoprotein, and polymerase protein of NM/12 strain were 2.56%, 4.69%, and 4.21%, respectively, relative to the original strain of MM2222. CONCLUSION: A novel BATV NM/12 strain from bovine serum collected in Inner Mongolia was isolated from cattle in China for the first time. Our findings elucidate the evolutionary status of the BATV NM/12 strain among different orthobunyavirus strains and may provide some clues to prevent the emergence of BATV infection in cattle and humans.

Project description:Arid grassland ecosystems have significant interannual variation in carbon exchange; however, it is unclear how environmental factors influence carbon exchange in different hydrological years. In this study, the eddy covariance technique was used to investigate the seasonal and interannual variability of CO? flux over a temperate desert steppe in Inner Mongolia, China from 2008 to 2010. The amounts and times of precipitation varied significantly throughout the study period. The precipitation in 2009 (186.4 mm) was close to the long-term average (183.9±47.6 mm), while the precipitation in 2008 (136.3 mm) and 2010 (141.3 mm) was approximately a quarter below the long-term average. The temperate desert steppe showed carbon neutrality for atmospheric CO? throughout the study period, with a net ecosystem carbon dioxide exchange (NEE) of -7.2, -22.9, and 26.0 g C m?² yr?¹ in 2008, 2009, and 2010, not significantly different from zero. The ecosystem gained more carbon in 2009 compared to other two relatively dry years, while there was significant difference in carbon uptake between 2008 and 2010, although both years recorded similar annual precipitation. The results suggest that summer precipitation is a key factor determining annual NEE. The apparent quantum yield and saturation value of NEE (NEE(sat)) and the temperature sensitivity coefficient of ecosystem respiration (R(eco)) exhibited significant variations. The values of NEE(sat) were -2.6, -2.9, and -1.4 µmol CO? m?² s?¹ in 2008, 2009, and 2010, respectively. Drought suppressed both the gross primary production (GPP) and R(eco), and the drought sensitivity of GPP was greater than that of R(eco). The soil water content sensitivity of GPP was high during the dry year of 2008 with limited soil moisture availability. Our results suggest the carbon balance of this temperate desert steppe was not only sensitive to total annual precipitation, but also to its seasonal distribution.

Project description:BackgroundWe are profoundly ignorant about the diversity of viruses that infect the domain Archaea. Less than 100 have been identified and described and very few of these have had their genomic sequences determined. Here we report the genomic sequence of a previously undescribed archaeal virus.ResultsHaloarchaeal strains with 16S rRNA gene sequences 98% identical to Halorubrum saccharovorum were isolated from a hypersaline lake in Inner Mongolia. Two lytic viruses infecting these were isolated from the lake water. The BJ1 virus is described in this paper. It has an icosahedral head and tail morphology and most likely a linear double stranded DNA genome exhibiting terminal redundancy. Its genome sequence has 42,271 base pairs with a GC content ofapproximately 65 mol%. The genome of BJ1 is predicted to encode 70 ORFs, including one for a tRNA. Fifty of the seventy ORFs had no identity to data base entries; twenty showed sequence identity matches to archaeal viruses and to haloarchaea. ORFs possibly coding for an origin of replication complex, integrase, helicase and structural capsid proteins were identified. Evidence for viral integration was obtained.ConclusionThe virus described here has a very low sequence identity to any previously described virus. Fifty of the seventy ORFs could not be annotated in any way based on amino acid identities with sequences already present in the databases. Determining functions for ORFs such as these is probably easier using a simple virus as a model system.

Project description:Asian dust comprises a large portion of the northern hemisphere atmospheric dust load, thereby exerting substantial influence on the Earth's climate, global biogeochemistry and hydrological cycle through accelerated snow and ice melt. Dust deposited on alpine glaciers encodes information on broad scale atmospheric-environmental processes. The (234U/238U) values of dust fine particulates can reflect the comminuting time and intermediate processes; thus, it provides a new method for the provenance of aeolian dust in the glacial snowpack/cryoconite. Here we present results from a comprehensive survey of uranium isotopic concentrations in dust collected from cryoconites on the glaciers of the northern Tibetan Plateau (TP). These results indicate significant spatial heterogeneity in the (234U/238U) values associated with snowpack/cryoconite dust over a broad range of glaciers in the northern TP. The values of the (234U/238U) ratio in the glaciers of western Qilian Mountains (Qiyi Glacier, Shiyi Glacier, Laohugou Glacier No.12) were the highest, followed by the Tanggula (Dunkemadi Glacier) and Kunlun Mountains (Yuzhufeng Glacier), whereas these values were the lowest in the eastern Qilian Mountains (Jingyangling Snowpack, Dabanshan Snowpack, Lenglongling Glacier). By including the analysis of Sr-Nd isotopic compositions, we find the spatial isotopic distribution reflects a combination of local dust, which is associated with short comminuting times, and dust transported over long ranges. Meteorological data indicate that the dust production in the west and north TP, Alxa arid lands and Gobi Deserts, may have a significant impact on the TP glaciers. Moreover, U-Sr-Nd isotopic composition and end-member mixing models (EMMA) were used in our study to find out the relative contribution of distinct Asian dust sources to the dust budget in the TP glaciers. The results reveal that snowpack/cryoconite dust is derived from both local sources (low comminution signatures) as well as other dust sources in the Asian region. Our study demonstrates the potential of U isotope composition as a dust tracing method. In particular, by investigating this composition on dust collected from glacier snowpack and cryoconite holes, we arrive at a map of the distribution characteristics of (234U/238U) values in different regions of the TP. Our study is the first to deploy uranium comminution age for Tibetan dust source tracing, and the results are important to elucidate the multiple origins and dynamics of dust in the Tibetan Plateau.

Project description:Chronic arsenic exposure via drinking water has become a worldwide public health concern. In humans, inorganic arsenic (iAs) is metabolized to monomethylarsonic acid (MMA) and dimethylarsinic acid (DMA) mainly mediated by arsenic (+3 oxidation state) methyltransferase (As3MT). We reported recently that N-6 adenine-specific DNA methyltransferase 1 (N6AMT1) was involved in arsenic metabolism, and examined its interactive effect with As3MT on arsenic metabolism in vitro To further evaluate the interactive effect of N6AMT1 and As3MT on arsenic biomethylation in humans, we conducted a human population-based study including 289 subjects living in rural villages in Inner Mongolia, China, and assessed their urinary arsenic metabolites profiles in relation to genetic polymorphisms and haplotypes of N6AMT1 and As3MT Five N6AMT1 single nucleotide polymorphisms (SNPs; rs1003671, rs7282257, rs2065266, rs2738966, rs2248501) and the N6AMT1 haplotype 2_GGCCAT were significantly associated with the percentage of iAs (% iAs) in urine (e.g., for rs7282257, mean was 9.62% for TT, 6.73% for AA). Rs1003671 was also in a significant relationship with urinary MMA and DMA (the mean of %MMA was 24.95% for GA, 31.69% for GG; the mean of % DMA was 69.21% for GA, 59.82% for GG). The combined effect of N6AMT1 haplotype 2_GGCCAT and As3MT haplotype 2_GCAC showed consistence with the additive significance of each haplotype on % iAs: the mean was 5.47% and 9.36% for carriers with both and null haplotypes, respectively. Overall, we showed that N6AMT1 genetic polymorphisms were associated with arsenic biomethylation in the Chinese population, and its interaction with As3MT was observed in specific haplotype combinations.

Project description:Alkaline lakes are a special aquatic ecosystem that act as important water and alkali resource in the arid-semiarid regions. The primary aim of the study is to explore how environmental factors affect community diversity and structure, and to find whether there are key microbes that can indicate changes in environmental factors in alkaline lakes. Therefore, four sediment samples (S1, S2, S3, and S4) were collected from Hamatai Lake which is an important alkali resource in Ordos' desert plateau of Inner Mongolia. Samples were collected along the salinity and alkalinity gradients and bacterial community compositions were investigated by Illumina Miseq sequencing. The results revealed that the diversity and richness of bacterial community decreased with increasing alkalinity (pH) and salinity, and bacterial community structure was obviously different for the relatively light alkaline and hyposaline samples (LAHO; pH < 8.5; salinity < 20‰) and high alkaline and hypersaline samples (HAHR; pH > 8.5; salinity > 20‰). Firmicutes, Proteobacteria and Bacteriodetes were observed to be the dominant phyla. Furthermore, Acidobacteria, Actinobacteria, and low salt-tolerant alkaliphilic nitrifying taxa were mainly distributed in S1 with LAHO characteristic. Firmicutes, Clostridia, Gammaproteobacteria, salt-tolerant alkaliphilic denitrifying taxa, haloalkaliphilic sulfur cycling taxa were mainly distributed in S2, S3 and S4, and were well adapted to haloalkaline conditions. Correlation analysis revealed that the community diversity (operational taxonomic unit numbers and/or Shannon index) and richness (Chao1) were significantly positively correlated with ammonium nitrogen (r = 0.654, p < 0.05; r = 0.680, p < 0.05) and negatively correlated with pH (r = -0.924, p < 0.01; r = -0.800, p < 0.01; r = -0.933, p < 0.01) and salinity (r = -0.615, p < 0.05; r = -0.647, p < 0.05). A redundancy analysis and variation partitioning analysis revealed that pH (explanation degrees of 53.5%, pseudo-F = 11.5, p < 0.01), TOC/TN (24.8%, pseudo-F = 10.3, p < 0.05) and salinity (9.2%, pseudo-F = 9.5, p < 0.05) were the most significant factors that caused the variations in bacterial community structure. The results suggested that alkalinity, nutrient salt and salinity jointly affect bacterial diversity and community structure, in which one taxon (Acidobacteria), six taxa (Cyanobacteria, Nitrosomonadaceae, Nitrospira, Bacillus, Lactococcus and Halomonas) and five taxa (Desulfonatronobacter, Dethiobacter, Desulfurivibrio, Thioalkalivibrio and Halorhodospira) are related to carbon, nitrogen and sulfur cycles, respectively. Classes Clostridia and Gammaproteobacteria might indicate changes of saline-alkali conditions in the sediments of alkaline lakes in desert plateau.
