Project description:Purpose: Foxp2 is the first and for now the only gene connected to speech and language in humans. Two aminoacid substitutions took place in this protein during recent human evolution, after our split from the last common ancestor with chimpanzees, and are most likely to have undergone positive selection in human lineage (Enard et al., 2002). Methods: Transgenic mice in which the wild-type (murine) version of Foxp2 was replaced with the one bearing two human-specific amino acid substitutions (i.e. "humanized" Foxp2) - Foxp2hum/hum, have been compared to their wild-type (WT) counterparts in terms of behavior, electrophysiology and striatal gene expression. The latter was analyzed through RNA-sequencing performed on pooled indexed libraries on three flow cells on Illumina GAIIx. The reads were mapped to mouse genome (mm9) by TopHat 1.4.1 and were counted using Bedtools. mRNA profiles were obtained with more than 20 million reads for every sample. Differential gene expression was analyzed with DESeq using multifactor model (Anders and Huber, 2010). Results: Wild-type and Foxp2hum/hum mice did not show any significant differences in expression at individual gene level, neither in dorsomedial nor in dorsolateral striatum. However, when genes were grouped into functional categories and analyzed accordingly, this revealed a significant downregulation of functional categories related to synaptic signalling and plasticity in dorsomedial striatum of Foxp2hum/hum mice.

Project description:It is well established that acute stress can influence memory function, yet its influence may differ across memory systems. Whereas stress sometimes exerts a negative influence on declarative learning, it does not necessarily harm learning in general, as demonstrated in the case of procedural learning. Probabilistic category learning is mediated by the striatum, but delaying feedback by a few seconds shifts learning to become more hippocampal-dependent. Here, we examined the influence of acute stress on this type of learning, under different conditions that favor either procedural-based (immediate feedback) vs. declarative-based (delayed feedback) learning. Sixty-two participants randomly assigned to either stress or non-stress groups, performed a probabilistic category learning task, in which they were instructed to learn associations between cues and outcomes under different feedback conditions (immediate feedback, short-delayed feedback, and long-delayed feedback). Acute stress was induced by the Maastricht Acute Stress Test (MAST), and stress levels were gauged by Galvanic Skin Response (GSR) measures and a self-reported questionnaire. Results showed that although the MAST was effective in inducing stress, this did not harm learning in either of the feedback conditions. These findings suggest that not all hippocampal-based learning types are negatively influenced by stress.

Project description:Declarative memory and procedural memory are known to be two fundamentally different kinds of memory that are dissociable in their psychological characteristics and measurement (explicit vs. implicit) and in the neural systems that subserve each kind of memory. Declarative memory abilities are known to improve from childhood through young adulthood, but the developmental maturation of procedural memory is largely unknown. We compared 10-year-old children and young adults on measures of declarative memory and working memory capacity and on four measures of procedural memory that have been strongly dissociated from declarative memory (mirror tracing, rotary pursuit, probabilistic classification, and artificial grammar). Children had lesser declarative memory ability and lesser working memory capacity than adults, but children exhibited learning equivalent to adults on all four measures of procedural memory. Therefore, declarative memory and procedural memory are developmentally dissociable, with procedural memory being adult-like by age 10years and declarative memory continuing to mature into young adulthood.

Project description:According to the Declarative/Procedural Model, the lexicon depends on declarative memory while grammar relies on procedural memory. Furthermore, procedural memory underlies the sequential processing of language. Thus, this system is important for predicting the next item in a sentence. Verb processing represents a good candidate to test this assumption. Semantic representations of verbs include information about the protagonists in the situations they refer to. This semantic knowledge is acquired implicitly and used during verb processing, such that the processing of a verb preactivates its typical patients (e.g., the window for break). Thus, determining how the patient typicality effect appears during children's cognitive development could provide evidence about the memory system that is dedicated to this effect. Two studies are presented in which French children aged 6-10 and adults made grammaticality judgments on 80 auditorily presented sentences. In Experiment 1, the verb was followed by a typical patient or by a less typical patient. In Experiment 2, grammatical sentences were constructed such that the verb was followed either by a typical patient or by a noun that could not be a patient of that verb. The typicality effect occurs in younger children and is interpreted in terms of developmental invariance. We suggest that this effect may depend on procedural memory, in line with studies that showed that meaning is necessary to allow procedural memory to learn the sequence of words in a sentence.

Project description:The retrogenesis hypothesis proposes that the order of breakdown of cognitive abilities in older adults is the reverse of the developmental order of children. Declarative and procedural memory systems, however, have been empirically understudied regarding this issue. The current study aimed to investigate whether retrogenesis occurs in the developmental and decline order of the declarative and procedural memory systems. Besides, we further investigated whether retrogenesis occurs in declarative memory, which was tested through the recognition of familiar and unfamiliar items. Both questions were investigated by looking at 28 Chinese younger adults and 27 cognitively healthy Chinese older adults. The recognition memory task and the Serial Reaction Time Task were administered on two consecutive days in order to measure their declarative and procedural memory, respectively. The results showed older adults performed significantly worse than younger adults for both tasks on both days, suggesting a decline in both declarative and procedural memory. Moreover, older adults exhibited relatively preserved declarative memory compared to procedural memory. This does not follow the expectations of the retrogenesis hypothesis. However, older adults demonstrated superior performance and a steeper rate of forgetting for recognizing familiar items than unfamiliar items. This reverses the developmental order of different patterns in the declarative memory system. Overall, we conclude that retrogenesis occurs in the declarative memory system, while does not in the decline order of the two memory systems; this understanding can better help inform our broader understanding of memory aging.

Project description:The study of declarative and procedural knowledge makes it possible to ascertain what cognitive processes are like during motor learning. This study aimed to compare, according to the methodology, gender and experience (football practise), and the levels of declarative and procedural knowledge after the implementation of two intervention programmes on school football including one based on the tactics learning and the other on the technique learning. A total of 41 students in the 5th year of primary education from a state school from Spain, distributed in two class groups, participated in the study. Each class group participated in a different intervention programme. The sample of subjects was equal (tactical programme (n = 20) and technical programme (n = 21)). A panel of 13 experts validated both programmes. Levels of knowledge were measured using the Tactical Knowledge Assessment test in football. A descriptive analysis was performed to characterise the sample. Moreover, a t-test for independent samples, a t-test for related samples, and a 2 × 2 ANOVA (analysis of variance) were performed to compare the levels of knowledge between the pre-test and the post-test, according to the methodology, gender, and experience of the students. Results indicate that both intervention programmes induced higher levels of declarative and procedural knowledge in the post-test. Similarly, there were no significant differences with regard to the applied methodology. This fact is due to the heterogeneous character of the class groups with gender and experience showing effects on the levels of knowledge. The boys possessed greater experience and a higher level of knowledge compared to the girls.

Project description:The acquisition of language and speech is uniquely human, but how genetic changes might have adapted the nervous system to this capacity is not well understood. Two human-specific amino acid substitutions in the transcription factor forkhead box P2 (FOXP2) are outstanding mechanistic candidates, as they could have been positively selected during human evolution and as FOXP2 is the sole gene to date firmly linked to speech and language development. When these two substitutions are introduced into the endogenous Foxp2 gene of mice (Foxp2(hum)), cortico-basal ganglia circuits are specifically affected. Here we demonstrate marked effects of this humanization of Foxp2 on learning and striatal neuroplasticity. Foxp2(hum/hum) mice learn stimulus-response associations faster than their WT littermates in situations in which declarative (i.e., place-based) and procedural (i.e., response-based) forms of learning could compete during transitions toward proceduralization of action sequences. Striatal districts known to be differently related to these two modes of learning are affected differently in the Foxp2(hum/hum) mice, as judged by measures of dopamine levels, gene expression patterns, and synaptic plasticity, including an NMDA receptor-dependent form of long-term depression. These findings raise the possibility that the humanized Foxp2 phenotype reflects a different tuning of corticostriatal systems involved in declarative and procedural learning, a capacity potentially contributing to adapting the human brain for speech and language acquisition.

Project description:Neuroimaging techniques applied to a variety of organisms-from zebrafish, to rodents to humans-can offer valuable insights into neuronal network properties and their dysfunction in epilepsy. A wide range of imaging methods used to monitor neuronal circuits and networks during evoked seizures in animal models and advances in functional magnetic resonance imaging (fMRI) applied to patients with epilepsy were discussed during the XIV Workshop on Neurobiology of Epilepsy (XIV WONOEP) organized in 2017 by the Neurobiology Commission of the International League Against Epilepsy (ILAE). We review the growing number of technological approaches developed, as well as the current state of knowledge gained from studies applying these advanced imaging approaches to epilepsy research.

Project description:Many studies investigating sleep and memory consolidation have evaluated full-night sleep rather than alternative sleep periods such as daytime naps. This multi-centre study followed up on, and was compared with, an earlier full-night study (Schabus et al., 2004) investigating the relevance of daytime naps for the consolidation of declarative and procedural memory. Seventy-six participants were randomly assigned to a nap or wake group, and performed a declarative word-pair association or procedural mirror-tracing task. Performance changes from before to after a 90-min retention interval filled with sleep or quiet wakefulness were evaluated between groups. Associations between performance changes, sleep architecture, spindles, and slow oscillations were investigated. For the declarative task we observed a trend towards stronger forgetting across a wake period compared with a nap period, and a trend towards memory increase over the full-night. For the procedural task, accuracy was significantly decreased following daytime wakefulness, showed a trend to increase with a daytime nap, and significantly increased across full-night sleep. For the nap protocol, neither sleep stages, spindles, nor slow oscillations predicted performance changes. A direct comparison of day and nighttime sleep revealed that daytime naps are characterized by significantly lower spindle density, but higher spindle activity and amplitude compared with full-night sleep. In summary, data indicate that daytime naps protect procedural memories from deterioration, whereas full-night sleep improves performance. Given behavioural and physiological differences between day and nighttime sleep, future studies should try to characterize potential differential effects of full-night and daytime sleep with regard to sleep-dependent memory consolidation.
