Project description:Genetic bottlenecks resulting from human-induced population declines make alarming symbols for the irreversible loss of our natural legacy worldwide. The grey wolf (Canis lupus) is an iconic example of extreme declines driven by anthropogenic factors. Here, we assessed the genetic signatures of 150 years of wolf persecution throughout the Western Palaearctic by high-throughput mitochondrial DNA sequencing of historical specimens in an unprecedented spatio-temporal framework. Despite Late Pleistocene bottlenecks, we show that historical genetic variation had remained high throughout Europe until the last several hundred years. In Western Europe, where wolves nearly got fully exterminated, diversity dramatically collapsed at the turn of the twentieth century and recolonization from few homogeneous relict populations induced drastic shifts of genetic composition. By contrast, little genetic displacement and steady levels of diversity were maintained in Eastern European regions, where human persecution had lesser effects on wolf demography. By comparing prehistoric, historic and modern patterns of genetic diversity, our study hence traces the timeframe and the active human role in the decline of the grey wolf, an emblematic yet controversial animal which symbolizes the complex relationship between human societies and nature conservation.

Project description:Pigmentation of the skin, hair, and eyes varies both within and between human populations. Identifying the genes and alleles underlying this variation has been the goal of many candidate gene and several genome-wide association studies (GWAS). Most GWAS for pigmentary traits to date have been based on subjective phenotypes using categorical scales. But skin, hair, and eye pigmentation vary continuously. Here, we seek to characterize quantitative variation in these traits objectively and accurately and to determine their genetic basis. Objective and quantitative measures of skin, hair, and eye color were made using reflectance or digital spectroscopy in Europeans from Ireland, Poland, Italy, and Portugal. A GWAS was conducted for the three quantitative pigmentation phenotypes in 176 women across 313,763 SNP loci, and replication of the most significant associations was attempted in a sample of 294 European men and women from the same countries. We find that the pigmentation phenotypes are highly stratified along axes of European genetic differentiation. The country of sampling explains approximately 35% of the variation in skin pigmentation, 31% of the variation in hair pigmentation, and 40% of the variation in eye pigmentation. All three quantitative phenotypes are correlated with each other. In our two-stage association study, we reproduce the association of rs1667394 at the OCA2/HERC2 locus with eye color but we do not identify new genetic determinants of skin and hair pigmentation supporting the lack of major genes affecting skin and hair color variation within Europe and suggesting that not only careful phenotyping but also larger cohorts are required to understand the genetic architecture of these complex quantitative traits. Interestingly, we also see that in each of these four populations, men are more lightly pigmented in the unexposed skin of the inner arm than women, a fact that is underappreciated and may vary across the world.

Project description:IntroductionThe diagnosis of endogenous Cushing syndrome is often challenging and requires multiple repeated blood, urine, and saliva tests to detect elevated cortisol levels. Hair cortisol concentration has been described as a marker of long-term exposure to systemic cortisol in patients with Cushing syndrome. Like hemoglobin A1c is used to detect serum glucose exposure over months, segmental hair cortisol can help identify patients with milder forms of and/or periodic or cyclical Cushing syndrome, which may reduce time and costs associated with collection of urine, salivary, and serum cortisol.Areas coveredSuccess of hair cortisol in detection of Cushing syndrome will be discussed in context of current literature, including differences between total or segmental hair cortisol in accurately determining timeline of cortisol exposure. Optimal methods of hair collection, storage, processing, and analysis and efforts toward standardization will be a major focus.Expert commentaryRecent evidence suggests increased sensitivity and specificity of hair cortisol in detecting Cushing syndrome. Future guidelines should consider this test as a routine part of the repertoire of screening tests for Cushing syndrome. Possible confounders to explain discrepant results in the literature will be discussed.

Project description:Several cross-sectional studies have shown hair cortisol concentration to be associated with adiposity, but the relationship between hair cortisol concentration and longitudinal changes in measures of adiposity are largely unknown. We included 786 adults from the NoHoW trial, who had achieved a successful weight loss of ≥5% and had a body mass index of ≥25 kg/m2 prior to losing weight. Hair cortisol concentration (pg/mg hair) was measured at baseline and after 12 months. Body weight and body fat percentage were measured at baseline, 6-month, 12-month and 18-month visits. Participants weighed themselves at home ≥2 weekly using a Wi-Fi scale for the 18-month study duration, from which body weight variability was estimated using linear and non-linear approaches. Regression models were conducted to examine log hair cortisol concentration and change in log hair cortisol concentration as predictors of changes in body weight, change in body fat percentage and body weight variability. After adjustment for lifestyle and demographic factors, no associations between baseline log hair cortisol concentration and outcome measures were observed. Similar results were seen when analysing the association between 12-month concurrent development in log hair cortisol concentration and outcomes. However, an initial 12-month increase in log hair cortisol concentration was associated with a higher subsequent body weight variability between month 12 and 18, based on deviations from a nonlinear trend (β: 0.02% per unit increase in log hair cortisol concentration [95% CI: 0.00, 0.04]; P=0.016). Our data suggest that an association between hair cortisol concentration and subsequent change in body weight or body fat percentage is absent or marginal, but that an increase in hair cortisol concentration during a 12-month weight loss maintenance effort may predict a slightly higher subsequent 6-months body weight variability.Clinical trial registrationISRCTN registry, identifier ISRCTN88405328.

Project description:Simple Summary Prosocial behaviour is shown by a variety of mammals. Prosocial behaviours can also be part of social feedback mechanisms. For group-living mammals, group coordination and group cohesion are crucial factors for survival. Therefore, group-living mammals display a broad variety of social behaviours towards their group members. Canidae are known to be socially organised mammals. Wolves are one of the most cooperative and social canine species. In this case study, we investigated social feedback mechanisms in two European grey wolf groups. The social feedback was observed after novel object interaction, bold behaviour, and the individual behaviours of single individuals. The purpose of this paper is to focus on prosocial behaviour that might serve as social reward. We assume that there is social reward behaviour as a category that falls within social feedback in grey wolves. Abstract Prosociality occurs in many species and is likely to be a crucial factor for the survival of group-living animals. Social feedback is an important mechanism for the coordination of group decisions. Since group-living animals with specific personality axes, i.e., boldness, are known to provide certain benefits for their group, bold actions might receive more prosocial feedback than other actions. Our case study aims to determine whether bold behaviour, i.e., novel object interaction (Nobj), might be answered more frequently with prosocial behaviours. We investigated the differences in the frequency of occurrence in prosocial behaviours after three different individual actions in two groups of grey wolves. We aim to outline the development of a social reward behavioural category as part of social feedback mechanisms. We used Markov chain models for probability analyses, and a non-parametric ANOVA to test for differences between the influences of individual behaviours on the probability of a prosocial behaviour chain. We additionally tested for the potential influences of age, sex and personality on the frequency of Nobj. Our results suggest that bold interactions are more often responded to with prosocial behaviour. Bold behaviour might be more often socially rewarded because of its benefits for group-living animals. More research is needed to investigate whether bold behaviour is more frequently responded to prosocially, and to investigate the social reward phenomenon.

Project description:Dysregulation of the hypothalamic-pituitary-adrenal (HPA) axis is one mechanism through which chronic stress during pregnancy and parenthood may affect parental, child, and family health. Hair cortisol concentration (HCC) may be well-suited to elucidate associations between chronic stress and HPA axis regulation because HCC reflects cortisol output over several months. However, most previous studies that examine chronic stress in conjunction with cortisol in pregnant individuals or mothers use measures that reflect HPA axis output over a relatively limited time. We report findings from two longitudinal studies that tested associations between chronic stress and HCC in women during the perinatal period (Study 1; n = 144) and mothers of young children (Study 2; n = 102). Both studies measured chronic stress with a measure developed to comprehensively assess chronic stressors in community samples. Hair samples were collected three times in Study 1 (mid-pregnancy, one month postpartum, and 12 months postpartum) and twice in Study 2, approximately one year apart. Chronic stress was associated with higher HCC in both studies. Exploratory analyses indicated that the strength of associations between chronic stress and HCC differed as a function of the life domain of chronic stress. Chronic work and family demands were associated with higher postpartum HCC in Study 1 whereas neighbourhood/housing and discrimination chronic stress were associated with higher HCC in Study 2. These findings provide evidence of a biological pathway through which chronic stress may influence health in mothers and support the utility of hair cortisol as a neuroendocrine measure of chronic stress during pregnancy and parenthood.

Project description:BACKGROUND:While it is generally accepted that patterns of intra-specific genetic differentiation are substantially affected by glacial history, population genetic processes occurring during Pleistocene glaciations are still poorly understood. In this study, we address the question of the genetic consequences of Pleistocene glaciations for European grey wolves. Combining our data with data from published studies, we analysed phylogenetic relationships and geographic distribution of mitochondrial DNA haplotypes for 947 contemporary European wolves. We also compared the contemporary wolf sequences with published sequences of 24 ancient European wolves. RESULTS:We found that haplotypes representing two haplogroups, 1 and 2, overlap geographically, but substantially differ in frequency between populations from south-western and eastern Europe. A comparison between haplotypes from Europe and other continents showed that both haplogroups are spread throughout Eurasia, while only haplogroup 1 occurs in contemporary North American wolves. All ancient wolf samples from western Europe that dated from between 44,000 and 1,200 years B.P. belonged to haplogroup 2, suggesting the long-term predominance of this haplogroup in this region. Moreover, a comparison of current and past frequencies and distributions of the two haplogroups in Europe suggested that haplogroup 2 became outnumbered by haplogroup 1 during the last several thousand years. CONCLUSIONS:Parallel haplogroup replacement, with haplogroup 2 being totally replaced by haplogroup 1, has been reported for North American grey wolves. Taking into account the similarity of diets reported for the late Pleistocene wolves from Europe and North America, the correspondence between these haplogroup frequency changes may suggest that they were associated with ecological changes occurring after the Last Glacial Maximum.

Project description:Hair cortisol is a novel biomarker of chronic stress. The objective of this study was to determine the effect of hair color and length, as well as, animal age on hair cortisol concentration in beef feedlot steers. Nineteen beef crossbred steers used for nutrition research and housed in a small feedlot setting were used for this study. Seven of the steers (680 ± 4.5 kg; ~9 years of age) were fitted with ruminal fistulas and duodenal cannulas. The other 12 steers (473 ± 3.1 kg; ~2.5 years of age) were fitted with only ruminal fistulas. Hair samples from each steer were collected throughout a period of 6 weeks from six different areas and analyzed for cortisol concentrations. One pre-determined area was shaved each week for 5 weeks (Weeks 1-5). During week 6, all five, previously shaved areas and an additional area was shaved to collect hair samples of various lengths. Hair length was recorded prior to the collection of each hair sample. Only data from the last week (Week 6) of collection were included in the analyses. Steers were categorized into one of three groups: old with black hair (OB, n = 3); old with white hair (OW, n = 3); young with black hair (YB, n = 12). Older steers exhibited greater hair cortisol concentrations than younger steers (P < 0.001). Hair cortisol concentration was not impacted by duration of growth (P = 0.33). Cortisol concentrations exhibited a weak, positive correlation with hair length (r = 0.33, P-value = 0.01). The average hair growth per week of beef steers in the winter months was calculated to be 0.90 mm. Further research should be performed to improve our understanding of the effect of hair characteristics, sampling methodologies and analysis techniques on hair cortisol concentrations.

Project description:BackgroundMothers and their children demonstrate dyadic synchrony of hypothalamic-pituitary-adrenal (HPA) axis function, likely influenced by shared genetic or environmental factors. Although evidence has shown that chronic stress exposure has physiologic consequences for individuals-including on the HPA axis-minimal research has explored how unmet social needs such as food and housing instability may be associated with chronic stress and HPA axis synchrony in mother-child dyads.MethodsWe conducted a secondary analysis of data from 364 mother-child dyads with low-income recruited during a randomized trial conducted in an urban pediatric clinic. We used latent profile analysis (LPA) to identify subgroups based on naturally occurring patterns of within-dyad hair cortisol concentration (HCC). A logistic regression model predicted dyadic HCC profile membership as a function of summative count of survey-reported unmet social needs, controlling for demographic and health covariates.ResultsLPA of HCC data from dyads revealed a 2-profile model as the best fit. Comparisons of log HCC for mothers and children in each profile group resulted in significantly "higher dyadic HCC" versus "lower dyadic HCC" profiles (median log HCC for mothers: 4.64 vs 1.58; children: 5.92 vs 2.79, respectively; P < .001). In the fully adjusted model, each one-unit increase in number of unmet social needs predicted significantly higher odds of membership in the higher dyadic HCC profile when compared to the lower dyadic HCC profile (odds ratio = 1.13; 95% confidence interval [1.04-1.23]; P = .01).ConclusionMother-child dyads experience synchronous patterns of physiologic stress, and an increasing number of unmet social needs is associated with a profile of higher dyadic HCC. Interventions aimed at decreasing family-level unmet social needs or maternal stress are, therefore, likely to affect pediatric stress and related health inequities; efforts to address pediatric stress similarly may affect maternal stress and related health inequities. Future research should explore the measures and methods needed to understand the impact of unmet social needs and stress on family dyads.

Project description:ObjectivesPrior studies found increased hair cortisol concentration (a surrogate marker for hypothalamic-pituitary-adrenal axis activation) in users of the levonorgestrel intrauterine device (LNG 52 mg IUD). We evaluated change in hair cortisol and psychometric tests in women initiating a copper (CuT380 IUD) or LNG 52 mg IUD.Study designWe prospectively enrolled healthy women initiating an LNG 52 mg IUD or CuT380 IUD. Participants provided hair and blood samples and completed psychometric inventories (Patient Health Questionnaire-9, Positive and Negative Affect Schedule, and Psychological General Well-Being Index) after IUD insertion and at 6 and 12 months. We used liquid chromatography with tandem mass spectrometry to measure hair cortisol concentrations. We compared hair cortisol concentrations and psychometric test changes from baseline to 6 and 12 months using independent two-sample t tests.ResultsWe enrolled 39 of our targeted 86 participants (LNG 52 mg IUD 26, CuT380 IUD 13). Thirty-eight subjects (LNG 52 mg IUD 25, CuT380 IUD 13) completed 6 months of follow-up. We found no difference between cohorts in the mean change in hair cortisol concentrations at 6 months (LNG 52 mg IUD n = 21 [-0.01 pg/mg (95% CI -1.26, 1.23); CuT380 IUD n = 13 [-1.31 pg/mg (-3.36, 0.73)]). While psychometric inventory results remained within normal ranges, LNG 52 mg IUD users reported a trend toward more favorable changes over time.ConclusionsWe did not find clinically important differences in hair cortisol concentrations following initiation of a CuT380 IUD or LNG 52 mg IUD; psychometric inventories demonstrated no adverse effect of hormonal IUDs on mood.ImplicationsOur findings of similar hair cortisol concentrations following the initiation of either the LNG 52 mg IUD or CuT380 IUD suggest that hormonal IUDs do not increase cortisol concentrations or alter stress reactivity, and favorable effects on psychometric inventories provide further reassurance that the LNG 52 mg IUD has no adverse impact on mood.Trial registration numberClinicalTrials.gov NCT03499379.
