Language of fungi derived from their electrical spiking activity.

ABSTRACT: Fungi exhibit oscillations of extracellular electrical potential recorded via differential electrodes inserted into a substrate colonized by mycelium or directly into sporocarps. We analysed electrical activity of ghost fungi (Omphalotus nidiformis), Enoki fungi (Flammulina velutipes), split gill fungi (Schizophyllum commune) and caterpillar fungi (Cordyceps militaris). The spiking characteristics are species specific: a spike duration varies from 1 to 21 h and an amplitude from 0.03 to 2.1 mV. We found that spikes are often clustered into trains. Assuming that spikes of electrical activity are used by fungi to communicate and process information in mycelium networks, we group spikes into words and provide a linguistic and information complexity analysis of the fungal spiking activity. We demonstrate that distributions of fungal word lengths match that of human languages. We also construct algorithmic and Liz-Zempel complexity hierarchies of fungal sentences and show that species S. commune generate the most complex sentences.

Electrical spiking in bacterial biofilms.

Project description:In nature, biofilms are the most common form of bacterial growth. In biofilms, bacteria display coordinated behaviour to perform specific functions. Here, we investigated electrical signalling as a possible driver in biofilm sociobiology. Using a multi-electrode array system that enables high spatio-temporal resolution, we studied the electrical activity in two biofilm-forming strains and one non-biofilm-forming strain. The action potential rates monitored during biofilm-forming bacterial growth exhibited a one-peak maximum with a long tail, corresponding to the highest biofilm development. This peak was not observed for the non-biofilm-forming strain, demonstrating that the intensity of the electrical activity was not linearly related to the bacterial density, but was instead correlated with biofilm formation. Results obtained indicate that the analysis of the spatio-temporal electrical activity of bacteria during biofilm formation can open a new frontier in the study of the emergence of collective microbial behaviour.

Terpenes a common language between fungi and bacteria

Project description:Previously, we investigated the effect of fungal VOCs on the behavior of phylogenetically different soil bacteria (Schmidt et al 2015). In these experiments we showed that VOCs emitted by several fungi can lead to phenotypical responses in bacteria, for example, by inducing a change in motility (Schmidt et al 2015). We observed that the plant pathogenic fungus Fusarium culmorum produced a unique cluster of VOCs consisting primarily of terpenes. When exposed to the VOCs emitted by this fungus, the rhizobacterium Serratia plymuthica PRI-2C responded with an induction of motility. It is plausible that in soil, microorganisms sense changes in their environments via shifts in VOCs blend and adapt their behavior accordingly (Garbeva et al 2014). Although several studies indicated that VOCs can be used as signaling molecules in microbial inter-species interactions, the following questions remain unanswered as how are VOCs perceived as signals by the microorganisms and which regulatory pathways and genes are involved in the response? To answer these questions, the rhizosphere isolate S. plymuthica PRI-2C was grown alone or exposed to VOCs emitted by F. culmorum. The bacterial transcriptome and proteome were analyzed under each situation to identify the molecular basis of the bacterial response to fungal VOCs.

Probabilistic mapping of language networks from high frequency activity induced by direct electrical stimulation.

Project description:Direct electrical stimulation (DES) at 50 Hz is used as a gold standard to map cognitive functions but little is known about its ability to map large-scale networks and specific subnetwork. In the present study, we aim to propose a new methodological approach to evaluate the specific hypothesis suggesting that language errors/dysfunction induced by DES are the result of large-scale network modification rather than of a single cortical region, which explains that similar language symptoms may be observed after stimulation of different cortical regions belonging to this network. We retrospectively examined 29 patients suffering from focal drug-resistant epilepsy who benefitted from stereo-electroencephalographic (SEEG) exploration and exhibited language symptoms during a naming task following 50 Hz DES. We assessed the large-scale language network correlated with behavioral DES-induced responses (naming errors) by quantifying DES-induced changes in high frequency activity (HFA, 70-150 Hz) outside the stimulated cortical region. We developed a probabilistic approach to report the spatial pattern of HFA modulations during DES-induced language errors. Similarly, we mapped the pattern of after-discharges (3-35 Hz) occurring after DES. HFA modulations concurrent to language symptoms revealed a brain network similar to our current knowledge of language gathered from standard brain mapping. In addition, specific subnetworks could be identified within the global language network, related to different language processes, generally described in relation to the classical language regions. Spatial patterns of after-discharges were similar to HFA induced during DES. Our results suggest that this new methodological DES-HFA mapping is a relevant approach to map functional networks during SEEG explorations, which would allow to shift from "local" to "network" perspectives.

Representational Pattern Similarity of Electrical Brain Activity Reveals Rapid and Specific Prediction during Language Comprehension.

Project description:Predicting upcoming events is a critical function of the brain, and language provides a fertile testing ground for studying prediction, as comprehenders use context to predict features of upcoming words. Many aspects of the mechanisms of prediction remain elusive, partly due to a lack of methodological tools to probe prediction formation in the moment. To elucidate what features are neurally preactivated and when, we used representational similarity analysis on previously collected sentence reading data. We compared EEG activity patterns elicited by expected and unexpected sentence final words to patterns from the preceding words of the sentence, in both strongly and weakly constraining sentences. Pattern similarity with the final word was increased in an early time window following the presentation of the pre-final word, and this increase was modulated by both expectancy and constraint. This was not seen at earlier words, suggesting that predictions were precisely timed. Additionally, pre-final word activity-the predicted representation-had negative similarity with later final word activity, but only for strongly expected words. These findings shed light on the mechanisms of prediction in the brain: rapid preactivation occurs following certain cues, but the predicted features may receive reduced processing upon confirmation.

Marine-Derived Fungi as a Valuable Resource for Amylases Activity Screening.

Project description:Marine microbial enzymes including amylases are important in different industrial production due to their properties and applications. This study was focused on the screening of marine-derived fungi for amylase activities. First, we isolated a number of fungi from the sediments of the South China Sea. By the method of dish screening (in vitro), we subsequently obtained a series of amylase-producing fungal strains. The cell-lysate activities of amylases produced by marine fungi toward starch hydrolysis were achieved with the dinitrosalyicylic acid (DNS) method. In addition, the effect of pH and temperature on amylase activities, including thermal and pH stability were discussed. Results showed that out of the 57 isolates with amylase-producing activities, fungi Aspergillus flavus 9261 was found to produce amylase with the best activity of 10.7482 U/mg (wet mycelia). The amylase of Aspergillus flavus 9261 exhibited remarkable thermostability and pH stability with no activity loss after incubation at 50 °C and pH 5.0 for 1 h, respectively. The results provide advances in discovering enzymes from marine-derived fungi and their biotechnology relevance.

Electrical stimulation in a spiking neural network model of monkey superior colliculus.

Project description:The superior colliculus (SC) generates saccades by recruiting a population of cells in its topographically organized motor map. Supra-threshold electrical stimulation in the SC produces a normometric saccade with little effect of the stimulation parameters. Moreover, the kinematics of electrically evoked saccades strongly resemble natural, visual-evoked saccades. These findings support models in which the saccade vector is determined by a center-of-gravity computation of activated neurons, while trajectory and kinematics arise in brainstem-cerebellar feedback circuits. Recent single-unit recordings, however, have indicated that the SC population also specifies the instantaneous saccade kinematics, supporting an alternative model, in which the saccade trajectory results from dynamic summation of movement effects of all SC spike trains. Here we reconcile the linear summation model with stimulation results, by assuming that the electric field directly activates a relatively small set of neurons around the electrode tip, which subsequently sets up a large population response through lateral synaptic interactions.

Electrical stimulation enhances neuronal cell activity mediated by Schwann cell derived exosomes.

Project description:Electrical stimulation (ES) therapy has good effects in patients with nervous system injury-related diseases. ES promotes nerve cell regeneration and stimulates Schwann cells to express neurotrophic factors. The incidence of stress urinary incontinence (SUI) among elderly people is increasing. Some studies suggest that damage to the pudendal nerve is closely related to the pathogenesis of SUI. It has also been found that pelvic ES can reduce SUI symptoms in a rat model of SUI caused by pudendal nerve injury. Clinically, pelvic floor electrical stimulation is effective in patients with mild to moderate SUI. These studies indicate that ES may ameliorate damage to the pudendal nerve and thus achieve the goal of SUI treatment, although the mechanism of action of this treatment remains unclear. Therefore, the purpose of the present study was to clarify the relationships among ES, neural cells and Schwann cells at the cellular level. We applied ES to nerve cells at 100 mV/mm or 200 mV/mm for 0, 0.5, 1, or 2 h to investigate changes in nerve cell activity. We then co-cultured the nerve cells with Schwann cells to explore the influence of single-culture and co-culture conditions on the nerve cells. Compared to non-ES, ES of the nerve cells increased their activity. Compared to those in single culture, co-cultured nerve cells exhibited an additional increase in activity. We also found that Schwann cell derived exosomes could promote the activity of nerve cells, with glutamate and calcium ions playing a potential role in this process. These results suggest that the mutual regulation of neural cells and Schwann cells plays an important role in the process by which ES ameliorates neurological function, which may provide a basis for subsequent studies.

An integrated model of electrical spiking, bursting, and calcium oscillations in GnRH neurons.

Project description:The plasma membrane electrical activities of neurons that secrete gonadotropin-releasing hormone (GnRH) have been studied extensively. A couple of mathematical models have been developed previously to explain different aspects of these activities. The goal of this article is to develop a single model that accounts for the previously modeled experimental results and some more recent results that have not been accounted for. The latter includes two types of membrane potential bursting mechanisms and their associated cytosolic calcium oscillations. One bursting mechanism has not been reported in experiments and is thus regarded as a model prediction. Although the model is mainly based on data collected in immortalized GnRH cell lines, it is capable of explaining some properties of GnRH neurons observed in several other preparations including mature GnRH neurons in hypothalamic slices. We present a spatial model that incorporates a detailed description of calcium dynamics in a three-dimensional cell body with the ion channels evenly distributed on the cell surface. A phenomenological reduction of the spatial model into a simplified form is also presented. The simplified model will facilitate the study of the roles of plasma membrane electrical activities in the pulsatile release of GnRH.

Gap junctions between striatal fast-spiking interneurons regulate spiking activity and synchronization as a function of cortical activity.

Project description:Striatal fast-spiking (FS) interneurons are interconnected by gap junctions into sparsely connected networks. As demonstrated for cortical FS interneurons, these gap junctions in the striatum may cause synchronized spiking, which would increase the influence that FS neurons have on spiking by the striatal medium spiny (MS) neurons. Dysfunction of the basal ganglia is characterized by changes in synchrony or periodicity, thus gap junctions between FS interneurons may modulate synchrony and thereby influence behavior such as reward learning and motor control. To explore the roles of gap junctions on activity and spike synchronization in a striatal FS population, we built a network model of FS interneurons. Each FS connects to 30-40% of its neighbors, as found experimentally, and each FS interneuron in the network is activated by simulated corticostriatal synaptic inputs. Our simulations show that the proportion of synchronous spikes in FS networks with gap junctions increases with increased conductance of the electrical synapse; however, the synchronization effects are moderate for experimentally estimated conductances. Instead, the main tendency is that the presence of gap junctions reduces the total number of spikes generated in response to synaptic inputs in the network. The reduction in spike firing is due to shunting through the gap junctions; which is minimized or absent when the neurons receive coincident inputs. Together these findings suggest that a population of electrically coupled FS interneurons may function collectively as input detectors that are especially sensitive to synchronized synaptic inputs received from the cortex.

Biotransformation of Thiochroman Derivatives Using Marine-Derived Fungi: Isolation, Characterization, and Antimicrobial Activity.

Project description:Thiochroman derivatives are highly versatile molecules widely used for the synthesis of novel heterocycles and bioactive compounds. In our study, we conducted the biotransformation of thiochroman-4-ol (1) and 6-chlorothiochroman-4-ol (1a) using the marine-derived fungal strains Emericellopsis maritima BC17 and Purpureocillium lilacinum BC17-2. Biotransformations yielded ten known thiochroman derivatives along with the compound 1-(5-chloro-2-(methylthio)phenyl)propane-1,3-diol (6a), which was described for the first time. Moreover, we successfully characterized the stereoisomers of sulfoxides 3 and 3a. Their structures and absolute configurations were established though comprehensive analyses of NMR, HR ESI-MS, and ECD spectra, as well as by using Mosher's method. Antimicrobial activity of the isolated metabolites was evaluated against bacterial and fungal human pathogens, specifically Staphylococcus aureus ATCC 29213, Escherichia coli ATCC25922, and Candida albicans HPM-1922816.
