Leaf Photosynthesis and Its Temperature Response Are Different between Growth Stages and N Supplies in Rice Plants.

ABSTRACT: Leaf photosynthesis is highly correlated with CO2-diffusion capacities, which are determined by both leaf anatomical traits and environmental stimuli. In the present study, leaf photosynthetic rate (A), stomatal conductance (gs), mesophyll conductance (gm) and the related leaf anatomical traits were studied on rice plants at two growth stages and with two different N supplies, and the response of photosynthesis to temperature (T) was also studied. We found that gm was significantly higher at mid-tillering stage and at high N treatment. The larger gm was related to a larger chloroplast surface area facing intercellular air spaces and a thinner cell wall in comparison with booting stage and zero N treatment. At mid-tillering stage and at high N treatment, gm showed a stronger temperature response. The modelling of the gm-T relationships suggested that, in comparison with booting stage and zero N treatment, the stronger temperature response of gm was related to the higher activation energy of the membrane at mid-tillering stage and at high N treatment. The findings in the present study can enhance our knowledge on the physiological and environmental determinants of photosynthesis.

Project description:Photosynthetic performance and biomass at different growth stages of the salt-sensitive KDML105 rice cultivar, three improved lines (RD73, CSSL8-94, and TSKC1-144), and the salt-tolerant standard genotype (Pokkali) were investigated under non-saline, semi-saline, and the heavy-saline field conditions in the northeast of Thailand. In the non-saline field, net photosynthesis rates (Pn) of all genotypes remained high from the early vegetative stage to the milky stage and then dramatically reduced at maturity. In contrast, in both saline fields, Pn was the highest at the early vegetative stage and continuously declining until maturity. Leaf chlorophyll content remained high from the early vegetative to milky stage then reduced at maturity for all three field conditions. During the reproductive phase, Pn of KDML105 and the improved lines were reduced by 4-17% in the heavy-saline field, while that of Pokkali was increased (11-19% increase over that of the non-saline). Pokkali also showed a prominent increase in water use efficiency (WUE) under salinity. Nevertheless, rice leaves under saline conditions maintained the PSII integrity, as indicated by the pre-dawn values of maximum quantum yield of PSII photochemistry (Fv/Fm) of higher than 0.8. Pokkali under the semi-saline and the heavy-saline conditions exhibited 51% and 27% increases in final biomass, and 64% and 42% increases in filled grain weight plant-1, respectively. In the semi-saline condition, RD73, TSKC1-144, CSSL8-94, and KDML105 showed moderate salt tolerance by displaying 24%, 18.6%, 15%, and 11.3% increases in final biomass, and 24%, 4%, 13%, and 6% increases in filled grain weight plant-1, respectively. In contrast, in the heavy-saline field, final biomass of RD73, KDML105, CSSL8-94, and TSKC1-144 showed 48%, 45%, 38%, and 36% reductions from that in the non-saline field, while the filled grain weight plant-1 were reduced by 45%, 58%, 35%, and 32%, respectively. This indicated that the improved lines carrying drought- and/or salt-tolerance genes achieved an increased salt tolerance level than the parental elite cultivar, KDML105.

Project description:Background and aimsThe C4 perennial grass miscanthus has been found to be less sensitive to cold than most other C4 species, but still emerges later in spring than C3 species. Genotypic differences in miscanthus were investigated to identify genotypes with a high cold tolerance at low temperatures and quick recovery upon rising temperatures to enable them to exploit the early growing season in maritime cold climates. Suitable methods for field screening of cold tolerance in miscanthus were also identified.MethodsFourteen genotypes of M. sacchariflorus, M. sinensis, M. tinctorius and M. × giganteus were selected and grown under warm (24 °C) and cold (14 °C) conditions in a controlled environment. Dark-adapted chlorophyll fluorescence, specific leaf area (SLA) and net photosynthetic rate at a photosynthetically active radiation (PAR) of 1000 μmol m(-2) s(-1) (A1000) were measured. Photosynthetic light and CO2 response curves were obtained from 11 of the genotypes, and shoot growth rate was measured under field conditions.Key resultsA positive linear relationship was found between SLA and light-saturated photosynthesis (Asat) across genotypes, and also between shoot growth rate under cool field conditions and A1000 at 14 °C in a climate chamber. When lowering the temperature from 24 to 14 °C, one M. sacchariflorus exhibited significantly higher Asat and maximum photosynthetic rate in the CO2 response curve (Vmax) than other genotypes at 14 °C, except M × giganteus 'Hornum'. Several genotypes returned to their pre-chilling A1000 values when the temperature was increased to 24 °C after 24 d growth at 14 °C.ConclusionsOne M. sacchariflorus genotype had similar or higher photosynthetic capacity than M × giganteus, and may be used for cultivation together with M × giganteus or for breeding new interspecies hybrids with improved traits for temperate climates. Two easily measured variables, SLA and shoot growth rate, may be useful for genotype screening of productivity and cold tolerance.

Project description:Tomato (Solanum lycopersicum L.) is an important vegetable crop that grows easily under controlled conditions, such as in greenhouses and hydroponics. To overcome freshwater scarcity, researchers are searching for alternatives to groundwater sources such as desalinated water (saline water) for irrigation. High salinity in irrigation water alters physiological functions and crop development, thereby reducing the yield. Best management practices and the use of grafted tomato plants on salt-tolerant rootstocks can alleviate salinity stress. The present study was conducted to address the impact of salinity stress on leaf transpiration (Tr), stomatal conductance (gs), photosynthesis (Pn), leaf chlorophyll content, proline content, and yield of hydroponically cultivated tomato plants. Saline (NaCl) water was used for the preparation of nutrient solution with three salinity levels, electrical conductivity (EC, dS m-1) of 2.5 (control), 6.0, and 9.5. Three commercial tomato cultivars (Valouro-RZ, Ghandora-F1, and Feisty-Red) were used. Both self-rooted plants and plants grafted onto Maxifort rootstocks were transplanted onto a perlite substrate. The recorded data revealed that all studied cultivars were critically affected by higher salinity (≈ 9.5 dS m-1) compared to low (≈ 2.5 dS m-1) and medium (≈ 6.0 dS m-1) salinity levels. The Variations in Tr, Pn, gs, chlorophyll content of leaf, and yield between medium and high salinity trials were reported at 3%, 5%, 9%, 5%, and 7.1%, respectively, whereas no significant differences were observed between low and medium salinities. However, at medium salinity levels, grafted plants performed better in photosynthesis than non-grafted plants. This is due to the accumulation of leaf proline, which maintains osmotic regulation and photosynthetic activity by preventing cell damage at medium salinities. Hence, this study confirmed the use of saline water for growing tomatoes under hydroponic conditions up to an EC of 6.0 dS m-1 including the EC of nutrient fertilizers.
