Confocal 3D reflectance imaging through multimode fiber without wavefront shaping.

ABSTRACT: Imaging through optical multimode fiber (MMF) has the potential to enable hair-thin endoscopes that reduce the invasiveness of imaging deep inside tissues and organs. Active wavefront shaping and fluorescent labeling have recently been exploited to overcome modal scrambling and enable MMF imaging. Here, we present a computational approach that circumvents the need for active wavefront control and exogenous fluorophores. We demonstrate the reconstruction of depth-gated confocal images through MMF using a raster-scanned, focused input illumination at the fiber proximal end. We show the compatibility of this approach with quantitative phase, dark-field, and polarimetric imaging. Computational imaging through MMF opens a new pathway for minimally invasive imaging in medical diagnosis and biological investigations.

Speckle-correlation imaging through a kaleidoscopic multimode fiber.

Project description:Speckle-correlation imaging techniques are widely used for noninvasive imaging through complex scattering media. While light propagation through multimode fibers and scattering media share many analogies, reconstructing images through multimode fibers from speckle correlations remains an unsolved challenge. Here, we exploit a kaleidoscopic memory effect emerging in square-core multimode fibers and demonstrate fluorescence imaging with no prior knowledge on the fiber. Experimentally, our approach simply requires to translate random speckle patterns at the input of a square-core fiber and to measure the resulting fluorescence intensity with a bucket detector. The image of the fluorescent object is then reconstructed from the autocorrelation of the measured signal by solving an inverse problem. This strategy does not require the knowledge of the fragile deterministic relation between input and output fields, which makes it promising for the development of flexible minimally invasive endoscopes.

Shaping the light amplified in a multimode fiber.

Project description:Propagation of light in multimode optical fibers usually gives a spatial and temporal randomization of the transmitted field similar to the propagation through scattering media. Randomization still applies when scattering or multimode propagation occurs in gain media. We demonstrate that appropriate structuration of the input beam wavefront can shape the light amplified by a rare-earth-doped multimode fiber. Profiling of the wavefront was achieved by a deformable mirror in combination with an iterative optimization process. We present experimental results and simulations showing the shaping of a single sharp spot at different places in the output cross-section of an ytterbium-doped fiber amplifier. Cleaning and narrowing of the amplifier far-field pattern was realized as well. Tailoring the wavefront to shape the amplified light can also serve to improve the effective gain. The shaping approach still works under gain saturation, showing the robustness of the method. Modeling and experiments attest that the shaping is effective even with a highly multimode fiber amplifier carrying up to 127 modes.

Mitigating stimulated Brillouin scattering in multimode fibers with focused output via wavefront shaping.

Project description:The key challenge for high-power delivery through optical fibers is overcoming nonlinear optical effects. To keep a smooth output beam, most techniques for mitigating optical nonlinearities are restricted to single-mode fibers. Moving out of the single-mode paradigm, we show experimentally that wavefront-shaping of coherent input light to a highly multimode fiber can increase the power threshold for stimulated Brillouin scattering (SBS) by an order of magnitude, whilst simultaneously controlling the output beam profile. The SBS suppression results from an effective broadening of the Brillouin spectrum under multimode excitation, without broadening of transmitted light. Strongest suppression is achieved with selective mode excitation that gives the broadest Brillouin spectrum. Our method is efficient, robust, and applicable to continuous waves and pulses. This work points toward a promising route for mitigating detrimental nonlinear effects in optical fibers, enabling further power scaling of high-power fiber systems for applications to directed energy, remote sensing, and gravitational-wave detection.

Non-invasive and noise-robust light focusing using confocal wavefront shaping.

Project description:Wavefront-shaping is a promising approach for imaging fluorescent targets deep inside scattering tissue despite strong aberrations. It enables focusing an incoming illumination into a single spot inside tissue, as well as correcting the outgoing light scattered from the tissue. Previously, wavefront shaping modulations have been successively estimated using feedback from strong fluorescent beads, which have been manually added to a sample. However, such algorithms do not generalize to neurons whose emission is orders of magnitude weaker. We suggest a wavefront shaping approach that works with a confocal modulation of both the illumination and imaging arms. Since the aberrations are corrected in the optics before the detector, the low photon budget is directed into a single sensor spot and detected with high signal-noise ratio. We derive a score function for modulation evaluation from mathematical principles, and successfully use it to image fluorescence neurons, despite scattering through thick tissue.

In vivo imaging of oral neoplasia using a miniaturized fiber optic confocal reflectance microscope.

Project description:The purpose of this study was to determine whether in vivo images of oral mucosa obtained with a fiber optic confocal reflectance microscope could be used to differentiate normal and neoplastic tissues. We imaged 20 oral sites in eight patients undergoing surgery for squamous cell carcinoma. Normal and abnormal areas within the oral cavity were identified clinically, and real-time videos of each site were obtained in vivo using a fiber optic confocal reflectance microscope. Following imaging, each site was biopsied and submitted for histopathologic examination. We identified distinct features, such as nuclear irregularity and spacing, which can be used to qualitatively differentiate between normal and abnormal tissue. Representative confocal images of normal, pre-neoplastic, and neoplastic oral tissue are presented. Previous work using much larger microscopes has demonstrated the ability of confocal reflectance microscopy to image cellular and tissue architecture in situ. New advances in technology have enabled miniaturization of imaging systems for in vivo use.

Swept-source multimode fiber imaging.

Project description:High-resolution compressive imaging via a flexible multimode fiber is demonstrated using a swept-laser source and wavelength dependent speckle illumination. An in-house built swept-source allowing for independent control of bandwidth and scanning range is used to explore and demonstrate a mechanically scan-free approach for high-resolution imaging through an ultrathin and flexible fiber probe. The computational image reconstruction is shown by utilizing a narrow sweeping bandwidth of [Formula: see text] nm while acquisition time is decreased by 95% compared to conventional raster scanning endoscopy. Demonstrated narrow-band illumination in the visible spectrum is vital for the detection of fluorescence biomarkers in neuroimaging applications. The proposed approach yields device simplicity and flexibility for minimally invasive endoscopy.

Non-Invasive Imaging Through Scattering Medium by Using a Reverse Response Wavefront Shaping Technique.

Project description:Fundamental challenge of imaging through a scattering media has been resolved by various approaches in the past two decades. Optical wavefront shaping technique is one such method in which one shapes the wavefront of light entering a scattering media using a wavefront shaper such that it cancels the scattering effect. It has been the most effective technique in focusing light inside a scattering media. Unfortunately, most of these techniques require direct access to the scattering medium or need to know the scattering properties of the medium beforehand. Through the novel scheme presented on this paper, both the illumination module and the detection are on the same side of the inspected object and the imaging process is a real time fast converging operation. We model the scattering medium being a biological tissue as a matrix having mathematical properties matched to the physical and biological aspects of the sample. In our adaptive optics scheme, we aim to estimate the scattering function and thus to encode the intensity of the illuminating laser light source using DMD (Digital Micromirror Device) with an inverse scattering function of the scattering medium, such that after passing its scattering function a focused beam is obtained. We optimize the pattern to be displayed on the DMD using Particle Swarm Algorithm (PSO) which eventually help in retrieving a 1D object hidden behind the media.

NeuWS: Neural wavefront shaping for guidestar-free imaging through static and dynamic scattering media.

Project description:Diffraction-limited optical imaging through scattering media has the potential to transform many applications such as airborne and space-based imaging (through the atmosphere), bioimaging (through skin and human tissue), and fiber-based imaging (through fiber bundles). Existing wavefront shaping methods can image through scattering media and other obscurants by optically correcting wavefront aberrations using high-resolution spatial light modulators-but these methods generally require (i) guidestars, (ii) controlled illumination, (iii) point scanning, and/or (iv) statics scenes and aberrations. We propose neural wavefront shaping (NeuWS), a scanning-free wavefront shaping technique that integrates maximum likelihood estimation, measurement modulation, and neural signal representations to reconstruct diffraction-limited images through strong static and dynamic scattering media without guidestars, sparse targets, controlled illumination, nor specialized image sensors. We experimentally demonstrate guidestar-free, wide field-of-view, high-resolution, diffraction-limited imaging of extended, nonsparse, and static/dynamic scenes captured through static/dynamic aberrations.

Phase contrast reflectance confocal brain imaging at 1650 nm.

Project description:SignificanceThe imaging depth of microscopy techniques is limited by the ability of light to penetrate biological tissue. Recent research has addressed this limitation by combining a reflectance confocal microscope with the NIR-II (or shortwave infrared) spectrum. This approach offers significant imaging depth, is straightforward in design, and remains cost-effective. However, the imaging system, which relies on intrinsic signals, could benefit from adjustments in its optical design and post-processing methods to differentiate cortical cells, such as neurons and small blood vessels.AimWe implemented a phase contrast detection scheme to a reflectance confocal microscope using NIR-II spectral range as illumination.ApproachWe analyzed the features retrieved in the images while testing the imaging depth. Moreover, we introduce an acquisition method for distinguishing dynamic signals from the background, allowing the creation of vascular maps similar to those produced by optical coherence tomography.ResultsThe phase contrast implementation is successful to retrieve deep images in the cortex up to 800 μm using a cranial window. Vascular maps were retrieved at similar cortical depth and the possibility of combining multiple images can provide a vessel network.ConclusionsPhase contrast reflectance confocal microscopy can improve the outlining of cortical cell bodies. With the presented framework, angiograms can be retrieved from the dynamic signal in the biological tissue. Our work presents an optical implementation and analysis techniques from a former microscope design.

High-gain and high-speed wavefront shaping through scattering media.

Project description:Wavefront shaping (WFS) is emerging as a promising tool for controlling and focusing light in complex scattering media. The shaping system's speed, the energy gain of the corrected wavefronts, and the control degrees of freedom (DOF) are the most important metrics for WFS, especially for highly scattering and dynamic samples. Despite recent advances, current methods suffer from trade-offs that limit satisfactory performance to only one or two of these metrics. Here, we report a WFS technique that simultaneously achieves high speed, high energy gain, and high control DOF. By combining photorefractive crystal-based analog optical phase conjugation (AOPC) and stimulated emission light amplification, our technique achieves an energy gain approaching unity, more than three orders of magnitude larger than conventional AOPC. The response time of ~10 μs with about 106 control modes corresponds to an average mode time of about 0.01 ns/mode, which is more than 50 times lower than some of the fastest WFS systems to date. We anticipate that this technique will be instrumental in overcoming the optical diffusion limit in photonics and translate WFS techniques to real-world applications.
