Identification and Characterization of the Phosphate-Solubilizing Bacterium Pantoea sp. S32 in Reclamation Soil in Shanxi, China.

Project description:Phosphate solubilizing bacteria (PSB) can convert insoluble forms of phosphorus (P) to accessible forms. Five highly efficient PSB strains, H22, Y11, Y14, Y34, and S32, were screened and isolated from an alfalfa rhizosphere in heavy metal-contaminated reclamation area in Shanxi Province, China. Based on morphological observations, 16S rRNA sequencing, cellular fatty acid composition analysis, and the BIOLOG test, H22, Y11, and Y34 were identified as Pseudomonas sp., while Y14 and S32 were identified as Pantoea sp. Among them, S32 showed the highest P-solubilizing efficiency in culture medium containing Ca3(PO4)2, lecithin, and powered phosphate rock. The culture medium conditions to obtain the highest P-solubilization efficiency were optimized as follows: the culture temperature was 30°C; the incubation time was 5 days; the initial pH was 7.0; and glucose served as the carbon source. Furthermore, the P-solubilization efficiency of S32 in media containing CaHPO4, lecithin, phosphate rock (PR), FePO4, or AlPO4 was determined to be 18.38, 3.07, 0.16, 0.51, or 2.62%, respectively. In addition, the acid and alkali phosphatase activities of S32 were tested as 6.94 U/100 mL and 4.12 U/100 mL, respectively. The soil inoculation experiment indicated that inoculation with S32 resulted in an obvious improvement in the available P of both the experimental and reclaimed soil. The rice seedling growth experiment also suggested that the application of S32 significantly increased the plant height, biomass, root growth, and P uptake of rice in both experimental and reclaimed soil. Taken together, the isolated S32 strain showed high P-solubilization capacity for both Pi and Po, and its ameliorative effect on reclaimed soil recovery provides the theoretical basis for crop development in the reclaimed soil of mine field.

Complete genome sequence of Nguyenibacter sp. L1, a phosphate solubilizing bacterium isolated from Lespedeza bicolor rhizosphere.

Project description:Phosphorus (P) deficiency is a predominant constraint on plant growth in acidified soils, largely due to the sequestration of P by toxic aluminum (Al) compounds. Indigenous phosphorus-solubilizing bacteria (PSBs) capable of mobilizing Al-P in these soils hold significant promise. A novel Al-P-solubilizing strain, Al-P Nguyenibacter sp. L1, was isolated from the rhizosphere soil of healthy Lespedeza bicolor plants indigenous to acidic terrains. However, our understanding of the genomic landscape of bacterial species within the genus Nguyenibacter remains in its infancy. To further explore its biotechnological potentialities, we sequenced the complete genome of this strain, employing an amalgamation of Oxford Nanopore ONT and Illumina sequencing platforms. The resultant genomic sequence of Nguyenibacter sp. L1 manifests as a singular, circular chromosome encompassing 4,294,433 nucleotides and displaying a GC content of 66.73%. The genome was found to host 3,820 protein-coding sequences, 12 rRNAs, and 55 tRNAs. Intriguingly, annotations derived from the eggNOG and KEGG databases indicate the presence of genes affiliated with phosphorus solubilization and nitrogen fixation, including iscU, glnA, and gltB/D associated with nitrogen fixation, and pqqBC associated with inorganic phosphate dissolution. Several bioactive secondary metabolite genes in the genome, including pqqCDE, phytoene synthase and squalene synthase predicted by antiSMASH. Moreover, we uncovered a complete metabolic pathway for ammonia, suggesting an ammonia-affinity property inherent to Nguyenibacter sp. L1. This study verifies the nitrogen-fixing and phosphate-dissolving abilities of Nguyenibacter sp. L1 at the molecular level through genetic screening and analysis. The insights gleaned from this study offer strategic guidance for future strain enhancement and establish a strong foundation for the potential incorporation of this bacterium into agricultural practices.

Phosphate-Solubilizing Bacillus sp. Modulate Soil Exoenzyme Activities and Improve Wheat Growth.

Project description:Phosphorus (P) is a vital mineral nutrient in agriculture and its deficiency results in reduced growth, yield, and grain quality in cereals. Much of the applied P in agriculture becomes fixed in soils, limiting its accessibility to plants. Thus, investigating sustainable strategies to release fixed P resources and enhance plant uptake is crucial. This study explored how plant-associated bacteria employ phosphate solubilizing mechanisms to improve P availability. The growth patterns of four bacterial strains, namely Bacillus subtilis ZE15 and ZR3, along with Bacillus megaterium ZE32 and ZR19, were examined in Pikovskaya's broth culture with and without the addition of insoluble phosphorus (P). In the absence of P amendment, most strains reached a stationary growth phase by the fourth day. However, their responses diverged when exposed to P-amended media. Particularly, ZE15 demonstrated the highest P solubilization capability, achieving up to 130 µg mL-1 solubilization in vitro. All strains produced organic acids in Pikovskaya's broth culture. A comparison of the influence of Ca3(PO4)2 revealed significantly greater organic acid quantities in the presence of insoluble P. Notably, strain ZE15 exhibited the highest phosphate esterase activity (3.65 nmol g-1 dry matter), while strain ZE32 showed the highest ß-D glucosidase activity (2.81 nmol g-1 dry matter) in the presence of insoluble P. The ability of Bacillus species to solubilize P in combination with increased exoenzyme activity in the rhizosphere could be used in future studies to support P uptake through enhanced solubilization and mineralization.

Strain AK 1817 biotransforms the ergostane triterpenoid antcin K from the fungus Antrodia cinnamomea. The genome was sequenced using the PacBio RS II platform and consists of one chromosome of 4,096,020 bp, comprising 3,907 protein-coding genes, 75 tRNAs, 30 rRNAs, 5 noncoding RNAs (ncRNAs), and 100 pseudogenes.

Genome-resolved metagenomics identifies the particular genetic traits of phosphate-solubilizing bacteria in agricultural soil

Project description:Bacteria play a key role in phosphate solubilization, but related genome-centric research on agricultural microbiomes is scarce. Here, we reconstructed 472 metagenome-assembled genomes (MAGs) covering agricultural soils from six long-term field trials across China. A total of 79 MAGs contained gcd encoding quinoprotein glucose dehydrogenase (GCD), which is the key biomarker for phosphate solubilization. Our findings showed that all GCD-MAGs represent potentially novel species, with gcd copy numbers varying from 1 to 10 per genome. Large genome size, a high ratio of glycosyl hydrolase genes, and increased capacity for carbohydrate utilization were specific traits of GCD-MAGs. Notably, the gcd copy number showed a significant and positive correlation with genome size. Generated using a machine learning approach, our findings were validated in a dataset of 692 genotypes covering the 18 bacterial families to which the 79 GCD-MAGs belong. Our results improve the knowledge of both the diversity and the genetic composition of phosphate-solubilizing bacteria. In particular, they reveal a genomic link between phosphate solubilization capacity and increased potential for carbohydrate metabolism, which may accelerate targeted engineering and improve management practices for sustainable agriculture.
