Effect of TiO₂ on Selected Pathogenic and Opportunistic Intestinal Bacteria.

ABSTRACT: Food-grade titanium dioxide (TiO₂) containing a nanoparticle fraction (TiO₂ NPs-nanoparticles) is widely used as a food additive (E171 in the EU). In recent years, questions concerning its effect on the gastrointestinal microbiota have been raised. In the present study, we examined interactions between bacteria and TiO₂. The study involved six pathogenic/opportunistic bacterial strains and four different-sized TiO₂ types: three types of food-grade E171 compounds and TiO₂ NPs (21 nm). Each bacterial strain was exposed to four concentrations of TiO₂ (60, 150, 300, and 600 mg/L TiO₂). The differences in the growth of the analyzed strains, caused by the type and concentration of TiO₂, were observed. The growth of a majority of the strains was shown to be inhibited after exposure to 300 and 600 mg/L of the food-grade E171 and TiO₂ NPs.

Leprous lesion presents enrichment of opportunistic pathogenic bacteria.

Project description:Leprosy is a chronic infectious disease that remains a major challenge to public health in endemic countries. Increasing evidence has highlighted the importance of microbiota for human general health and, as such, the study of skin microbiota is of interest. But while studies are continuously revealing the complexity of human skin microbiota, the microbiota of leprous cutaneous lesions has not yet been characterized. Here we used Sanger and massively parallel small sub-unit rRNA (SSU) rRNA gene sequencing to characterize the microbiota of leprous lesions, and studied how it differs from the bacterial skin composition of healthy individuals previously described in the literature. Taxonomic analysis of leprous lesions revealed main four phyla: Proteobacteria, Firmicutes, Bacteroidetes, and Actinobacteria, with Proteobacteria presenting the highest diversity. There were considerable differences in the distribution of Proteobacteria, Bacteroidetes, Firmicutes, and Actinobacteria, with the first two phyla enriched and the other markedly diminished in the leprous lesions, when compared with healthy skin. Propionibacterium, Corynebacterium and Staphylococcus, resident and abundant in healthy skin, were underrepresented in skin from leprous lesions. Most of the taxa found in skin from leprous lesions are not typical in human skin and potentially pathogenic, with the Burkholderia, Pseudomonas and Bacillus genera being overrepresented. Our data suggest significant shifts of the microbiota with emergence and competitive advantage of potentially pathogenic bacteria over skin resident taxa.

Multidrug-resistant Opportunistic and Pathogenic Bacteria Contaminate Algerian Banknotes Currency.

Project description:Currency is one of the most exchanged items in human communities as it is used daily in exchange for goods and services. It is handled by persons with different hygiene standards and can transit in different environments. Hence, money can constitute a reservoir for different types of human pathogens. This study aimed to evaluate the potential of Algerian banknotes to shelter opportunistic pathogenic and multiresistant bacteria. To that end, 200 circulating notes of four different denominations were collected from various places and analyzed for their bacterial loads and contents. Besides, predominant strains were identified and characterized by biochemical and molecular methods, and their resistance profiles against 34 antibiotics were determined. Our results indicated that 100% of the studied banknotes were contaminated with bacteria. The total bacterial concentrations were relatively high, and different bacterial groups were grown, showing important diversity. In total, 48 predominant strains were identified as belonging to 17 genera. Staphylococcus and Micrococcus were the most prevalent genera, followed by Bacillus, Pseudomonas, and Acinetobacter. Antibiotic susceptibility testing showed that all the isolates harbored resistance to at least two molecules, and worrying resistance levels were observed. These findings prove that Algerian currency harbors opportunistic multiresistant bacteria and could potentially act as a vehicle for the spread of bacterial diseases and as a reservoir for antibiotic resistance genes among the community. Therefore, no cash payment systems should be developed and generalized to minimize cash handling and subsequent potential health risks.

Photocatalytic effect of N-TiO2 conjugated with folic acid against biofilm-forming resistant bacteria.

Project description:Antibiotic resistance challenges the treatment of bacterial biofilm-related infections, but the use of nanoparticles as a treatment is a promising strategy to overcome bacterial infections. This study applied nitrogen-doped titanium dioxide (N-TiO2) conjugated with folic acid (FA) on biofilm-forming resistant bacteria. The photocatalytic effect of TiO2 nanoparticles (NPs) was studied under ultraviolet (UV), visible light, and dark conditions at 60, 120, and 180 min against planktonic cells and biofilms of Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), and Pseudomonas aeruginosa. TiO2 NPs were in the anatase phase, spherical shaped with sizes of 10-13 nm, and effectively doped and conjugated with N and FA. The FA-conjugated nanoparticles (N-TiO2-FA and FA-TiO2) were shown to have a bactericidal effect on all bacteria between 60 and 180 min under UV and visible light conditions. Concerning biofilms, N-TiO2-FA was shown to have a highly disruptive effect on all bacterial biofilms under UV irradiation at 180 min. Meanwhile, the nanoparticles did not show DNA damaging potential and they had no cytostatic effect, indicating that these NPs are biocompatible. In sum, nanoparticle conjugation with FA promoted photocatalytic effectiveness, revealing the promise this nanomaterial holds as a biocompatible antimicrobial agent.

TiO2/Karaya Composite for Photoinactivation of Bacteria.

Project description:TiO2/Karaya composite was synthesized by the sol-gel method for the photoinactivation of pathogens. This is the first time that we have reported this composite for an antimicrobial approach. The structure, morphology, and optical properties were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive X-rays (EDS), Fourier transform infrared spectroscopy (FTIR), and diffuse reflectance, and the surface area was characterized by the BET method. The XRD and EDS results showed that the TiO2/Karaya composite was successfully stabilized by the crystal structure and pore diameter distribution, indicating a composite of mesoporous nature. Furthermore, antibacterial experiments showed that the TiO2/Karaya composite under light was able to photoinactivate bacteria. Therefore, the composite is a promising candidate for inhibiting the growth of bacteria.

Photo-reduction of heavy metal ions and photo-disinfection of pathogenic bacteria under simulated solar light using photosensitized TiO2 nanofibers.

Project description:We report the photosensitization of electrospun titania nanofibers, with a mean diameter of 195 nm, by low bandgap silver sulfide nanoparticles of 11-23 nm mean size with the aim of treating heavy metal ions and pathogenic bacteria simultaneously under simulated solar light irradiation. The 17 nm Ag2S/TiO2 nanofibers showed 90% photocatalytic reduction of Cr(vi) at pH of 3 with a pseudo-first order rate constant of 0.016 min-1 which is significantly better than the previously reported for Ag-Ag2S/TiO2 composite particles. The antibacterial capability of the Ag2S/TiO2 nanofibers was evaluated via photo-disinfection of the Gram-positive and Gram-negative bacterial strains. The smallest sized 11 nm Ag2S/TiO2 nanofiber showed the best bactericidal efficiency of 100% and 90.6% against Gram-negative E. coli and Gram-positive S. aureus after 1 h of irradiation, respectively, whereas, only 50% E. coli and 41% S. aureus were found to be inactivated in dark. Furthermore, a UV-O3 treatment of the 11 nm Ag2S/TiO2 nanofibers remarkably enhanced the antibacterial activity where 89% E. coli and 81% S. aureus were inactivated in just 10 min of the irradiation. Enhanced photocatalytic activity is attributed to the efficient charge separation and transfer and reduced electron-hole recombination induced by the effective heterojunction formation between TiO2 and the optimally sized Ag2S nanoparticles. The disinfection nature of the Ag2S nanoparticles, role of the generated hydroxyl species under irradiation, and the cell wall damage mechanism is also discussed. This study demonstrates the potential use of these multifunctional composite TiO2 nanofibers for water remediation.

Bee Pollen Extracts: Chemical Composition, Antioxidant Properties, and Effect on the Growth of Selected Probiotic and Pathogenic Bacteria.

Project description:This paper evaluated the chemical and biological properties of bee pollen samples from Romania. Firstly, the bee pollen alcoholic extracts (BPEs) were obtained from raw bee pollen harvested by Apis mellifera carpatica bees. The chemical composition of BPE was obtained by determination of total phenol content and total flavonoid content, UHPLC-DAD-ESI/MS analysis of phenolic compounds, and GC-MS analysis of fatty acids, esters, and terpenes. Additionally, the antioxidant activity was evaluated by the Trolox Equivalent Antioxidant Capacity method. Furthermore, the biological properties of BPE were evaluated (antimicrobial and cytotoxic activity). The raw BP samples studied in this paper had significant phenolic acid and flavonoid content, and moderate fatty acid, ester, and terpene content. P1, P2, and P4 have the highest TPC and TFC levels, and the best antioxidant activity. All BPEs studied had antimicrobial activity on pathogenic strains isolated from the clinic or standard strains. A synergistic antimicrobial effect of the BPEs was observed along with the soluble compounds of L. rhamnosus MF9 and E. faecalis 2M17 against some pathogenic (clinical) strains and, considering the tumour proliferation inhibitory activity, makes BP a potential prebiotic and antitumour agent for the gut environment.

Photo-catalyzed TiO2 inactivates pathogenic viruses by attacking viral genome.

Project description:Previous observations have been reported that viruses were inactivated using strong irradiation. Here, new evidence was disclosed by studying the effects of nanosized TiO2 on viral pathogens under a low irradiation condition (0.4 mW/cm2 at UVA band) that mimics the field setting. We showed that photo-activated TiO2 efficiently inhibits hepatitis C virus infection, and weak indoor light with intensity of 0.6 mW/cm2 at broad-spectrum wavelength and around 0.15 mW/cm2 of UVA band also lead to partial inhibition. Mechanistic studies demonstrated that hydroxyl radicals produced by photo-activated TiO2 do not destroy virion structure and contents, but attack viral RNA genome, thus inactivating the virus. Furthermore, we showed that photo-activated TiO2 inactivates a broad range of human viral pathogens, including SARS-CoV-2, a novel coronavirus responsible for the ongoing COVID-19 pandemic. In conclusion, we showed that photo-catalyzed nanosized TiO2 inactivates pathogenic viruses, paving a way to its field application in control of viral infectious diseases.

The Influence of Different Forms of Silver on Selected Pathogenic Bacteria.

Project description:The application of silver nanoparticles as an antibacterial agent is becoming more common. Unfortunately, their effect on microorganisms is still not fully understood. Therefore, this paper attempts to investigate the influence of silver ions, biologically synthesized silver nanoparticles and nanoparticles functionalized with antibiotics on molecular bacteria profiles. The initial stage of research was aimed at the mechanism determination involved in antibiotics sorption onto nanoparticles' surface. For this purpose, the kinetics study was performed. Next, the functionalized formulations were characterized by Fourier transform infrared spectroscopy (FT-IR), dynamic light scattering (DLS) and a zeta potential study. The results reveal that functionalization is a complex process, but does not significantly affect the stability of biocolloids. Furthermore, the antimicrobial assays, in most cases, have shown no increases in antibacterial activity after nanoparticle functionalization, which suggests that the functionalization process does not always generate the improved antimicrobial effect. Finally, the matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) technique was employed to characterize the changes in the molecular profile of bacteria treated with various antibacterial agents. The recorded spectra proved many differences in bacterial lipids and proteins profiles compared to untreated cells. In addition, the statistical analysis of recorded spectra revealed the strain-dependent nature of stress factors on the molecular profile of microorganisms.

The DH31/CGRP enteroendocrine peptide triggers intestinal contractions favoring the elimination of opportunistic bacteria.

Project description:The digestive tract is the first organ affected by the ingestion of foodborne bacteria. While commensal bacteria become resident, opportunistic or virulent bacteria are eliminated from the gut by the local innate immune system. Here we characterize a new mechanism of defense, independent of the immune system, in Drosophila melanogaster. We observed strong contractions of longitudinal visceral muscle fibers for the first 2 hours following bacterial ingestion. We showed that these visceral muscle contractions are induced by immune reactive oxygen species (ROS) that accumulate in the lumen and depend on the ROS-sensing TRPA1 receptor. We then demonstrate that both ROS and TRPA1 are required in a subset of anterior enteroendocrine cells for the release of the DH31 neuropeptide which activates its receptor in the neighboring visceral muscles. The resulting contractions of the visceral muscles favors quick expulsion of the bacteria, limiting their presence in the gut. Our results unveil a precocious mechanism of defense against ingested opportunistic bacteria, whether they are Gram-positive like Bacillus thuringiensis or Gram-negative like Erwinia carotovora carotovora. Finally, we found that the human homolog of DH31, CGRP, has a conserved function in Drosophila.

Intestinal Microecology of Mice Exposed to TiO2 Nanoparticles and Bisphenol A.

Project description:Exposure to titanium dioxide nanoparticles (TiO2 NPs) and bisphenol A (BPA) is ubiquitous, especially through dietary and other environmental pathways. In the present study, adult C57BL/6J mice were exposed to TiO2 NPs (100 mg/kg), BPA (0, 5, and 50 mg/kg), or their binary mixtures for 13 weeks. The 16S rDNA amplification sequence analysis revealed that co-exposure to TiO2 NPs and BPA altered the intestinal microbiota; however, this alteration was mainly caused by TiO2 NPs. Faecal metabolomics analysis revealed that 28 metabolites and 3 metabolic pathways were altered in the co-exposed group. This study is the first to reveal the combined effects of TiO2 NPs and BPA on the mammalian gut microbial community and metabolism dynamics, which is of great value to human health. The coexistence of TiO2 NPs and BPA in the gut poses a potential health risk due to their interaction with the gut microbiota.
