Prevalence of Tick-Borne Pathogens in Questing Ixodes ricinus and Dermacentor reticulatus Ticks Collected from Recreational Areas in Northeastern Poland with Analysis of Environmental Factors.

ABSTRACT: Ticks, such as Ixodes ricinus and Dermacentor reticulatus, act as vectors for multiple pathogens posing a threat to both human and animal health. As the process of urbanization is progressing, those arachnids are being more commonly encountered in urban surroundings. In total, 1112 I. ricinus (n = 842) and D. reticulatus (n = 270) ticks were collected from several sites, including recreational urban parks, located in Augustów and Białystok, Poland. Afterwards, the specimens were examined for the presence of Borrelia spp., Babesia spp., Anaplasma phagocytophilum, Rickettsia spp., Bartonella spp., and Coxiella burnetii using the PCR method. Overall obtained infection rate reached 22.4% (249/1112). In total, 26.7% (225/842) of I. ricinus was infected, namely with Borrelia spp. (25.2%; 212/842), Babesia spp. (2.0%; 17/842), and A. phagocytophilum (1.2%; 10/842). Among D. reticulatus ticks, 8.9% (24/270) were infected, specifically with Babesia spp. (7.0%; 19/270), A. phagocytophilum (1.1%; 3/270), and Borrelia burgdorferi s.l. (0.7%; 2/270). No specimen tested positively for Rickettsia spp., Bartonella spp., or Coxiella burnetii. Co-infections were detected in 14 specimens. Results obtained in this study confirm that I. ricinus and D. reticulatus ticks found within the study sites of northeastern Poland are infected with at least three pathogens. Evaluation of the prevalence of pathogens in ticks collected from urban environments provides valuable information, especially in light of the growing number of tick-borne infections in humans and domesticated animals.

Project description:BackgroundLyme borreliosis and other tick-borne diseases emerge from increased interactions between humans, other animals, and infected ticks. The risk of acquiring a tick-borne infection varies across space and time, so knowledge of the occurrence and prevalence of pathogens in ticks can facilitate disease diagnosis in a specific area and the implementation of mitigation measures and awareness campaigns. Here we identify the occurrence and prevalence of several pathogens in Ixodes ricinus ticks in Wester Ross, Northwest Scotland, a region of high tourism and tick exposure, yet data-poor in terms of tick-borne pathogens.MethodsQuesting I. ricinus nymphs (n = 2828) were collected from 26 sites in 2018 and 2019 and tested for the presence of tick-borne pathogens using PCR-based methods. Prevalence was compared with other regions of Scotland, England, Wales, and the Netherlands.ResultsAnaplasma phagocytophilum (4.7% prevalence), Borrelia burgdorferi sensu lato (s.l.) (2.2%), Babesia from clade X (0.2%), Rickettsia helvetica (0.04%), and Spiroplasma ixodetis (0.4%) were detected, but no Neoehrlichia mikurensis, Borrelia miyamotoi, or Babesia microti. Typing of A. phagocytophilum using a fragment of the GroEL gene identified the presence of both ecotype I and ecotype II. Genospecies identification of Borrelia burgdorferi s.l. revealed B. afzelii (53% of infected nymphs), B. garinii (9%), B. burgdorferi sensu stricto (7%), and B. valaisiana (31%). We found similar prevalence of A. phagocytophilum in Wester Ross as in the Netherlands, but higher than in other parts of Great Britain. We found lower B. burgdorferi s.l. prevalence than in England or the Netherlands, and similar to some other Scottish studies. We found higher prevalence of B. valaisiana and lower prevalence of B. garinii than in other Scottish studies. We found S. ixodetis at much lower prevalence than in the Netherlands, and R. helvetica at much lower prevalence than in England and the Netherlands.ConclusionsAs far as we know, this is the first description of S. ixodetis in Great Britain. The results are relevant for disease surveillance and management for public and veterinary health. The findings can also aid in designing targeted public health campaigns and in raising awareness among outdoor recreationists and professionals.

Project description:BackgroundOver the last decades, climatic and environmental changes have led to an expanding seasonal activity pattern and increasing distribution of ticks across Europe. In particular, Dermacentor reticulatus is now commonly found on dogs in central Europe. The present study compared attachment sites between Ixodes spp. and Dermacentor reticulatus ticks collected by veterinarians from dogs and cats, and investigated risk factors associated with tick infestation intensity and engorgement duration.ResultsThe dataset comprised 6,335 dogs harbouring 10,287 ticks (8,095 Ixodes ricinus, 1,860 D. reticulatus, 218 Ixodes hexagonus/Ixodes canisuga, 114 of other tick species) respectively 4,248 cats harbouring 8,005 ticks (7,344 I. ricinus, 56 D. reticulatus, 505 I. hexagonus/I. canisuga, 100 of other tick species). Differing sites of tick attachment were not only found between the different host and tick species, but also between the tick developmental stages. Regarding the risk of infestation with multiple ticks, dogs and cats living in rural areas harboured significantly more often multiple than single specimens. Further, a long coat in cats was associated with a higher probability of multiple infestation, while this was not observed in dogs. However, there was a tendency towards a potential influence of the density of the undercoat (p = 0.051). In dogs, a tall to very tall body size as well as folded ears increased the risk of multiple infestation, while in cats, increasing age and increasing body size were negatively associated with multiple infestations. Ticks with an engorgement duration of > 48 h were found significantly more often on senior dogs and cats than on younger individuals, as well as on working/utility dog breeds, while engorgement duration was negatively correlated with infestation intensity in dogs. In cats, female gender and a rural residence were significantly associated with longer attachment duration.ConclusionsIndividual as well as breed specific characteristics can lead to a higher tick infestation intensity or longer engorgement duration. The knowledge of tick attachment sites and specific risk factors can help to raise awareness among owners concerning the importance of tick control with licensed acaricides, as recommended e.g. by the European Scientific Counsel Companion Animal Parasites (ESCCAP), and may aid in early tick removal to decrease the risk of pathogen transmission to dogs and cats whose owners nonetheless refuse acaricidal drugs.
