Directional growth of octacalcium phosphate using micro-flow reactor mixing and subsequent aging.

ABSTRACT: Well-defined belt-shaped particles of octacalcium phosphate were prepared by mixing aqueous solutions of calcium acetate and that of sodium phosphate monobasic with the aid of a micro-flow reactor. Higher crystallinity and narrower particle size distribution were achieved by the micro-flow reactor if compared with the results of the batch reaction using the same solutions. The width of the belt was controlled by the mixing temperature (0.8 and 2.3 μm for the preparation at 50 and 70 °C, respectively). Post mixing aging at 50 °C, resulted in the directional growth of belt-shaped particles to obtain particles with the length of 17 μm (aspect ratio of 53). XRD and TEM analysis indicated that the micro-flow reactor could separate nucleation and growth allowing preferential growth along the a-direction.

Reactive mixing performance for a nanoparticle precipitation in a swirling vortex flow reactor.

Project description:Mixing performance for a consecutive competing reaction system has been investigated in a swirling vortex flow reactor (SVFR). The direct quadrature method of moments combined with the interaction by exchange with the mean (DQMOM-IEM) method was employed to model such reacting flows. This type of reactors is able to generate a strong swirling flow with a great shear gradient in the radial direction. Firstly, mixing at both macroscale and microscale was assessed by mean mixture fraction and its variance, respectively. It is found that macromixing can be rapidly achieved throughout the whole reactor chamber due to its swirling feature. However, micromixing estimated by Bachelor length scale is sensitive to turbulence. Moreover, the additional introduction of ultrasound irradiation can significantly improve the mixing uniformity, namely, free of any stagnant zone presented in the reactor chamber on a macroscale, and little variance deviating from the mean environment value can be observed on a microscale. Secondly, reaction progress variable and the reactant conversion serve as indicators for the occurrence of side reaction. It is found that strong turbulence and a relatively fast micromixing process compared to chemical reaction can greatly reduce the presence of by-product, which will then provide homogenous environment for particle precipitation. Moreover, due to the generation of cavitation bubbles and their subsequent collapse, ultrasound irradiation can further intensify turbulence, creating rather even environment for chemical reactions. Low conversion rate was observed and little by-products were generated consequently. Therefore, it is suggested that the SVFR especially intensified by ultrasound irradiation has the ability to provide efficient mixing performance for the fine-particle synthesis process.

Synthesis of Micro-/Nanohydroxyapatite Assisted by the Taylor-Couette Flow Reactor.

Project description:Hydroxyapatite (HAP) has received increasing attention as an essential chemical product with good biocompatibility and adsorption properties. Generally, amorphous calcium phosphate (ACP) was generated first in the reactor and transformed into HAP after a period of crystallization. In this work, a series of Taylor-Couette flow reactors with different inner diameters were designed to assist in synthesizing HAP micro-/nanocrystals. ACP was obtained in a Taylor-Couette flow reactor at Re = 247 and successfully transformed into needle-like HAP crystals with a length of about 200 nm and a uniform particle size distribution after crystallization transformation. The yield of a single reactor can reach 2.16 kg per day. The finite element analysis results and time-space diagram of flow pattern variation showed that the Taylor-Couette flow reactor could improve the mixing behavior and the flow field distribution. The Taylor-Couette flow reactor provides a valuable reference for synthesizing inorganic micro-/nanomaterials.

Renewable Wood Pulp Paper Reactor with Hierarchical Micro/Nanopores for Continuous-Flow Nanocatalysis.

Project description:Continuous-flow nanocatalysis based on metal nanoparticle catalyst-anchored flow reactors has recently provided an excellent platform for effective chemical manufacturing. However, there has been limited progress in porous structure design and recycling systems for metal nanoparticle-anchored flow reactors to create more efficient and sustainable catalytic processes. In this study, traditional paper is used for a highly efficient, recyclable, and even renewable flow reactor by tailoring the ultrastructures of wood pulp. The "paper reactor" offers hierarchically interconnected micro- and nanoscale pores, which can act as convective-flow and rapid-diffusion channels, respectively, for efficient access of reactants to metal nanoparticle catalysts. In continuous-flow, aqueous, room-temperature catalytic reduction of 4-nitrophenol to 4-aminophenol, a gold nanoparticle (AuNP)-anchored paper reactor with hierarchical micro/nanopores provided higher reaction efficiency than state-of-the-art AuNP-anchored flow reactors. Inspired by traditional paper materials, successful recycling and renewal of AuNP-anchored paper reactors were also demonstrated while high reaction efficiency was maintained.

Tooth enamel proteins enamelin and amelogenin cooperate to regulate the growth morphology of octacalcium phosphate crystals.

Project description:To examine the hypothetical cooperative role of enamelin and amelogenin in controlling the growth morphology of enamel crystals in the post-secretory stage, we applied a cation selective membrane system for the growth of octacalcium phosphate (OCP) in the truncated recombinant porcine amelogenin (rP148) with and without the 32kDa enamelin fragment. Enamelin alone inhibited the growth in the c-axis direction more than rP148, yielding OCP crystals with the smallest aspect ratio of all conditions tested. When enamelin was added to the amelogenin "gel-like matrix", the inhibitory action of the protein mixture on the growth of OCP in the c-axis direction was diminished, while that in the b-axis direction was increased. As a result, the length to width ratio (aspect ratio) of OCP crystal was markedly increased. Addition of enamelin to amelogenin enhanced the potential of amelogenin to stabilize the amorphous calcium phosphate (ACP) transient phase. The ratio of enamelin and amelogenin was crucial for stabilization of ACP and the growth of OCP crystals with larger aspect ratio. The cooperative regulatory action of enamelin and amelogenin was attained, presumably, through co-assembling of enamelin and amelogenin. These results have important implications in understanding the growth mechanism of enamel crystals with large aspect ratio.

Ultrafast and Reproducible Synthesis of Tailor-Made Octacalcium Phosphate.

Project description:Octacalcium phosphate (OCP) has excellent bone formation ability and a good resorption rate as compared to the commercial bone substitutes [e.g., Bio-Oss (Geistlich Pharma AG) and MBCP+ (Biomatlante)], as well as synthesized biomaterials (hydroxyapatite and tricalcium phosphate). The synthesis approach to obtain phase-pure OCP possesses a great challenge due to its complex reaction mechanism and narrow synthesis window. Thus, the current study aimed to overcome the synthesis challenges and to define the precise reaction conditions required for controllable and reproducible synthesis of OCP. Using the principles of the coprecipitation method, novel synthesis protocols were developed ensuring ultrafast synthesis of OCP within minutes. XRD analysis confirmed that phase-pure OCP was obtained. These processing schemes enabled the synthesis of tailor-made OCP with specific surface areas ranging from 16 to 91 m2/g. The synthesized OCPs exhibited plate-like morphology. The interaction of the synthesized OCPs with MC3T3-E1 cells was found to be nontoxic, confirming their cytocompatibility. The synthesis approach developed in this study indicates that challenges such as the reaction volume, stirring rate, and flow rate need to be addressed in the future to upscale the technology.

Adsorption of Serum Albumin onto Octacalcium Phosphate in Supersaturated Solutions Regarding Calcium Phosphate Phases.

Project description:Octacalcium phosphate (OCP) has been shown to enhance new bone formation, coupled with its own biodegradation, through osteoblasts and osteoclast-like cell activities concomitant with de novo hydroxyapatite (HA) formation and serum protein accumulation on its surface. However, the nature of the chemical environment surrounding OCP and how it affects its metabolism and regulates protein accumulation is unknown. The present study examined how the degree of supersaturation (DS) affects the bovine serum albumin (BSA) adsorption onto OCP in 150 mM Tris-HCl buffer at 37 °C and pH 7.4, by changing the Ca2+ ion concentration. The amount of BSA adsorbed onto OCP increased as the DS increased. In addition, the amount of newly formed calcium phosphate, which could be OCP, was increased, not only by increases in DS, but also at lower equilibrium concentrations of BSA. The increased adsorption capacity of BSA was likely related to the formation of calcium phosphate on the adsorbed OCP. Together the results suggested that the formation of new calcium phosphate crystals is dependent on both the DS value and the adsorbate protein concentration, which may control serum protein accumulation on the OCP surface in vivo.

Surface Properties of Octacalcium Phosphate Nanocrystals Are Crucial for Their Bioactivities.

Project description:The fundamental structure-biofunction relationship of calcium phosphates (CaPs) remains unclear despite their clinical successes as important biomaterials. Herein, a series of CaP coatings with gradual change of topography and crystallinity is constructed by electrochemical deposition, and the roles of the two basic physicochemical properties are scrutinized for further understanding the mechanism behind the superior bioactivities of octacalcium phosphate (OCP). We observe a distinct modulation on cell proliferation on the prepared CaP coatings for different cells. The magnitude of the modulation seems to depend on the cellular size, and the effect is attributed mainly to the microstructure of the coatings. On the other hand, the crystallinity manifests its significance for the osteogenic property of the OCP coatings. Further transmission electron microscopy analysis and density functional theory calculations reveal a surface rich in HPO4 2- for the high-crystalline OCP nanocrystals. The results highlight that the nanocrystal surface properties of the OCP coatings, including the periodic structure and the HPO4 2- composition, may play significant roles surpassing the ion release effect in determining its osteogenic property, probably via surface spatial and/or chemical recognitions. The present findings shed light on the fundamental understanding of the structure-biofunction relationship for CaP biomaterials.

Understanding the Steric Structures of Dicarboxylate Ions Incorporated in Octacalcium Phosphate Crystals.

Project description:Octacalcium phosphate (OCP) can incorporate various dicarboxylate ions in the interlayer spaces of its layered structure. Although not proven, these incorporated ions are believed to have a linear structure. In this study, the steric structures of twelve different dicarboxylate ions incorporated into OCP were investigated by comparing the experimentally determined interlayer distance of the OCP with the distance estimated using the molecular sizes of dicarboxylic acids calculated by considering their steric structures. The results revealed that the incorporated succinate, glutarate, adipate, pimelate, suberate, and aspartate ions possessed linear structures, whereas the incorporated azelate, sebacate, methylsuccinate, and malate ions exhibited bent structures. Further, the incorporated mercaptosuccinate ions featured linear, bent, other types of structures. Moreover, the steric structure of the incorporated malonate ion significantly differed from those of other dicarboxylate ions. The computational approach employed in this study is expected to deepen our understanding of the steric structures of dicarboxylate ions incorporated in the OCP interlayer spaces.

Ammonium-to-sodium ion-exchange process at the interlayer of octacalcium phosphate.

Project description:Octacalcium phosphate (OCP) has been considered as the layer component of calcium phosphate, but whether it achieves the ionic-exchange ability of conventional layer components is unclear. As OCP is highly biocompatible, understanding its ionic-exchange properties would potentially expand its pharmaceutical and medical applications. Herein, we demonstrate that the substituted cations in ammonium (NH4)-substituted octacalcium phosphate (OCP-NH4) and sodium (Na)-containing ammonium phosphate solutions undergo ion exchanges with OCP interlayers. Replacing NH4 + with Na+ did not alter the crystal structure of OCP, confirming that a substituted cation exchange process similar to that in other layered compounds occurs in OCP.

3D Printed Gene-activated Octacalcium Phosphate Implants for Large Bone Defects Engineering.

Project description:The aim of the study was the development of three-dimensional (3D) printed gene-activated implants based on octacalcium phosphate (OCP) and plasmid DNA encoding VEGFA. The first objective of the present work involved design and fabrication of gene-activated bone substitutes based on the OCP and plasmid DNA with VEGFA gene using 3D printing approach of ceramic constructs, providing the control of its architectonics compliance to the initial digital models. X-ray diffraction, scanning electron microscopy (SEM), Fourier transform infrared spectroscopy, and compressive strength analyses were applied to investigate the chemical composition, microstructure, and mechanical properties of the experimental samples. The biodegradation rate and the efficacy of plasmid DNA delivery in vivo were assessed during standard tests with subcutaneous implantation to rodents in the next stage. The final part of the study involved substitution of segmental tibia and mandibular defects in adult pigs with 3D printed gene-activated implants. Biodegradation, osteointegration, and effectiveness of a reparative osteogenesis were evaluated with computerized tomography, SEM, and a histological examination. The combination of gene therapy and 3D printed implants manifested the significant clinical potential for effective bone regeneration in large/critical size defect cases.
