A novel ionic AIE smart responsive material with multiple structural transformations.

ABSTRACT: A novel ionic liquid [TPE-Dim-DMe] (Br)₂ was successfully synthesiszed. With the change of temperature, quadruple transformations of structure were observed in the [TPE-Dim-DMe] (Br)₂ crystal, and it simultaneously displayed mechanochromic phenomena. In addition, its excellent solubility in water makes it a novel bioimaging agent.

Smart Material Hydrogel Transfer Devices Fabricated with Stimuli-Responsive Silk-Elastin-Like Proteins.

Project description:Three-dimensional organoid tissue culture models are a promising approach for the study of biological processes including diseases. Advances in these tissue culture technologies improve in vitro analysis compared to standard 2D cellular approaches and are more representative of the physiological environment. However, a major challenge associated with organoid systems stems from the laborious processing involved in the analysis of large numbers of organoids. Here the design, characterization, and application of silk-elastin-like protein-based smart carrier arrays for processing organoids is presented. Fabrication of hydrogel-based carrier systems at room temperature result in organized arrays of organoids that maintain tissue culture plate orientation and could be processed simultaneously for histology. The system works by transfer of the organoids to the hydrogel arrays after which the material is subjected to 65 °C to induce hydrogel contraction to secure the organoids, resulting in multisample constructs and allowing for placement on a microscope slide. Histological processing and immunostaining of these arrayed cerebral organoids analyzed within the contracted silk-elastin-like proteins (SELP) show retention of native organoid features compared to controls without the hydrogel carrier system, thus avoiding any artifacts. These SELP carriers present a useful approach for improving efficiency of scaled organoid screening and processing.

Multi-Stimuli-Responsive Tadpole-like Polymer/Lipid Janus Microrobots for Advanced Smart Material Applications.

Project description:Microrobots are of significant interest due to their smart transport capabilities, especially for precisely targeted delivery in dynamic environments (blood, cell membranes, tumor interstitial matrixes, blood-brain barrier, mucosa, and other body fluids). To perform a more complex micromanipulation in biological applications, it is highly desirable for microrobots to be stimulated with multiple stimuli rather than a single stimulus. Herein, the biodegradable and biocompatible smart micromotors with a Janus architecture consisting of PrecirolATO 5 and polycaprolactone compartments inspired by the anisotropic geometry of tadpoles and sperms are newly designed. These bioinspired micromotors combine the advantageous properties of polypyrrole nanoparticles (NPs), a high near-infrared light-absorbing agent with high photothermal conversion efficiency, and magnetic NPs, which respond to the magnetic field and exhibit multistimulus-responsive behavior. By combining both fields, we achieved an "on/off" propulsion mechanism that can enable us to overcome complex tasks and limitations in liquid environments and overcome the limitations encountered by single actuation applications. Moreover, the magnetic particles offer other functions such as removing organic pollutants via the Fenton reaction. Janus-structured motors provide a broad perspective not only for biosensing, optical detection, and on-chip separation applications but also for environmental water treatment due to the catalytic activities of multistimulus-responsive micromotors.

Hybrid hydrogels derived from renewable resources as a smart stimuli responsive soft material for drug delivery applications.

Project description:The design and synthesis of amphiphilic molecules play a crucial role in fabricating smart functional materials via self-assembly. Especially, biologically significant natural molecules and their structural analogues have inspired chemists and made a major contribution to the development of advanced smart materials. In this report, a series of amphiphilic N-acyl amides were synthesized from natural precursors using a simple synthetic protocol. Interestingly, the self-assembly of amphiphiles 6a and 7a furnished a hydrogel and oleogel in vegetable oils. Morphological analysis of gels revealed the existence of a 3-dimensional fibrous network. Thermoresponsive and thixotropic behavior of these gels were evaluated using rheological analysis. A composite gel prepared by the encapsulation of curcumin in the hydrogel formed from 7a displayed a gel-sol transition in response to pH and could act as a dual channel responsive drug carrier.

Biochemically responsive smart surface.

Project description:A design of smart surfaces responsive to biochemical analytes is demonstrated in the example of mixed monolayers of biotin/fluorocarbon. The contact angle of aqueous solutions on such surfaces decreases upon streptavidin binding and can be used in detecting this protein. The specificity of the effect is confirmed by the lack of a contact angle change by streptavidin blocked with biotin and by bovine serum albumin.

Structural characterization of IrisFP, an optical highlighter undergoing multiple photo-induced transformations.

Project description:Photoactivatable fluorescent proteins (FPs) are powerful fluorescent highlighters in live cell imaging and offer perspectives for optical nanoscopy and the development of biophotonic devices. Two types of photoactivation are currently being distinguished, reversible photoswitching between fluorescent and nonfluorescent forms and irreversible photoconversion. Here, we have combined crystallography and (in crystallo) spectroscopy to characterize the Phe-173-Ser mutant of the tetrameric variant of EosFP, named IrisFP, which incorporates both types of phototransformations. In its green fluorescent state, IrisFP displays reversible photoswitching, which involves cis-trans isomerization of the chromophore. Like its parent protein EosFP, IrisFP also photoconverts irreversibly to a red-emitting state under violet light because of an extension of the conjugated pi-electron system of the chromophore, accompanied by a cleavage of the polypeptide backbone. The red form of IrisFP exhibits a second reversible photoswitching process, which may also involve cis-trans isomerization of the chromophore. Therefore, IrisFP displays altogether 3 distinct photoactivation processes. The possibility to engineer and precisely control multiple phototransformations in photoactivatable FPs offers exciting perspectives for the extension of the fluorescent protein toolkit.

Smart responsive organic microlasers with multiple emission states for high-security optical encryption.

Project description:Modern high-security cryptography and optical communication call for covert bit sequences with high coding capacity and efficient authentication. Stimuli-responsive lasing emissions with easily distinguishable readout are promising in the coding field as a novel cryptographic primitive, while the application is frequently restricted by the limited number of emission states. Here, we report a strategy of achieving multiple competitive lasing signals in responsive organic microspheres where a donor-acceptor pair was introduced. The competitive lasing from the donor and acceptor was reversibly switched by modulating the competition between the radiative rate of the donor and the rate of energy transfer, and the generated multiple lasing signals enabled a quaternary coding for recognizable cryptographic implementation. Data encryption and extraction were demonstrated using a 4 × 4 microlaser array, showing vast prospects in avoiding the disclosure of security information. The results offer a comprehensive understanding of excited-state dynamics in organic composite materials, which may play a major role in high-security optical recording and information encryption.

Thermally responsive AIE-active polyurethanes based on a tetraaniline derivative.

Project description:Polyurethanes with different soft-hard segment ratios were successfully synthesized, with an aggregation-induced-emission (AIE)-active tetraaniline derivative (NH2-B3-Ani4-NH2) as the hard segment. The resulting polyurethanes exhibited typical AIE features. The fluorescence intensities of polyurethane films changed with heat treatments. The fluorescence intensities of the polyurethane films decreased sharply after quenching treatment, yet their fluorescence intensities exceeded the original intensities of the films after thermal annealing at 80 °C for 24 h. Differential Scanning Calorimetry (DSC) results implied that the melting peaks in polyurethane films disappeared after quenching treatment, but the melting peaks appeared again after thermal annealing. These results proved that the arrangement of the structure had an important effect on the AIE properties of the polyurethane films. Meanwhile, the fluorescence intensities of these polyurethanes decreased with the increase of temperature, indicating that all three polyurethanes exhibited temperature-dependent fluorescent characteristics. Based on the above investigations, the AIE-active polyurethanes may provide a platform for the development of stimuli-responsive fluorescent materials.

Smart vaults: thermally-responsive protein nanocapsules.

Project description:Synthetic modification of a recombinant protein cage called a vault with stimuli-responsive smart polymers provides access to a new class of biohybrid materials; the polymer nanocapsules retain the structure of the protein cage and exhibit the responsive nature of the polymer. Vaults are naturally occurring ubiquitous ribonucleoprotein particles 41 × 41 × 72.5 nm composed of a protein shell enclosing multiple copies of two proteins and multiple copies of one or more small untranslated RNAs. Recombinant vaults are structurally identical but lack the vault content. Poly(N-isopropylacrylamide) (pNIPAAm), a polymer responsive to heat, was conjugated to recombinant vaults that were composed of ~78 copies of the major vault protein (MVP) modified to contain a cysteine rich region at the N-terminus (CP-MVP). The polymer was synthesized using reversible addition-fragmentation chain transfer (RAFT) polymerization to have a dansyl group at the alpha end and modified to have a thiol-reactive pyridyl disulfide at the omega end, which readily coupled to CP-MVP vaults. The resulting vault nanocapsules underwent reversible aggregation upon heating above the lower critical solution temperature (LCST) of the polymer as determined by electron microscopy (EM), dynamic light scattering experiments, and UV-vis turbidity analysis. The vault structure remained entirely intact throughout the phase transition; suggesting its use in a myriad of biomedical and biotechnology applications.

Click Access to a Cyclodextrin-Based Spatially Confined AIE Material for Hydrogenase Recognition.

Project description:The spatial confinement of conjugated phenyl rotators is a compulsory requirement for the fluorescence enhancement of aggregation induced emission (AIE) molecules. This work reports a novel spatially confined AIE material by restricting several tetraphenylethylene (TPE) molecules around the primary face of β-cyclodextrin (CD) via a Cu(I) catalytic 1,3-dipolar cycloaddition reaction (click chemistry). The spatial confinement effect was found to significantly enhance the fluorescence emission when compared with a single TPE modified CD. In addition, the emission maxima took place with the dimethyl sulfoxide volume ratio of 30% in a water mixture, which is remarkably different from traditional AIE molecules. Benefiting from the CD's complexation effect, this material exhibits a selective fluorescence quenching property in certain hydrogenases and can be used as a fluorescence probe for hydrogenase sensing. This demonstrates the potential of the spatially confined AIECD for practical applications.
