Two case studies of very long-term retention.

ABSTRACT: Here, we present two case studies of extremely long-term retention. In the first, Richard C. Atkinson (RCA) had learned word sequences during experiments for his dissertation. Sixty-seven years later, RCA relearned the same words either in the original order or in a scrambled order. RCA reported no conscious awareness that the words were those used in the dissertation, but his relearning was considerably better for the words in the original order. In the second case study, Denis Cousineau had searched displays of objects for the presence of a target. The targets and foils had been novel at the beginning of training, and his search rate improved markedly over about 70 sessions. After 22 years, retraining showed retention of much of this gain in rate of search, and the rate was markedly faster than search for new objects with the same structure as the trained set. We consider interpretations of these case studies for our understanding of long-term retention.

Working memory limits severely constrain long-term retention.

Project description:There has been considerable controversy in recent years as to whether information held in working memory (WM) is rapidly forgotten or automatically transferred to long-term memory (LTM). Although visual WM capacity is very limited, we appear able to store a virtually infinite amount of information in visual LTM. Still, LTM retrieval often fails. Some view visual WM as a mental sketchpad that is wiped clean when new information enters, but not a consistent precursor of LTM. Others view the WM and LTM systems as inherently linked. Distinguishing between these possibilities has been difficult, as attempts to directly manipulate the active holding of information in visual WM has typically introduced various confounds. Here, we capitalized on the WM system's capacity limitation to control the likelihood that visual information was actively held in WM. Our young-adult participants (N = 103) performed a WM task with unique everyday items, presented in groups of two, four, six, or eight items. Presentation time was adjusted according to the number of items. Subsequently, we tested participants' LTM for items from the WM task. LTM was better for items presented originally within smaller WM set sizes, indicating that WM limitations contribute to subsequent LTM failures, and that holding items in WM enhances LTM encoding. Our results suggest that a limit in WM capacity contributes to an LTM encoding bottleneck for trial-unique familiar objects, with a relatively large effect size.

Probabilistic population forecasting: Short to very long-term.

Project description:Population forecasts are used by governments and the private sector for planning, with horizons up to about three generations (around 2100) for different purposes. The traditional methods are deterministic using scenarios, but probabilistic forecasts are desired to get an idea of accuracy, assess changes, and make decisions involving risks. In a significant breakthrough, since 2015, the United Nations has issued probabilistic population forecasts for all countries using a Bayesian methodology that we review here. Assessment of the social cost of carbon relies on long-term forecasts of carbon emissions, which in turn depend on even longer-range population and economic forecasts, to 2300. We extend the UN method to very-long range population forecasts by combining the statistical approach with expert review and elicitation. While the world population is projected to grow for the rest of this century, it will likely stabilize in the 22nd century and decline in the 23rd century.

Long term follow-up in two siblings with Sengers syndrome: Case report.

Project description:BackgroundSengers syndrome is characterized by congenital cataract, hypertrophic cardiomyopathy, mitochondrial myopathy, and lactic acidosis associated with mutations in AGK gene. Clinical course ranges from a severe fatal neonatal form, to a more benign form allowing survival into adulthood, to an isolated form of congenital cataract. Thus far few reported cases have survived the second decade at their latest examination, and no natural history data are available for the disease.Case presentationHere we provide a 20-year follow-up in two siblings with a benign form of Sengers syndrome, expanding the phenotypical spectrum of the disease by reporting a condition of ovarian agenesis.ConclusionTo our knowledge, this report provides the first longitudinal data of Sengers syndrome patients.

Practicing memory retrieval improves long-term retention in rats.

Project description:The view that the human mind is a repository of stored items dates at least to Aristotle and Plato and continues to dominate investigations of human memory. This view fits with our intuitions that we study information as the optimal method to store information in memory and that retrieval of information functions only to assess what information was previously stored. Yet modern research on human memory suggests that retrieving information during a test facilitates later memory of that information. Because human memory is intertwined with language, it is difficult to resist the conclusion that language is essential for this key aspect of human cognition. Here we show that practising memory retrieval improves long-term retention in a nonhuman species. We report evidence that rats' long-term memory performance is enhanced if they had previously retrieved specific items stored in memory.

Are values stable throughout adulthood? Evidence from two German long-term panel studies.

Project description:Previous studies have used cross-sectional or short-term longitudinal data, resulting in a truncated view of a phenomenon unfolding across the lifespan. We find that, contrary to the consensus in the literature, people's values continue developing in adulthood, albeit at a slower pace than in previous developmental stages. We use longitudinal data sources with two measurement instruments. We show their comparability using confirmatory MDS in Study 1 (N = 1,027). We examined value development using latent growth models in a convenience sample of highly educated German peace activists (Study 2, N = 1,209) and corroborated these with evidence from a representative sample from the German population (Study 3, N = 19,566). We find that all values change up to age 40 consistent with theoretical expectations. We observe that with age, self-transcendence and conservation values increase while self-enhancement values decrease. At the same time, we find a curvilinear pattern for openness to change in Study 2 and an overall decrease in Study 3. Moreover, the developmental trajectory of conservation and of self-enhancement in the German general population differ between those with tertiary and without tertiary education. We discuss the implication of the present findings for research on value development and for interventions.

Very poor long-term survival in past and more recent studies for relapsed AML patients: The ECOG-ACRIN experience.

Project description:This study examines the long-term OS of relapsed AML patients who were enrolled to 9 successive ECOG-ACRIN trials for newly diagnosed AML, during 1984-2008. The objectives were to examine whether there is a trend of improvement in the survival of relapsed AML patients in the more recent studies and to search for prognostic factors that are associated with long-term OS after relapse. A total of 3012 patients were enrolled, 1779 (59.1%) achieved CR1 and of these, 58.9% relapsed. The median follow-up was 9.7 years. The median OS from relapse was 0.5 years and the 5-year OS was 10 (±1)%. These results were similar even for the most recent studies. A multivariate model showed that age, cytogenetics at diagnosis, duration of CR1 and undergoing allogeneic transplantation were significantly associated with OS from relapse. Even among patients who relapsed with better prognostic factors; age < 40 and CR1 > 12 months, there was no significant OS difference between the studies. In conclusion, this large cohort appears to confirm that the survival of AML patients postrelapse continues to be dismal and has not improved during the past quarter of a century.

Exploring long-term retention and reactivation of micropollutant biodegradation capacity.

Project description:The factors limiting micropollutant biodegradation in the environment and how to stimulate this process have often been investigated. However, little information is available on the capacity of microbial communities to retain micropollutant biodegradation capacity in the absence of micropollutants or to reactivate micropollutant biodegradation in systems with fluctuating micropollutant concentrations. This study investigated how a period of 2 months without the addition of micropollutants and other organic carbon affected micropollutant biodegradation by a micropollutant-degrading microbial community. Stimulation of micropollutant biodegradation was performed by adding different types of dissolved organic carbon (DOC)-extracted from natural sources and acetate-increasing 10 × the micropollutant concentration, and inoculating with activated sludge. The results show that the capacity to biodegrade 3 micropollutants was permanently lost. However, the biodegradation activity of 2,4-D, antipyrine, chloridazon, and its metabolites restarted when these micropollutants were re-added to the community. Threshold concentrations similar to those obtained before the period of no substrate addition were achieved, but biodegradation rates were lower for some compounds. Through the addition of high acetate concentrations (108 mg-C/L), gabapentin biodegradation activity was regained, but 2,4-D biodegradation capacity was lost. An increase of bentazon concentration from 50 to 500 µg/L was necessary for biodegradation to be reactivated. These results provide initial insights into the longevity of micropollutant biodegradation capacity in the absence of the substance and strategies for reactivating micropollutant biodegrading communities.

Long-term retention of self-fertilization in a fish clade.

Project description:Among vertebrate animals, only the mangrove rivulus (Kryptolebias marmoratus) was known to self-fertilize. Here, we use microsatellite analyses to document a high selfing rate (97%) in a related nominal species, Kryptolebias ocellatus, which likewise is androdioecious (populations consist of males and hermaphrodites). In contrast, we find no evidence of self-fertilization in Kryptolebias caudomarginatus (an androdioecious species closely related to the marmoratus-ocellatus clade) or in Kryptolebias brasiliensis (a dioecious outgroup). These findings indicate that the initiation of self-fertilization predated the origin of the marmoratus-ocellatus clade. From mitochondrial DNA sequences and microsatellite data, we document a substantial genetic distance between Kryptolebias marmoratus and K. ocellatus, implying that the selfing capacity has persisted in these fishes for at least several hundred thousand years.

Preoperative buccal bone volume predicts long-term graft retention following augmentation in the esthetic zone: A retrospective case series.

Project description:ObjectivesBuccal bone augmentation in the esthetic zone is routinely used to achieve optimal clinical outcomes. Nonetheless, long-term data are sparse, and it is unknown how baseline buccal bone volume affects the retention of the augmented volume over time.Material and methodsThis is a long-term follow-up retrospective case series. After a preoperative computed tomography scan, implants were placed in the anterior maxilla following guided bone regeneration, autogenous block grafting, or both. At the follow-up, patients received a computed tomography scan and a clinical examination. Buccal bone volume was the primary outcome. Buccal bone thickness, peri-implant, and esthetic parameters were secondary outcomes.ResultsAfter a median follow-up of 6.7 years (interquartile range: 4.9-9.4), 28 implants in 19 patients (median age at augmentation: 43.3 years, interquartile range: 34.4-56.7, 53% female) were followed up. Preoperative buccal bone volume at baseline (V0 ) showed a moderate correlation to final buccal bone volume (Vt , rs = .43) but a strong correlation to the absolute volumetric change (ΔV = Vt -V0 , rs = -.80). A linear mixed model for Vt had a large intercept of 91.39 (p < .001) and a rather small slope of .11 for V0 (p = .11). Observed differences between treatments were not statistically significant in the mixed model. V0 above 105 mm3 predicted a negative volume change (ΔV < 0) with a specificity of 100% and a sensitivity of 96%.ConclusionsThe results suggest higher gains in sites with lower V0 and point to a cutoff V0 above which the augmented volume is not retained long-term.

Expression profiling of archival tissues for long-term health studies

Project description:Over 20 million archival tissue samples are stored annually in the United States as formalin-fixed, paraffin-embedded (FFPE) tissue blocks, but only recently has whole-genome expression profiling from these samples become technically feasible. Here, we introduce novel general methods for assessing, summarizing, and visualizing expression data quality from archival samples. We validated these methods in technical study of 144 clinical breast cancer and autopsy samples and in an overview of all current publicly available FFPE whole-genome expression data. We additionally performed a case study incorporating over 1,000 colorectal cancer (CRC) samples collected from patients across the United States over a period of more than 25 years, integrating clinicopathological information, tumor molecular data, and archival tissue gene expression on an unparalleled scale. Both large-scale clinical studies presented a much greater range of data quality than previous smaller studies, emphasizing the need for rigorous quality control in translational applications of archival tissue gene expression profiling. This series includes 1,003 FFPE-preserved CRC tumors, assayed by Illumina HumanRef-v3 WG-DASL microarray.
