Cytoarchitectonic parcellation and functional characterization of four new areas in the caudal parahippocampal cortex.

ABSTRACT: Brain areas at the parahippocampal gyrus of the temporal-occipital transition region are involved in different functions including processing visual-spatial information and episodic memory. Results of neuroimaging experiments have revealed a differentiated functional parcellation of this region, but its microstructural correlates are less well understood. Here we provide probability maps of four new cytoarchitectonic areas, Ph1, Ph2, Ph3 and CoS1 at the parahippocampal gyrus and collateral sulcus. Areas have been identified based on an observer-independent mapping of serial, cell-body stained histological sections of ten human postmortem brains. They have been registered to two standard reference spaces, and superimposed to capture intersubject variability. The comparison of the maps with functional imaging data illustrates the different involvement of the new areas in a variety of functions. Maps are available as part of Julich-Brain atlas and can be used as anatomical references for future studies to better understand relationships between structure and function of the caudal parahippocampal cortex.

Project description:A comprehensive concept of the biological basis of reward, social and emotional behavior, and language requires a deeper understanding of the microstructure and connectivity of the underlying brain regions. Such understanding could provide deeper insights into their role in functional networks, and form the anatomical basis of the functional segregation of this region as shown in recent in vivo imaging studies. Here, we investigated the cytoarchitecture of the lateral orbitofrontal cortex (lateral OFC) in serial histological sections of 10 human postmortem brains by image analysis and a statistically reproducible approach to detect borders between cortical areas. Profiles of the volume fraction of cell bodies were therefore extracted from digitized histological images, describing laminar changes from the layer I/layer II boundary to the white matter. As a result, four new areas, Fo4-7, were identified. Area Fo4 was mainly found in the anterior orbital gyrus (AOG), Fo5 anteriorly in the inferior frontal gyrus (IFG), Fo6 in the lateral orbital gyrus (LOG), and Fo7 in the lateral orbital sulcus. Areas differed in cortical thickness, abundance and size of pyramidal cells in layer III and degree of granularity in layer IV. A hierarchical cluster analysis was used to quantify cytoarchitectonic differences between them. The 3D-reconstructed areas were transformed into the single-subject template of the Montreal Neurological Institute (MNI), where probabilistic maps and a maximum probability map (MPM) were calculated as part of the JuBrain Cytoarchitectonic Atlas. These maps served as reference data to study the functional properties of the areas using the BrainMap database. The type of behavioral tasks that activated them was analyzed to get first insights of co-activation patterns of the lateral OFC and its contribution to cognitive networks. Meta-analytic connectivity modeling (MACM) showed that functional decoding revealed activation in gustatory perception in Fo4; reward and somesthetic perception in Fo5; semantic processing and pain perception in Fo6; and emotional processing and covert reading in Fo7. Together with existing maps of the JuBrain Cytoarchitectonic Atlas, the new maps can now be used as an open-source reference for neuroimaging studies, allowing to further decode brain function.

Project description:Although lesion studies over the past several decades have focused on functional dissociations in posterior parietal cortex (PPC) during arithmetic, no consistent view has emerged of its differential involvement in addition, subtraction, multiplication, and division. To circumvent problems with poor anatomical localization, we examined functional overlap and dissociations in cytoarchitectonically defined subdivisions of the intraparietal sulcus (IPS), superior parietal lobule (SPL) and angular gyrus (AG), across these four operations. Compared to a number identification control task, all operations except addition, showed a consistent profile of left posterior IPS activation and deactivation in the right posterior AG. Multiplication and subtraction differed significantly in right, but not left, IPS and AG activity, challenging the view that the left AG differentially subserves retrieval during multiplication. Although addition and multiplication both rely on retrieval, multiplication evoked significantly greater activation in right posterior IPS, as well as the prefrontal cortex, lingual and fusiform gyri, demonstrating that addition and multiplication engage different brain processes. Comparison of PPC responses to the two pairs of inverse operations: division versus multiplication and subtraction versus addition revealed greater activation of left lateral SPL during division, suggesting that processing inverse relations is operation specific. Our findings demonstrate that individual IPS, SPL and AG subdivisions are differentially modulated by the four arithmetic operations and they point to significant functional heterogeneity and individual differences in activation and deactivation within the PPC. Critically, these effects are related to retrieval, calculation and inversion, the three key cognitive processes that are differentially engaged by arithmetic operations. Our findings point to distribute representation of these processes in the human PPC and also help explain why lesion and previous imaging studies have yielded inconsistent findings.

Project description:Neuroimaging has revealed a core network of cortical regions that contribute to speech production, but the functional organization of this network remains poorly understood. Purpose We describe efforts to identify reliable boundaries around functionally homogenous regions within the cortical speech motor control network in order to improve the sensitivity of functional magnetic resonance imaging (fMRI) analyses of speech production and thus improve our understanding of the functional organization of speech production in the brain. Method We used a bottom-up, data-driven approach by pooling data from 12 previously conducted fMRI studies of speech production involving the production of monosyllabic and bisyllabic words and pseudowords that ranged from single vowels and consonant-vowel pairs to short sentences (163 scanning sessions, 136 unique participants, 39 different speech conditions). After preprocessing all data through the same pipeline and registering individual contrast maps to a common surface space, hierarchical clustering was applied to contrast maps randomly sampled from the pooled data set in order to identify consistent functional boundaries across subjects and tasks. Boundary completion was achieved by applying adaptive smoothing and watershed segmentation to the thresholded population-level boundary map. Hierarchical clustering was applied to the mean within-functional region of interest (fROI) response to identify networks of fROIs that respond similarly during speech. Results We identified highly reliable functional boundaries across the cortical areas involved in speech production. Boundary completion resulted in 117 fROIs in the left hemisphere and 109 in the right hemisphere. Clustering of the mean within-fROI response revealed a core sensorimotor network flanked by a speech motor planning network. The majority of the left inferior frontal gyrus clustered with the visual word form area and brain regions (e.g., anterior insula, dorsal anterior cingulate) associated with detecting salient sensory inputs and choosing the appropriate action. Conclusion The fROIs provide insight into the organization of the speech production network and a valuable tool for studying speech production in the brain by improving within-group and between-groups comparisons of speech-related brain activity.
