ABSTRACT: A hydrophobic diradical-platinum(ii) complex was solubilized in aqueous solutions by using bovine serum albumin and exhibited photothermal conversion under near-infrared (NIR) light irradiation. The complex was introduced into cancer cells and induced cell death upon absorption of NIR. These results imply that the complex can function as a photothermal therapeutic agent.

Study on cellular uptake of a hydrophobic near-infrared-absorbing diradical-platinum(II) complex solubilized by albumin using hyperspectral imaging, spectrophotometry, and spectrofluorimetry.

Project description:Owing to its biopenetrability and minimal invasiveness, near-infrared (NIR) light in the region between 700-1100 nm has attracted attention in cancer diagnosis and therapy. Our group previously reported that the hydrophobic diradical-platinum(II) complex PtL2 is a promising agent for cancer photothermal therapy (L = 3,5-dibromo-1,2-diiminobenzosemiquinonate radical). Because PtL2 does not fluoresce, its intercellular uptake of PtL2 cannot be observed with a fluorescence microscope. In this study, we clarified the uptake and intracellular behavior of PtL2 solubilized by bovine serum albumin (BSA) using hyperspectral imaging enabling spectrophotometric analysis of the image. The spectral changes in the obtained images indicated that the internalization of PtL2 was followed by crystallization of the complex during the long incubation period (> 4 h). Additionally, the binding constant Kb = 5.91 × 104 M-1 could be estimated upon fluorescence quenching analysis of BSA upon binding of PtL2; Kb is two orders of magnitude smaller than that of albumin-common drugs. Considering the small Kb and low solubility of PtL2 in water, we ultimately proposed the internalization path and fate of PtL2 in the cell: release of PtL2 from BSA near cellular membranes and subsequent cellular uptake via membrane permeation followed by saturation, resulting in crystallization.

NeoR, a near-infrared absorbing rhodopsin.

Project description:The Rhizoclosmatium globosum genome encodes three rhodopsin-guanylyl cyclases (RGCs), which are predicted to facilitate visual orientation of the fungal zoospores. Here, we show that RGC1 and RGC2 function as light-activated cyclases only upon heterodimerization with RGC3 (NeoR). RGC1/2 utilize conventional green or blue-light-sensitive rhodopsins (λmax = 550 and 480 nm, respectively), with short-lived signaling states, responsible for light-activation of the enzyme. The bistable NeoR is photoswitchable between a near-infrared-sensitive (NIR, λmax = 690 nm) highly fluorescent state (QF = 0.2) and a UV-sensitive non-fluorescent state, thereby modulating the activity by NIR pre-illumination. No other rhodopsin has been reported so far to be functional as a heterooligomer, or as having such a long wavelength absorption or high fluorescence yield. Site-specific mutagenesis and hybrid quantum mechanics/molecular mechanics simulations support the idea that the unusual photochemical properties result from the rigidity of the retinal chromophore and a unique counterion triad composed of two glutamic and one aspartic acids. These findings substantially expand our understanding of the natural potential and limitations of spectral tuning in rhodopsin photoreceptors.

Cobalt Phthalocyanine-Sensitized Graphene-ZnO Composite: An Efficient Near-Infrared-Active Photothermal Agent.

Project description:Herein, a promising near-infrared-responsive photothermal agent was designed by anchoring of rice grain-shaped ZnO particles over graphene (GR) nanosheets and subsequent sensitization with cobalt phthalocyanine (CoPc). Thus, produced GR-ZnO-CoPc was able to attain the temperature of 68 °C by irradiating to 980 nm laser for 7 min, which is extremely higher than the endurance temperature of cancer cells. The linear fashioned progression in the photothermal effect of GR nanosheets was conquered by immobilization of ZnO particles and successive sensitization with CoPc. The excellence found in the photothermal effect of GR-ZnO-CoPc was verified by estimation of its photothermal conversion efficiency. The photothermal conversion efficiency assessed for GR-ZnO-CoPc was higher than those for the popular gold- and CuS-based photothermal agents. In addition, it possessed significant stability against photobleaching and structural rupture. It was found that the photothermal effect of GR-ZnO-CoPc is proportional to its concentration. However, by replacement of a 980 nm laser system with 808 nm, the photothermal effect of GR-ZnO-CoPc was reduced, which could be due to lower absorption of GR-ZnO-CoPc at 808 nm compared to 980 nm. On account of its significance and important properties, GR-ZnO-CoPc could be an interesting photothermal agent to employ in future photothermal therapy for cancer.

Single-Step Photochemical Formation of Near-Infrared-Absorbing Gold Nanomosaic within PNIPAm Microgels: Candidates for Photothermal Drug Delivery.

Project description:This work demonstrates the dynamic potential for tailoring the surface plasmon resonance (SPR), size, and shapes of gold nanoparticles (AuNPs) starting from an Au(I) precursor, chloro(dimethyl sulfide)gold (I) (Au(Me2S)Cl), in lieu of the conventional Au(III) precursor hydrogen tetrachloroaurate (III) hydrate (HAuCl4). Our approach presents a one-step method that permits regulation of an Au(I) precursor to form either visible-absorbing gold nanospheres or near-infrared-window (NIRW)-absorbing anisotropic AuNPs. A collection of shapes is obtained for the NIR-absorbing AuNPs herein, giving rise to spontaneously formed nanomosaic (NIR-absorbing anisotropic gold nanomosaic, NIRAuNM) without a dominant geometry for the tesserae elements that comprise the mosaic. Nonetheless, NIRAuNM exhibited high stability; one test sample remains stable with the same SPR absorption profile 7 years post-synthesis thus far. These NIRAuNM are generated within thermoresponsive poly(N-isopropylacrylamide) (PNIPAm) microgels, without the addition of any growth-assisting surfactants or reducing agents. Our directed-selection methodology is based on the photochemical reduction of a light-, heat-, and water-sensitive Au(I) precursor via a disproportionation mechanism. The NIRAuNM stabilized within the thermoresponsive microgels demonstrates a light-activated size decrease of the microgels. On irradiation with a NIR lamp source, the percent decrease in the size of the microgels loaded with NIRAuNM is at least five times greater compared to the control microgels. The concept of photothermal shrinkage of hybrid microgels is further demonstrated by the release of a model luminescent dye, as a drug release model. The absorbance and emission of the model dye released from the hybrid microgels are over an order of magnitude higher compared to the absorbance and emission of the dye released from the unloaded-control microgels.

Highly stable organic photothermal agent based on near-infrared-II fluorophores for tumor treatment.

Project description:BackgroundThe aim to develop a highly stable near-infrared (NIR) photoinduced tumor therapy agent stems from its considerable potential for biological application. Due to its long wavelength, biological imaging exhibits a high signal-to-background ratio, deep tissue penetration and maximum permissible light power, which can minimize damage to an organism during photoinduced tumor therapy.ResultsA class of stable NIR-II fluorophores (NIR998, NIR1028, NIR980, NIR1030, and NIR1028-S) based on aza-boron-dipyrromethene (aza-BODIPY) dyes with donor-acceptor-donor structures have been rationally designed and synthesized by harnessing the steric relaxation effect and intramolecular photoinduced electron transfer (IPET). These fluorophores exhibit an intense range of NIR-II emission, large Stokes shift (≥ 100 nm), excellent photothermal conversion performance, and superior stability against photobleaching. Among the NIR-II fluorophores, NIR998 possesses better NIR-II emission and photothermal conversion performance. NIR998 nanoparticles (NIR998 NPs) can be encapsulated by liposomes. NIR998 NPs show superior stability in the presence of light, heat, and reactive oxygen nitrogen species than that of indocyanine green NPs, as well as a higher photothermal conversion ability (η = 50.5%) compared to other photothermal agents. Finally, under the guidance of photothermal imaging, NIR998 NPs have been proven to effectively eliminate tumors via their excellent photothermal conversion performance while presenting negligible cytotoxicity.ConclusionsUtilizing IPET and the steric relaxation effect can effectively induce NIR-II emission of aza-BODIPY dyes. Stable NIR998 NPs have excellent photothermal conversion performance and negligible dark cytotoxicity, so they have the potential to act as photothermal agents in biological applications.

Au-Silica nanowire nanohybrid as a hyperthermia agent for photothermal therapy in the near-infrared region.

Project description:Nanomaterial-based photothermal therapy has shown great potential for efficient cancer treatment. Here, we report a new hyperthermia agent, Au-silica nanowire nanohybrid (Au-SiNW nanohybrid) with tunable optical properties, for photothermal therapy. The unique feature of the synthetic method is no need of surface modification of SiNWs for the direct deposition of Au seeds, which can avoid complicated synthetic procedures and improve the reproducibility. The Au-SiNW nanohybrid can generate significant amount of heat upon irradiation in the near-infrared (NIR) region for inducing thermal cell death. Moreover, compared to reported hyperthermia nanomaterials, the new nanohybrid requires a much lower laser irradiation density of 0.3 W/cm(2) for destroying cancer cells. A549 lung cancer cells were used for in vitro photothermal study. The nanohybrid showed excellent in vitro biocompatibility by using a 96-nonradioactive-cell proliferation assay. Even at a high concentration of 0.500 mg/mL nanohybrid, over 80% cells were alive. In contrast, almost all the cells were killed when NIR irradiation was applied at a concentration of 0.100 mg/mL nanohybrid. The Au-SiNW nanohybrid may become a promising hyperthermia agent.

Photostable, hydrophilic, and near infrared quaterrylene-based dyes for photoacoustic imaging.

Project description:Novel near-infrared contrast agents based on the quaterrylene structure were strategically developed and tested for high photo-stability. Both a dendrimeric quaterrylene molecule, QR-G2-COOH, and a small molecule cationic quaterrylene dye, QR-4PyC4, remain optically stable and continue to generate a competitive photoacoustic response when irradiated by short near-infrared laser pulses for a relatively long time in an in-vitro cell study, unlike indocyanine green that rapidly decreases photoacoustic signal amplitude. The small molecule dye, QR-4PyC4 exhibits not only significantly higher cellular uptake rate than QR-G2-COOH and indocyanine green, but also low toxicity at a concentration of up to 10 μM. The dendrimeric dye, QR-G2-COOH that has surface functional groups available for conjugation with targeting and therapeutic agents shows the highest photoacoustic amplitude with high optical stability. Therefore, QR-4PyC4 can be a promising universal, sensitive and reliable photoacoustic contrast agent and QR-G2-COOH has great potential as a nano-platform with stable photoacoustic imaging capability.

Albumin tailoring fluorescence and photothermal conversion effect of near-infrared-II fluorophore with aggregation-induced emission characteristics.

Project description:Fluorophores with donor-acceptor-donor groups with the emission spanning the second near-infrared window (NIR-II) have recently received great attention for biomedical application. Yet, the mechanism underlying the equilibrium between fluorescence (radiative decay) and photothermal effect (non-radiative decay) of these fluorophores remains elusive. Here, we demonstrate that a lipophilic NIR-II fluorophore, BPBBT, possesses both twisted intramolecular charge transfer (TICT) and aggregation-induced emission (AIE) characteristics. Human serum albumin (HSA) binds to BPBBT, which changes the planarity of the fluorophore and restricts its intramolecular rotation. The binding results in alteration to the equilibrium between AIE and TICT state of BPBBT, tailoring its fluorescence and photothermal efficiency. Under the guidance of intraoperative NIR-II fluorescence image, the prepared HSA-bound BPBBT nanoparticles delineate primary orthotopic mouse colon tumor and metastatic lesions with dimensions as small as 0.5 mm × 0.3 mm, and offer photothermal ablation therapy with optimized timing, dosing and area of the laser irradiation.

Transparent organic photodetector using a near-infrared absorbing cyanine dye.

Project description:Organic photodetectors are interesting for low cost, large area optical sensing applications. Combining organic semiconductors with discrete absorption bands outside the visible wavelength range with transparent and conductive electrodes allows for the fabrication of visibly transparent photodetectors. Visibly transparent photodetectors can have far reaching impact in a number of areas including smart displays, window-integrated electronic circuits and sensors. Here, we demonstrate a near-infrared sensitive, visibly transparent organic photodetector with a very high average visible transmittance of 68.9%. The transmitted light of the photodetector under solar irradiation exhibits excellent transparency colour perception and rendering capabilities. At a wavelength of 850 nm and at -1 V bias, the photoconversion efficiency is 17% and the specific detectivity is 10(12) Jones. Large area photodetectors with an area of 1.6 cm(2) are demonstrated.
