Biguanide-transition metals complexes as potential drug for hyperglycemia treatment.

ABSTRACT: Coordination compounds of Cu(ii), Ni(ii), Co(ii), and Zn(ii) with a type of biguanide (known commercially as metformin) have been synthesized and characterized using spectroscopic techniques (FT-IR, UV/VIS), X-ray diffraction techniques and thermal analysis. For all compounds, single crystals were obtained for single-crystal X-ray diffraction. For the first time, an octahedral cobalt compound with the formula [Co(C₄H₁₁N₅)₃]Cl₂·2H₂O that crystallizes in the monoclinic space group C2/c with one molecule in the asymmetric unit has been obtained. Also, a novel nickel compound with the formula [Ni(C₄H₁₁N₅) (C₄H₁₀N₅)]Cl·H₂O that crystallizes in the monoclinic space group P2₁/c with two molecules in the asymmetric unit was obtained. Finally, we obtained copper and zinc compounds that crystallize in the monoclinic space groups P2₁/n and P2₁/c with the general formula [Cu(C₄H₁₁N₅)₂]Cl₂·H₂O and [Zn(C₄H₁₂N₅)Cl₃], respectively. A structural and supramolecular analysis was developed for all compounds using Hirshfeld surface analysis and electrostatic potential maps. The cell viability of the obtained compounds was evaluated in C2C12 (ATCCCRL-1772™) mouse muscle cells and HepG2 (ATCC HB-8065™) human liver carcinoma cells by the MTT assay to determine the potential of the compounds as new safe drugs. The results demonstrate that the compounds exhibit low cytotoxicity at doses less than 250 μg mL^-1 with a cell viability greater than 80%.

Project description:Hydrogen positions in hydrides play a key role in hydrogen storage materials and high-temperature superconductors. Our recently published study of five crystal structures of transition-metal-bound hydride complexes showed that using aspherical atomic scattering factors for Hirshfeld atom refinement (HAR) resulted in a systematic elongation of metal-hydrogen bonds compared to using spherical scattering factors with the Independent Atom Model (IAM). Even though only standard-resolution X-ray data was used, for the highest-quality data, we obtained excellent agreement between the X-ray and the neutron-derived bond lengths. We present an extended version of this study including 10 crystal structures of metal-organic complexes containing hydrogen atoms bonded to transition-metal atoms for which both X-ray and neutron data are available. The neutron structures were used as a benchmark, and the X-ray structures were refined by applying Hirshfeld atom refinement using various basis sets and DFT functionals in order to investigate the influence of the technical aspects on the length of metal-hydrogen bonds. The result of including relativistic effects in the Hamiltonian and using a cluster of multipoles simulating interactions with a crystal environment during wave function calculations was examined. The effect of the data quality on the final result was also evaluated. The study confirms that a high quality of experimental data is the key factor allowing us to obtain significant improvement in transition metal (TM)-hydrogen bond lengths from HAR in comparison with the IAM. Individual adjustments and better choices of the basis set can improve hydrogen positions. Average differences between TM-H bond lengths obtained with various DFT functionals upon including relativistic effects or between double-ζ and triple-ζ basis sets were not statistically significant. However, if all bonds formed by H atoms were considered, significant differences caused by different refinement strategies were observed. Finally, we examined the refinement of atomic thermal motions. Anisotropic refinement of hydrogen thermal motions with HAR was feasible only in some cases, and isotropically refined hydrogen thermal motions were in similar agreement with neutron values whether obtained with HAR or with the IAM.

Project description:The molecular engineering of organometallic complexes has recently attracted renewed interest on account of their potential technological applications for optoelectronics in general and optical data storage. The transition metal which induces control of enhanced nonlinear optical properties of functionalized organometallic complexes versus not only the intensity but also the polarization of the incident laser beam is original and important for all optical switching. This makes organometallic complexes valuable and suitable candidates for nonlinear optical applications. In the present work, we report the synthesis and full characterization of four organometallic complexes consisting of N, N-dibutylamine and azobenzene fragments but differ by auxiliary alkynyl ligands or metal cations. Thus, a ferrocenyl derivative 1 and three ruthenium complexes 2-4 have been prepared. The nonlinear optical properties of the four new azo-based ruthenium and iron organometallic complexes in the solid state, using polymethylmethacrylate as matrix, have been thoroughly studied. This concept is extended to computing the HOMO and LUMO energy levels of the considered complexes, dipole moment, first and second order hyperpolarizabilities using the 6-31 + G(d,p) + LANL2DZ mixed basis set. The second and third nonlinear optical properties of the resulting polymer composites were obtained by measuring SHG and THG response by means of the Maker fringe technique using a laser generating at 1,064 nm with a 30 ps pulse duration. The values of the second and third order NLO susceptibilities of the four organometallic complexes were found to be higher than the common references used. Theoretical calculation shows that the large first and second order hyperpolarizablities are caused by strong intramolecular charge transfer between the transition metal parts and the ligands though a conjugated transmitter. These results indicate that the present organometallic complexes are valuable candidates for optoelectronic and photonic applications.
