A network model of glymphatic flow under different experimentally-motivated parametric scenarios.

ABSTRACT: Flow of cerebrospinal fluid (CSF) through perivascular spaces (PVSs) in the brain delivers nutrients, clears metabolic waste, and causes edema formation. Brain-wide imaging cannot resolve PVSs, and high-resolution methods cannot access deep tissue. However, theoretical models provide valuable insight. We model the CSF pathway as a network of hydraulic resistances, using published parameter values. A few parameters (permeability of PVSs and the parenchyma, and dimensions of PVSs and astrocyte endfoot gaps) have wide uncertainties, so we focus on the limits of their ranges by analyzing different parametric scenarios. We identify low-resistance PVSs and high-resistance parenchyma as the only scenario that satisfies three essential criteria: that the flow be driven by a small pressure drop, exhibit good CSF perfusion throughout the cortex, and exhibit a substantial increase in flow during sleep. Our results point to the most important parameters, such as astrocyte endfoot gap dimensions, to be measured in future experiments.

Project description:BackgroundEstimations of causal effects from observational data are subject to various sources of bias. One method for adjusting for the residual biases in the estimation of treatment effects is through the use of negative control outcomes, which are outcomes not believed to be affected by the treatment of interest. The empirical calibration procedure is a technique that uses negative control outcomes to calibrate p-values. An extension of this technique calibrates the coverage of the 95% confidence interval of a treatment effect estimate by using negative control outcomes as well as positive control outcomes, which are outcomes for which the treatment of interest has known effects. Although empirical calibration has been used in several large observational studies, there is no systematic examination of its effect under different bias scenarios.MethodsThe effect of empirical calibration of confidence intervals was analyzed using simulated datasets with known treatment effects. The simulations consisted of binary treatment and binary outcome, with biases resulting from unmeasured confounder, model misspecification, measurement error, and lack of positivity. The performance of the empirical calibration was evaluated by determining the change in the coverage of the confidence interval and the bias in the treatment effect estimate.ResultsEmpirical calibration increased coverage of the 95% confidence interval of the treatment effect estimate under most bias scenarios but was inconsistent in adjusting the bias in the treatment effect estimate. Empirical calibration of confidence intervals was most effective when adjusting for the unmeasured confounding bias. Suitable negative controls had a large impact on the adjustment made by empirical calibration, but small improvements in the coverage of the outcome of interest were also observable when using unsuitable negative controls.ConclusionsThis work adds evidence to the efficacy of empirical calibration of the confidence intervals in observational studies. Calibration of confidence intervals is most effective where there are biases due to unmeasured confounding. Further research is needed on the selection of suitable negative controls.

Project description:Predicting runoff with precision holds immense importance for flood control, water resource management, and basin ecological dispatch. Deep learning, especially long short-term memory (LSTM) neural networks, has excelled in runoff prediction, often outperforming traditional hydrological models. Recent studies suggest that deep learning models employing the self-attention mechanism, such as Transformer and Informer, can achieve even better results than LSTM. However, research exploring the multi-step runoff prediction capabilities of these novel models across diverse scenarios remains scarce. In this investigation, we introduce a relative location coding-enhanced Informer model, termed Rel-Informer, and compare its performance in rainfall-runoff prediction against the standard Informer, Transformer, and LSTM models. The publicly available CAMELS dataset is utilized for training and validating the models, and four experiments are designed: (1) Individual rainfall-runoff modeling (one model per catchment); (2) Regional rainfall-runoff modeling (one model per region); (3) Fine-tuned regional rainfall-runoff modeling (fine-tuned from Experiment 2); (4) Large-scale rainfall-runoff modeling for ungauged catchments (one model for all catchments). The findings reveal that Rel-Informer consistently performs better than the other models, particularly in short-term runoff predictions (1-3 days ahead). Although regional modeling is less precise than individual modeling, it significantly benefits from fine-tuning. The large-scale regional Rel-Informer model effectively predicts runoff in ungauged catchments, showcasing its potential for widespread runoff prediction. This study underscores the influence of hydrological characteristics, such as snowmelt and baseflow indices, on prediction accuracy. In conclusion, the Rel-Informer model, enhanced with improved relative position encoding, emerges as a promising tool for runoff forecasting, especially in data-rich catchments.

Project description:IntroductionThe slow descent in TB burden, the COVID-19 pandemic, along with the rise of multidrug-resistant strains of Mycobacterium tuberculosis, seriously threaten TB control and the goals of the End TB strategy. To fight back, several vaccine candidates are under development, with some of them undergoing the phases 2B and 3 of the development pipeline. The impact of these vaccines on the general population needs to be addressed using disease-transmission models, and, in a country like China, which last year ranked third in number of cases worldwide, and where the population is aging at a fast pace, the impact of TB vaccination campaigns may depend heavily upon the age of targeted populations, the mechanistic descriptions of the TB vaccines and the coupling between TB dynamics and demographic evolution.MethodsIn this work, we studied the potential impact of a new TB vaccine in China targeting adolescents (15-19 y.o.) or older adults (60-64 y.o.), according to varying vaccine descriptions that represent reasonable mechanisms of action leading to prevention of disease, or prevention of recurrence, each of them targetting specific routes to TB disease. To measure the influence of the description of the coupling between transmission dynamics and aging in TB transmission models, we explored two different approaches to compute the evolution of the contact matrices, which relate to the spreading among different age strata.ResultsOur findings highlight the dependence of model-based impact estimates on vaccine profiles and the chosen modeling approach for describing the evolution of contact matrices. Our results also show, in line with previous modeling works, that older adult vaccination is a suitable option in China to reduce the incidence of TB as long as the vaccine is able to protect already exposed individuals.DiscussionThis study underscores the importance of considering vaccine characteristics and demographic dynamics in shaping TB control strategies. In this sense, older adult vaccination emerges as a promising avenue for mitigating TB transmission in China but also remarks the need for tailored intervention strategies aligned with demographic trends.
