Self-assembled quantum dot microstructure guided by a microemulsion approach for immunoassays.

ABSTRACT: Quantum dot microstructures were fabricated through a convenient microemulsion approach in this study. A polymer solution containing a stabilizer was mixed with a quantum dot aqueous solution, to prepare a reversed microemulsion, through shaking. Then, the microemulsion was cast on a solid substrate followed by evaporating steps, resulting in the formation of an ordered porous film. Interestingly, the quantum dot microstructure can be produced at the same time. The immunoassay experiment could be realized by the fluorescent microstructures. The green fluorescence microstructure specifically bound with antigens marked with red color quantum dots, resulting in the enhancement of red fluorescence domains and the decrease of green fluorescence. With the addition of unlabeled antigens, the green fluorescence microstructure was recovered. This strategy implies that the quantum dot pattern has potential on biochip, biosensor, and imaging analysis.

Misfit-guided self-organization of anticorrelated Ge quantum dot arrays on Si nanowires.

Project description:Misfit-strain guided growth of periodic quantum dot (QD) arrays in planar thin film epitaxy has been a popular nanostructure fabrication method. Engineering misfit-guided QD growth on a nanoscale substrate such as the small curvature surface of a nanowire represents a new approach to self-organized nanostructure preparation. Perhaps more profoundly, the periodic stress underlying each QD and the resulting modulation of electro-optical properties inside the nanowire backbone promise to provide a new platform for novel mechano-electronic, thermoelectronic, and optoelectronic devices. Herein, we report a first experimental demonstration of self-organized and self-limited growth of coherent, periodic Ge QDs on a one-dimensional Si nanowire substrate. Systematic characterizations reveal several distinctively different modes of Ge QD ordering on the Si nanowire substrate depending on the core diameter. In particular, Ge QD arrays on Si nanowires of around 20 nm diameter predominantly exhibit an anticorrelated pattern whose wavelength agrees with theoretical predictions. The correlated pattern can be attributed to propagation and correlation of misfit strain across the diameter of the thin nanowire substrate. The QD array growth is self-limited as the wavelength of the QDs remains unchanged even after prolonged Ge deposition. Furthermore, we demonstrate a direct kinetic transformation from a uniform Ge shell layer to discrete QD arrays by a postgrowth annealing process.

Culling a Self-Assembled Quantum Dot as a Single-Photon Source Using X-ray Microscopy.

Project description:Epitaxially grown self-assembled semiconductor quantum dots (QDs) with atom-like optical properties have emerged as the best choice for single-photon sources required for the development of quantum technology and quantum networks. Nondestructive selection of a single QD having desired structural, compositional, and optical characteristics is essential to obtain noise-free, fully indistinguishable single or entangled photons from single-photon emitters. Here, we show that the structural orientations and local compositional inhomogeneities within a single QD and the surrounding wet layer can be probed in a screening fashion by scanning X-ray diffraction microscopy and X-ray fluorescence with a few tens of nanometers-sized synchrotron radiation beam. The presented measurement protocol can be used to cull the best single QD from the enormous number of self-assembled dots grown simultaneously. The obtained results show that the elemental composition and resultant strain profiles of a QD are sensitive to in-plane crystallographic directions. We also observe that lattice expansion after a certain composition-limit introduces shear strain within a QD, enabling the possibility of controlled chiral-QD formation. Nanoscale chirality and compositional anisotropy, contradictory to common assumptions, need to be incorporated into existing theoretical models to predict the optical properties of single-photon sources and to further tune the epitaxial growth process of self-assembled quantum structures.

An Integrated Approach to Improve the Assay Performance of Quantum Dot-Based Lateral Flow Immunoassays by Using Silver Deposition.

Project description:Traditional quantum dot-based lateral flow immunoassay (QD-LFIA) is limited to signal loss in part by the blinking, photobleaching and oxidative quenching of QD probes. Inspired by the good application of silver deposition on QD surfaces in tissue imaging, and in the context of improving the assay performance without compromising the simplicity and practicality, we report that introducing the QD-silver combination to the LFIA system, has the advantages of accuracy improvement, signal enhancement and user friendliness promotion, but maintains the cost-effective property and commercial accessibility of QD-LFIA. The effect was shown by using CdSe/ZnS QD-LFIA coupled with anti-sodium pentachlorophenate antibody, which provided a 4-fold improvement in the signal, a 2.5-fold improvement in the detection limit and a zero false-negative rate for sodium pentachlorophenate analysis in chicken samples. The proposed LFIA integrates the possibilities of colorimetric and fluorometric detection with different detection limits (fluorometric at 10 ng/mL and colorimetric at 4 ng/mL) and with acceptable detection times (fluorometric at 12 min and colorimetric at 27 min). The current results indicate that this QD-silver combined LFIA is complementary to conventional fluorescence LFIA and could be an inexpensive, versatile, and sensitive alternative.

Self-Catalyzed InSb/InAs Quantum Dot Nanowires.

Project description:The nanowire platform offers great opportunities for improving the quality and range of applications of semiconductor quantum wells and dots. Here, we present the self-catalyzed growth of InAs/InSb/InAs axial heterostructured nanowires with a single defect-free InSb quantum dot, on Si substrates, by chemical beam epitaxy. A systematic variation of the growth parameters for the InAs top segment has been investigated and the resulting nanowire morphology analyzed. We found that the growth temperature strongly influences the axial and radial growth rates of the top InAs segment. As a consequence, we can reduce the InAs shell thickness around the InSb quantum dot by increasing the InAs growth temperature. Moreover, we observed that both axial and radial growth rates are enhanced by the As line pressure as long as the In droplet on the top of the nanowire is preserved. Finally, the time evolution of the diameter along the entire length of the nanowires allowed us to understand that there are two In diffusion paths contributing to the radial InAs growth and that the interplay of these two mechanisms together with the total length of the nanowires determine the final shape of the nanowires. This study provides insights in understanding the growth mechanisms of self-catalyzed InSb/InAs quantum dot nanowires, and our results can be extended also to the growth of other self-catalyzed heterostructured nanowires, providing useful guidelines for the realization of quantum structures with the desired morphology and properties.

Simultaneous detection of multiple viruses in their co-infected cells using multicolour imaging with self-assembled quantum dot probes

Project description:The simultaneous detection and evaluation of the coinfection of a cell by multiple viruses or even multiple subtypes still is a difficult challenge. The authors introduce a method for simultaneous imaging, detection and quantitative evaluation of multiple viruses in single cells by using multicolor quantum dot (QD) probes and in a single staining cycle. The multicolor QD probes were fabricated via interaction between QDs conjugated to Staph. aureus protein A (SpA-QDs) and virus-specific antibodies. A cocktail of differently colored QD-SpA-MAbs probes were loaded into the same cells containing multiple viruses, and this enabled the different viruses to be fluorescently imaged and analyzed simultaneously. Specifically, influenza A viruses of type H1N1, H3N2, and H9N2, as well as human adenovirus species B type 3 (HAdV-B3) were imaged and detected in virus-infected cells or in their co-infected cells. In our perception, the method provides a flexible platform for simultaneous detection of multiple viruses in co-infected cells. Hence, it offers new opportunities for the molecular diagnosis of virus coinfection and for studies on virus-cell interactions. Mono-Dispersed Microspheres Locally Assembled on Porous Substrates Formed through a Microemulsion Approach.

Project description:A cost-effective, simple, and time-saving method to fabricate mono-dispersed periodic microsphere structures on substrates with patterned sites is very meaningful due to their significance on various biological studies. Herein, a simple and facile method to fabricate mono-dispersed microsphere arrays on porous substrates was developed. The mixture of polystyrene and an organic stabilizer solution which contains aqueous solution, fabricated through shaking, was applied to prepare microemulsion solution. An ordered porous structure was produced by spreading and evaporating the solvent of microemulsion on a glass slide, accompanied by the enrichment of didodecylamine in the cavities. The porous cavities were further modified with polyacrylic acid and poly(diallyldimethylammonium chloride) which could immobilize the microspheres. The charged microspheres were incorporated into the cavities by an electrostatic interaction with the oppositely charged polyelectrolytes. The positive polyelectrolytes with abundant charges as well as a suitable content and dimension of microspheres, ensured the formation of mono-dispersed and ordered arrays. Considering that other charged particles were universally suitable for the present strategy, the reported approach opened an efficient way for the preparation of microsphere-based materials.

Quantum Dot Self-Assembly Enables Low-Threshold Lasing.

Project description:Perovskite quantum dots (QDs) are of interest for solution-processed lasers; however, their short Auger lifetime has limited lasing operation principally to the femtosecond temporal regime the photoexcitation levels to achieve optical gain threshold are up to two orders of magnitude higher in the nanosecond regime than in the femtosecond. Here the authors report QD superlattices in which the gain medium facilitates excitonic delocalization to decrease Auger recombination and in which the macroscopic dimensions of the structures provide the optical feedback required for lasing. The authors develope a self-assembly strategy that relies on sodiumd-an assembly director that passivates the surface of the QDs and induces self-assembly to form ordered three-dimensional cubic structures. A density functional theory model that accounts for the attraction forces between QDs allows to explain self-assembly and superlattice formation. Compared to conventional organic-ligand-passivated QDs, sodium enables higher attractive forces, ultimately leading to the formation of micron-length scale structures and the optical faceting required for feedback. Simultaneously, the decreased inter-dot distance enabled by the new ligand enhances exciton delocalization among QDs, as demonstrated by the dynamically red-shifted photoluminescence. These structures function as the lasing cavity and the gain medium, enabling nanosecond-sustained lasing with a threshold of 25 µJ cm-2 .

Ultra-sensitive, rapid gold nanoparticle-quantum dot plexcitonic self-assembled aptamer-based nanobiosensor for the detection of human cardiac troponin I.

Project description:Acute myocardial infarction (AMI) is one of the leading causes of death throughout the world. Usual methods for detecting AMI are expensive, time-consuming and using blood samples as biological samples. Therefore, creating an ultra-fast, sensitive and non-invasive diagnostic test is necessary. Herein, a novel ultra-sensitive, fluorescent, plasmon-exciton coupling hybrid of a gold nanoparticle-quantum dot (PQ)-based aptamer nanobiosensor is presented for the detection of human cardiac troponin I (cTnI), the golden biomarker of AMI, and a preclinical test is performed within saliva. The binding of the cTnI protein to aptamer leads to a fluorescence enhancement of the plexcitonic hybrid system. The response range of this nanobiosensor is 0.4-2500 fM and the limit of detection is 0.3 fM. It seems that this novel design of nanobiosensor in the form of the PQ plexcitonic hybrid system can presents new opportunities for nanobiosensor progress.

Efficient quantum dot-quantum dot and quantum dot-dye energy transfer in biotemplated assemblies.

Project description:CdSe semiconductor nanocrystal quantum dots are assembled into nanowire-like arrays employing microtubule fibers as nanoscale molecular "scaffolds." Spectrally and time-resolved energy-transfer analysis is used to assess the assembly of the nanoparticles into the hybrid inorganic biomolecular structure. Specifically, we demonstrate that a comprehensive study of energy transfer between quantum dot pairs on the biotemplate and, alternatively, between quantum dots and molecular dyes embedded in the microtubule scaffold comprises a powerful spectroscopic tool for evaluating the assembly process. In addition to revealing the extent to which assembly has occurred, the approach allows determination of particle-to-particle (and particle-to-dye) distances within the biomediated array. Significantly, the characterization is realized in situ, without need for further sample workup or risk of disturbing the solution-phase constructs. Furthermore, we find that the assemblies prepared in this way exhibit efficient quantum dot-quantum dot and quantum dot-dye energy transfer that affords faster energy-transfer rates compared to densely packed quantum dot arrays on planar substrates and to small-molecule-mediated quantum dot-dye couples, respectively.
