Seedless preparation of Au nanorods by hydroquinone assistant and red blood cell membrane camouflage.

ABSTRACT: Au nanorods (AuNRs) have attracted extensive attention in diagnosis and therapy, because of their excellent photothermal effects. Although many of attempts have been employed to prepare AuNRs with a good-size monodispersity, facile and feasible preparation methods are still welcomed in consideration of theranostics-related photothermal applications. In this work, AuNRs are prepared through a seedless method using hydroquinone as the reductant. The longitudinal located surface plasmon resonance peak of the as-prepared AuNRs can be tuned from 650 to 840 nm and the AuNRs indicate a good photothermal conversion efficiency. To lower the toxicity, natural red blood cell membranes are employed to coat the AuNRs. In the primary experiment in vitro, red blood cell membrane-coated AuNRs exhibit good biocompatibility and photothermal treatment effect.

Preparation and Gas Sensing Properties of In2O3/Au Nanorods for Detection of Volatile Organic Compounds in Exhaled Breath.

Project description:A series of In2O3/Au nanorods (NRs) were fabricated and characterized by scanning electron microscope (SEM), transmission electron microscope (TEM), X-ray diffractometer (XRD) and X-ray photoelectron spectroscopy (XPS). The length to diameter ratios of In2O3/Au NRs was periodically modulated in the range of 2.9-4.5 through controlling the initial content of indium salt and reaction time. Their gas sensing properties to volatile organic compounds (VOCs) were carefully studied and then applied in exhaled breath detection. The results demonstrate that In2O3/Au NRs gas sensor can effectively detect acetone at 250 °C and ethanol at 400 °C. The corresponding actual detection limit is as low as 0.1 ppm to acetone and 0.05 ppm to ethanol, respectively. Moreover, by using humidity compensation method, In2O3/Au NRs gas sensor can clearly distinguish the acetone and ethanol biomarkers in human breath. The main reason of the enhanced gas sensing properties was attributed to the "spillover effects" between Au and In2O3 NRs. The excellent sensing performance indicates that In2O3/Au NRs is a promising functional material to actual application in monitoring and detecting diabetes and safe driving area in a noninvasive and more accurate way.

Study on preparation, characterization and multidrug resistance reversal of red blood cell membrane-camouflaged tetrandrine-loaded PLGA nanoparticles.

Project description:The multidrug resistance in tumor (MDR) is a major barrier to efficient cancer therapy. Modern pharmacological studies have proven that tetrandrine (TET) has great potential in reversing MDR. However, it has a series of medication problems in clinic such as poor water solubility, low oral bioavailability and short half-life in vivo. Aiming at the above problems, red blood cell membrane-camouflaged TET-loaded poly(lactic-co-glycolic acid) (PLGA) nanoparticles (RPTNs) had been developed. The RPTNs had spherical shell-core double layer structure with average particle size of 164.1 ± 1.65 nm and encapsulation efficiency of 84.1% ± 0.41%. Compared with TET-PLGA nanoparticles (PTNs), the RPTNs reduced RAW 264.7 macrophages' swallowing by 32% due to its retention of natural membrane proteins. The cumulative drug release of RPTNs was 81.88% within 120 h. And pharmacokinetic study showed that the blood half-life of RPTNs was 19.38 h, which was 2.95 times of free drug. When RPTNs of 2 μg/mL TET were administered in combination with adriamycin (ADR), significant MDR reversal effect was observed in drug-resistant cells MCF-7/ADR. In a word, the RPTNs hold potential to improve its efficacy and broaden its clinical application.

Electrochemical impedance spectroscopy of single Au nanorods.

Project description:We propose monochromatic dark-field imaging microscopy (DFM) to measure the non-faradaic electrochemical impedance spectroscopy (EIS) of single Au nanorods (AuNRs). DFM was utilized to monitor the plasmonic scattering of monochromatic incident light by surface-immobilized individual AuNRs. When modulating the surface potential at a certain frequency, non-faradaic charging and discharging of AuNRs altered their electron density, leading to periodical fluctuations in the scattering intensity. Analysis of the amplitude and phase of the optical intensity fluctuation as a function of modulation frequency resulted in the EIS of single AuNRs. High-frequency (>100 Hz) modulation allowed us to differentiate the intrinsic charging effect from other contributions such as the periodic migration and accumulation of counterions in the surrounding medium, because the latter occurred at a longer timescale. As a result, single nanoparticle EIS led to the surface capacitance of single AuNRs being closer to the theoretical value. Since interfacial capacitance has been proven sensitive to molecular interactions, the present work also offers a new platform for single nanoparticle sensing by measuring the single nanoparticle capacitance.

Red Blood Cell Membrane-Coated Nanoparticles Enable Incompatible Blood Transfusions.

Project description:Blood transfusions save lives and improve health every day. Despite the matching of blood types being stricter than it ever has been, emergency transfusions among incompatible blood types are still inevitable in the clinic when there is a lack of acceptable blood types for recipients. Here to overcome this, a counter measure nanoplatform consisting of a polymeric core coated by a red blood cell (RBC) membrane is developed. With A-type or B-type RBC membrane camouflaging, the nanoplatform is capable of specifically capturing anti-A or anti-B IgM antibodies within B-type or A-type whole blood, thereby decreasing the corresponding IgM antibody levels and then allowing the incompatible blood transfusions. In addition to IgM, the anti-RBC IgG antibody in a passive immunization murine model can likewise be neutralized by this nanoplatform, leading to prolonged circulation time of incompatible donor RBCs. Noteworthily, nanoplatform made by expired RBCs (>42 days stored hypothermically) and then subjected to lyophilization does not impair their effect on antibody neutralization. Most importantly, antibody-captured RBC-NP do not exacerbate the risk of inflammation, complement activation, and coagulopathy in an acute hemorrhagic shock murine model. Overall, this biomimetic nanoplatform can safely neutralize the antibody to enable incompatible blood transfusion.

One-Step Hydrothermal Synthesis, Thermochromic and Infrared Camouflage Properties of Vanadium Dioxide Nanorods.

Project description:Vanadium dioxide (VO2) has attracted interest from researchers because it undergoes a metal-insulator phase transition (MIT), which is accompanied by a reversible and remarkable change in both electrical and optical properties. VO2 exhibits numerous polymorphs and thus it is essential to control the growth of specific monoclinic VO2 (M) and rutile VO2 (R) phases. In this study, we developed a cost-effective and facile method for preparing VO2 nanorods with a highly crystalline monoclinic phase by one-step hydrothermal synthesis, in which only V2O5 and H2C2O4 are used as raw materials. The phase evolution of VO2 during the hydrothermal process was studied. The obtained VO2 nanorods were thoroughly mixed with fluorocarbon resin and homogeneous emulsifier in an ethanol solution to obtain a VO2 dispersion. To prepare VO2 films, screen printing was performed with a stainless steel screen mesh mask on glasses or fabric substrate. The VO2 coating had good thermochromic performance; the infrared transmittance change was greater than 20% @1.5 μm whilst keeping the visible transmittance greater than 50%. Meanwhile, the polyester base coating on the fabric had an emissivity change of up to 22%, which provides a solution for adaptive IR camouflage.

Red Blood Cell Membrane Processing for Biomedical Applications.

Project description:Red blood cells (RBC) are actually exploited as innovative drug delivery systems with unconventional and convenient properties. Because of a long in vivo survival and a non-random removal from circulation, RBC can be loaded with drugs and/or contrasting agents without affecting these properties and maintaining the original immune competence. However, native or drug-loaded RBC, can be modified decorating the membrane with peptides, antibodies or small chemical entities so favoring the targeting of the processed RBC to specific cells or organs. Convenient modifications have been exploited to induce immune tolerance or immunogenicity, to deliver antibodies capable of targeting other cells, and to deliver a number of constructs that can recognize circulating pathogens or toxins. The methods used to induce membrane processing useful for biomedical applications include the use of crosslinking agents and bifunctional antibodies, biotinylation and membrane insertion. Another approach includes the expression of engineered membrane proteins upon ex vivo transfection of immature erythroid precursors with lentiviral vectors, followed by in vitro expansion and differentiation into mature erythrocytes before administration to a patient in need. Several applications have now reached the clinic and a couple of companies that take advantage from these properties of RBC are already in Phase 3 with selected applications. The peculiar properties of the RBC and the active research in this field by a number of qualified investigators, have opened new exciting perspectives on the use of RBC as carriers of drugs or as cellular therapeutics.

Membrane compartmentalization in Southeast Asian ovalocytosis red blood cells.

Project description:Red blood cells (RBCs) from individuals with Southeast Asian ovalocytosis (SAO) contain a mutant band 3 protein that causes the formation of unique linear oligomers in the RBC membrane. We used single-particle tracking to measure the lateral diffusion of individual glycophorin C (GPC), band 3, and CD58 proteins in membranes of intact SAO RBCs and normal RBCs (nRBCs). GPC, an integral protein that binds with high affinity to the RBC membrane skeleton, showed oscillatory motion within confinement areas that were smaller in SAO RBCs than in nRBCs. The additional confinement in SAO RBCs could be due to membrane stiffening associated with the SAO phenotype. Band 3 in both SAO RBCs and nRBCs also showed confined motion over short times (ms) and distances (nm), and the area of confinement was smaller in SAO RBCs than in nRBCs. These data presumably reflect the constraints imposed by band 3 oligomerization. Similarly, the glycosylphosphatidylinositol-linked protein CD58 showed loosely confined diffusion in nRBCs and a substantially higher degree of confinement in SAO RBCs. Restricted protein mobility could contribute to the altered adherence of parasite-infected RBCs to vascular endothelium that is thought to protect individuals with SAO from severe manifestations of malaria.

Metabolic remodeling of the human red blood cell membrane.

Project description:The remarkable deformability of the human red blood cell (RBC) results from the coupled dynamic response of the phospholipid bilayer and the spectrin molecular network. Here we present quantitative connections between spectrin morphology and membrane fluctuations of human RBCs by using dynamic full-field laser interferometry techniques. We present conclusive evidence that the presence of adenosine 5'-triphosphate (ATP) facilitates non-equilibrium dynamic fluctuations in the RBC membrane that are highly correlated with the biconcave shape of RBCs. Spatial analysis of the fluctuations reveals that these non-equilibrium membrane vibrations are enhanced at the scale of spectrin mesh size. Our results indicate that the dynamic remodeling of the coupled membranes powered by ATP results in non-equilibrium membrane fluctuations manifesting from both metabolic and thermal energies and also maintains the biconcave shape of RBCs.

Dietary Fatty Acids Affect Red Blood Cell Membrane Composition and Red Blood Cell ATP Release in Dairy Cows.

Project description:Diets of dairy cows are often based on maize silage (MS), delivering lower amounts of n-3 fatty acids (FA) compared to grass silage-based diets. The fatty acid composition of the cell membrane can affect the cell function. We evaluated the effects of an MS-based diet on bovine red blood cell (RBC) membrane FA composition and dietary effects on controlled ATP release of RBC. In trial 1, German Holstein cows were fed an MS-based total mixed ration for 24 weeks. The FA composition of RBC membranes from repeatedly taken blood samples was analysed in addition to the abundance of the RBC membrane protein flotillin-1, which is involved in, for example, cell signalling. In trial 2, four rumen fistulated MS-fed cows were abomasally infused in a 4 × 4 Latin square model with three successively increasing lipid dosages (coconut oil, linseed-safflower oil mix (EFA; rich in n-3 FA), Lutalin®, providing conjugated linoleic acids (CLA) or the combination of the supplements, EFA + CLA) for six weeks, followed by a three-week washout period. In trial 2, we analysed RBC ATP release, flotillin-1, and the membrane protein abundance of pannexin-1, which is involved in ATP release as the last part of a signalling cascade. In trial 1, the total amount of n-3 FA in RBC membranes decreased and the flotillin-1 abundance increased over time. In trial 2, the RBC n-3 FA amount was higher after the six-week infusion period of EFA or EFA + CLA. Furthermore, depending on the dosage of FA, the ATP release from RBC increased. The abundance of flotillin-1 and pannexin-1 was not affected in trial 2. It is concluded that changes of the membrane FA composition influence the RBC function, leading to altered ATP release from intact bovine RBC.
