The association between inadequate sleep and accelerated brain ageing.

ABSTRACT: Numerous studies indicate large heterogeneity in brain ageing, which can be attributed to modifiable lifestyle factors, including sleep. Inadequate sleep has been previously linked to gray (GM) and white (WM) matter changes. However, the reported findings are highly inconsistent. By contrast to previous research independently characterizing patterns of either GM or WM changes, we used here linked independent component analysis (FLICA) to examine covariation in GM, and WM in a group of older adults (n = 50). Next, we employed a novel technique to estimate the brain age delta (difference between chronological and brain age assessed using neuroimaging data) and study its associations with sleep quality and sleep fragmentation, hypothesizing that inadequate sleep accelerates brain ageing. FLICA revealed a number of multimodal components, associated with age, sleep quality, and sleep fragmentation. Subsequently, we show significant associations between brain age delta and inadequate sleep, suggesting 2 years deviation above the chronological age. Our findings indicate sensitivity of multimodal approaches and brain age delta in detecting link between inadequate sleep and accelerated brain ageing.

Project description:Accelerated cognitive ageing (ACA) is an ageing co-morbidity in epilepsy that is diagnosed through the observation of an evident IQ decline of more than 1 standard deviation (15 points) around the age of 50 years old. To understand the mechanism of action of this pathology, we assessed brain dynamics with the use of resting-state fMRI data. In this paper, we present novel and promising methods to extract brain dynamics between large-scale resting-state networks: the emulative power, wavelet coherence, and granger causality between the networks were extracted in two resting-state sessions of 24 participants (10 ACA, 14 controls). We also calculated the widely used static functional connectivity to compare the methods. To find the best biomarkers of ACA, and have a better understanding of this epilepsy co-morbidity we compared the aforementioned between-network neurodynamics using classifiers and known machine learning algorithms; and assessed their performance. Results show that features based on the evolutionary game theory on networks approach, the emulative powers, are the best descriptors of the co-morbidity, using dynamics associated with the default mode and dorsal attention networks. With these dynamic markers, linear discriminant analysis could identify ACA patients at 82.9% accuracy. Using wavelet coherence features with decision-tree algorithm, and static functional connectivity features with support vector machine, ACA could be identified at 77.1% and 77.9% accuracy respectively. Granger causality fell short of being a relevant biomarker with best classifiers having an average accuracy of 67.9%. Combining the features based on the game theory, wavelet coherence, Granger-causality, and static functional connectivity- approaches increased the classification performance up to 90.0% average accuracy using support vector machine with a peak accuracy of 95.8%. The dynamics of the networks that lead to the best classifier performances are known to be challenged in elderly. Since our groups were age-matched, the results are in line with the idea of ACA patients having an accelerated cognitive decline. This classification pipeline is promising and could help to diagnose other neuropsychiatric disorders, and contribute to the field of psychoradiology.

Project description:BackgroundHealthy sleep is vital for maintaining optimal mental and physical health. Accumulating evidence suggests that sleep loss and disturbances play a significant role in the biological aging process, early onset of disease, and reduced lifespan. While numerous studies have explored the association between biological aging and its drivers, only a few studies have examined its relationship with sleep quality. In this study, we investigated the associations between sleep quality and epigenetic age acceleration using whole blood samples from a cohort of 692 Korean adults. Sleep quality of each participant was assessed using the validated Pittsburgh Sleep Quality Index (PSQI), which encompassed seven domains: subjective sleep quality, sleep latency, sleep duration, habitual sleep efficiency, sleep disturbance, use of sleep medication, and daytime dysfunction. Four epigenetic age accelerations (HorvathAgeAccel, HannumAgeAccel, PhenoAgeAccel, and GrimAgeAccel) and the pace of aging, DunedinPACE, were investigated for epigenetic aging estimates.ResultsAmong the 692 participants (good sleepers [n = 441, 63.7%]; poor sleepers [n = 251, 36.3%]), DunedinPACE was positively correlated with PSQI scores in poor sleepers ( γ =0.18, p < 0.01). GrimAgeAccel ( β =0.18, p = 0.02) and DunedinPACE ( β =0.01, p < 0.01) showed a statistically significant association with PSQI scores only in poor sleepers by multiple linear regression. In addition, every one-point increase in PSQI was associated with a 15% increase in the risk of metabolic syndrome (MetS) among poor sleepers (OR = 1.15, 95% CI = 1.03-1.29, p = 0.011). In MetS components, a positive correlation was observed between PSQI score and fasting glucose ( γ = 0.19, p < 0.01).ConclusionsThis study suggests that worsening sleep quality, especially in poor sleepers, is associated with accelerated epigenetic aging for GrimAgeAccel and DundinePACE with risk of metabolic syndrome. This finding could potentially serve as a promising strategy for preventing age-related diseases in the future.

Project description:IntroductionThe process of population ageing that is occurring in developed societies represents a major challenge for the health system. The aim of this study is to analyse factors that have an influence on early vascular ageing (EVA), estimated by carotid-femoral pulse wave velocity (cf-PWV) and Cardio Ankle Vascular Index (CAVI), and to determine differences by gender in a Spanish population.Methods and analysisAn observational, descriptive, cross-sectional study.Study populationFrom the population assigned to the participating healthcare centres, a cluster random sampling stratified by age and gender will be performed to obtain 500 participants aged between 35 and 75. Those who meet the inclusion criteria and give written informed consent will be included in the study.MeasurementsMain dependent variables: cf-PWV determined using the SphygmoCor System and CAVI estimated using VASERA. Secondary dependent variables: telomere length, carotid intima-media thickness, central and peripheral augmentation index, ankle-brachial pulse wave velocity, ankle-brachial index, retinal arteriovenous index, and renal and cardiac organ damage.Independent variableslifestyles (physical activity, adherence to the Mediterranean diet, alcohol and tobacco consumption); psychological factors (depression, anxiety and chronic stress); inflammatory factors and oxidative stress.Ethics and disseminationThe study has been approved by the clinical research ethics committee of the healthcare area of Salamanca. All study participants will sign an informed consent form agreeing to participate in the study in compliance with the Declaration of Helsinki and the WHO standards for observational studies.
