DABCO-catalyzed Knoevenagel condensation of aldehydes with ethyl cyanoacetate using hydroxy ionic liquid as a promoter.

ABSTRACT: N-(2-Hydroxy-ethyl)-pyridinium chloride ([HyEtPy]Cl) was synthesized and explored as a novel promoter for 1,4-diazabicyclo [2.2.2] octane (DABCO)-catalyzed Knoevenagel condensation reactions, which showed better catalytic activity compared to other ionic liquid (IL) that had no hydroxyl group attached to the IL scaffold. The effect of hydrogen bond formation between the hydroxyl group of [HyEtPy]Cl and the carbonyl group of aldehyde played an important role in the Knoevenagel condensation reaction. In the [HyEtPy]Cl-H₂O-DABCO composite system, Knoevenagel condensation reactions proceeded smoothly and cleanly, and the corresponding Knoevenagel condensation products were obtained in good to excellent yields in all cases examined. This protocol provides a versatile solvent-catalyst system, which has notable advantages such as being eco-friendly, ease of work-up and convenient reuse of the ionic liquid.

Novel Pyridinium Based Ionic Liquid Promoter for Aqueous Knoevenagel Condensation: Green and Efficient Synthesis of New Derivatives with Their Anticancer Evaluation.

Project description:Herein, a distinctive dihydroxy ionic liquid ([Py-2OH]OAc) was straightforwardly assembled from the sonication of pyridine with 2-chloropropane-1,3-diol by employing sodium acetate as an ion exchanger. The efficiency of the ([Py-2OH]OAc as a promoter for the sono-synthesis of a novel library of condensed products through DABCO-catalyzed Knoevenagel condensation process of adequate active cyclic methylenes and ninhydrin was next investigated using ultimate greener conditions. All of the reactions studied went cleanly and smoothly, and the resulting Knoevenagel condensation compounds were recovered in high yields without detecting the aldol intermediates in the end products. Compared to traditional strategies, the suggested approach has numerous advantages including mild reaction conditions with no by-products, eco-friendly solvent, outstanding performance in many green metrics, and usability in gram-scale synthesis. The reusability of the ionic liquid was also studied, with an overall retrieved yield of around 97% for seven consecutive runs without any substantial reduction in the performance. The novel obtained compounds were further assessed for their in vitro antitumor potential toward three human tumor cell lines: Colo-205 (colon cancer), MCF-7 (breast cancer), and A549 (lung cancer) by employing the MTT assay, and the findings were evaluated with the reference Doxorubicin. The results demonstrated that the majority of the developed products had potent activities at very low doses. Compounds comprising rhodanine (5) or chromane (12) moieties exhibited the most promising cytotoxic effects toward three cell lines, particularly rhodanine carboxylic acid derivative (5c), showing superior cytotoxic effects against the investigated cell lines compared to the reference drug. Furthermore, automated docking simulation studies were also performed to support the results obtained.

Unexpected polymorphism during a catalyzed mechanochemical Knoevenagel condensation.

Project description:The transformation of a base-catalyzed, mechano-assisted Knoevenagel condensation of mono-fluorinated benzaldehyde derivatives (p-, m-, o-benzaldehyde) with malonodinitrile was investigated in situ and in real time. Upon milling, the para-substituted product was found to crystallize initially into two different polymorphic forms, depending on the quantity of catalyst used. For low catalyst concentrations, a mechanically metastable phase (monoclinic) was initially formed, converting to the mechanically stable phase (triclinic) upon further grinding. Instead, higher catalyst concentrations crystallize directly as the triclinic product. Inclusion of catalyst in the final product, as evidenced by mass spectrometric analysis, suggests this complex polymorphic pathway may be due to seeding effects. Multivariate analysis for the in situ Raman spectra supports this complex formation pathway, and offers a new approach to monitoring multi-phase reactions during ball milling.

Chitosan nanofiber-catalyzed highly selective Knoevenagel condensation in aqueous methanol.

Project description:A chitosan nanofiber (CsNF)-catalyzed Knoevenagel reaction in green solvent, namely aqueous methanol, was investigated. CsNFs solely catalyzed the desired C-C bond formations in high yield with high selectivity, while conventional small-molecule amines, such as n-hexylamine and triethylamine, inevitably promoted transesterification to produce a large amount of solvolysis byproducts. Structural and chemical analyses of CsNFs suggested that the unique nanoarchitecture, in which chitosan molecules were bundled to ensure the high accessibility of substrates to catalytic sites, was critical to the highly efficient Knoevenagel condensation. The products were obtained in high purity without solvent-consuming purification, and the CsNF catalyst was easily removed and recycled. This study highlights a novel and promising function of CsNFs in green catalysis as emerging polysaccharide-based nanofibers.

A novel composite of ionic liquid-containing polymer and metal-organic framework as an efficient catalyst for ultrasonic-assisted Knoevenagel condensation.

Project description:1-Butyl-3-vinylimidazolium chloride was synthesized and polymerized with acrylamide to furnish an ionic liquid-containing polymer, which was then used for the formation of a composite with iron-based metal-organic framework. The resultant composite was characterized with XRD, TGA, FE-SEM, FTIR, EDS and elemental mapping analyses and its catalytic activity was appraised for ultrasonic-assisted Knoevenagel condensation. The results confirmed that the prepared composite could promote the reaction efficiently to furnish the corresponding products in high yields in very short reaction times. Moreover, the composite exhibited high recyclability up to six runs. It was also established that the activity of the composite was higher compared to pristine metal-organic framework or polymer.

Composites of Palladium-Nickel Alloy Nanoparticles and Graphene Oxide for the Knoevenagel Condensation of Aldehydes with Malononitrile.

Project description:Herein, we have described uniformly dispersed palladium-nickel nanoparticles furnished on graphene oxide (GO-supported PdNi nanoparticles) as a powerful heterogeneous nanocatalyst for the promotion of Knoevenagel reaction between malononitrile and aromatic aldehydes under mild reaction conditions. The successful characterization of PdNi nanoparticles on the GO surface was shown by X-ray diffraction, X-ray photoelectron spectroscopy, high-resolution transmission electron microscopy (HR-TEM), and TEM. GO-supported PdNi nanoparticles, which are used as highly efficient, stable, and durable catalysts, were used for the first time for the Knoevenagel condensation reaction. The data obtained here showed that the GO-supported PdNi nanocatalyst had a unique catalytic activity and demonstrated that it could be reused five times without a significant decrease in the catalytic performance. The use of this nanocatalyst results in a very short reaction time under mild reaction conditions, high recyclability, excellent catalytic activity, and a straightforward work-up procedure for Knoevenagel condensation of malononitrile and aromatic aldehydes.

Project description:An efficient, convenient, and eco-friendly biocatalytic approach was developed for the synthesis of quinoline derivatives via the α-chymotrypsin-catalyzed Friedländer reaction. Interestingly, α-chymotrypsin exhibited higher catalytic activity in an ionic liquid (IL) aqueous solution as compared to that observed in our previous relevant study, which was conducted using an organic solvent, and a series of substrates gave similar excellent yields at lower reaction temperature and under reduced enzyme-loading conditions.

Mechanochemical Knoevenagel condensation investigated in situ.

Project description:The mechanochemical Knoevenagel condensation of malononitrile with p-nitrobenzaldehyde was studied in situ using a tandem approach. X-ray diffraction and Raman spectroscopy were combined to yield time-resolved information on the milling process. Under solvent-free conditions, the reaction leads to a quantitative conversion to p-nitrobenzylidenemalononitrile within 50 minutes. The in situ data indicate that the process is fast and proceeds under a direct conversion. After stopping the milling process, the reaction continues until complete conversion. The continuous and the stopped milling process both result in crystalline products suitable for single crystal X-ray diffraction.

DABCO-promoted photocatalytic C-H functionalization of aldehydes.

Project description:Herein we present a direct application of DABCO, an inexpensive and broadly accessible organic base, as a hydrogen atom transfer (HAT) abstractor in a photocatalytic strategy for aldehyde C-H activation. The acyl radicals generated in this step were arylated with aryl bromides through a well stablished nickel cross-coupling methodology, leading to a variety of interesting aryl ketones in good yields. We also performed computational calculations to shine light in the HAT step energetics and determined an optimized geometry for the transition state, showing that the hydrogen atom transfer between aldehydes and DABCO is a mildly endergonic, yet sufficiently fast step. The same calculations were performed with quinuclidine, for comparison of both catalysts and the differences are discussed.

Enantioselective Multicomponent Condensation Reactions of Phenols, Aldehydes, and Boronates Catalyzed by Chiral Biphenols.

Project description:Chiral diols and biphenols catalyze the multicomponent condensation reaction of phenols, aldehydes, and alkenyl or aryl boronates. The condensation products are formed in good yields and enantioselectivities. The reaction proceeds via an initial Friedel-Crafts alkylation of the aldehyde and phenol to yield an ortho-quinone methide that undergoes an enantioselective boronate addition. A cyclization pathway was discovered while exploring the scope of the reaction that provides access to chiral 2,4-diaryl chroman products, the core of which is a structural motif found in natural products.

A facile, rapid procedure for Knoevenagel condensation reaction catalyzed by efficient amino-bifunctional frameworks under mild conditions.

Project description:A series of bifunctional hexagonal MOFs have been successfully constructed by the introduction of various amine functional groups within the unsaturated Cu-based MOF, HKUST, to access amino-modified frameworks. The prepared compounds are cost-effective and display high chemical and thermal stability. They were effectually exploited as efficacious and superb heterogeneous catalysts in rapid and facile Knoevenagel condensation reactions for a variety of substrates containing different electron-donating and electron-withdrawing substituents with very high conversion, good reusability under mild conditions, and very short reaction time. The contaminant presence of Lewis acid and basic sites resulted in efficient condensation reactions by the prepared catalysts.
