The influence of Pseudomonas syringae on water freezing and ice melting.

ABSTRACT: Pseudomonas syringae is a widely spread plant pathogen known to have ice-nucleating proteins that serve as crystallization sites promoting ice growth at near-zero temperatures. Three temperatures that characterize water freezing and ice melting are (i) the freezing point of water, (ii) the temperature of coexistence of ice and water, and (iii) the melting point of ice. Here we show the influence of different concentrations of P. syringae on these three parameters. P. syringae appears to affect both the freezing point of water and the temperature of the coexistence of ice and water. Additionally, we propose a research technique for studying the freezing/melting process that is simple and requires no complex equipment.

High-speed cryo-microscopy reveals that ice-nucleating proteins of <i>Pseudomonas syringae</i> trigger freezing at hydrophobic interfaces.

Project description:Ice-nucleating proteins (INpro) trigger the freezing of supercooled water droplets relevant to atmospheric, biological, and technological applications. The high ice nucleation activity of INpro isolated from the bacteria Pseudomonas syringae could be linked to the aggregation of proteins at the bacterial membrane or at the air-water interface (AWI) of droplets. Here, we imaged freezing onsets, providing direct evidence of these proposed mechanisms. High-speed cryo-microscopy identified the onset location of freezing in droplets between two protein-repellent glass slides. INpro from sterilized P. syringae (Snomax) statistically favored nucleation at the AWI of the droplets. Removing cellular fragments by filtration or adding surfactants increased the frequency of nucleation events at the AWI. On the other hand, cultivated intact bacteria cells or lipid-free droplets nucleated ice without an affinity to the AWI. Overall, we provide visual evidence that INpro from P. syringae trigger freezing at hydrophobic interfaces, such as the AWI or the bacterial membrane, with important mechanistic implications for applications of INpro.

Direct observations of melting, freezing, and ocean circulation in an ice shelf basal crevasse.

Project description:Ocean conditions near the grounding zones of Antarctica's ice shelves play a key role in controlling the outflow and mass balance of the ice sheet. However, ocean observations in these regions are largely absent. Here, we present a detailed spatial survey collected with an underwater vehicle in a basal crevasse located in the ocean cavity at the Ross Ice Shelf grounding zone. The observations depict fine-scale variability in ocean forcing that drives asymmetric melting along the lower crevasse sidewalls and freezing in the upper reaches of the crevasse. Freshwater release from melting at depth and salt rejection from freezing above drives an overturning circulation. This vertical circulation pattern overlays a dominant throughflow jet, which funnels water parallel to the coastline, orthogonal to the direction of tidal currents. Importantly, these data reveal that basal crevasses influence ocean circulation and mixing at ice shelf grounding zones to an extent previously unknown.

A sudden-melting event during water freezing inside a copper well.

Project description:We studied the freezing of super-cooled water inside a millimeter-sized copper well by confocal microscopy. During freezing, we surprisingly observed a novel melting scenario, which we call a 'sudden-melting event': the ice directly above the bottom substrate suddenly melts in the late stage of the freezing process, while the system is continuously being cooled. After this event, an empty gap around 10 μm to 20 μm between the substrate and the bulk ice is formed. Because this gap occupies the majority of the area of the bottom substrate, the adhesion between the bulk ice and the substrate is greatly reduced: the adhesion force decreases by more than 50% compared with the flat-substrate situation. We further discovered that air dissolved in water plays a crucial role in this melting event: the air excluded by water freezing produces inter-connecting channels in the bulk ice, which transport the warm water produced by latent heat to the substrate which causes the sudden melting event. Because this event makes the contact between ice and substrate very poor, and greatly reduces ice adhesion, our observation may lead to a promising anti-icing method on solid substrates. Compared to the prevalent super-hydrophobic surface technique, our approach only requires millimeter-sized wells instead of complex microscopic textures. Therefore, it is much easier and cheaper to produce, as well as much more robust for large-scale practical applications.

Modeling Pseudomonas syringae ice-nucleation protein as a beta-helical protein.

Project description:Antifreeze proteins (AFPs) inhibit the growth of ice, whereas ice-nucleation proteins (INPs) promote its formation. Although the structures of several AFPs are known, the structure of INP has been modeled thus far because of the difficulty in determining membrane protein structures. Here, we present a novel model of an INP structure from Pseudomonas syringae based on comparison with two newly determined insect AFP structures. The results suggest that both this class of AFPs and INPs may have a similar beta-helical fold and that they could interact with water through the repetitive TXT motif. By theoretical arguments, we show that the distinguishing feature between an ice inhibitor and an ice nucleator lies in the size of the ice-interacting surface. For INPs, the larger surface area acts as a template that is larger than the critical ice embryo surface area required for growth. In contrast, AFPs are small enough so that they bind to ice and inhibit further growth without acting as a nucleator.

Liquid water on cold exo-Earths via basal melting of ice sheets.

Project description:Liquid water is a critical component of habitability. However, the production and stability of surficial liquid water can be challenging on planets outside the Habitable Zone and devoid of adequate greenhouse warming. On such cold, icy exo-Earths, basal melting of regional/global ice sheets by geothermal heat provides an alternative means of forming liquid water. Here, we model the thermophysical evolution of ice sheets to ascertain the geophysical conditions that allow liquid water to be produced and maintained at temperatures above the pressure-controlled freezing point of water ice on exo-Earths. We show that even with a modest, Moon-like geothermal heat flow, subglacial oceans of liquid water can form at the base of and within the ice sheets on exo-Earths. Furthermore, subglacial oceans may persist on exo-Earths for a prolonged period due to the billion-year half-lives of heat-producing elements responsible for geothermal heat. These subglacial oceans, often in contact with the planet's crust and shielded from the high energy radiation of their parent star by thick ice layers, may provide habitable conditions for an extended period.

Thermodynamics of freezing and melting.

Project description:Although the freezing of liquids and melting of crystals are fundamental for many areas of the sciences, even simple properties like the temperature-pressure relation along the melting line cannot be predicted today. Here we present a theory in which properties of the coexisting crystal and liquid phases at a single thermodynamic state point provide the basis for calculating the pressure, density and entropy of fusion as functions of temperature along the melting line, as well as the variation along this line of the reduced crystalline vibrational mean-square displacement (the Lindemann ratio), and the liquid's diffusion constant and viscosity. The framework developed, which applies for the sizable class of systems characterized by hidden scale invariance, is validated by computer simulations of the standard 12-6 Lennard-Jones system.

Metastable water at several compression rates and its freezing kinetics into ice VII.

Project description:Water can be dynamically over-compressed well into the stability field of ice VII. Whether water then transforms into ice VII, vitreous ice or a metastable novel crystalline phase remained uncertain. We report here the freezing of over-compressed water to ice VII by time-resolved X-ray diffraction. Quasi-isothermal dynamic compression paths are achieved using a dynamic-piezo-Diamond-Anvil-Cell, with programmable pressure rise time from 0.1 ms to 100 ms. By combining the present data set with those obtained on various ns-dynamical platforms, a complete evolution of the solidification pressure of metastable water versus the compression rate is rationalized within the classical nucleation theory framework. Also, when crystallization into ice VII occurs in between 1.6 GPa and 2.0 GPa, that is in the stability field of ice VI, a structural evolution over few ms is then observed into a mixture of ice VI and ice VII that seems to resolve apparent contradictions between previous results.

Methane excess in Arctic surface water-triggered by sea ice formation and melting.

Project description:Arctic amplification of global warming has led to increased summer sea ice retreat, which influences gas exchange between the Arctic Ocean and the atmosphere where sea ice previously acted as a physical barrier. Indeed, recently observed enhanced atmospheric methane concentrations in Arctic regions with fractional sea-ice cover point to unexpected feedbacks in cycling of methane. We report on methane excess in sea ice-influenced water masses in the interior Arctic Ocean and provide evidence that sea ice is a potential source. We show that methane release from sea ice into the ocean occurs via brine drainage during freezing and melting i.e. in winter and spring. In summer under a fractional sea ice cover, reduced turbulence restricts gas transfer, then seawater acts as buffer in which methane remains entrained. However, in autumn and winter surface convection initiates pronounced efflux of methane from the ice covered ocean to the atmosphere. Our results demonstrate that sea ice-sourced methane cycles seasonally between sea ice, sea-ice-influenced seawater and the atmosphere, while the deeper ocean remains decoupled. Freshening due to summer sea ice retreat will enhance this decoupling, which restricts the capacity of the deeper Arctic Ocean to act as a sink for this greenhouse gas.

Survival and ice nucleation activity of Pseudomonas syringae strains exposed to simulated high-altitude atmospheric conditions.

Project description:Pseudomonas syringae produces highly efficient biological ice nuclei (IN) that were proposed to influence precipitation by freezing water in clouds. This bacterium may be capable of dispersing through the atmosphere, having been reported in rain, snow, and cloud water samples. This study assesses its survival and maintenance of IN activity under stressing conditions present at high altitudes, such as UV radiation within clouds. Strains of the pathovars syringae and garcae were compared to Escherichia coli. While UV-C effectively inactivated these cells, the Pseudomonas were much more tolerant to UV-B. The P. syringae strains were also more resistant to radiation from a solar simulator, composed of UV-A and UV-B, while only one of them suffered a decline in IN activity at -5 °C after long exposures. Desiccation at different relative humidity values also affected the IN, but some activity at -5 °C was always maintained. The pathovar garcae tended to be more resistant than the pathovar syringae, particularly to desiccation, though its IN were found to be generally more sensitive. Compared to E. coli, the P. syringae strains appear to be better adapted to survival under conditions present at high altitudes and in clouds.
