Investigating cell autonomy in microorganisms.

ABSTRACT: Cell-cell signaling in microorganisms is still poorly characterized. In this Methods paper, we describe a genetic procedure for detecting cell-nonautonomous genetic effects, and in particular cell-cell signaling, termed the chimeric colony assay (CCA). The CCA measures the effect of a gene on a biological response in a neighboring cell. This assay can measure cell autonomy for range of biological activities including transcript or protein accumulation, subcellular localization, and cell differentiation. To date, the CCA has been used exclusively to investigate colony patterning in the budding yeast Saccharomyces cerevisiae. To demonstrate the wider potential of the assay, we applied this assay to two other systems: the effect of Grr1 on glucose repression of GAL1 transcription in yeast and the effect of rpsL on stop-codon translational readthrough in Escherichia coli. We also describe variations of the standard CCA that address specific aspects of cell-cell signaling, and we delineate essential controls for this assay. Finally, we discuss complementary approaches to the CCA. Taken together, this Methods paper demonstrates how genetic assays can reveal and explore the roles of cell-cell signaling in microbial processes.

Project description:Cell adhesion molecules (CAMs) provide identifying cues by which neural architecture is sculpted. The Down Syndrome Cell Adhesion Molecule (DSCAM) is required for many neurodevelopmental processes in different species and also has several potential mechanisms of activity, including homophilic adhesion, homophilic repulsion and heterophilic interactions. In the mouse retina, Dscam is expressed in many, but not all neuronal subtypes. Mutations in Dscam cause the fasciculation of dendrites of neighboring homotypic neurons, indicating a role in self-avoidance among cells of a given type, a disruption of the non-random patterning of their cell bodies, and a decrease in developmental cell death in affected cell populations. In order to address how DSCAM facilitates retinal pattering, we developed a conditional allele of Dscam to use alongside existing Dscam mutant mouse strains. Conditional deletion of Dscam reproduces cell spacing, cell number and dendrite arborization defects. Inducible deletion of Dscam and retinal ganglion cell depletion in Brn3b mutant retinas both indicate that these DSCAM-mediated phenotypes can occur independently. In chimeric retinas, in which wild type and Dscam mutant cells are comingled, Dscam mutant cells entangle adjacent wild type cells of the same type, as if both cells were lacking Dscam, consistent with DSCAM-dependent cell spacing and neurite arborization being mediated through homophilic binding cell-to-cell. Deletion of Dscam in specific cell types causes cell-type-autonomous cell body spacing defects, indicating that DSCAM mediates arborization and spacing by acting within given cell types. We also examine the cell autonomy of DSCAM in laminar stratification and find that laminar disorganization can be caused in a non-cell autonomous fashion. Finally, we find Dscam dosage-dependent defects in developmental cell death and amacrine cell spacing, relevant to the increased cell death and other disorders observed in Down syndrome mouse models and human patients, in which Dscam is present in three copies.

Project description:Our eating behaviors are highly influenced by those of individuals surrounding us and the groups we belong to. The first goal of this experiment was to determine how social norms that encourage (pro-) vs. discourage (anti-) unhealthy eating influence people’s intentions and motivations to eat unhealthily. Since these norms can be conveyed by one’s group in a manner that either promotes group members’ autonomy (i.e., collective autonomy support), or pressures them into eating certain foods (i.e., collective control), the experiment also tests which of these types of messages promotes the highest conformity to group norms. Hence, the second goal of this experiment was to investigate this synergetic effect of pro- vs anti-unhealthy eating norms and of collective autonomy support vs. collective control on participants’ unhealthy eating intentions and their motivations for unhealthy eating. An experimental study (N = 341) using a 2 (eating norm: pro-unhealthy eating norm vs. anti-unhealthy eating norm) x 3 (type of group support: collective autonomy support vs. collective control vs. no support) design was conducted. Results showed that pro-unhealthy eating norms increased participants’ intentions to eat salty and fatty food, but also their amotivation (i.e., lack of motivation) for unhealthy eating relative to anti-unhealthy eating norms. In addition, when pro-unhealthy eating was encouraged in a controlling (vs. in an autonomy supportive) manner, participants reported higher intentions to eat tofu tacos. Finally, when pro-unhealthy eating was promoted by supporting group members’ autonomy, participants reported higher integrated regulation, i.e., a highly internalized motivation, for unhealthy eating. These results demonstrate that eating norms do not impact all types of unhealthy food consumption in the same manner, and that collective control may be motivating in uncertain contexts; furthermore, when individuals’ autonomy is supported and promoted by other group members, they are more susceptible to integrate unhealthy eating in their life.

Project description:The morphogenetic process of germ band retraction in Drosophila embryos involves coordinated movements of two epithelial tissues - germ band and amnioserosa. The germ band shortens along its rostral-caudal or head-to-tail axis, widens along its perpendicular dorsal-ventral axis, and uncurls from an initial 'U' shape. The amnioserosa mechanically assists this process by pulling on the crook of the U-shaped germ band. The amnioserosa may also provide biochemical signals that drive germ band cells to change shape in a mechanically autonomous fashion. Here, we use a finite-element model to investigate how these two contributions reshape the germ band. We do so by modeling the response to laser-induced wounds in each of the germ band's spatially distinct segments (T1-T3, A1-A9) during the middle of retraction when segments T1-A3 form the ventral arm of the 'U', A4-A7 form its crook, and A8-A9 complete the dorsal arm. We explore these responses under a range of externally applied stresses and internal anisotropy of cell edge tensions - akin to a planar cell polarity that can drive elongation of cells in a direction parallel to the minimum edge tension - and identify regions of parameter space (edge-tension anisotropy versus stress anisotropy) that best match previous experiments for each germ band segment. All but three germ band segments are best fit when the applied stress anisotropy and the edge-tension anisotropy work against one another - i.e., when the isolated effects would elongate cells in perpendicular directions. Segments in the crook of the germ band (A4-A7) have cells that elongate in the direction of maximum external stress, i.e., external stress anisotropy is dominant. In most other segments, the dominant factor is internal edge-tension anisotropy. These results are consistent with models in which the amnioserosa pulls on the crook of the germ band to mechanically assist retraction. In addition, they suggest a mechanical cue for edge-tension anisotropy whereby cells do not globally orient their internal elongation axis towards the amnioserosa, but instead orient this axis perpendicular to the local principal stress direction.

Project description:BackgroundIn the process of education reform, carrying out continuous educational environment measurement is necessary. This study aimed to evaluate the perceptions of the medical education environment and learning autonomy in blended learning in pediatrics among clinical medicine students, as well as the relationship between these two constructs.MethodsA cross-sectional study was conducted on clinical medical students from the First Clinical School of Wuhan University who had completed the blended learning courses of pediatrics. Perceptions of the medical education environment and learning autonomy were surveyed using the Dundee Ready Education Environment Measure (DREEM) and the Autonomy in Learning Rating Scale for college students (ALRS), respectively. Independent samples t-test, one-way ANOVA, correlation, and multiple linear regression analyses were conducted.ResultsA total of 487 students participated in the survey (valid response rate: 83.53%). Students' perceptions of the current medical education environment and learning autonomy were generally positive. Among the average scoring rates of DREEM, students' perception of teachers (SPT) > students' perception of learning (SPL) > students' perception of atmosphere (SPA) > students' academic self-perceptions (SASP) > students' social self-perception (SSSP), while learning strategies > learning motivation > self-regulation > content and environment among the average scoring rates of ALRS. Correlation analyses revealed that all subscales of the two were significantly associated with each other (P < 0.05). Regression analyses showed that SPA and SASP were significant factors for students' learning autonomy.ConclusionClinical medicine students who experienced blended learning in pediatrics had positive perceptions of the medical education environment, and their learning autonomy was good and closely related to the perceptions of the medical education environment. In the implementation of blended learning, continuous optimization of teaching content and proactive enhancement of the medical education environment will help enhance clinical medicine students' satisfaction with the educational environment and their learning autonomy.Clinical trial numberNot applicable.

Project description:Background The proportion of women surgeons is increasing, but studies show that women in surgical residency are granted less autonomy than men. Objective We utilized the Surgical Autonomy Program (SAP), an educational framework, to evaluate gender differences in self-reported autonomy, attending-reported autonomy, and operative feedback among US neurosurgical residents. Methods The SAP tracks resident progression and guides teaching in neurosurgery. Surgeries are divided into zones of proximal development (opening, exposure, critical portion, and closure). Postoperatively, resident autonomy is rated on a 4-point scale by the resident and the attending for each part of the case, or zone. We utilized data from July 2017 to February 2024 from 8 institutions. Ordinal regression was used to evaluate the odds of self- and attending-evaluated autonomy, accounting for gender, training year, case difficulty, and institution. Differences between attending assessment and self-assessment were calculated across time. Chi-square analyses were used to measure any differences in feedback given to men and women. Results From 128 residents (32 women, 25%), 11894 cases were included. Women were granted less autonomy (OR 0.81; 95% CI 0.74-0.89; P<.001) and self-evaluated as having less autonomy (OR 0.73; 95% CI 0.67-0.80; P<.001). The odds of women operating at higher autonomy were similar to the odds of operating on a hard case compared to average difficulty (OR 0.77; 95% CI 0.71-0.84; P<.001). Men's and women's self-assessment became closer to attending assessment over time, with women improving more quickly for the critical portions of surgeries. Women residents received meaningful postoperative feedback on fewer cases (women: 74.2%, men: 80.5%; X2=31.929; P<.001). Conclusions Women operated with lower autonomy by both attending and self-assessment, but the assessment gap between genders decreased over time. Women also received less feedback from their attendings.
