The complete mitochondrial genome of Ornithomya biloba (Diptera, Hippoboscidae).

ABSTRACT: The mitochondrial genome (mitogenome) of Ornithomya biloba (Dufour 1827) was first sequenced and annotated in this study as the first representative of the genus Ornithomya. The complete mitogenome is 18,654 bp in length and contains 37 genes (13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and control region). The phylogenetic analysis based on 13 PCGs in IQ-TREE supports the monophyly of Hippoboscidae, which was a sister group of Streblidae. Families Hippoboscidae and Streblidae formed the monophyletic Hippoboscoidea clade.

Project description:The complete mitochondrial genome (15,573 bp) of an understudied sheep parasite Melophagus ovinus was sequenced and characterized. Its organization and characteristics, including the size, structure, gene order, start/stop codon usage and gene overlaps, are largely typical for Diptera. It exhibits very high A + T bias (81%). Posterior probability values in the inferred phylogenetic dendrogram were very high, but Oestroidea and Muscoidea superfamilies were both paraphyletic. The sequence was nested within the Oestridae clade, thus also rendering the family paraphyletic. A larger number of Hippoboscoidea mitogenomes will have to be available to achieve a better phylogenetic resolution.

Complete mitochondrial genome of a potential vector louse fly, Lipoptena grahami (Diptera, Hippoboscidae).

Project description:Lipoptena grahami Bequaert, 1942 (Diptera, Hippoboscidae) was first described in China almost 80 years ago. Species of Lipoptena were obligate blooding-feeding insects and commonly reported as vectors of wild animals of Cervinae. The complete mitochondrial genome of L. grahami was assembled to 16,953 bp in length. The AT content of L. grahami mitogenome is 80.59%. In total, 22 tRNAs, 2 rRNAs, and 13 protein-coding genes (PCGs) were annotated from L. grahami's mitogenome. The typical clover-leaf structure of tRNAs was also analyzed and confirmed except the tRNA-Ser (AGN). A phylogenetic tree was constructed based on L. grahami with some other fly species.

Project description:The complete mitochondrial genome of Limnophyes minimus (Meigen 1818) was sequenced and annotated, and its general features and base composition were analyzed. The phylogenetic relationships of the families Chironomidae, Simuliidae, Sciaridae and Culicidae based on 25 metagenomes were reconstructed using maximum likelihood (ML) methods based on the concatenated nucleotide sequences, the phylogenetic analysis showed that L. minimus belongs family Chironomidae, which is consistent with the traditional morphological classification.

The complete mitochondrial genome of Graphomya rufitibia (Diptera: Muscidae).

Project description:Graphomya rufitibia (Diptera: Muscidae) was distributed worldwide. In this study, the complete mitogenome of G. rufitibia was sequenced and annotated, and the full-length was a 15,374 bp fragment, consisting of A (40.64%), G (8.96%), T (36.80%), and C (13.59%). The mitogenome is composed of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and a non-coding AT-rich region. Most PCGs used the canonical putative codon to start and stop, 21 tRNAs are folded into the classic clover-leaf structure, with the one exception of tRNA-Ser (AGN). A total length of 11,182 bp encoding 3727 amino acids, and account for 72.7% of the whole mitogenome. Phylogenetic analyses showed that G. rufitibia belongs to family Mydaeinae.

Project description:Sarcophaga brevicornis (Diptera: Sarcophagidae) was of utmost forensic importance due to their wide distribution, ubiquitous, and synanthropic nature. The complete mitochondrial genome (mitogenome) of S. brevicornis was first sequenced and assembled in this study. The length of circular mitogenome was 15,152 bp, which showed in a typical arthropod genome, including 13 protein-coding genes (13 PCGs), two ribosomal RNA (two rRNA) genes, 22 transfer RNA (22 tRNA) genes, and an AT-rich region. Its nucleotide composition was A 39.0%, C 12.7%, G 10.6%, and T 37.7%. Furthermore, phylogenetic relationships of S. brevicornis and the published sarcophagid species were evaluated based on 13 PCGs. The results indicated that S. brevicornis was clearly separated from the other sarcophagid species, but it was closed to the species of Sarcophaga similis. This study provided a significant database reference for genetic structure and phylogenetic analysis of Sarcophagidae.

The complete mitochondrial genome (mitogenome) of S. pterygota was first sequenced and annotated. The circle DNA is composed of 13 protein-coding genes (13 PCGs), 2 ribosomal RNA (2 rRNAs), 22 transfer RNA (22 tRNAs) and a AT-rich region. It shows that arrangement of the genes is similar with the classical metazoan. The size of mitogenome is 15,236 bp containing A (40.0%), G (9.3%), T (36.6%), and C (14.1%). Moreover, phylogenetic analysis reveals that the branch of S. pterygota is clustered separately. This study enriches the mitogenome database of flesh flies and represents progress for analyzing of phylogenetic relationships.

Project description:The complete mitochondrial genome of Compsilura concinnata (Meigen), was analyzed by next-generation sequencing approach. Its mitogenome is 15,841 bp totally, which consists of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and one non-coding control region. The nucleotide composition biases toward A and T, the overall A + T% was up to 80.9% of the entire mitogenome. Phylogenetic analysis supported the sister relationship between Goniini and Blondeliini. The result also suggested that the monophyly of the Exoristinae.
