Controllable synthesis and stabilization of Tamarix aphylla-mediated copper oxide nanoparticles for the management of Fusarium wilt on musk melon.

ABSTRACT: Excessive use of pesticides and mineral fertilizers poses a serious threat to ecoenvironment sustainability and human health. Nano pesticides or Nano fungicides have attained great attention in the field of agriculture due to their unique characteristics, by improving crop growth with enhancing pathogenesis-related defense system. However, there is a need to develop a sustainable mechanism for the synthesis of fungicides which replace the chemical pesticides to avoid their hazardous impact. Here in, Tamarix aphylla mediated CuO-Nanoparticles (NPs) were synthesized, characterized and their activity was evaluated under in-vitro and in-vivo conditions. The structural and elemental analysis of NPs were carried out by using X-ray powder diffraction (XRD), Fourier transformed infrared spectroscopy (FTIR), UV-visible spectrophotometer, Scanning electron microscope (SEM) and Transmission electron microscope (TEM). In the greenhouse, at an optimum concentration of 50 mg/L reduced disease severity very effectively and enhanced plant growth. Application of NPs also assisted in the induction of systemic response of defense-related genes in melon. Under In vitro condition at 100 mg/L significantly reduced mycelial growth (84.5%) by directly acting on the pathogenic cell wall. Our work confirmed that dosedependent concentration of T. aphylla extract based biological CuO-NPs enhance plant growth and help to effectively resist against F. oxysporum infection.

Project description:Tamarix aphylla is a Saudi herb, which possesses antimicrobial properties and potentially introduces a solution to the subsequent dilemma caused by agrochemicals and antifungal misuse. The current study aimed to assess the fungicidal properties of water and ethanolic extracts of T. aphylla leaves against Macrophomina phaseolina, Curvularia spicifera, and Fusarium spp. The chemical composition of T. aphylla was evaluated by gas chromatography/mass spectrometry technique (GC-MS) and Fourier-transform infrared spectroscopy (FTIR). The antifungal assay assessed the fungal growth inhibition using the poisoned food technique. Scanning and transmission electron microscopy (SEM and TEM) were used to evaluate the structural changes induced in the fungal species post-treatment by T. aphylla. FTIR and GC-MS analysis revealed that T. aphylla extracts were rich in aromatic and volatile compounds, such as Benzeneselenol, Gibberellic acid, and Triaziquone, which proved multiple antifungal properties. The results showed significant inhibition in the growth of all species (p < 0.05) except for F. moniliforme, where the water extract induced the highest mycelial growth inhibition at the dose of 30%. The highest inhibition was for M. phaseolina treated with the water extract (36.25 ± 1.06 mm, p < 0.001) and C. spicifera, treated with the ethanolic extract (27.25 ± 1.77 mm, p < 0.05), as compared to the untreated control and the positive control of Ridomol. SEM and TEM revealed some ultrastructural changes within the fungal growth of treated M. phaseolina, which included the thickening and mild rupture of mycelia. Those findings suggested the robust antifungal properties of T. aphylla against some filamentous fungi. The phenolic composition illustrated the potential fungicidal properties of T. aphylla. Additional studies are required to focus on more antimicrobial properties of T. aphylla against other species, particularly those that might benefit the medical field.

Project description:Disease-suppressive soils are soils in which specific soil-borne plant pathogens cause only limited disease although the pathogen and susceptible host plants are both present. Suppressiveness is in most cases of microbial origin. We conducted a comparative metabarcoding analysis of the taxonomic diversity of fungal and bacterial communities from suppressive and non-suppressive (conducive) soils as regards Fusarium wilts sampled from the Châteaurenard region (France). Bioassays based on Fusarium wilt of flax confirmed that disease incidence was significantly lower in the suppressive soil than in the conducive soil. Furthermore, we succeeded in partly transferring Fusarium wilt-suppressiveness to the conducive soil by mixing 10% (w/w) of the suppressive soil into the conducive soil. Fungal diversity differed significantly between the suppressive and conducive soils. Among dominant fungal operational taxonomic units (OTUs) affiliated to known genera, 17 OTUs were detected exclusively in the suppressive soil. These OTUs were assigned to the Acremonium, Chaetomium, Cladosporium, Clonostachys, Fusarium, Ceratobasidium, Mortierella, Penicillium, Scytalidium, and Verticillium genera. Additionally, the relative abundance of specific members of the bacterial community was significantly higher in the suppressive and mixed soils than in the conducive soil. OTUs found more abundant in Fusarium wilt-suppressive soils were affiliated to the bacterial genera Adhaeribacter, Massilia, Microvirga, Rhizobium, Rhizobacter, Arthrobacter, Amycolatopsis, Rubrobacter, Paenibacillus, Stenotrophomonas, and Geobacter. Several of the fungal and bacterial genera detected exclusively or more abundantly in the Fusarium wilt-suppressive soil included genera known for their activity against F. oxysporum. Overall, this study supports the potential role of known fungal and bacterial genera in Fusarium wilt suppressive soils from Châteaurenard and pinpoints new bacterial and fungal genera for their putative role in Fusarium wilt suppressiveness.
