Miau, a microbalance autosampler.

ABSTRACT: Powder weighing is an essential but tedious activity in many branches of science. Here I describe a MIcrobalance AUtosampler (miau) that transfers solids in the sub-mg range to a microbalance. Miau is a pick-and-place machine which moves a gripper with dual function: 1) move tin capsules; 2) deliver powder from a container to tin capsules. In our laboratory we routinely use miau to prepare working standards for quality control of elemental and isotopic analyses. In a test, miau produced standards between 0.3 and 1.1 mg, which is a useful range in our laboratory. Failure to produce a weighed standard happened in 5% of the cases. A comparison with manual measurements demonstrated that obtained amounts for automated samples were as accurate and precise as manually prepared ones. Setup for daily use is simple, and the microbalance can be easily used alternately with or without miau. Miau is a low-cost device that can work with microbalances from many manufacturers, and can be readily adopted by many laboratories.

Low-cost composite autosampler for wastewater sampling.

Project description:Wastewater monitoring is a proven method for detecting viruses, illicit drug consumption, pharmaceutical use/abuse, water pollution, and the occurrence of antimicrobial resistance genes. Analyzing sewage water for pathogens like the SARS-Cov-2 virus could enable early detection of infected areas or virus variants. We developed a low-cost (∼US$1000), open-source, composite automatic water sampler with operational flexibility and simple data logging. Sample quantity is measured and cut off appropriately by continuously weighing the incoming liquid. Our system is customizable in both its hardware and firmware (via options and code modification) which allows for the device to be tailored easily to each application. For example, the user can input sampling quantities from 10 g per sample every 1.5 min to a single 2900 g sample after up to a 16-day delay prior to sampling. The outer diameter and height of the sampler are less than 0.5 m to fit in human-entry openings. We present experimental data for sample sizes of 10 g and 100 g at suction heights between 0.65 and 4.75 m.

Portable open-source autosampler for shallow waters.

Project description:Automated water sampling can be very useful, but open-source choices are limited. Here I present an autosampler which consists of a gantry robot that delivers water from a syringe pump to 24 capped 40 ml vials. The autosampler is controlled using an Android tablet automatized using Macrodroid. Three rinsing cycles ensure negligible carryover between consecutive samples. Hourly sampling from a creek under rainy conditions suggested that total organic carbon in water was diluted by the rain. Some important limitations: 1) the autosampler must be on a steady, flat, horizontal surface; 2) unattended sampling can only last as long as the batteries powering the tablet and the motors; 3) distance from the syringe pump to water cannot exceed ~2 m in height and ~4 m in length for 3 mm tubing; 4) sampling frequency does not exceed one sample every eleven minutes. However, because of its open design, the autosampler can be modified and improved to not only overcome these limitations, but also potentially expand its scope to more demanding sampling if necessary.

micrIO: an open-source autosampler and fraction collector for automated microfluidic input-output.

Project description:Microfluidic devices are an enabling technology for many labs, facilitating a wide range of applications spanning high-throughput encapsulation, molecular separations, and long-term cell culture. In many cases, however, their utility is limited by a 'world-to-chip' barrier that makes it difficult to serially interface samples with these devices. As a result, many researchers are forced to rely on low-throughput, manual approaches for managing device input and output (IO) of samples, reagents, and effluent. Here, we present a hardware-software platform for automated microfluidic IO (micrIO). The platform, which is uniquely compatible with positive-pressure microfluidics, comprises an 'AutoSipper' for input and a 'Fraction Collector' for output. To facilitate widespread adoption, both are open-source builds constructed from components that are readily purchased online or fabricated from included design files. The software control library, written in Python, allows the platform to be integrated with existing experimental setups and to coordinate IO with other functions such as valve actuation and assay imaging. We demonstrate these capabilities by coupling both the AutoSipper and Fraction Collector to two microfluidic devices: a simple, valved inlet manifold and a microfluidic droplet generator that produces beads with distinct spectral codes. Analysis of the collected materials in each case establishes the ability of the platform to draw from and output to specific wells of multiwell plates with negligible cross-contamination between samples.

RotoMate: An open-source, 3D printed autosampler for use with benchtop nuclear magnetic resonance spectrometers.

Project description:Benchtop nuclear magnetic resonance (NMR) spectrometers are versatile analytic instruments with low acquisition and operation cost. However, in the basic version, samples must be manually measured one after the other. We herein describe the open-source autosampler RotoMate that allows the automated operation of such instruments. The hardware is easily assembled from 3D-printed and inexpensive off-the-shelf parts, and is controlled by an Arduino Uno. A software package interlinks the operation of the autosampler with the software of the NMR spectrometer and the software for the processing of the spectra. Experiments for up to 30 samples can be inserted into an interactive sample list. The autosampler automatically inserts and ejects the samples, initiates measurements on the spectrometer according to parameters specified in the sample list, and interacts with a common NMR software in the processing and visualization of the obtained spectroscopic raw data. If an internal standard is present, conversions and yields of chemical reactions are automatically calculated, enabling e.g. the monitoring of reactions. The device was fitted to a Magritek Spinsolve instrument and can interact with a free academic version of ACD NMR software to process the spectra, but can likely be adapted to similar instruments and spectroscopy software packages.

HPLC-Assisted Automated Oligosaccharide Synthesis: Implementation of the Autosampler as a Mode of the Reagent Delivery.

Project description:The development of a useful methodology for simple, scalable, and transformative automation of oligosaccharide synthesis that easily interfaces with existing methods is reported. The automated synthesis can now be performed using accessible equipment where the reactants and reagents are delivered by the pump or the autosampler and the reactions can be monitored by the UV detector. The HPLC-based platform for automation is easy to setup and adapt to different systems and targets.

The Resistance-Amplitude-Frequency Effect of In-Liquid Quartz Crystal Microbalance.

Project description:Due to the influence of liquid load, the equivalent resistance of in-liquid quartz crystal microbalance (QCM) increases sharply, and the quality factor and resonant frequency decreases. We found that the change in the resonant frequency of in-liquid QCM consisted of two parts: besides the frequency changes due to the mass and viscous load (which could be equivalent to motional inductance), the second part of frequency change was caused by the increase of motional resistance. The theoretical calculation and simulation proved that the increases of QCM motional resistance may indeed cause the decreases of resonant frequency, and revealed that the existence of static capacitance was the root cause of this frequency change. The second part of frequency change (due to the increases of motional resistance) was difficult to measure accurately, and may cause great error for in-liquid QCM applications. A technical method to reduce the interference caused by this effect is presented. The study contributes to the accurate determination of the frequency and amplitude change of in-liquid QCM caused by liquid load, which is significant for the QCM applications in the liquid phase.

Quantification of Desiccated Extracellular Vesicles by Quartz Crystal Microbalance.

Project description:Extracellular vesicle (EV) quantification is a procedure through which the biomedical potential of EVs can be used and their biological function can be understood. The number of EVs isolated from cell culture media depends on the cell status and is especially important in studies on cell-to-cell signaling, disease modeling, drug development, etc. Currently, the methods that can be used to quantify isolated EVs are sparse, and each have limitations. In this report, we introduce the application of a quartz crystal microbalance (QCM) as a biosensor for quantifying EVs in a small drop of volatile solvent after it evaporates and leaves desiccated EVs on the surface of the quartz crystal. The shifts in the crystal's resonant frequency were found to obey Sauerbrey's relation for EV quantities up to 6 × 107, and it was determined that the biosensors could resolve samples that differ by at least 2.7 × 105 EVs. A ring-shaped pattern enriched in EVs after the samples had dried on the quartz crystal is also reported and discussed. QCM technology is highly sensitive and only requires small sample volumes and is significantly less costly compared with the approaches that are currently used for EV quantification.

Investigating Adsorbing Viscoelastic Fluids Using the Quartz Crystal Microbalance.

Project description:There is little research on using the quartz crystal microbalance (QCM) with adsorbing viscoelastic fluids. These fluids are widely encountered but often difficult to study as many are opaque and highly viscous. Since the QCM does not involve any scattering or reflection of input radiation, it has the potential to study these complex fluids to determine the relative viscoelasticity of the bulk fluid and surface adsorption of active species onto different substrates. In the current study, both Newtonian (sucrose) and viscoelastic (sodium polystyrene sulfonate (NaPSS)) fluids were introduced into the QCM, and the sensor responses were compared. QCM responses of Newtonian sucrose solutions matched the Kanazawa and Gordon model (KG model), as expected. The QCM responses with viscoelastic NaPSS solutions were well below those described by the KG model. A viscoelastic model was used to determine the fluid viscosity and shear modulus at a very high frequency. It was found that the viscosity of NaPSS did not change much compared with low-frequency rheometer measurements, but a significant increase in the shear modulus of several orders of magnitude was found at the QCM frequencies. Modifying the KG model frequency shifts by multiplying by the QCM shear wave decay length ratio, X = δV/δN, we were able to match the measured QCM values in viscoelastic NaPSS solutions. The QCM dissipation values for NaPSS were matched in a similar way by multiplying the KG model by X 1/3. By changing the QCM sensor from silica (no NaPSS adsorption) to alumina (NaPSS adsorption), it was shown that the adsorption isotherm of NaPSS on alumina could be recovered and fitted with a Langmuir isotherm despite the frequency response being only a small fraction of the total measured QCM signal.

Quartz Crystal Microbalance Platform for SARS-CoV-2 Immuno-Diagnostics.

Project description:Rapid and accurate serological analysis of SARS-CoV-2 antibodies is important for assessing immune protection from vaccination or infection of individuals and for projecting virus spread within a population. The quartz crystal microbalance (QCM) is a label-free flow-based sensor platform that offers an opportunity to detect the binding of a fluid-phase ligand to an immobilized target molecule in real time. A QCM-based assay was developed for the detection of SARS-CoV-2 antibody binding and evaluated for assay reproducibility. The assay was cross-compared to the Roche electrochemiluminescence assay (ECLIA) Elecsys® Anti-SARS-CoV-2 serology test kit and YHLO's chemiluminescence immunoassay (CLIA). The day-to-day reproducibility of the assay had a correlation of r2 = 0.99, p < 0.001. The assay linearity was r2 = 0.96, p < 0.001, for dilution in both serum and buffer. In the cross-comparison analysis of 119 human serum samples, 59 were positive in the Roche, 52 in the YHLO, and 48 in the QCM immunoassay. Despite differences in the detection method and antigen used for antibody capture, there was good coherence between the assays, 80-100% for positive and 96-100% for negative test results. In summation, the QCM-based SARS-CoV-2 IgG immunoassay showed high reproducibility and linearity, along with good coherence with the ELISA-based assays. Still, factors including antibody titer and antigen-binding affinity may differentially affect the various assays' responses.
