The Potential of Sewage Sludge to Predict and Evaluate the Human Chemical Exposome.

ABSTRACT: Chemicals are part of our daily lives, and we are exposed to numerous chemicals through multiple pathways. Relevant scientific evidence contributing to the regulation of hazardous chemicals require a holistic approach to assess simultaneous exposure to multiple compounds. Biomonitoring provides an accurate estimation of exposure to chemicals through very complex and costly sampling campaigns. Finding efficient proxies to predict the risk of chemical exposure in humans is an urgent need to cover large areas and populations at a reasonable cost. We conducted an exploratory study to characterize the human chemical exposome in maternal blood and placenta samples of a population-based birth cohort in Barcelona (2018-2021). Ultimate HRMS-based approaches were applied including wide-scope target, suspect, and nontarget screening. Forty-two chemicals were identified including pesticides, personal care products, or industrial compounds, among others, in the range of ng/mL and ng/g. In parallel, sewage sludge from the wastewater treatment plants serving the residence areas of the studied population were also screened, showing correlations with the type and concentrations of chemicals found in humans. Our findings were suggestive for the potential use of sewage sludge as a proxy of the human exposure and its application in early warning systems to prevent bioaccumulation of hazardous chemicals.

Project description:Sewage sludges generation and their disposal have become one of the greatest challenges of the 21st century. They have great microbial diversity that may impact wastewater treatment plant (WWTP) efficiency and soil quality whether used as fertilizers. Therefore, this research aimed to characterize microbial community diversity and structure of 19 sewage sludges from São Paulo, Brazil, as well as to draw their relations to sludge sources [domestic and mixed (domestic+industrial)], biological treatments (redox conditions and liming), and chemical attributes, using molecular biology as a tool. All sludges revealed high bacterial diversity, but their sources and redox operating conditions as well as liming did not consistently affect bacterial community structures. Proteobacteria was the dominant phylum followed by Bacteroidetes and Firmicutes; whereas Clostridium was the dominant genus followed by Treponema, Propionibacterium, Syntrophus, and Desulfobulbus. The sludge samples could be clustered into six groups (C1 to C6) according their microbial structure similarities. Very high pH (≥11.9) was the main sludge attribute segregating C6, that presented very distinct microbial structure from the others. Its most dominant genera were Propionibacterium > > Comamonas > Brevundimonas > Methylobacterium ∼Stenotrophomonas ∼Cloacibacterium. The other clusters' dominant genera were Clostridium > > Treponema > Desulfobulbus ∼Syntrophus. Moreover, high Fe and S were important modulators of microbial structure in certain sludges undertaking anaerobic treatment and having relatively low N-Kj, B, and P contents (C5). However, high N-Kj, B, P, and low Fe and Al contents were typical of domestic, unlimed, and aerobically treated sludges (C1). In general, heavy metals had little impact on microbial community structure of the sludges. However, our sludges shared a common core of 77 bacteria, mostly Clostridium, Treponema, Syntrophus, and Comamonas. They should dictate microbial functioning within WWTPs, except by SS12 and SS13.

Project description:BackgroundDue to the ubiquitous use of chemicals in modern society, humans are increasingly exposed to thousands of chemicals that contribute to a major portion of the human exposome. Should a comprehensive and risk-based human exposome database be created, it would be conducive to the rapid progress of human exposomics research. In addition, once a xenobiotic is biotransformed with distinct half-lives upon exposure, monitoring the parent compounds alone may not reflect the actual human exposure. To address these questions, a comprehensive and risk-prioritized human exposome database is needed.ObjectivesOur objective was to set up a comprehensive risk-prioritized human exposome database including physicochemical properties as well as risk prediction and develop a graphical user interface (GUI) that has the ability to conduct searches for content associated with chemicals in our database.MethodsWe built a comprehensive risk-prioritized human exposome database by text mining and database fusion. Subsequently, chemicals were prioritized by integrating exposure level obtained from the Systematic Empirical Evaluation of Models with toxicity data predicted by the Toxicity Estimation Software Tool and the Toxicological Priority Index calculated from the ToxCast database. The biotransformation half-lives (HLBs) of all the chemicals were assessed using the Iterative Fragment Selection approach and biotransformation products were predicted using the previously developed BioTransformer machine-learning method.ResultsWe compiled a human exposome database of >20,000 chemicals, prioritized 13,441 chemicals based on probabilistic hazard quotient and 7,770 chemicals based on risk index, and provided a predicted biotransformation metabolite database of >95,000 metabolites. In addition, a user-interactive Java software (Oracle)-based search GUI was generated to enable open access to this new resource.DiscussionOur database can be used to guide chemical management and enhance scientific understanding to rapidly and effectively prioritize chemicals for comprehensive biomonitoring in epidemiological investigations. https://doi.org/10.1289/EHP7722.

Project description:Sewage sludge and livestock (chicken, swine and cattle) manure samples were collected from the Yanmenguan Cattle Herbivorous Livestock Area to compare the potential ecological and human health risks caused by heavy metals contained in them. In this study, the Class II level of Quality Control of Imported Organic Fertilizers is selected as the limit standard value of heavy metals. Based on the mean content values, no heavy metal in cattle manure was higher than the limit standard value; the content of Cu in swine manure was higher than the limit of Cu; the content of Zn in sewage sludge, chicken manure and swine manure were all higher than the limit of Zn; and the content of Cr in sewage sludge and chicken manure were all higher than the limit of Cr. Results indicated that sewage sludge and livestock manure all had high contents of Zn, Cu and Cr. The mean pollution index (PI) suggested that Cu, Zn, As and Cr in sewage sludge and livestock manures all induced potential ecological risks. According to the mean Nemerow's synthetic pollution index (PN) values, swine manure had the highest potential ecological risk for agricultural use. Daily exposure to Cu, Zn and Cr was higher than other heavy metals from sewage sludge and livestock manures, and heavy metal exposure was always higher for children than adults, with ingestion as the main pathway. Non-carcinogenic risk was caused mainly by Cu and Cr, based on the higher hazard quotient (HQ) values for adults and children. There was no non-carcinogenic risk for all people, except exposure of Cu from swine manure for children, which was 1.76 times higher than the threshold value of 1. According to the mean hazard index (HI) values, only swine manure had a non-carcinogenic risk for children. As the carcinogenic risk index (Risk) values were continuously greater for As than Cd, As had a higher carcinogenic risk than Cd. There was no carcinogenic risk for any single heavy metal, although As exposure from sewage sludge was found to have an inapparent carcinogenic risk for both adults and children. Regarding the RISK value, sewage sludge had an unacceptable carcinogenic risk for adults and children, and swine manure had an unacceptable risk for children only. In general, for both non-carcinogenic and carcinogenic risks, ingestion was the main pathway, and children were more sensitive than adults. Comparing the four kinds of organic waste, cattle manure was the safest for agricultural use in terms of ecological and human health risks. In multiple comparisons, swine manure was significantly different regarding potential ecological risk and non-carcinogenic risk, and sewage sludge was significantly different regarding carcinogenic risk.
