First Trifluoromethylated Phenanthrolinediamides: Synthesis, Structure, Stereodynamics and Complexation with Ln(III).

ABSTRACT: The first examples of 1,10-phenanthroline-2,9-diamides bearing CF₃-groups on the side amide substituents were synthesized. Due to stereoisomerism and amide rotation, such complexes have complicated behavior in solutions. Using advanced NMR techniques and X-ray analysis, their structures were completely elucidated. The possibility of the formation of complex compounds with lanthanoids nitrates was shown, and the constants of their stability are quantified. The results obtained are explained in terms of quantum-chemical calculations.

Synthesis of Trifluoromethylated Monoterpene Amino Alcohols.

Project description:For the first time, monoterpene trifluoromethylated β-hydroxy-benzyl-O-oximes were synthesized in 81-95% yields by nucleophilic addition of the Ruppert-Prakash reagent (TMSCF3) to the corresponding β-keto-benzyl-O-oximes based on (+)-nopinone, (-)-verbanone and (+)-camphoroquinone. Trifluoromethylation has been determined to entirely proceed chemo- and stereoselective at the C=O rather than C=N bond. Trifluoromethylated benzyl-O-oximes were reduced to the corresponding α-trifluoromethyl-β-amino alcohols in 82-88% yields. The structure and configuration of the compounds obtained have been established.

Heterometallic Octanuclear NiII 4LnIII 4 (Ln = Y, Gd, Tb, Dy, Ho, Er) Complexes Containing NiII 2LnIII 2O4 Distorted Cubane Motifs: Synthesis, Structure, and Magnetic Properties.

Project description:The reaction of 2-methoxy-6-[{2-(2-hydroxyethylamino)ethylimino}methyl] phenol (LH3) with lanthanide metal salts followed by the addition of nickel acetate allowed isolation of a family of octanuclear complexes, [Ni4Ln4(μ2-OH)2(μ3-OH)4(μ-OOCCH3)8(LH2)4]·(OH)2·xH2O. Single crystal X-ray diffraction studies of these complexes reveal that their central metallic core consists of two tetranuclear [Ni2Ln2O4] cubane subunits fused together by acetate and hydroxide bridges. The magnetic study of these complexes reveals a ferromagnetic interaction between the LnIII and the NiII center. The magnitude of exchange coupling between the NiII and LnIII center, parametrized from the magnetic data of the Gd analogue, gives J = +0.86 cm-1. The magneto caloric effect, studied for the NiII 4GdIII 4 complex, shows a maximum of magnetic entropy change, -ΔS m = 22.58 J kg-1 K-1 at 3 K for an applied external field of 5 T.

Molecular Ln(III)-H-E(II) Linkages (Ln=Y, Lu; E=Ge, Sn, Pb).

Project description:Following the alkane-elimination route, the reaction between tetravalent aryl tintrihydride Ar*SnH3 and trivalent rare-earth-metallocene alkyls [Cp*2 Ln(CH{SiMe3 }2 )] gave complexes [Cp*2 Ln(μ-H)2 SnAr*] implementing a low-valent tin hydride (Ln=Y, Lu; Ar*=2,6-Trip2 C6 H3 , Trip=2,4,6-triisopropylphenyl). The homologous complexes of germanium and lead, [Cp*2 Ln(μ-H)2 EAr*] (E = Ge, Pb), were accessed via addition of low-valent [(Ar*EH)2 ] to the rare-earth-metal hydrides [(Cp*2 LnH)2 ]. The lead compounds [Cp*2 Ln(μ-H)2 PbAr*] exhibit H/D exchange in reactions with deuterated solvents or dihydrogen.

First Example of Fluorinated Phenanthroline Diamides: Synthesis, Structural Study, and Complexation with Lanthanoids.

Project description:An efficient approach to the synthesis of diamides of 4,7-difluoro-1,10-phenanthroline-2,9-dicarboxylic acid was elaborated. Direct nucleophilic substitution with 4,7-dichloro-1,10-phenanthroline precursors opened access to difluoro derivatives in high yield. As a result, four new fluorinated ligands were prepared in up to 88% yield. Their structure was proved by a combination of spectral methods and X-ray data. A set of lanthanoid complexes was prepared to demonstrate the utility of new ligands. The structure of the complexes was studied in solid state (IR-spectroscopy, X-ray diffraction) and in solution (NMR-spectroscopy).

Structure and bonding patterns in heterometallic organometallics with linear Ln-Pd-Ln motifs.

Project description:Complexes with short intermetallic distances between transition metal fragments and lanthanide (Ln) fragments are fascinating objects of study, owing to the ambiguity of the nature of the interaction. The addition of the divalent lanthanide fragments Cp*2Ln(OEt2) (Ln = Sm or Yb) to a Pd(ii) complex bearing the deprotonated form of the redox-active, non-symmetrical ligand, 2-pyrimidin-2-yl-1H-benzimidazole (Hbimpm), leads to two isostructural complexes, of the general formula (Cp*2Ln)2[μ-Pd(pyridyl)2] (Ln = Sm (4) and Yb (5)). These adducts have interesting features, such as unique linear Ln-Pd-Ln arrangements and short Ln-Pd distances, which deviate from the expected lanthanide contraction. A mixed computational and spectroscopic study into the formation of these adducts gathers important clues as to their formation. At the same time, thorough characterization of these complexes establishes the +3 oxidation state of all the involved Ln centers. Detailed theoretical computations demonstrate that the apparent deviation from lanthanide contraction is not due to any difference in the intermetallic interaction between the Pd and the Ln, but that the fragments are joined together by electrostatic interactions and dispersive forces. This conclusion contrasts with the findings about a third complex, Cp*2Yb(μ-Me)2PdCp* (6), formed during the reaction, which also possesses a short Yb-Pd distance. Studies at the CASSCF level of theory on this complex show several orbitals containing significant interactions between the 4f and 4d manifolds of the metals. This demonstrates the need for methodical and careful analyses in gauging the intermetallic interaction and the inadequacy of empirical metrics in describing such phenomena.

Antiparasitic Behavior of Trifluoromethylated Pyrazole 2-Amino-1,3,4-thiadiazole Hybrids and Their Analogues: Synthesis and Structure-Activity Relationship.

Project description:A series of trifluoromethylated pyrazole thiosemicarbazone, trifluromethylated pyrazole isothiosemicarbazone, and trifluoromethylated pyrazole 2-amino-1,3,4-thiadiazole hybrids were synthesized and evaluated in vitro against the promastigote form of Leishmania amazonensis and the epimastigote form of Trypanosoma cruzi, the pathogens causing the neglected tropical diseases leishmaniasis and Chagas disease, respectively. The results show the potential of these compounds regarding their antiparasitic properties. Studies on the structure-activity relationship demonstrated that compounds containing a bulky group at the para position of the phenyl ring attached to the 5-position of the pyrazole core had better antiparasitic effects. Among the substituents attached at the 3-position of the pyrazole ring, the insertion of the 2-amino-1,3,4-thiadiazole nucleus led to the most potent compounds compared to the thiosemicarbazone derivative.

NMR and TRLFS studies of Ln(iii) and An(iii) C5-BPP complexes.

Project description:C5-BPP is a highly efficient N-donor ligand for the separation of trivalent actinides, An(iii), from trivalent lanthanides, Ln(iii). The molecular origin of the selectivity of C5-BPP and many other N-donor ligands of the BTP-type is still not entirely understood. We present here the first NMR studies on C5-BPP Ln(iii) and An(iii) complexes. C5-BPP is synthesized with 10% 15N labeling and characterized by NMR and LIFDI-MS methods. 15N NMR spectroscopy gives a detailed insight into the bonding of C5-BPP with lanthanides and Am(iii) as a representative for trivalent actinide cations, revealing significant differences in 15N chemical shift for coordinating nitrogen atoms compared to Ln(iii) complexes. The temperature dependence of NMR chemical shifts observed for the Am(iii) complex indicates a weak paramagnetism. This as well as the observed large chemical shift for coordinating nitrogen atoms show that metal-ligand bonding in Am(C5-BPP)3 has a larger share of covalence than in lanthanide complexes, confirming earlier studies. The Am(C5-BPP)3 NMR sample is furthermore spiked with Cm(iii) and characterized by time-resolved laser fluorescence spectroscopy (TRLFS), yielding important information on the speciation of trace amounts of minor complex species.

Design of A3B-Porphyrin Conjugates with Terpyridine as Potential Theranostic Agents: Synthesis, Complexation with Fe(III), Gd(III), and Photodynamic Activity.

Project description:This paper reports on the design and synthesis of new multifunctional porphyrin-based therapeutic agents for potential therapeutic and diagnostic applications. Zinc complexes of A3B-type meso-arylporphyrins containing OH- and COOH- groups were modified with chelating ligands based on 4'-(4-methylphenyl)-2,2':6',2″-terpyridine derivatives in high yields. Novel complexes with Gd(III), Fe(III) were obtained for these conjugates. Aggregation behaviour in solutions of different solubilisers was studied to inform the selection of the optimal solubilising platform for the porphyrins obtained; their photophysical and photochemical properties were also characterised. Micellar Pluronic F127 formulation was found to be the most effective solubiliser for stabilising the fluorescence-active monomolecular form of the photosensitisers (PS). In vitro cytotoxicity of the compounds was studied on the HEP-2 cell line with and without irradiation for 1.5 and 24 h. As a result, the IC50 of compounds 12 and 14 at an irradiation dose of 8.073 J/cm2 was shown to be 1.87 ± 0.333 and 1.4 ± 0.152 μM, respectively; without irradiation, the compound had no toxic effect within the studied concentration range (1.5 h). A test for the inhibition of metabolic cooperation or promoter activity was also performed for the abovementioned compounds, showing the efficacy and safety of the conjugates obtained. Preliminary data have indicated the high potential of the new type of PS to be promising molecular theranostic agents.

A Biocatalytic Platform for Synthesis of Chiral α-Trifluoromethylated Organoborons.

Project description:There are few biocatalytic transformations that produce fluorine-containing molecules prevalent in modern pharmaceuticals. To expand the scope of biocatalysis for organofluorine synthesis, we have developed an enzymatic platform for highly enantioselective carbene B-H bond insertion to yield versatile α-trifluoromethylated (α-CF3) organoborons, an important class of organofluorine molecules that contain stereogenic centers bearing both CF3 and boron groups. In contrast to current "carbene transferase" enzymes that use a limited set of simple diazo compounds as carbene precursors, this system based on Rhodothermus marinus cytochrome c (Rma cyt c) can accept a broad range of trifluorodiazo alkanes and deliver versatile chiral α-CF3 organoborons with total turnovers up to 2870 and enantiomeric ratios up to 98.5:1.5. Computational modeling reveals that this broad diazo scope is enabled by an active-site environment that directs the alkyl substituent on the heme CF3-carbene intermediate toward the solvent-exposed face, thereby allowing the protein to accommodate diazo compounds with diverse structural features.
