Corpus-based typology: applications, challenges and some solutions.

ABSTRACT: Over the last few years, the number of corpora that can be used for language comparison has dramatically increased. The corpora are so diverse in their structure, size and annotation style, that a novice might not know where to start. The present paper charts this new and changing territory, providing a few landmarks, warning signs and safe paths. Although no corpus at present can replace the traditional type of typological data based on language description in reference grammars, corpora can help with diverse tasks, being particularly well suited for investigating probabilistic and gradient properties of languages and for discovering and interpreting cross-linguistic generalizations based on processing and communicative mechanisms. At the same time, the use of corpora for typological purposes has not only advantages and opportunities, but also numerous challenges. This paper also contains an empirical case study addressing two pertinent problems: the role of text types in language comparison and the problem of the word as a comparative concept.

5G support for Industrial IoT Applications - Challenges, Solutions, and Research gaps.

Project description:Industrial IoT has special communication requirements, including high reliability, low latency, flexibility, and security. These are instinctively provided by the 5G mobile technology, making it a successful candidate for supporting Industrial IoT (IIoT) scenarios. The aim of this paper is to identify current research challenges and solutions in relation to 5G-enabled Industrial IoT, based on the initial requirements and promises of both domains. The methodology of the paper follows the steps of surveying state-of-the art, comparing results to identify further challenges, and drawing conclusions as lessons learned for each research domain. These areas include IIoT applications and their requirements; mobile edge cloud; back-end performance tuning; network function virtualization; and security, blockchains for IIoT, Artificial Intelligence support for 5G, and private campus networks. Beside surveying the current challenges and solutions, the paper aims to provide meaningful comparisons for each of these areas (in relation to 5G-enabled IIoT) to draw conclusions on current research gaps.

Integration of federated learning with IoT for smart cities applications, challenges, and solutions.

Project description:In the past few years, privacy concerns have grown, making the financial models of businesses more vulnerable to attack. In many cases, it is hard to emphasize the importance of monitoring things in real-time with data from Internet of Things (IoT) devices. The people who make the IoT devices and those who use them face big problems when they try to use Artificial Intelligence (AI) techniques in real-world applications, where data must be collected and processed at a central location. Federated learning (FL) has made a decentralized, cooperative AI system that can be used by many IoT apps that use AI. It is possible because it can train AI on IoT devices that are spread out and do not need to share data. FL allows local models to be trained on local data and share their knowledge to improve a global model. Also, shared learning allows models from all over the world to be trained using data from all over the world. This article looks at the IoT in all of its forms, including "smart" businesses, "smart" cities, "smart" transportation, and "smart" healthcare. This study looks at the safety problems that the federated learning with IoT (FL-IoT) area has brought to market. This research is needed to explore because federated learning is a new technique, and a small amount of work is done on challenges faced during integration with IoT. This research also helps in the real world in such applications where encrypted data must be sent from one place to another. Researchers and graduate students are the audience of our article.

Intracochlear PLGA based implants for dexamethasone release: Challenges and solutions.

Project description:The effective treatment of diseases of the inner ear is currently an unmet medical need. Local controlled drug delivery to the cochlea is challenging due to the hidden location, small volume and high sensitivity of this organ. A local intracochlear delivery of drugs would avoid the problems of intratympanic (extracochlear) drug application, but is more invasive. The requirements for such a delivery system include a small size and appropriate flexibility. The delivery device must be rigid enough for surgical handling but also flexible to avoid traumatizing cochlear structures. We developed biodegradable dexamethasone loaded PLGA extrudates for the controlled intracochlear release. In order to achieve the desired flexibility, Polyethylene glycol (PEG) was used as a plasticizer. In addition to the drug release, the extrudates were characterized in vitro by differential scanning calorimetry (DSC) and texture analysis. Simulation of the pharmacokinetics of the inner ear support the expectation that a constant perilymph drug level is obtained after few hours and retained over several weeks. Ex vivo implantation of the extrudates into a guinea pig cochlea indicate that PEG containing extrudates have the desired balance between mechanical strength and flexibility for direct implantation into the cochlea. The location of the implant was visualized by computer tomography. In summary, we postulate that intracochlear administration of drug releasing biodegradable implants is a new and promising approach to achieve local drug delivery to the cochlea for an extended time.

CRISPR-based therapeutics: current challenges and future applications.

Project description:The discovery, only a decade ago, of the genome editing power of clustered regularly interspaced short palindromic repeats (CRISPR)-associated nucleases is already reinventing the therapeutic process, from how new drugs are discovered to novel ways to treat diseases. CRISPR-based screens can aid therapeutic development by quickly identifying a drug's mechanism of action and escape mutants. Additionally, CRISPR-Cas has advanced emerging ex vivo therapeutics, such as cell replacement therapies. However, Cas9 is limited as an in vivo therapeutic due to ineffective delivery, unwanted immune responses, off-target effects, unpredictable repair outcomes, and cellular stress. To address these limitations, controls that inhibit or degrade Cas9, biomolecule-Cas9 conjugates, and base editors have been developed. Herein, we discuss CRISPR-Cas systems that advance both conventional and emerging therapeutics.

Insole-Based Systems for Health Monitoring: Current Solutions and Research Challenges.

Project description:Wearable health monitoring devices allow for measuring physiological parameters without restricting individuals' daily activities, providing information that is reflective of an individual's health and well-being. However, these systems need to be accurate, power-efficient, unobtrusive and simple to use to enable a reliable, convenient, automatic and ubiquitous means of long-term health monitoring. One such system can be embedded in an insole to obtain physiological data from the plantar aspect of the foot that can be analyzed to gain insight into an individual's health. This manuscript provides a comprehensive review of insole-based sensor systems that measure a variety of parameters useful for overall health monitoring, with a focus on insole-based PPD measurement systems developed in recent years. Existing solutions are reviewed, and several open issues are presented and discussed. The concept of a fully integrated insole-based health monitoring system and considerations for future work are described. By developing a system that is capable of measuring parameters such as PPD, gait characteristics, foot temperature and heart rate, a holistic understanding of an individual's health and well-being can be obtained without interrupting day-to-day activities. The proposed device can have a multitude of applications, such as for pathology detection, tracking medical conditions and analyzing gait characteristics.

Challenges and solutions in proteomics.

Project description:The accelerated growth of proteomics data presents both opportunities and challenges. Large-scale proteomic profiling of biological samples such as cells, organelles or biological fluids has led to discovery of numerous key and novel proteins involved in many biological/disease processes including cancers, as well as to the identification of novel disease biomarkers and potential therapeutic targets. While proteomic data analysis has been greatly assisted by the many bioinformatics tools developed in recent years, a careful analysis of the major steps and flow of data in a typical highthroughput analysis reveals a few gaps that still need to be filled to fully realize the value of the data. To facilitate functional and pathway discovery for large-scale proteomic data, we have developed an integrated proteomic expression analysis system, iProXpress, which facilitates protein identification using a comprehensive sequence library and functional interpretation using integrated data. With its modular design, iProXpress complements and can be integrated with other software in a proteomic data analysis pipeline. This novel approach to complex biological questions involves the interrogation of multiple data sources, thereby facilitating hypothesis generation and knowledge discovery from the genomic-scale studies and fostering disease diagnosis and drug development.

Intelligent Biopolymer-Based Films: Promising New Solutions for Food Packaging Applications.

Project description:The development of biopolymer-based films represents a promising direction in the packaging industry that responds to stringent needs for sustainability, reducing the ecological impact. Traditional fossil-derived polymers present major concerns because of their long decomposition time and their significant contribution to the pollution of the environment. On the contrary, biopolymers such as chitosan, PVA, and PLA offer viable alternatives. This study aimed to obtain an innovative pH indicator for smart packaging using a synthetic non-toxic anthocyanin analogue dye incorporated in bio-based films to indicate meat freshness and quality. The pH-responsive color-changing properties of the dye make it suitable for developing intelligent films to monitor food freshness. The obtained polymeric films were characterized by FT-IR and UV-VIS spectroscopy, and their thermal properties were assessed using thermogravimetric methods. Moisture content, swelling capacity, and water solubility of the polymeric films were also evaluated. The sensitivity of the biopolymer-flavylium composite films to pH variations was studied in the pH range of 2 to 12 and noticeable color variations were observed, allowing the monitoring of the meat's quality damage through pH changes. The pH-responsive films were applied directly on the surface or in the proximity of pork and chicken meat samples, to evaluate their colorimetric response to fresh and spoilt meat. This study can be the starting point for creating more durable packaging solutions leading to a circular economy.

Targeting T cell malignancies using CAR-based immunotherapy: challenges and potential solutions.

Project description:Chimeric antigen receptor (CAR) T cell therapy has been successful in treating B cell malignancies in clinical trials; however, fewer studies have evaluated CAR T cell therapy for the treatment of T cell malignancies. There are many challenges in translating this therapy for T cell disease, including fratricide, T cell aplasia, and product contamination. To the best of our knowledge, no tumor-specific antigen has been identified with universal expression on cancerous T cells, hindering CAR T cell therapy for these malignancies. Numerous approaches have been assessed to address each of these challenges, such as (i) disrupting target antigen expression on CAR-modified T cells, (ii) targeting antigens with limited expression on T cells, and (iii) using third party donor cells that are either non-alloreactive or have been genome edited at the T cell receptor α constant (TRAC) locus. In this review, we discuss CAR approaches that have been explored both in preclinical and clinical studies targeting T cell antigens, as well as examine other potential strategies that can be used to successfully translate this therapy for T cell disease.

Hate speech detection: Challenges and solutions.

Project description:As online content continues to grow, so does the spread of hate speech. We identify and examine challenges faced by online automatic approaches for hate speech detection in text. Among these difficulties are subtleties in language, differing definitions on what constitutes hate speech, and limitations of data availability for training and testing of these systems. Furthermore, many recent approaches suffer from an interpretability problem-that is, it can be difficult to understand why the systems make the decisions that they do. We propose a multi-view SVM approach that achieves near state-of-the-art performance, while being simpler and producing more easily interpretable decisions than neural methods. We also discuss both technical and practical challenges that remain for this task.

Managing eosinophilic esophagitis: challenges and solutions.

Project description:Eosinophilic esophagitis (EoE) is a chronic and progressive immune-mediated condition defined by symptoms of esophageal dysfunction and dense eosinophilic infiltration of the esophageal mucosa. Therapies consist of anti-eosinophilic medications and specialized diets aimed to decrease the progression of EoE and alleviate its symptoms, namely, dysphagia and food impaction. Assessing response to therapy remains challenging, as treatment end points are not well defined and currently consist of clinical, histologic, and endoscopic features. Newer validated measures may help standardize treatment end points. Emerging data support the use of maintenance therapy, which may reduce disease progression. Optimal dosages, delivery techniques, and duration of treatment need to be determined. When features of fibrostenosis develop, esophageal dilation is a safe and effective adjunctive strategy for improving symptoms. In EoE cases refractory to conventional treatments, newer therapies targeting inflammatory mediators and cytokines are on the horizon.
