A comparative analysis of biopolymer production by microbial and bioelectrochemical technologies.

ABSTRACT: Production of biopolymers from renewable carbon sources provides a path towards a circular economy. This review compares several existing and emerging approaches for polyhydroxyalkanoate (PHA) production from soluble organic and gaseous carbon sources and considers technologies based on pure and mixed microbial cultures. While bioplastics are most often produced from soluble sources of organic carbon, the use of carbon dioxide (CO₂) as the carbon source for PHA production is emerging as a sustainable approach that combines CO₂ sequestration with the production of a value-added product. Techno-economic analysis suggests that the emerging approach of CO₂ conversion to carboxylic acids by microbial electrosynthesis followed by microbial PHA production could lead to a novel cost-efficient technology for production of green biopolymers.

Emerging bioelectrochemical technologies for biogas production and upgrading in cascading circular bioenergy systems.

Project description:Biomethane is suggested as an advanced biofuel for the hard-to-abate sectors such as heavy transport. However, future systems that optimize the resource and production of biomethane have yet to be definitively defined. This paper assesses the opportunity of integrating anaerobic digestion (AD) with three emerging bioelectrochemical technologies in a circular cascading bioeconomy, including for power-to-gas AD (P2G-AD), microbial electrolysis cell AD (MEC-AD), and AD microbial electrosynthesis (AD-MES). The mass and energy flow of the three bioelectrochemical systems are compared with the conventional AD amine scrubber system depending on the availability of renewable electricity. An energy balance assessment indicates that P2G-AD, MEC-AD, and AD-MES circular cascading bioelectrochemical systems gain positive energy outputs by using electricity that would have been curtailed or constrained (equivalent to a primary energy factor of zero). This analysis of technological innovation, aids in the design of future cascading circular biosystems to produce sustainable advanced biofuels.

Microbial community analysis of a methane-producing biocathode in a bioelectrochemical system.

Project description:A methane-producing biocathode that converts CO(2) into methane was studied electrochemically and microbiologically. The biocathode produced methane at a maximum rate of 5.1 L CH(4)/m(2) projected cathode per day (1.6 A/m(2)) at -0.7 V versus NHE cathode potential and 3.0 L CH(4)/m(2) projected cathode per day (0.9 A/m(2)) at -0.6 V versus NHE cathode potential. The microbial community at the biocathode was dominated by three phylotypes of Archaea and six phylotypes of bacteria. The Archaeal phylotypes were most closely related to Methanobacterium palustre and Methanobacterium aarhusense. Besides methanogenic Archaea, bacteria seemed to be associated with methane production, producing hydrogen as an intermediate. Biomass density varied greatly with part of the carbon electrode covered with a dense biofilm, while only clusters of cells were found on other parts. Based on our results, we discuss how inoculum enrichment and changing operational conditions may help to increase biomass density and to select for microorganisms that produce methane.

Microbial electricity-driven anaerobic phenol degradation in bioelectrochemical systems.

Project description:Microbial electrochemical technologies have been extensively employed for phenol removal. Yet, previous research has yielded inconsistent results, leaving uncertainties regarding the feasibility of phenol degradation under strictly anaerobic conditions using anodes as sole terminal electron acceptors. In this study, we employed high-performance liquid chromatography and gas chromatography-mass spectrometry to investigate the anaerobic phenol degradation pathway. Our findings provide robust evidence for the purely anaerobic degradation of phenol, as we identified benzoic acid, 4-hydroxybenzoic acid, glutaric acid, and other metabolites of this pathway. Notably, no typical intermediates of the aerobic phenol degradation pathway were detected. One-chamber reactors (+0.4 V vs. SHE) exhibited a phenol removal rate of 3.5 ± 0.2 mg L-1 d-1, while two-chamber reactors showed 3.6 ± 0.1 and 2.6 ± 0.9 mg L-1 d-1 at anode potentials of +0.4 and + 0.2 V, respectively. Our results also suggest that the reactor configuration certainly influenced the microbial community, presumably leading to different ratios of phenol consumers and microorganisms feeding on degradation products.

Microbial reduction of organosulfur compounds at cathodes in bioelectrochemical systems.

Project description:Organosulfur compounds, present in e.g. the pulp and paper industry, biogas and natural gas, need to be removed as they potentially affect human health and harm the environment. The treatment of organosulfur compounds is a challenge, as an economically feasible technology is lacking. In this study, we demonstrate that organosulfur compounds can be degraded to sulfide in bioelectrochemical systems (BESs). Methanethiol, ethanethiol, propanethiol and dimethyl disulfide were supplied separately to the biocathodes of BESs, which were controlled at a constant current density of 2 A/m2 and 4 A/m2. The decrease of methanethiol in the gas phase was correlated to the increase of dissolved sulfide in the liquid phase. A sulfur recovery, as sulfide, of 64% was found over 5 days with an addition of 0.1 mM methanethiol. Sulfur recoveries over 22 days with a total organosulfur compound addition of 1.85 mM were 18% for methanethiol and ethanethiol, 17% for propanethiol and 22% for dimethyl disulfide. No sulfide was formed in electrochemical nor biological control experiments, demonstrating that both current and microorganisms are required for the conversion of organosulfur compounds. This new application of BES for degradation of organosulfur components may unlock alternative strategies for the abatement of anthropogenic organosulfur emissions.

Phenothiazine derivative-accelerated microbial extracellular electron transfer in bioelectrochemical system.

Project description:In bioelectrochemical system (BES) the extracellular electron transfer (EET) from bacteria to anode electrode is recognized as a crucial step that governs the anodic reaction efficiency. Here, we report a novel approach to substantially enhance the microbial EET by immobilization of a small active phenothiazine derivative, methylene blue, on electrode surface. A comparison of the currents generated by Shewanella oneidensis MR-1 and its mutants as well as the electrochemical analytical results reveal that the accelerated EET was attributed to enhanced interactions between the bacterial outer-membrane cytochromes and the immobilized methylene blue. A further investigation into the process using in situ Raman spectro-electrochemical method coupled with density functional theory calculations demonstrates that the electron shuttling was achieved through the change of the molecule conformation of phenothiazine in the redox process. These results offer useful information for engineering BES.

Graphene oxide and H2 production from bioelectrochemical graphite oxidation.

Project description:Graphene oxide (GO) is an emerging material for energy and environmental applications, but it has been primarily produced using chemical processes involving high energy consumption and hazardous chemicals. In this study, we reported a new bioelectrochemical method to produce GO from graphite under ambient conditions without chemical amendments, value-added organic compounds and high rate H2 were also produced. Compared with abiotic electrochemical electrolysis control, the microbial assisted graphite oxidation produced high rate of graphite oxide and graphene oxide (BEGO) sheets, CO2, and current at lower applied voltage. The resultant electrons are transferred to a biocathode, where H2 and organic compounds are produced by microbial reduction of protons and CO2, respectively, a process known as microbial electrosynthesis (MES). Pseudomonas is the dominant population on the anode, while abundant anaerobic solvent-producing bacteria Clostridium carboxidivorans is likely responsible for electrosynthesis on the cathode. Oxygen production through water electrolysis was not detected on the anode due to the presence of facultative and aerobic bacteria as O2 sinkers. This new method provides a sustainable route for producing graphene materials and renewable H2 at low cost, and it may stimulate a new area of research in MES.

Recycling urine for bioelectrochemical hydrogen production using a MoS<sub>2</sub> nano carbon coated electrode in a microbial electrolysis cell.

Project description:In this study, a novel molybdenum disulfide (MoS2) nano-carbon (NC) coated cathode was developed for hydrogen production in a microbial electrolysis cell (MEC), while treating simulated urine with 2-6 times dilution (conductivity <20 mS cm-1). MoS2 nanoparticles were electrodeposited on the NC coated cathodes at -100, -150 and -200 μA cm-2 and their performances were evaluated in the MEC. The chronopotentiometry (CP) tests showed the improved catalytic activity of MoS2-NC cathodes with much lower cathode overpotential than non-MoS2 coated electrodes. The MoS2-NC200 cathode, electrodeposited at -200 μA cm-2, showed the maximum hydrogen production rate of 0.152 ± 0.002 m3 H2 m-2 d-1 at 0.9V of Eap, which is comparable to the previously reported Pt electrodes. It was found that high solution conductivity over 20 mS cm-1 (>600 mg L-1 NH3-N) can adversely affect the biofilm architecture and the bacterial activity at the anode of the MEC. Exoelectrogenic bacteria for this system at the anode were identified as Tissierella (Clostridia) and Bacteroidetes taxa. Maximum ammonia-nitrogen (NH3-N) and phosphorus (PO4 3--P) removal were 68.7 and 98.6%, respectively. This study showed that the newly fabricated MoS2-NC cathode can be a cost-effective alternative to the Pt cathode for renewable bioelectrochemical hydrogen production from urine.

Changes in the microbial consortium during dark hydrogen fermentation in a bioelectrochemical system increases methane production during a two-stage process.

Project description:BackgroundBioelectrochemical systems (BESs) are an innovative technology developed to influence conventional anaerobic digestion. We examined the feasibility of applying a BES to dark hydrogen fermentation and its effects on a two-stage fermentation process comprising hydrogen and methane production. The BES used low-cost, low-reactivity carbon sheets as the cathode and anode, and the cathodic potential was controlled at - 1.0 V (vs. Ag/AgCl) with a potentiostat. The operation used 10 g/L glucose as the major carbon source.ResultsThe electric current density was low throughout (0.30-0.88 A/m2 per electrode corresponding to 0.5-1.5 mM/day of hydrogen production) and water electrolysis was prevented. At a hydraulic retention time of 2 days with a substrate pH of 6.5, the BES decreased gas production (hydrogen and carbon dioxide contents: 52.1 and 47.1%, respectively), compared to the non-bioelectrochemical system (NBES), although they had similar gas compositions. In addition, a methane fermenter (MF) was applied after the BES, which increased gas production (methane and carbon dioxide contents: 85.1 and 14.9%, respectively) compared to the case when the MF was applied after the NBES. Meta 16S rRNA sequencing revealed that the BES accelerated the growth of Ruminococcus sp. and Veillonellaceae sp. and decreased Clostridium sp. and Thermoanaerobacterium sp., resulting in increased propionate and ethanol generation and decreased butyrate generation; however, unknowingly, acetate generation was increased in the BES.ConclusionsThe altered redox potential in the BES likely transformed the structure of the microbial consortium and metabolic pattern to increase methane production and decrease carbon dioxide production in the two-stage process. This study showed the utility of the BES to act on the microbial consortium, resulting in improved gas production from carbohydrate compounds.

Energy generation through bioelectrochemical degradation of pentachlorophenol in microbial fuel cell.

Project description:Bio-electrochemical degradation of pentachlorophenol was carried out in single as well as dual chambered microbial fuel cell (MFC) with simultaneous production of electricity. The maximum cell potential was recorded to be 787 and 1021 mV in single and dual chambered systems respectively. The results presented nearly 66 and 89% COD removal in single and dual chambered systems with corresponding power densities of 872.7 and 1468.85 mW m-2 respectively. The highest coulombic efficiency for single and dual chambered counterparts was found to be 33.9% and 58.55%. GC-MS data revealed that pentachlorophenol was more effectively degraded under aerobic conditions in dual-chambered MFC. Real-time polymerase chain reaction showed the dominance of exoelectrogenic Geobacter in the two reactor systems with a slightly higher concentration in the dual-chambered system. The findings of this work suggested that the aerobic treatment of pentachlorophenol in cathodic compartment of dual chambered MFC is better than its anaerobic treatment in single chambered MFC in terms of chemical oxygen demand (COD) removal and output power density.
