Effects of dandelion addition on antioxidant property, sensory characteristics and inhibitory activity against xanthine oxidase of beer.

ABSTRACT: The effects of dandelion addition (DA) on the physiochemical properties, antioxidant activity, inhibitory activity against xanthine oxidase (XOD) and flavor of craft beer were investigated. It was found that DA changed the pH value, total acid content, thiobarbituric-acid-value, sugar content and color of beer, and increased the contents of total polyphenols and flavonoids and thus the antioxidant activity of beer. HPLC analysis showed that DA provided beer with chlorogenic, caffeic, ferulic, and chicoric acid, contributing to the inhibition activity against XOD that is a key enzyme in uric acid production. GC-MS analysis showed that 3-methyl-1-butanol, isopentyl acetate and ethyl caprylate were main aroma components of all samples. Although DA introduced the special aroma component of azulene, it did not significantly affect the appearance, bubble, aroma and taste evaluation of beer. Conclusively, DA potentially improved the beer properties of antioxidant and inhibition of uric acid production without changing its sensory characteristics.

Project description:Alocasia longiloba, locally known as 'Keladi Candik', has been used traditionally to treat wounds, furuncle and joint inflammations. A. longiloba can be a new source of herbal medicine against hyperuricemia by inhibiting the activity of xanthine oxidase enzyme, the enzyme which is responsible for the development of hyperuricemia in human. Existing xanthine oxidase inhibitors (XOI drugs) show several side effects on gout patients. Therefore, an alternative herbal medicine from plants, with high therapeutic property and free of side effects, are greatly needed. This study was conducted to evaluate XO inhibitory activity, chemical composition, antioxidant activity and GC-MS profile of A. longiloba. Our results showed that ethanolic petiole extract exhibited the highest XO inhibitory activity (70.40 ± 0.05%) with IC50 value of 42.71 μg/mL, followed by ethanolic fruit extracts (61.44 ± 1.24%) with the IC50 value of 51.32 μg/mL. In a parallel study, the phytochemical analysis showed the presence of alkaloid, flavonoid, terpenoids, glycoside and saponin in petiole and fruit extracts, as well as higher total phenolic and flavonoid contents and strong scavenging activity on DPPH and ABTS antioxidant assay. The GC-MS analysis of fruit and petiole extracts revealed the presence of various compounds belonging to different chemical nature, among them are limonen-6-ol, α-DGlucopyranoside, paromomycin, aziridine, phenol, Heptatriacotanol, Phen-1,2,3-dimethyl and Betulin found in ethanolic fruit extract, and Phen-1,4-diol,2,3-dimethyl-, 1-Ethynyl-3,trans(1,1-dimethylethyl), Phenol,2,6-dimethoxy-4-(2-propenyl)- and 7-Methyl-Z-tetradecen-1-olacetate found in ethanolic petiole extract. Some compounds were documented as potent anti-inflammatory and arthritis related diseases by other researchers. In this study, the efficiency of solvents to extract bioactives was found to be ethanol > water, methanol > hexane > chloroform. Together, our results suggest the prospective utilization of fruit and petiole of A. longiloba to inhibit the activity of XO enzyme.

Project description:Syzygium cumini (L.) Skeels, commonly known as the Jamun or Indian blackberry, is a tropical evergreen tree native to the Indian subcontinent, and it belongs to the Myrtaceae family. This research aimed to assess the antibacterial properties of the extracts derived from S. cumini seed kernels and evaluate their total flavonoid content, total phenol content, total carbohydrate content, antioxidant capacity, and inhibitory effects on xanthine oxidase. Cold maceration was chosen for its ability to preserve thermolabile compounds and efficiently extract bioactive constituents with minimal energy and equipment requirement, with hexane and methanol employed as extraction solvents. The methanolic seed kernel extract of S. cumini showed the highest flavonoid (127.78 μg quercetin equivalent/mg dried extract vs. 21.24 μg quercetin equivalent/mg in hexane dried extract) and polyphenol content (153.81 μg gallic acid equivalent/mg dried extract vs. 38.89 μg gallic acid equivalent/mg in hexane dried extract), along with significant carbohydrate content (475.61 μg glucose equivalent/mg dried extract vs. 5.57 μg GE/mg in hexane dried extract). It also demonstrated potent antioxidant activity (IC50: 9.23 μg/mL; ascorbic acid: 5.10 μg/mL) and xanthine oxidase inhibition (IC50: 14.88 μg/mL), comparable to the standard drug allopurinol (IC50: 6.54 μg/mL), suggesting its therapeutic potential. Moreover, the methanolic extract of seed kernels exhibited strong antibacterial activity, with inhibition zones of 19.00 mm against S. epidermidis, higher than the standard antibiotic (gentamicin: 18.33 mm) against K. pneumonia (ciprofloxacin: 33.66 mm). The lowest minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values of 0.32 mg/mL and 0.52 mg/mL, respectively, were observed for the same extract against S. epidermis. In conclusion, this study demonstrated the remarkable antibacterial effects of S. cumini methanolic seed kernel extract against various pathogenic microorganisms as well as significant inhibitory effects on xanthine oxidase and antioxidant activity.
