Tracing the geographic origin of velvet antlers in China via stable isotope analyses.

ABSTRACT: Multielement (H, C, N, O) stable isotope ratio analysis was used to discriminate the geographical origin of velvet antlers (VAs) of deer from ten provinces in China. Ratios of ²H/¹H, ¹³C/¹²C, ¹⁵N/¹⁴N, and ¹⁸O/¹⁶O in the VA samples were measured using isotope ratio mass spectrometry. The results showed that there were highly significant differences in the mean isotopic values and in four isotopic ratios between VA samples from the ten provinces. The most significant difference among the four isotope ratios was in δ ²H ratio of VA samples; regions with a more humid climate and higher average ambient temperatures had higher δ ²H ratios than those with dry climates and lower temperatures. These results demonstrate that the multiple stable isotopic ratio approach is a powerful tool to help trace the geographical origin of VAs, and could be adopted by government officials to help protect consumer interests from improper labeling in VA markets.

Tracing the geographical origin of tobacco at two spatial scales by stable isotope and element analyses with chemometrics.

Project description:Tobacco is a widely cultivated cash crop, but it is often smuggled and sold illegally. Unfortunately, there is currently no way to verify the origin of tobacco in China. In an effort to address this issue, we conducted a study using stable isotopes and elements from 176 tobacco samples at both provincial and municipal scales. Our findings revealed significant differences in δ13C, K, Cs, and 208/206Pb at the provincial-level, and Sr, Se, and Pb at the municipal level. We created a heat map at the municipal level, which showed a similar cluster classification to geographic grouping and provided an initial assessment of tobacco origins. Using OPLS-DA modeling, we achieved a 98.3% accuracy rate for the provincial scale and 97.6% for the municipal scale. It is worth noting that the importance of rankings of variables varied depending on the spatial scale of the evaluation. This study offers the first traceability fingerprint dataset of tobacco and has the potential to combat mislabeling and fraudulent conduct by identifying the geographical origin of tobacco.

Stable isotope tracing to assess tumor metabolism in vivo.

Project description:Cancer cells undergo diverse metabolic adaptations to meet the energetic demands imposed by dysregulated growth and proliferation. Assessing metabolism in intact tumors allows the investigator to observe the combined metabolic effects of numerous cancer cell-intrinsic and -extrinsic factors that cannot be fully captured in culture models. We have developed methods to use stable isotope-labeled nutrients (e.g., [13C]glucose) to probe metabolic activity within intact tumors in vivo, in mice and humans. In these methods, the labeled nutrient is introduced to the circulation through an intravenous catheter prior to surgical resection of the tumor and adjacent nonmalignant tissue. Metabolism within these tissues during the infusion transfers the isotope label into metabolic intermediates from pathways supplied by the infused nutrient. Extracting metabolites from surgical specimens and analyzing their isotope labeling patterns provides information about metabolism in the tissue. We provide detailed information about this technique, from introduction of the labeled tracer through data analysis and interpretation, including streamlined approaches to quantify isotope labeling in informative metabolites extracted from tissue samples. We focus on infusions with [13C]glucose and the application of mass spectrometry to assess isotope labeling in intermediates from central metabolic pathways, including glycolysis, the tricarboxylic acid cycle and nonessential amino acid synthesis. We outline practical considerations to apply these methods to human subjects undergoing surgical resections of solid tumors. We also discuss the method's versatility and consider the relative advantages and limitations of alternative approaches to introduce the tracer, harvest the tissue and analyze the data.

Global Stable-isotope Tracing Metabolomics Reveals System-wide Metabolic Alternations in Aging Drosophila

Project description:System-wide metabolic homeostasis is crucial for maintaining physiological functions of living organisms. Stable-isotope tracing metabolomics allows to unravel metabolic activity quantitatively by measuring the isotopically labeled metabolites, but has been largely restricted by coverage. Yet, delineating system-wide metabolic homeostasis at the whole-organism level remains non-trivial. Here, we develop a global isotope tracing metabolomics technology to measure labeled metabolites with a metabolome-wide coverage. Using Drosophila as an aging model organism, we probe the in vivo tracing kinetics with quantitative information on labeling patterns, extents and rates on a metabolome-wide scale. We curate a system-wide metabolic network to characterize metabolic homeostasis and disclose a system-wide loss of metabolic coordinations that impacts both intra- and inter-tissue metabolic homeostasis significantly during Drosophila aging. Importantly, we reveal an unappreciated metabolic diversion from glycolysis to serine metabolism and purine metabolism as Drosophila aging. The developed technology facilitates a system-level understanding of metabolic regulation in living organisms.

Oxygen and hydrogen stable isotope ratios of bulk needles reveal the geographic origin of Norway spruce in the European Alps.

Project description:BackgroundTracking timber is necessary in order to prevent illegal logging and protect local timber production, but there is as yet no suitable analytical traceability method. Stable isotope ratios in plants are known to reflect geographical variations. In this study we analysed four stable isotope ratios in order to develop a model able to identify the geographic origin of Norway spruce in the European Alps.Methodology and principal findingsδ18O, δ2H, δ13C and δ15N were measured in bulk needles of Picea abies sampled in 20 sites in and around the European Alps. Environmental and spatial variables were found to be related to the measured isotope ratios. An ordinary least squares regression was used to identify the most important factor in stable isotope variability in bulk needles. Spatial autocorrelation was tested for all isotope ratios by means of Moran's I. δ18O, δ2H and δ15N values differed significantly between sites. Distance from the coast had the greatest influence on δ2H, while latitude and longitude were strongly related to δ18O. δ13C values did not appear to have any relationship with geographical position, while δ15N values were influenced by distance from the motorway. The regression model improved the explanatory power of the spatial and environmental variables. Positive spatial autocorrelations were found for δ18O and δ2H values.ConclusionsThe δ 18O, δ2H and δ15N values in P. abies bulk needles are a suitable proxy to identify geographic origin as they vary according to geographical position. Although the regression model showed the explanatory variables to have significant power and stability, we conclude that our model might be improved by multivariate spatial interpolation of the δ 18O and δ2H values.

Tracing mobility patterns through the 6th-5th millennia BC in the Carpathian Basin with strontium and oxygen stable isotope analyses.

Project description:The complexity of Neolithic population movements and their interpretation through material culture have been the subject of archaeological research for decades. One of the dominant narratives proposes that groups from the Starčevo-Körös-Criş complex spread from the central towards the northern Balkans in the Early Neolithic and eventually brought the Neolithic lifestyle into present-day Hungary. Broad geographical migrations were considered to shape the continuous expansion of Neolithic groups and individuals. However, recent archaeological research, aDNA, and isotope analyses challenged the synchronous appearance of specific material culture distributions and human movement dynamics through emphasizing communication networks and socio-cultural transformation processes. This paper seeks to retrace the complexity of Neolithic mobility patterns across Hungary by means of strontium and oxygen stable isotope analyses, which were performed on a total of 718 human dental enamel samples from 55 Neolithic sites spanning the period from the Starčevo to the Balaton-Lasinja culture in Transdanubia and from the Körös to the Tiszapolgár cultural groups on the Great Hungarian Plain (Alföld). This study presents the largest strontium and oxygen isotope sample size for the Neolithic Carpathian Basin and discusses human mobility patterns on various geographical scales and throughout archaeological cultures, chronological periods, and sex and gender categories in a multiproxy analysis. Based on our results, we discuss the main stages of the Neolithisation processes and particularly trace individual movement behaviour such as exogamy patterns within extensive social networks. Furthermore, this paper presents an innovative differentiation between mobility patterns on small, micro-regional, and supra-regional scales, which provides new insights into the complex organisation of Neolithic communities.

Preliminary study on the geographical origin of Chinese 'Cuiguan' pears using integrated stable isotope and multi-element analyses.

Project description:Distinguish the geographical origin of the pear is important due to the increasingly valued brand protection and reducing the potential food safety risks. In this study, the profiles of stable isotopes (δ13C, δ15N, δ2H, δ18O) and the contents of 16 elements in pear peer from four production areas were analyzed. The δ13C, δ15N, δ2H, δ18O and 12 elements were significantly different (p < 0.05) in the four production areas. Chemometrics analysis including principal component analysis (PCA), orthogonal partial least squares discriminant analysis (OPLS-DA) and linear discriminant analysis (LDA) were exploited for geographical origin classification of samples. OPLS-DA analysis showed that crucial variables (δ13C, δ18O, δ2H, Ni, Cd, Ca, δ15N, Sr and Ga) are more relevant for the discrimination of the samples. OPLS-DA achieved pear origin accuracy rates of 87.76 % by combining stable isotope ratios and elemental contents. LDA had a higher accuracy rate than OPLS-DA, and the LDA analysis showed that the original discrimination rate reached to 100 %, while the cross-validated rate reached to 95.7 %. These studies indicated that this method could be used to assess the geographical discrimination of pear from different producing areas and could potentially control the fair trade of pear in fruit markets.

Generalized Tree Structure to Annotate Untargeted Metabolomics and Stable Isotope Tracing Data.

Without prior knowledge of the chemical identity or formula, computational organization and interpretation of these ions is challenging, which is the deficit of previous software tools that perform the task using network algorithms. We propose here a generalized tree structure to annotate ions in relationships to the original compound and infer neutral mass. An algorithm is presented to convert mass distance networks to this tree structure with high fidelity. This method is useful for both regular untargeted metabolomics and stable isotope tracing experiments. It is implemented as a Python package (khipu) and provides a JSON format for easy data exchange and software interoperability. By generalized preannotation, khipu makes it feasible to connect metabolomics data with common data science tools and supports flexible experimental designs.
