Project description:Herein, we developed a novel fluorescent assay using spherical identification probes and toehold-mediated strand displacement reaction-initiated silver nanoclusters (AgNCs) "on-off" signal switch. In this strategy, the target was captured by the spherical probes to induce the activity of exonuclease III (Exo III), catalyzing the cyclic cleavage of substrates to produce a mass of trigger strands. After magnetic bead separation, the intermediates in the supernatant activated downstream toehold-mediated strand displacement reaction to change the structure of silver nanocluster templates, leading to fluorescence intensity reduction. Furthermore, it is demonstrated that the application of spherical identification probes could reduce the signal leakage and the limit of detection. In addition, AgNCs with perfect optical property were ingeniously combined to realize signal output, which reduced the cost and time of synthesis. Under the optimal conditions, the sensing method displayed a good linear range from 250 pM to 25 nM with a detectable minimum concentration of 250 pM. And the practical application potential in complex biological matrices was also evaluated. Considering these advantages, this constructed strategy opens a new path for nucleic acid detection with better performance. A simple, label- and hairpin-free fluorescent system based on spherical identification probe and toehold-mediated strand displacement reaction-initiated silver nanoclusters (AgNCs) "on-off" signal switch was successfully constructed to detect target DNA.

Project description:The widespread use of silver nanoparticles (Ag NPs) has raised substantial health risks to environmental and human beings. Although various negative effects had been reported, little is known about the epigenetic toxicity induced by Ag+ and Ag NPs. This study characterized physiological and lncRNA profiles to explore the toxic effects and epigenetic mechanisms in Tetrahymena thermophila on exposure to Ag+ and three types of Ag NPs. We found that both of Ag+ and Ag NPs exhibited strong growth-inhibiting effects on T. thermophila. The toxicity was mainly caused by high levels of reactive oxygen species (ROS), which subsequently led to lipid peroxidation and mitochondrial dysfunction. As a defense mechanism against oxidative stress, the protist elicited an antioxidant response. Importantly, 1250 lncRNAs were differentially expressed under Ag+ or Ag NPs exposure relative to the non-exposure control, which were clustered into 15 expression modules in weighted gene co-expression network analysis. These gene modules exhibited toxicant-specific expression patterns, indicating that they play various regulatory roles, including cell growth inhibition, cell membrane cation channel activation, and oxidoreductase activity promotion. Taken together, this research illuminates how post-transcriptional mechanisms of a ciliated protozoa regulate responses to Ag+ and Ag NPs toxicities.

Project description:Isomerization is highly important in all aspects of science, yet it is rarely observed in nanoscience. Here, we synthesized a unique triple core-shell Ag84 nanocluster displaying isomerism, which is controlled by different carboxylic acids and a one-way transformation (SD/Ag84a → SD/Ag84b). The innermost core is a rare Ag10 nanocluster which comprises an Ag6 octahedral unit as seen in face-centred cubic (fcc) silver metal and four capped Ag atoms. It templates two crescent-shaped polyoxometalate (W7O26)10- shells which are then enclosed in a shell of silver shaped as rugby balls. The organic ligands (iPrS-, n PrCOO- and PhCOO-) finally shield the metallic clusters. Due to slight differences in structure at two poles and the steric hindrance of n PrCOO- and PhCOO-, SD/Ag84a and SD/Ag84b adopt the shapes of flat-headed and cuspidal prolate spheres, respectively. Interestingly, PhCOOH is dominant over n PrCOOH whereby crystals of SD/Ag84b were isolated if PhCOOH is added during the synthesis of SD/Ag84a. This demonstrates that PhCOOH not only alters the organic coats but also induces metal shell re-organization. This work reveals carboxylate-controlled skeletal isomerism in silver nanoclusters for the first time, thus deepening the understanding of silver nanocluster assembly, flexibility and reactivity.

Project description:Theoretical study of structural, optical, and photovoltaic properties of novel bio-nano hybrids (dye-nanocluster), as well as at TiO2 surface model support is presented in the context of the application for dye-sensitized solar cells (DSSC). A group of anthocyanidin dyes (pelargonidin, cyanidin, delphinidin, peonidin, petunidin, and malvidin) represented by cyanidin covalently bound to silver nanoclusters (NCs) with even or odd number of valence electrons have been investigated using DFT and TDDFT approach. The key role of nanoclusters as acceptors in hybrids cyanidin-NC has been shown. The nanoclusters with an even number of valence electrons are suitable as acceptors in hybrids. The interaction of bio-nano (cyanidin-NC) hybrid with the TiO2 surface model has been investigated in the context of absorption in near-infrared (NIR) and charge separation due to donor and acceptor subunits. Altogether, the theoretical concept serves to identify the key steps in the design of novel solar cells based on bio-nano hybrids at TiO2 surface for DSSC application.

Project description:For the first time site-specific doping of silver into a spherical Au25 nanocluster has been achieved in [Au19Ag6(MeOPhS)17(PPh3)6] (BF4)2 (Au19Ag6) through a dual-ligand coordination strategy. Single crystal X-ray structural analysis shows that the cluster has a distorted centered icosahedral Au@Au6Ag6 core of D 3 symmetry, in contrast to the I h Au@Au12 kernel in the well-known [Au25(SR)18]- (R = CH2CH2Ph). An interesting feature is the coexistence of [Au2(SPhOMe)3] dimeric staples and [P-Au-SPhOMe] semi-staples in the title cluster, due to the incorporation of PPh3. The observation of only one double-charged peak in ESI-TOF-MS confirms the ordered doping of silver atoms. Au19Ag6 is a 6e system showing a distinct absorption spectrum from [Au25(SR)18]-, that is, the HOMO-LUMO transition of Au19Ag6 is optically forbidden due to the P character of the superatomic frontier orbitals.

Project description:The emerging promise of few-atom metal catalysts has driven the need for developing metal nanoclusters (NCs) with ultrasmall core size. However, the preparation of metal NCs with single-digit metallic atoms and atomic precision is a major challenge for materials chemists, particularly for Ag, where the structure of such NCs remains unknown. In this study, we developed a shape-controlled synthesis strategy based on an isomeric dithiol ligand to yield the smallest crystallized Ag NC to date: [Ag9(1,2-BDT)6]3- (1,2-BDT = 1,2-benzenedithiolate). The NC's crystal structure reveals the self-assembly of two Ag square pyramids through preferential pyramidal vertex sharing of a single metallic Ag atom, while all other Ag atoms are incorporated in a motif with thiolate ligands, resulting in an elongated body-centered Ag9 skeleton. Steric hindrance and arrangement of the dithiolated ligands on the surface favor the formation of an anisotropic shape. Time-dependent density functional theory based calculations reproduce the experimental optical absorption features and identify the molecular orbitals responsible for the electronic transitions. Our findings will open new avenues for the design of novel single-digit metal NCs with directional self-assembled building blocks.

Project description:The abnormal activation of epidermal growth factor receptor (EGFR) is strongly associated with a variety of human cancers but the underlying molecular mechanism is not fully understood. By using direct stochastic optical reconstruction microscopy (dSTORM), we find that EGFR proteins form nanoclusters in the cell membrane of both normal lung epithelial cells and lung cancer cells, but the number and size of clusters significantly increase in lung cancer cells. The formation of EGFR clusters is mediated by the ionic interaction between the anionic lipid phosphatidylinositol-4,5-bisphosphate (PIP2) in the plasma membrane and the juxtamembrane (JM) region of EGFR. Disruption of EGFR clustering by PIP2 depletion or JM region mutation impairs EGFR activation and downstream signaling. Furthermore, JM region mutation in constitutively active EGFR mutant attenuates its capability of cell transformation. Collectively, our findings highlight the key roles of anionic phospholipids in EGFR signaling and function, and reveal a novel mechanism to explain the aberrant activation of EGFR in cancers.
