A two-photon lysosome-targeted probe for endogenous formaldehyde in living cells.

ABSTRACT: Formaldehyde (FA) is a gaseous signaling molecule that plays a vital role in various biological processes as well as neurodegenerative diseases. Therefore, it is of great practical significance to develop effective and reliable chemical sensors for the monitoring of endogenous FA. Here, we designed and synthesized a two-photon (810 nm) turn-on chemosensor AMNT (aminomorpholine naphthalimide) that accurately localizes lysosomes in cells for imaging of cellular endogenous FA. The fluorescence emission peak of AMNT was at ∼540 nm, with a slight blue shift (∼528 nm) in response to FA, while the green fluorescence intensity increased. The probe exhibits excellent selectivity for FA among other biological interference species and a fast response time for FA. It is worth mentioning that the probe successfully imaged endogenous FA in cells in two-photon mode, making the probe an effective research tool in the biomedical field to study diseases related to abnormal FA expression.

A New Two-Photon Ratiometric Fluorescent Probe for Detecting Alkaline Phosphatase in Living Cells.

Project description:Alkaline phosphatase (ALP) is an important diagnostic indicator of many human diseases. To quantitatively track ALP in biosystems, herein, for the first time, we report an efficient two-photon ratiometric fluorescent probe, termed probe 1 and based on classic naphthalene derivatives with a donor-π-acceptor (D-π-A) structure and deprotection of the phosphoric acid moiety by ALP. The presence of ALP causes the cleave of the phosphate group from naphthalene derivatives and the phosphate group changes the ability of the intramolecular charge transfer (ICT) and remarkably alters the probe's photophysical properties, thus an obvious ratiometric signal with an isoemissive point is observed. The fluorescence intensity ratio displayed a linear relationship against the concentration of ALP in the concentration range from 20 to 180 U/L with the limit of detection of 2.3 U/L. Additionally, the probe 1 is further used for fluorescence imaging of ALP in living cells under one-photon excitation (405 nm) or two-photon excitation (720 nm), which showed a high resolution imaging, thus demonstrating its practical application in biological systems.

A coumarin Schiff's base two-photon fluorescent probe for hypochlorite in living cells and zebrafish.

Project description:Selective and sensitive fluorescent probes for ClO- are desirable due to the importance of ClO- in biological processes. Here, a coumarin Schiff's base, compound 1, has been developed and successfully used as a one- and two-photon fluorescent probe for ClO- with high selectivity. This probe can recognize ClO- with obvious color change from yellow-green to colorless and green to blue fluorescence emission, which can be observed by the naked eye. The properties of low cytotoxicity and good cell permeability allow it to be used for ClO- detection in living cells and zebrafish by both one- and two-photon microscopy imaging. All these results indicate that the compound is a sensitive probe with potential for analysis of ClO- in biological samples. The mechanism by which probe 1 recognizes ClO- is possibly nucleophilic addition followed by hydrolysis.

A turn-on endoplasmic reticulum-targeted two-photon fluorescent probe for hydrogen sulfide and bio-imaging applications in living cells, tissues, and zebrafish.

Project description:As one of the important gas signal molecules, hydrogen sulfide (H2S) is associated with many important physiological processes in living organisms. Organelles, especially endoplasmic reticulum (ER), play a crucial role in the cell metabolism. Accordingly, the detection of H2S in the ER is of high interest. Toward this goal, we have described the development of the first ER-targeted fluorescent H2S probe (Na-H 2 S-ER). The new probe exhibited favorable features, such as a large turn-on fluorescence signal (45-fold fluorescence enhancement), high sensitivity and selectivity. The probe was successfully employed for imaging exogenous and endogenous H2S in the living HeLa cells. Significantly, the new probe Na-H 2 S-ER was employed to visualize H2S in the ER of living cells for the first time. In addition, the probe was also successfully used for imaging H2S in the living tissues up to a depth of 100 μm and in the living zebrafish.

Imaging of fluoride ion in living cells and tissues with a two-photon ratiometric fluorescence probe.

Project description:A reaction-based two-photon (TP) ratiometric fluorescence probe Z2 has been developed and successfully applied to detect and image fluoride ion in living cells and tissues. The Z2 probe was designed designed to utilize an ICT mechanism between n-butylnaphthalimide as a fluorophore and tert-butyldiphenylsilane (TBDPS) as a response group. Upon addition of fluoride ion, the Si-O bond in the Z2 would be cleaved, and then a stronger electron-donating group was released. The fluorescent changes at 450 and 540 nm, respectively, made it possible to achieve ratiometric fluorescence detection. The results indicated that the Z2 could ratiometrically detect and image fluoride ion in living cells and tissues in a depth of 250 μm by two-photon microscopy (TPM).

A FRET Based Two-Photon Fluorescent Probe for Visualizing Mitochondrial Thiols of Living Cells and Tissues.

Project description:Glutathione (GSH) is the main component of the mitochondrial thiol pool and plays key roles in the biological processes. Many evidences have suggested that cysteine and homocysteine also exist in mitochondria and are interrelated with GSH in biological systems. The fluctuation of the levels of mitochondrial thiols has been linked to many diseases and cells' dysfunction. Therefore, the monitoring of mitochondrial thiol status is of great significance for clinical studies. We report here a novel fluorescence resonance energy transfer based two-photon probe MT-1 for mitochondrial thiols detection. MT-1 was constructed by integrating the naphthalimide moiety (donor) and rhodamine B (accepter and targeting group) through a newly designed linker. MT-1 shows a fast response, high selectivity, and sensitivity to thiols, as well as a low limit of detection. The two-photon property of MT-1 allows the direct visualization of thiols in live cells and tissues by two-photon microscopy. MT-1 can serve as an effective tool to unravel the diverse biological functions of mitochondrial thiols in living systems.

A Two-Photon Fluorescent Probe for the Visual Detection of Peroxynitrite in Living Cells and Zebrafish.

Project description:Peroxynitrite (ONOO-), as an important reactive oxygen species (ROS), holds great potential to react with a variety of biologically active substances, leading to the occurrence of various diseases such as cancer and neurodegenerative diseases. In this work, we developed a novel mitochondria-localized fluorescent probe, HDBT-ONOO-, which was designed as a mitochondria-targeting two-photon fluorescence probe based on 1,8-naphthylimide fluorophore and the reactive group of 4-(bromomethyl)-benzene boronic acid pinacol ester. More importantly, the probe exhibited good biocompatibility, sensitivity, and selectivity, enabling its successful application in imaging the generation of intracellular and extracellular ONOO-. Furthermore, exogenous and endogenous ONOO- products in live zebrafish were visualized. It is greatly expected that the designed probe can serve as a useful imaging tool for clarifying the distribution and pathophysiological functions of ONOO- in cells and zebrafish.

Lysosome-Targeted Single Fluorescence Probe for Two-Channel Imaging Intracellular SO₂ and Biothiols.

Project description:As the members of reactive sulfur species, SO₂ and biothiols play a significant role in physiological and pathological processes and directly influence numerous diseases. Furthermore, SO₂ and biothiols can provide a reductive environment for lysosomes to carry out their optimal functionality. To this end, the development of single fluorescent probes for imaging SO₂ and biothiols from different emission channels is highly desirable for understanding their physiological nature. Here, a lysosome-targeted fluorescent probe (BPO-DNSP) with a dual reaction site for SO₂ and biothiols was presented. BPO-DNSP can sensitively and selectively respond to SO₂ in the green channel with a large Stokes shift over 105 nm, and to biothiols in the near-infrared emission channel with a large Stokes shift over 109 nm. The emission shift for the two channels was as high as 170 nm. Colocalization experiments verified that BPO-DNSP can selectively enrich lysosomes. Notably, BPO-DNSP can not only be used to image intracellular SO₂ and biothiols from two different channels, but also to monitor the conversion of biothiols to SO₂ without adding exogenous enzymes in living HeLa cells.

Multicolor two-photon imaging of endogenous fluorophores in living tissues by wavelength mixing.

Project description:Two-photon imaging of endogenous fluorescence can provide physiological and metabolic information from intact tissues. However, simultaneous imaging of multiple intrinsic fluorophores, such as nicotinamide adenine dinucleotide(phosphate) (NAD(P)H), flavin adenine dinucleotide (FAD) and retinoids in living systems is generally hampered by sequential multi-wavelength excitation resulting in motion artifacts. Here, we report on efficient and simultaneous multicolor two-photon excitation of endogenous fluorophores with absorption spectra spanning the 750-1040 nm range, using wavelength mixing. By using two synchronized pulse trains at 760 and 1041 nm, an additional equivalent two-photon excitation wavelength at 879 nm is generated, and achieves simultaneous excitation of blue, green and red intrinsic fluorophores. This method permits an efficient simultaneous imaging of the metabolic coenzymes NADH and FAD to be implemented with perfect image co-registration, overcoming the difficulties associated with differences in absorption spectra and disparity in concentration. We demonstrate ratiometric redox imaging free of motion artifacts and simultaneous two-photon fluorescence lifetime imaging (FLIM) of NADH and FAD in living tissues. The lifetime gradients of NADH and FAD associated with different cellular metabolic and differentiation states in reconstructed human skin and in the germline of live C. Elegans are thus simultaneously measured. Finally, we present multicolor imaging of endogenous fluorophores and second harmonic generation (SHG) signals during the early stages of Zebrafish embryo development, evidencing fluorescence spectral changes associated with development.

A mitochondria-targeted ratiometric two-photon fluorescent probe for biological zinc ions detection.

Project description:A mitochondria-targeted ratiometric two-photon fluorescent probe (Mito-MPVQ) for biological zinc ions detection was developed based on quinolone platform. Mito-MPVQ showed large red shifts (68 nm) and selective ratiometric signal upon Zn(2+) binding. The ratio of emission intensity (I488 nm/I420 nm) increases dramatically from 0.45 to 3.79 (ca. 8-fold). NMR titration and theoretical calculation confirmed the binding of Mito-MPVQ and Zn(2+). Mito-MPVQ also exhibited large two-photon absorption cross sections (150 GM) at nearly 720 nm and insensitivity to pH within the biologically relevant pH range. Cell imaging indicated that Mito-MPVQ could efficiently located in mitochondria and monitor mitochondrial Zn(2+) under two-photon excitation with low cytotoxicity.
