The mitochondrial genome of a wild edible mushroom, Russula rosea.

ABSTRACT: Russula rosea is a common wild edible ectomycorrhizal fungus, which is widely distributed all over the world. We assembled the complete mitochondrial genome of R. rosea with the total length was 54177 bp and the GC content of 22.34%. It contains a total of 57 genes, including 14 standard protein-coding genes, one conserved ribosomal protein S3 gene (rps3), two rRNA genes, 24 tRNA genes, 15 free-standing open reading frames (ORFs) and one DNA polymerase gene (dpo). Mitochondrial genome found a close evolutionary relationship between Russula rosea and Russula lepida, which was helpful to study the genetic evolutionary relationship of edible fungi.

Project description:Russula griseocarnosa is a wild edible ectomycorrhizal mushroom in southern China. In this study, we assembled the complete mitochondrial genome of R. griseocarnosa. Its total length was 60995 bp with a GC content of 21% and contained a total of 52 genes, including 14 standard protein-coding genes, two rRNA genes, 21 tRNA genes and 15 free-standing open reading frames (ORFs). Phylogenetic analysis reflected that the evolutionary processes between R. griseocarnosa and some agaricomycetes.

Project description:In the present article, we report data on the whole genome sequence of a wild edible and medicinal ectomycorrhizal fungus Russula griseocarnosa. The R. griseocarnosa genome consists of 64.81 Mb with a GC-pair content of 49.41%. The genome assembly consists of 471 scaffolds and 16128 coding protein genes. The coding protein genes was annotated in different databases (GO, KEGG and CAZys), respectively. The whole genome sequence and functional annotation provide important information for ectomycorrhizal fungus, which can be used as a basis for cultivation and breeding of R. griseocarnosa. The Whole Genome project of Russula griseocarnosa has been deposited at DDBJ/ENA/GenBank under the accession RMVF00000000. The version described is RMVF01000000. The mitochondrial genome of a wild toxic mushroom, Russula subnigricans.

Project description:Russula subnigricans is a wild toxic mushroom in the world. In this study, we present the 60,949 bp mitochondrial genome of R. subnigricans with a GC content of 21%. The mtDNA assembly consists of 60 genes, which including 14 standard protein-coding genes, two rRNA genes, 25 tRNA genes and 19 free-standing open reading frames (ORFs). Phylogenetic analysis was performed using 14 protein-coding genes, the results showed that the R. subnigricans had high homology with other Russula species.

Chemical Constituents with Inhibitory Activity of NO Production from a Wild Edible Mushroom, Russula vinosa Lindbl, May Be Its Nutritional Ingredients.

Project description:Russula vinosa Lindbl is a wild edible mushroom that is usually used for original material of food and soup and has rich nutritional value. What are the nutritional ingredients? In order to answer this question, we investigated the chemical constituents of this wild functional food. Six new compounds (1-6), together with nine known ones (7-15), were isolated from R. vinosa. The six new compounds were named as vinosane (1), rulepidadione C (2), (24E)-3,4-seco-cucurbita-4,24-diene-26,29-dioic acid-3-methyl ester (3), (24E)-3,4-seco-cucurbita-4,24-diene-26-oic acid-3-ethyl ester (4), (24E)-3β-hydroxycucurbita-5,24-diene-26,29-dioic acid (5), and (2S,3S,4R,2'R)-2-(2'-hydroxydocosanoylamino)eicosane-1,3,4-triol (6). Their structures were determined based on spectroscopic methods including HR-ESI-MS, 1D, and 2D NMR. Moreover, a cell counting kit-8 (CCK-8 kit) was used to screen for the cytotoxicity of compounds 1-5 and 7-13 on mouse macrophage RAW 264.7 cells. The results showed that compounds 1-5 and 7-13 had no obvious cytotoxicity. In addition, the inhibitory effects on nitric oxide (NO) production in lipopolysaccharide (LPS)-activated mouse macrophage RAW 264.7 cells were evaluated. Compounds 1, 3, 4, 7, 12, and 13 showed moderate inhibitory activity on NO production.

Complete mitochondrial genome of the edible mushroom Singerocybe alboinfundibuliformis (Clitocybaceae, Agaricales).

Project description:Singerocybe alboinfundibuliformis (Seok et al.) Yang, Qin & Takah 2014 is an edible mushroom distributed in several East or Southeast Asian countries. Herein, we report the mitochondrial genome of S. alboinfundibuliformis based on Illumina sequencing data. The overall length of the mitochondrial genome is 64,279 bp, with a GC content of 29.0%. It contains 14 typical protein-coding genes, 27 tRNA genes, two rRNA genes, and 13 intergenic ORFs. Most of these genes (39 out of 56) are transcribed at the forward strand, and few (17 out of 56) are transcribed at the reverse strand. Among these genes, only the rnl gene is invaded by an intron, and all other genes are intron-free. Phylogenetic analysis based on mitochondrial amino acid sequences supports the phylogenetic position of S. alboinfundibuliformis in Clitocybaceae, being close to Lepista sordida (Schumach.) Singer 1951. This study serves as a springboard for future investigation on fungal evolution in Clitocybaceae.

SMRT-based mitochondrial genome of the edible mushroom Morchella conica.

Project description:Morchella conica Pers. is a highly-prized mushroom for its edible and medical values. In this study, we determined the complete mitochondrial genome of M. conica combining both PacBio and Illumina sequencing technologies. The complete mitochondrial genome is 280,763 bp in length with a GC content of 39.88%. We identified a total of 14 core conserved protein-coding genes, 127 non-conserved open reading frames (ncORFs) and 30 tRNA genes in the M. conica mitogenome. However, no large or small rRNA subunits (rnl or rns) were identified in this mitogenome. In addition, we detected two mitochondrial RNase P (rnpB) genes and one ribosomal protein genes (rps3). Phylogenetic analysis was performed among M. conica and 18 other representative fungi from Ascomycota, Basidiomycota and Mucoromycota. Our results showed that M. conica was most closely related to M. importuna. The availability of the M. conica mitochondrial genome will form the basis of genetic breeding program and enhance our understanding of the evolution of this species.

The complete mitochondrial genome of the edible mushroom Grifola frondosa.

Project description:The culinary-medicinal mushroom Grifola frondosa is widely cultivated in East Asia. In this study, the complete mitochondrial genome of G. frondosa was determined using Illumina sequencing. The circular molecule was 197,486 bp in length with a content of 25.01% GC, which was one of the largest mitochondrial genomes in the order Polyporales. A total of 39 known genes encoding 13 common mitochondrial genes, 24 tRNA genes, 1 ribosomal protein s3 gene (rps3), and 1 DNA polymerase gene (dpo) were predicted in this genome. The phylogenetic analysis showed that G. frondosa clustered together with Sparassis crispa, Laetiporus sulphureus, Wolfiporia cocos, and Taiwanofungus camphoratus. The complete mitochondrial genome reported here may provide new insight into genetic information and evolution for further studies.

The complete mitochondrial genome of an edible mushroom, Sparassis crispa.

Project description:Sparassis crispa, also known as cauliflower mushroom, is a widely used medicinal mushroom in traditional Chinese medicine due to the presence of bioactive substances with pharmacological activity. Here, we report a complete mitochondrial genome sequence of S. crispa consisting of 139,253 bp containing 47 genes including 15 protein-coding genes, 27 transfer RNA, and 5 ribosomal RNA genes obtained from 40.406 Mb genome containing 18,917 predicted contigs using raw data of next-generation sequencing having 85.4% Q30. The overall base composition of S. crispa was 26.47% G-C and 73.53% A-T. The phylogenetic tree based on atp6 sequence data showed its close relationship with Sparassis radicata. The complete mitochondrial genome sequence of S. crispa provides an essential and important DNA molecular data for further phylogenetic and evolutionary analysis of S. crispa.

The complete mitochondrial genome of the Agrocybe aegerita, an edible mushroom.

Project description:Agrocybe aegerita is a medicinally and nutritionally important edible basidiomycete. Despite previous phylogenetic studies, the taxonomy of A. aegerita complex remains unclear due to lacking of resolutive data. Herein, the complete mitochondrial genome of A. aegerita is reported and analyzed. The mitogenome length was 116,329 bp, with a GC content of 27.6%, include 17 typical protein-coding genes, two ribosomal protein genes (rps3), two ribosomal RNA genes and a set of 32 transfer RNA genes. A phylogenetic analyses using complete mitogenome in Agaricales showed that A. aegerita is closely related to the genus Pleurotus and represents a clade clearly independent from other Agaricales species.
