Efficacy of a Modified Live Porcine Reproductive and Respiratory Syndrome Virus 1 (PRRSV-1) Vaccine against Experimental Infection with PRRSV AUT15-33 in Weaned Piglets.

ABSTRACT: In this study, the efficacy of the commercial modified live PRRSV-1 vaccine “Ingelvac PRRSFLEX® EU” was assessed in weaned piglets experimentally infected with PRRSV strain AUT15-33. Seventy-four weaned piglets were allocated to five groups. Vaccinated (groups 1, 2, and 5) and non-vaccinated piglets (groups 3 and 4), infected with either a low dose (103 TCID50/dose; groups 2 and 4) or a high dose (105 TCID50/dose; groups 1 and 3) of the virus, were compared regarding clinical signs, average daily weight gain (ADG), lung lesions, viral load in serum, oral swabs, and tissue samples. In comparison to vaccinated animals, coughing increased notably in the second week after challenge in non-vaccinated piglets. During the same time period, vaccinated, high-dose-infected piglets showed significantly higher ADG (p < 0.05) than non-vaccinated, high-dose-infected animals. All infected piglets reached approximately the same viremia levels, but vaccinated animals showed both a significantly reduced viral load in oral fluid (p < 0.05) and tissue samples and significantly reduced lung lesions (p < 0.05). In conclusion, vaccination was able to increase ADG, reduce the amount of viral shedding via oral fluids, and reduce the severity of lung lesions and the viral load in tissue samples under experimental conditions.

Project description:Cell-mediated immunity (CMI), IL-10, and the protective efficacy of modified-live porcine reproductive and respiratory syndrome virus (PRRSV) vaccines (MLV) against co-challenge with PRRSV-1 and PRRSV-2 (HP-PRRSV) were investigated. Seventy, PRRSV-free, 3-week old, pigs were allocated into 7 groups. Six groups were intramuscularly vaccinated with MLV, including Porcilis (PRRSV-1 MLV, MSD Animal Health, The Netherlands), Amervac (PRRSV-1 MLV, Laboratorios Hipra, Spain), Fostera (PRRSV-2 MLV, Zoetis, USA), Ingelvac PRRS MLV and Ingelvac PRRS ATP (PRRSV-2, Boehringer Ingelheim, USA), and Prime Pac PRRS (PRRSV-2 MLV, MSD Animal Health, The Netherlands). Unvaccinated pigs were left as control. Lymphocyte proliferative response, IL-10 and IFN-γ production were determined. At 35 days post-vaccination (DPV), all pigs were inoculated intranasally with 2 ml of each PRRSV-1 (105.4 TCID50/ml) and PRRSV-2 (105.2 TCID50/ml, HP-PRRSV). Following challenge, sera were quantitatively assayed for PRRSV RNA. Pigs were necropsied at 7 days post-challenge. Viremia, macro- and microscopic lung lesion together with PRRSV antigen presence were evaluated in lung tissues. The results demonstrated that, regardless of vaccine genotype, CMI induced by all MLVs was relatively slow. Increased production of IL-10 in all vaccinated groups was observed at 7 and 14 DPV. Pigs in Amervac, Ingelvac MLV and Ingelvac ATP groups had significantly higher levels of IL-10 compared to Porcilis, Fostera and Prime Pac groups at 7 and 14 DPV. Following challenge, regardless to vaccine genotype, vaccinated pigs had significantly lower lung lesion scores and PRRSV antigens than those in the control group. Both PRRSV-1 and PRRSV-2 RNA were significantly reduced. Prime Pac pigs had lowest PRRSV-1 and PRRSV-2 RNA in serum, and micro- and macroscopic lung lesion scores (p < 0.05) compared to other vaccinated groups. In conclusion, PRRSV MLVs, regardless of vaccine genotype, can reduce viremia and lung lesions following co-challenge with PRRSV-1 and PRRSV-2 (HP-PRRSV). The main difference between PRRSV MLV is the production of IL-10 following vaccination.

Project description:BackgroundIn 2020, a highly virulent PRRSV-1 strain emerged in Spain and rapidly spread across the country. The purpose of the present study was to test in a piglet model whether a commercial PRRSV-1 modified live vaccine was able to confer protection against strain R1, a representative of the emerging clade. For that purpose, two groups of 26 piglets were either vaccinated intradermally or kept as controls; 42 days later, half of the animals in each group were intranasally challenged with the R1 strain. Then, animals were followed to assess the development of clinical signs (until 14 days post-challenge), lung lesions (10- and 35-days post-challenge), weight gains, viremia and nasal shedding and the immune response (anti PRRS virus nucleoprotein antibodies) by ELISA and virus specific-interferon-γ secreting cells by ELISPOT).ResultsChallenge of naïve pigs resulted in high fever (up to 41.9 °C), lethargy and severely retarded growth (0.748 kg/day). In contrast, vaccinated/challenged pigs had less fever and for a shorter period, lower clinical scores and a higher average daily weight gain (0.940 kg/day), comparable to the unchallenged animals. At 10 days-post challenge, in naïve animals on average 49.1% of the lung was pneumonic (range 8-81%) while in vaccinated animals the average was 15.7% (4-41%). Duration of viremia was reduced in vaccinated animals and after 14 days post-challenge, most were negative by RT-qPCR. In contrast, 50% of the naïve/challenged pigs remained viremic at 35 days post-challenge. Vaccination induced rapid seroconversion and challenge of naïve animals resulted in 100% of ELISA-positive pigs by day 14 post-challenge. Regarding the development of IFN-γ responses, for vaccinated animals the frequencies increased until day 35 post-vaccination. After challenge, in vaccinated pigs, the peak of the R1-specific IFN-γ response was reached at 14 days and then the viremia ceased, although nasal shedding persisted in some vaccinated animals.ConclusionsIn the present trial, vaccination resulted in improved clinical course, better weight gain and reduced viremia. At the peak of the infection, lung lesions were reduced in most animals although some individuals still had extensive pneumonia. In summary, vaccination was shown to provide partial but significant protection against the highly virulent R1 strain.
