Constitutive expression of a pea apyrase, psNTP9, increases seed yield in field-grown soybean.

ABSTRACT: To address the demand for food by a rapidly growing human population, agricultural scientists have carried out both plant breeding and genetic engineering research. Previously, we reported that the constitutive expression of a pea apyrase (Nucleoside triphosphate, diphosphohydrolase) gene, psNTP9, under the control of the CaMV35S promoter, resulted in soybean plants with an expanded root system architecture, enhanced drought resistance and increased seed yield when they are grown in greenhouses under controlled conditions. Here, we report that psNTP9-expressing soybean lines also show significantly enhanced seed yields when grown in multiple different field conditions at multiple field sites, including when the gene is introgressed into elite germplasm. The transgenic lines have higher leaf chlorophyll and soluble protein contents and decreased stomatal density and cuticle permeability, traits that increase water use efficiency and likely contribute to the increased seed yields of field-grown plants. These altered properties are explained, in part, by genome-wide gene expression changes induced by the transgene.

Project description:Increased temperature means and fluctuations associated with climate change are predicted to exert profound effects on the seed yield of soybean. We conducted an experiment to evaluate the impacts of global warming on the phenology and yield of two determinate soybean cultivars in a temperate region (37.27°N, 126.99°E; Suwon, South Korea). These two soybean cultivars, Sinpaldalkong [maturity group (MG) IV] and Daewonkong (MG VI), were cultured on various sowing dates within a four-year period, under no water-stress conditions. Soybeans were kept in greenhouses controlled at the current ambient temperature (AT), AT+1.5°C, AT+3.0°C, and AT+5.0°C throughout the growth periods. Growth periods (VE-R7) were significantly prolonged by the elevated temperatures, especially the R1-R5 period. Cultivars exhibited no significant differences in seed yield at the AT+1.5°C and AT+3.0°C treatments, compared to AT, while a significant yield reduction was observed at the AT+5.0°C treatment. Yield reductions resulted from limited seed number, which was due to an overall low numbers of pods and seeds per pod. Heat stress conditions induced a decrease in pod number to a greater degree than in seed number per pod. Individual seed weight exhibited no significant variation among temperature elevation treatments; thus, seed weight likely had negligible impacts on overall seed yield. A boundary line analysis (using quantile regression) estimated optimum temperatures for seed number at 26.4 to 26.8°C (VE-R5) for both cultivars; the optimum temperatures (R5-R7) for single seed weight were estimated at 25.2°C for the Sinpaldalkong smaller-seeded cultivar, and at 22.3°C for the Daewonkong larger-seeded cultivar. The optimum growing season (VE-R7) temperatures for seed yield, which were estimated by combining the two boundary lines for seed number and seed weight, were 26.4 and 25.0°C for the Sinpaldalkong and Daewonkong cultivars, respectively. Considering the current soybean growing season temperature, which ranges from 21.7 (in the north) to 24.6°C (in the south) in South Korea, and the temperature response of potential soybean yields, further warming of less than approximately 1°C would not become a critical limiting factor for soybean production in South Korea.

Project description:Key messageSimulations demonstrated that estimates of realized genetic gain from linear mixed models using regional trials are biased to some degree. Thus, we recommend multiple selected models to obtain a range of reasonable estimates. Genetic improvements of discrete characteristics are obvious and easy to demonstrate, while quantitative traits require reliable and accurate methods to disentangle the confounding genetic and non-genetic components. Stochastic simulations of soybean [Glycine max (L.) Merr.] breeding programs were performed to evaluate linear mixed models to estimate the realized genetic gain (RGG) from annual multi-environment trials (MET). True breeding values were simulated under an infinitesimal model to represent the genetic contributions to soybean seed yield under various MET conditions. Estimators were evaluated using objective criteria of bias and linearity. Covariance modeling and direct versus indirect estimation-based models resulted in a substantial range of estimated values, all of which were biased to some degree. Although no models produced unbiased estimates, the three best-performing models resulted in an average bias of [Formula: see text] kg/ha[Formula: see text]/yr[Formula: see text] ([Formula: see text] bu/ac[Formula: see text]/yr[Formula: see text]). Rather than relying on a single model to estimate RGG, we recommend the application of several models with minimal and directional bias. Further, based on the parameters used in the simulations, we do not think it is appropriate to use any single model to compare breeding programs or quantify the efficiency of proposed new breeding strategies. Lastly, for public soybean programs breeding for maturity groups II and III in North America, the estimated RGG values ranged from 18.16 to 39.68 kg/ha[Formula: see text]/yr[Formula: see text] (0.27-0.59 bu/ac[Formula: see text]/yr[Formula: see text]) from 1989 to 2019. These results provide strong evidence that public breeders have significantly improved soybean germplasm for seed yield in the primary production areas of North America.

Project description:Crop yield is a highly complex quantitative trait. Historically, successful breeding for improved grain yield has led to crop plants with improved source capacity, altered plant architecture, and increased resistance to abiotic and biotic stresses. To date, transgenic approaches towards improving crop grain yield have primarily focused on protecting plants from herbicide, insects, or disease. In contrast, we have focused on identifying genes that, when expressed in soybean, improve the intrinsic ability of the plant to yield more. Through the large scale screening of candidate genes in transgenic soybean, we identified an Arabidopsis thaliana B-box domain gene (AtBBX32) that significantly increases soybean grain yield year after year in multiple transgenic events in multi-location field trials. In order to understand the underlying physiological changes that are associated with increased yield in transgenic soybean, we examined phenotypic differences in two AtBBX32-expressing lines and found increases in plant height and node, flower, pod, and seed number. We propose that these phenotypic changes are likely the result of changes in the timing of reproductive development in transgenic soybean that lead to the increased duration of the pod and seed development period. Consistent with the role of BBX32 in A. thaliana in regulating light signaling, we show that the constitutive expression of AtBBX32 in soybean alters the abundance of a subset of gene transcripts in the early morning hours. In particular, AtBBX32 alters transcript levels of the soybean clock genes GmTOC1 and LHY-CCA1-like2 (GmLCL2). We propose that through the expression of AtBBX32 and modulation of the abundance of circadian clock genes during the transition from dark to light, the timing of critical phases of reproductive development are altered. These findings demonstrate a specific role for AtBBX32 in modulating soybean development, and demonstrate the validity of expressing single genes in crops to deliver increased agricultural productivity.

Project description:BackgroundGenetic engineering has been effective in altering cell walls for biofuel production in the bioenergy crop, switchgrass (Panicum virgatum). However, regulatory issues arising from gene flow may prevent commercialization of engineered switchgrass in the eastern United States where the species is native. Depending on its expression level, microRNA156 (miR156) can reduce, delay, or eliminate flowering, which may serve to decrease transgene flow. In this unique field study of transgenic switchgrass that was permitted to flower, two low (T14 and T35) and two medium (T27 and T37) miR156-overexpressing 'Alamo' lines with the transgene under the control of the constitutive maize (Zea mays) ubiquitin 1 promoter, along with nontransgenic control plants, were grown in eastern Tennessee over two seasons.ResultsmiR156 expression was positively associated with decreased and delayed flowering in switchgrass. Line T27 did not flower during the 2-year study. Line T37 did flower, but not all plants produced panicles. Flowering was delayed in T37, resulting in 70.6% fewer flowers than controls during the second field year with commensurate decreased seed yield: 1205 seeds per plant vs. 18,539 produced by each control. These results are notable given that line T37 produced equivalent vegetative aboveground biomass to the controls. miR156 transcript abundance of field-grown plants was congruent with greenhouse results. The five miR156 SQUAMOSA PROMOTER BINDING PROTEIN-LIKE (SPL) target genes had suppressed expression in one or more of the transgenic lines. Line T27, which had the highest miR156 overexpression, showed significant downregulation for all five SPL genes. On the contrary, line T35 had the lowest miR156 overexpression and had no significant change in any of the five SPL genes.ConclusionsBecause of the research field's geographical features, this study was the first instance of any genetically engineered trait in switchgrass, in which experimental plants were allowed to flower in the field in the eastern U.S.; USDA-APHIS-BRS regulators allowed open flowering. We found that medium overexpression of miR156, e.g., line T37, resulted in delayed and reduced flowering accompanied by high biomass production. We propose that induced miR156 expression could be further developed as a transgenic switchgrass bioconfinement tool to enable eventual commercialization.
