Salvianolic Acid Modulates Physiological Responses and Stress-Related Genes That Affect Osmotic Stress Tolerance in Glycine max and Zea mays.

ABSTRACT: Drought is a serious threat worldwide to soybean and maize production. This study was conducted to discern the impact of salvianolic acid treatment on osmotic-stressed soybean (Glycine max L.) and maize (Zea mays L.) seedlings from the perspective of physiochemical and molecular reactions. Examination of varied salvianolic acid concentrations (0, 0.1, 1, 5, 10, and 25 μM) on soybean and maize seedling growth confirmed that the 0.1 and 1 μM concentrations, respectively, showed an improvement in agronomic traits. Likewise, the investigation ascertained how salvianolic acid application could retrieve osmotic-stressed plants. Soybean and maize seedlings were irrigated with water or 25% PEG for 8 days. The results indicated that salvianolic acid application promoted the survival of the 39-day-old osmotic-stressed soybean and maize plants. The salvianolic acid-treated plants retained high photosynthetic pigments, protein, amino acid, fatty acid, sugar, and antioxidant contents, and demonstrated low hydrogen peroxide and lipid contents under osmotic stress conditions. Gene transcription pattern certified that salvianolic acid application led to an increased expression of GmGOGAT, GmUBC2, ZmpsbA, ZmNAGK, ZmVPP1, and ZmSCE1d genes, and a diminished expression of GmMIPS2, GmSOG1, GmACS, GmCKX, ZmPIS, and ZmNAC48 genes. Together, our results indicate the utility of salvianolic acid to enhance the osmotic endurance of soybean and maize plants.

Project description:RNA helicases are enzymes that are thought to unwind double-stranded RNA molecules in an energy-dependent fashion through the hydrolysis of NTP. RNA helicases are associated with all processes involving RNA molecules, including nuclear transcription, editing, splicing, ribosome biogenesis, RNA export, and organelle gene expression. The involvement of RNA helicase in response to stress and in plant growth and development has been reported previously. While their importance in Arabidopsis and Oryza sativa has been partially studied, the function of RNA helicase proteins is poorly understood in Zea mays and Glycine max. In this study, we identified a total of RNA helicase genes in Arabidopsis and other crop species genome by genome-wide comparative in silico analysis. We classified the RNA helicase genes into three subfamilies according to the structural features of the motif II region, such as DEAD-box, DEAH-box and DExD/H-box, and different species showed different patterns of alternative splicing. Secondly, chromosome location analysis showed that the RNA helicase protein genes were distributed across all chromosomes with different densities in the four species. Thirdly, phylogenetic tree analyses identified the relevant homologs of DEAD-box, DEAH-box and DExD/H-box RNA helicase proteins in each of the four species. Fourthly, microarray expression data showed that many of these predicted RNA helicase genes were expressed in different developmental stages and different tissues under normal growth conditions. Finally, real-time quantitative PCR analysis showed that the expression levels of 10 genes in Arabidopsis and 13 genes in Zea mays were in close agreement with the microarray expression data. To our knowledge, this is the first report of a comparative genome-wide analysis of the RNA helicase gene family in Arabidopsis, Oryza sativa, Zea mays and Glycine max. This study provides valuable information for understanding the classification and putative functions of the RNA helicase gene family in crop growth and development.

Project description:Maize production and productivity are affected by drought stress in tropical and subtropical ecologies, as the majority of the area under maize cultivation in these ecologies is rain-fed. The present investigation was conducted to study the physiological and biochemical effects of 24-Epibrassinolide (EBR) as a plant hormone on drought tolerance in maize. Two maize hybrids, Vivek hybrid 9 and Bio 9637, were grown under three different conditions: (i) irrigated, (ii) drought, and (iii) drought+EBR. A total of 2 weeks before the anthesis, irrigation was discontinued to produce a drought-like condition. In the drought+EBR treatment group, irrigation was also stopped, and in addition, EBR was applied as a foliar spray on the same day in the drought plots. It was observed that drought had a major influence on the photosynthesis rate, membrane stability index, leaf area index, relative water content, and leaf water potential; this effect was more pronounced in Bio 9637. Conversely, the activities of antioxidant enzymes such as catalase (CAT), ascorbate peroxidase (APX), and superoxide dismutase (SOD) increased in both hybrids under drought conditions. Specifically, Vivek hybrid 9 showed 74% higher CAT activity under drought conditions as compared to the control. Additionally, EBR application further enhanced the activity of this enzyme by 23% compared to plants under drought conditions. Both hybrids experienced a significant reduction in plant girth due to drought stress. However, it was found that exogenously applying EBR reduced the detrimental effects of drought stress on the plant, and this effect was more pronounced in Bio 9637. In fact, Bio 9637 treated with EBR showed an 86% increase in proline content and a 70% increase in glycine betaine content compared to untreated plants under drought conditions. Taken together, our results suggested EBR enhanced tolerance to drought in maize hybrids. Hence, pre-anthesis foliar application of EBR might partly overcome the adverse effects of flowering stage drought in maize.

Project description:Maize (Zea mays L.) is the second most commonly produced and consumed crop after wheat globally and is adversely affected by high heat, which is a significant abiotic stress factor. This study was carried out to determine the physiological and biochemical responses of hybrid corn varieties under heat stress ('HS') compared to control ('C') conditions during the 2020 and 2021 growing seasons. The experiment was conducted under natural conditions in the Southeastern region of Turkey, where the most intense temperatures are experienced. This experiment used split plots in randomized blocks with three replications, with 'HS' and 'C' growing conditions applied to the main plots and the different hybrid corn varieties (FAO 650) planted on the sub plots. Mean values of days to 50% tasseling (DT, day), grain yield (GY, kg ha-1), leaf water potential (LWP, %), chlorophyll-a (Chl-a, mg g-1), cell membrane damage (CMD, %), and total phenol content (TPC, μg g-1) were significantly different between years, growing conditions, and hybrid corn varieties. Changes in the climate played a significant role in the differences between the years and growing conditions (GC), while the genetic characteristics of the different corn varieties explained the differences in outcomes between them. The values of DT, GY, LWP, Chl-a, CMD, and TPC ranged from 49.06-53.15 days, 9,173.0-10,807.2 kg ha-1, 78.62-83.57%, 6.47-8.62 mg g-1, 9.61-13.54%, and 232.36-247.01 μg g-1, respectively. Significant correlations were recorded between all the parameters. Positive correlations were observed between all the variables except for CMD. The increased damage to cell membranes under 'HS' caused a decrease in the other measured variables, especially GY. In contrast, the GY increased with decreased CMD. CMD was important in determining the stress and tolerance level of corn varieties under 'HS' conditions. The GY and other physiological parameters of ADA 17.4 and SYM-307 candidate corn varieties surpassed the control hybrid corn cultivars. The results revealed that the ADA 17.4 and SYM-307 cultivars might have 'HS'-tolerate genes.
