Neutral Red-carbon nanodots for selective fluorescent DNA sensing.

ABSTRACT: Carbon nanodots modified with Neutral Red covalently inserted in the nanostructure (NR-CDs) have been prepared by a simple synthesis method based on microwave irradiation under controlled temperature and pressure. The synthetized NR-CDs have been characterized by different techniques, demonstrating the covalent bonding of Neutral Red molecules to the carbon dots nanostructure. Fluorescence activity of the prepare NR-CDs has been explored showing different interaction pathways with singled and doubled stranded DNA. These studies have been successfully applied to develop a new fluorescence DNA hybridization assay to the detection of a specific DNA sequence of Escherichia coli bacteria.

Absorption and emission of light in red emissive carbon nanodots.

Project description:The structure-function relationship, especially the origin of absorption and emission of light in carbon nanodots (CNDs), has baffled scientists. The multilevel complexity arises due to the large number of by-products synthesized during the bottom-up approach. By performing systematic purification and characterization, we reveal the presence of a molecular fluorophore, quinoxalino[2,3-b]phenazine-2,3-diamine (QXPDA), in a large amount (∼80% of the total mass) in red emissive CNDs synthesized from o-phenylenediamine (OPDA), which is one of the well-known precursor molecules used for CND synthesis. The recorded NMR and mass spectra tentatively confirm the structure of QXPDA. The close resemblance of the experimental vibronic progression and the mirror symmetry of the absorption and emission spectra with the theoretically simulated spectra confirm an extended conjugated structure of QXPDA. Interestingly, QXPDA dictates the complete emission characteristics of the CNDs; in particular, it showed a striking similarity of its excitation independent emission spectra with that of the original synthesized red emissive CND solution. On the other hand, the CND like structure with a typical size of ∼4 nm was observed under a transmission electron microscope for a blue emissive species, which showed both excitation dependent and independent emission spectra. Interestingly, Raman spectroscopic data showed the similarity between QXPDA and the dot structure thus suggesting the formation of the QXPDA aggregated core structure in CNDs. We further demonstrated the parallelism in trends of absorption and emission of light from a few other red emissive CNDs, which were synthesized using different experimental conditions.

Super-strong and Intrinsically Fluorescent Silkworm Silk from Carbon Nanodots Feeding.

Project description:Fluorescent silk is fundamentally important for the development of future tissue engineering scaffolds. Despite great progress in the preparation of a variety of colored silks, fluorescent silk with enhanced mechanical properties has yet to be explored. In this study, we report on the fabrication of intrinsically super-strong fluorescent silk by feeding Bombyx mori silkworm carbon nanodots (CNDs). The CNDs were incorporated into silk fibroin, hindering the conformation transformation, confining crystallization, and inducing orientation of mesophase. The resultant silk exhibited super-strong mechanical properties with breaking strength of 521.9 ± 82.7 MPa and breaking elongation of 19.2 ± 4.3%, improvements of 55.1% and 53.6%, respectively, in comparison with regular silk. The CNDs-reinforced silk displayed intrinsic blue fluorescence when exposed to 405 nm laser and exhibited no cytotoxic effect on cells, suggesting that multi-functional silks would be potentially useful in bioimaging and other applications.

Carbon Nanodots with Nearly Unity Fluorescent Efficiency Realized via Localized Excitons.

Project description:Carbon nanodots (CDs) have emerged as an alternative option for traditional nanocrystals due to their excellent optical properties and low toxicity. Nevertheless, high emission efficiency is a long-lasting pursuit for CDs. Herein, CDs with near-unity emission efficiency are prepared via atomic condensation of doped pyrrolic nitrogen, which can highly localize the excited states thus lead to the formation of bound excitons and the symmetry break of the π-electron conjugation. The short radiative lifetimes (<8 ns) and diffusion lengths (<50 nm) of the CDs imply that excitons can be efficiently localized by radiative recombination centers for a defect-insensitive emission of CDs. By incorporating the CDs into polystyrene, flexible light-converting films with a high solid-state quantum efficiency of 84% and good resistance to water, heating, and UV light are obtained. With the CD-polymer films as light conversion layers, CD-based white light-emitting diodes (WLEDs) with a luminous efficiency of 140 lm W-1 and a flat-panel illumination system with lighting sizes of more than 100 cm2 are achieved, matching state-of-the-art nanocrystal-based LEDs. These results pave the way toward carbon-based luminescent materials for solid-state lighting technology.

Methylene Blue functionalized carbon nanodots combined with different shape gold nanostructures for sensitive and selective SARS-CoV-2 sensing.

Project description:The development of DNA-sensing platforms based on new synthetized Methylene Blue functionalized carbon nanodots combined with different shape gold nanostructures (AuNs), as a new pathway to develop a selective and sensitive methodology for SARS-CoV-2 detection is presented. A mixture of gold nanoparticles and gold nanotriangles have been synthetized to modify disposable electrodes that act as an enhanced nanostructured electrochemical surface for DNA probe immobilization. On the other hand, modified carbon nanodots prepared a la carte to contain Methylene Blue (MB-CDs) are used as electrochemical indicators of the hybridization event. These MB-CDs, due to their structure, are able to interact differently with double and single-stranded DNA molecules. Based on this strategy, target sequences of the SARS-CoV-2 virus have been detected in a straightforward way and rapidly with a detection limit of 2.00 aM. Moreover, this platform allows the detection of the SARS-CoV-2 sequence in the presence of other viruses, and also a single nucleotide polymorphism (SNPs). The developed approach has been tested directly on RNA obtained from nasopharyngeal samples from COVID-19 patients, avoiding any amplification process. The results agree well with those obtained by RT-qPCR or reverse transcription quantitative polymerase chain reaction technique.

Facile synthesis of dual-emission fluorescent carbon nanodots for a multifunctional probe.

Project description:In this study, we developed a facile method for synthesizing dual-emission carbon nanodots (CDs) through trimesic acid and o-phenylenediamine through electrolysis for 2 h. The synthesized CDs were mainly 3-7 nm in size, with an average size of 5.17 nm. The dual-emission fluorescent property of these CDs could be observed under two different excitation wavelengths. The green emission of the CDs could be quenched after the addition of mercury ions or copper ions, and the blue emission of the CDs could be inhibited using hydroxychloroquine (HCQ). Furthermore, the quenched fluorescence of CDs/Cu2+ could be recovered through the addition of glyphosate. We developed a multifunctional chemical sensor by using these special fluorescence materials. Under optimal conditions, the detection limits of mercury ions, glyphosate, and HCQ were 0.42 μM, 1.1 mg L-1, and 0.14 μM, respectively. Moreover, this method can be used to detect mercury ions, glyphosate, and HCQ in environmental water, cereals, and urine samples, respectively.

Efficient Red/Near-Infrared-Emissive Carbon Nanodots with Multiphoton Excited Upconversion Fluorescence.

Project description:Red/near-infrared (NIR) emissive carbon nanodots (CNDs) with photoluminescence (PL) quantum yield (QY) of 57% are prepared via an in situ solvent-free carbonization strategy for the first time. 1-Photon and 2-photon cellular imaging is demonstrated by using the CNDs as red/NIR fluorescence agent due to the high PL QY and low biotoxicity. Further study shows that the red/NIR CNDs exhibit multiphoton excited (MPE) upconversion fluorescence under excitation of 800-2000 nm, which involves three NIR windows (NIR-I, 650-950 nm; NIR-II, 1100-1350; NIR-III, 1600-1870 nm). 2-Photon, 3-photon, and 4-photon excited fluorescence of the CNDs under excitation of different wavelengths is achieved. This study develops an in situ solvent-free carbonization method for efficient red/NIR emissive CNDs with MPE upconversion fluorescence, which may push forward the application of the CNDs in bioimaging.

Intracellular ratiometric temperature sensing using fluorescent carbon dots.

Project description:Highly sensitive non-invasive temperature sensing is critical for studying fundamental biological processes and applications in medical diagnostics. Nanoscale-based thermometers are promising non-invasive probes for precise temperature sensing with subcellular resolution. However, many of these systems have limitations as they rely on fluorescence intensity changes, deconvolution of peaks, or the use of hybrid systems to measure thermal events. To address this, we developed a fluorescence-based ratiometric temperature sensing approach using carbon dots prepared via microwave synthesis. These dots possess dual fluorescence signatures in the blue and red regions of the spectrum. We observed a linear response as a function of temperature in the range of 5-60 °C with a thermal resolution of 0.048 K-1 and thermal sensitivity of 1.97% C-1. Temperature-dependent fluorescence was also observed in HeLa cancer cells over a range of 32-42 °C by monitoring changes in the red-to-blue fluorescence signatures. We demonstrate that the ratiometric approach is superior to intensity-based thermal sensing because it is independent of the intracellular concentration of the optical probe. These findings suggest that dual-emitting carbon dots can be an effective tool for in vitro and possibly in vivo fluorescence nanothermometry.

Ultraviolet phosphorescent carbon nanodots.

Project description:Phosphorescent carbon nanodots (CNDs) have generated enormous interest recently, and the CND phosphorescence is usually located in the visible region, while ultraviolet (UV) phosphorescent CNDs have not been reported thus far. Herein, the UV phosphorescence of CNDs was achieved by decreasing conjugation size and in-situ spatial confinement in a NaCNO crystal. The electron transition from the px to the sp2 orbit of the N atoms within the CNDs can generate one-unit orbital angular momentum, providing a driving force for the triplet excitons population of the CNDs. The confinement caused by the NaCNO crystal reduces the energy dissipation paths of the generated triplet excitons. By further tailoring the size of the CNDs, the phosphorescence wavelength can be tuned to 348 nm, and the room temperature lifetime of the CNDs can reach 15.8 ms. As a demonstration, the UV phosphorescent CNDs were used for inactivating gram-negative and gram-positive bacteria through the emission of their high-energy photons over a long duration, and the resulting antibacterial efficiency reached over 99.9%. This work provides a rational design strategy for UV phosphorescent CNDs and demonstrates their novel antibacterial applications.

Fluorescent calix[4]triazole for selective fluoride anion sensing.

Project description:Fluoride ions (F-) play an important role in preventing cavities and treating osteoporosis, but excessive exposure can lead to serious health problems such as fluorosis and kidney damage. These dual characteristics highlight the need for selective and sensitive methods to detect fluoride ions for health monitoring. Accordingly, in this study, we investigated the anion-binding ability of Py-CT4, a fluorescent chemosensor in which pyrene is linked to calix[4]triazole via an ester linker. Notably, Py-CT4 exhibited significant fluorescence quenching for F- compared to other anions, and its fluorescence intensity gradually decreased with increasing F- concentration. This phenomenon is driven by electron transfer from calix[4]triazole to pyrene, initiated by hydrogen bonding with F- and followed by F--induced deprotonation of calix[4]triazole. The selectivity of Py-CT4 for F- appears to stem from its relatively flexible structure and low acidity compared to the previously reported Py-CT4+. Py-CT4 thus represents the first macrocyclic receptor based on charge-neutral 1,2,3-triazole that selectively recognizes F- through fluorescence quenching. Compared to traditional detection methods, Py-CT4 utilizes the advantages of fluorescent detection, such as higher sensitivity, faster response times, and ease of use, for fluoride ion detection. Py-CT4 also demonstrates excellent selectivity for F- even in the presence of competing anions. These features make Py-CT4 a promising tool for monitoring fluoride ions in biological and environmental systems, providing valuable insights into public health and safety.

Investigation into Red Emission and Its Applications: Solvatochromic N-Doped Red Emissive Carbon Dots with Solvent Polarity Sensing and Solid-State Fluorescent Nanocomposite Thin Films.

Project description:In this work, a NIR emitting dye, p-toluenesulfonate (IR-813) was explored as a model precursor to develop red emissive carbon dots (813-CD) with solvatochromic behavior with a red-shift observed with increasing solvent polarity. The 813-CDs produced had emission peaks at 610 and 698 nm, respectively, in water with blue shifts of emission as solvent polarity decreased. Subsequently, 813-CD was synthesized with increasing nitrogen content with polyethyleneimine (PEI) to elucidate the change in band gap energy. With increased nitrogen content, the CDs produced emissions as far as 776 nm. Additionally, a CD nanocomposite polyvinylpyrrolidone (PVP) film was synthesized to assess the phenomenon of solid-state fluorescence. Furthermore, the CDs were found to have electrochemical properties to be used as an additive doping agent for PVP film coatings.
