Extended the depth of field and zoom microscope with varifocal lens.

ABSTRACT: Extending the depth of field (DOF) is especially essential in thick and 3D sample imaging. However, it's difficult to achieve both large DOF and high resolution in a zoom microscope. Currently, the use of optical sectioning to expand DOF still has the problem of inconstant magnification. Here, we develop an extended the depth of field (EDOF) and zoom microscope, which can realize EDOF with constant magnification and high resolution. Besides, the proposed microscope can achieve optical axial scanning at different NA and magnifications in real time without any mechanical movement. The proposed varifocal lens is employed to realize optical axial scanning, zooming, and keeping constant magnification when extending the DOF. Experimental results show that the proposed microscope can realize a continuous optical zoom of 10-40×, NA from 0.14 to 0.54, and the DOF of microscope can be extended to 1.2 mm.

Deep learning extended depth-of-field microscope for fast and slide-free histology.

Project description:Microscopic evaluation of resected tissue plays a central role in the surgical management of cancer. Because optical microscopes have a limited depth-of-field (DOF), resected tissue is either frozen or preserved with chemical fixatives, sliced into thin sections placed on microscope slides, stained, and imaged to determine whether surgical margins are free of tumor cells-a costly and time- and labor-intensive procedure. Here, we introduce a deep-learning extended DOF (DeepDOF) microscope to quickly image large areas of freshly resected tissue to provide histologic-quality images of surgical margins without physical sectioning. The DeepDOF microscope consists of a conventional fluorescence microscope with the simple addition of an inexpensive (less than $10) phase mask inserted in the pupil plane to encode the light field and enhance the depth-invariance of the point-spread function. When used with a jointly optimized image-reconstruction algorithm, diffraction-limited optical performance to resolve subcellular features can be maintained while significantly extending the DOF (200 µm). Data from resected oral surgical specimens show that the DeepDOF microscope can consistently visualize nuclear morphology and other important diagnostic features across highly irregular resected tissue surfaces without serial refocusing. With the capability to quickly scan intact samples with subcellular detail, the DeepDOF microscope can improve tissue sampling during intraoperative tumor-margin assessment, while offering an affordable tool to provide histological information from resected tissue specimens in resource-limited settings.

A lab-on-phone instrument with varifocal microscope via a liquid-actuated aspheric lens (LAL).

Project description:In this paper, we introduce a novel concept of liquid-actuated aspheric lens (LAL) with a built-in aspheric polydimethylsiloxane lens (APL) to enable the design of compact optical systems with varifocal microscopic imaging. The varifocal lens module consists of a sandwiched structures such as 3d printed syringe pump functionally serves as liquid controller. Other key components include two acrylic cylinders, a rigid separator, a APL/membrane composite (APLMC) embedded PDMS membrane. In functional operation, the fluidic controller was driven to control the pressure difference and ALPMC deformation. The focal length can be changed through the pressure difference. This is achieved by the adjustment of volume change of injected liquid such that a widely tunable focal length. The proposed LAL can transform to 3 modes: microscopic mode (APLMC only), convex-concave mode and biconcave mode. It is noticeable that LAL in the operation of microscopic mode is tunable in focus via the actuation of APLMC (focal length is from 4.3 to 2.3 mm and magnification 50X) and can rival the images quality of commercial microscopes. A new lab-on-phone device is economically feasible and functionally versatile to offer a great potential in the point of care applications.

Alvarez varifocal X-ray lens.

Project description:Visible light optical elements such as lenses and mirrors have counterparts for X-rays. In the visible regime, a variable focusing power can be achieved by an Alvarez lens which consists of a pair of inline planar refractors with a cubic thickness profile. When the two refractors are laterally displaced in opposite directions, the parabolic component of the wavefront is changed resulting in a longitudinal displacement of the focus. This paper reports an implementation of this concept for X-rays using two planar microfabricated refractive elements. The Alvarez X-ray lens can vary the focal distance of an elliptical X-ray mirror or a planar compound X-ray lens over several millimetres. The study presents the first demonstration of an Alvarez X-ray lens which adaptively corrects defocus and astigmatism aberrations of X-ray optics. In addition, the Alvarez X-ray lens eliminates coma aberration in an elliptical mirror, to the lowest order, when combining the lens with an adjustment of the pitch angle of the mirror.

Triboelectric effect-modulated varifocal liquid lens.

Project description:Electrically modulated varifocal liquid lenses, which are usually modulated by an external high voltage power source, have attracted much attention due to their bright application prospects in artificial optical systems. Here, a triboelectric nanogenerator (TENG)-based varifocal liquid lens (TVLL) has been demonstrated, in which the focal length can be directly modulated by external mechanical sliding. A dielectrophoretic force is generated by the TENG through the transfer of triboelectric charges in the asymmetric electrodes, which is used to continuously change the shape of the air-liquid interface between concave and convex without any complicated boost converter. Moreover, a triboelectric magnifying glass based on the accurate modulation effect of the TVLL on a light beam has been demonstrated. In this work, the TENG is used as a medium to modulate and accurately control the focal length of the liquid lens by an external mechanical stimulus, which may have great applications in micro-optical-electro-mechanical systems (MOEMS), human-machine interaction, artificial vision systems, etc.

E2E-BPF microscope: extended depth-of-field microscopy using learning-based implementation of binary phase filter and image deconvolution.

Project description:Several image-based biomedical diagnoses require high-resolution imaging capabilities at large spatial scales. However, conventional microscopes exhibit an inherent trade-off between depth-of-field (DoF) and spatial resolution, and thus require objects to be refocused at each lateral location, which is time consuming. Here, we present a computational imaging platform, termed E2E-BPF microscope, which enables large-area, high-resolution imaging of large-scale objects without serial refocusing. This method involves a physics-incorporated, deep-learned design of binary phase filter (BPF) and jointly optimized deconvolution neural network, which altogether produces high-resolution, high-contrast images over extended depth ranges. We demonstrate the method through numerical simulations and experiments with fluorescently labeled beads, cells and tissue section, and present high-resolution imaging capability over a 15.5-fold larger DoF than the conventional microscope. Our method provides highly effective and scalable strategy for DoF-extended optical imaging system, and is expected to find numerous applications in rapid image-based diagnosis, optical vision, and metrology.

Extended depth of field in augmented reality.

Project description:The 3D display device shows an image with depth information. Conventional 3D display devices based on binocular parallax can focus accurately only on the depth of a specific screen. Because the human eye has a narrow depth of field (DOF) under normal circumstances, 3D displays that provide a relatively wide range of virtual depth areas have limitations on the DOF where clear 3D images are seen. To resolve this problem, it is necessary to find the optical conditions to extend the DOF and analyze the phenomena related to it. For this, by using the Rayleigh criterion and the Strehl ratio, a criterion for this extension of the DOF is suggested. A practical optical structure that can effectively extend the DOF is devised using a flat panel display. This optical structure could be applied to AR, VR, and MR in the field of near-eye displays. From the results of this research, the fundamental optical conditions and standards are proposed for 3D displays that will provide 3D images with extended DOF in the future. Furthermore, it is also expected that these conditions and criteria can be applied to optical designs for the required performance in the development of 3D displays in various fields.

Large depth-of-field ultra-compact microscope by progressive optimization and deep learning.

Project description:The optical microscope is customarily an instrument of substantial size and expense but limited performance. Here we report an integrated microscope that achieves optical performance beyond a commercial microscope with a 5×, NA 0.1 objective but only at 0.15 cm3 and 0.5 g, whose size is five orders of magnitude smaller than that of a conventional microscope. To achieve this, a progressive optimization pipeline is proposed which systematically optimizes both aspherical lenses and diffractive optical elements with over 30 times memory reduction compared to the end-to-end optimization. By designing a simulation-supervision deep neural network for spatially varying deconvolution during optical design, we accomplish over 10 times improvement in the depth-of-field compared to traditional microscopes with great generalization in a wide variety of samples. To show the unique advantages, the integrated microscope is equipped in a cell phone without any accessories for the application of portable diagnostics. We believe our method provides a new framework for the design of miniaturized high-performance imaging systems by integrating aspherical optics, computational optics, and deep learning.

Assessment of imaging with extended depth-of-field by means of the light sword lens in terms of visual acuity scale.

Project description:We present outcomes of an imaging experiment using the refractive light sword lens (LSL) as a contact lens in an optical system that serves as a simplified model of the presbyopic eye. The results show that the LSL produces significant improvements in visual acuity of the simplified presbyopic eye model over a wide range of defocus. Therefore, this element can be an interesting alternative for the multifocal contact and intraocular lenses currently used in ophthalmology. The second part of the article discusses possible modifications of the LSL profile in order to render it more suitable for fabrication and ophthalmological applications.

Visual performance of an extended depth of focus intraocular lens for treatment selection.

Project description:PurposeTo evaluate the visual performance of an extended depth of focus (EDOF) intraocular lens (IOL) for patient selection.MethodsThis prospective non-comparative case series evaluated fifty-two eyes (26 patients) after bilateral implantation of the Tecnis Symfony IOL. Exclusion criteria were previous ocular surgeries, corneal astigmatism of >1.5 diopter, and ocular pathologies or corneal abnormalities. Uncorrected (UCVA) and distance-corrected (DCVA) visual acuity (VA) in 4 m, 80 cm, 60 cm and 40 cm (logMAR), defocus curve, contrast sensitivity (CS) and a questionnaire on subjective quality of vision (QoV) were assessed after 3 months.ResultsUCVA was -0.02 ± 0.071 logMAR at 4 m, 0.01 ± 0.077 logMAR at 80 cm, 0.08 ± 0.187 logMAR at 60 cm and 0.22 ± 0.153 logMAR at 40 cm. Defocus curve testing showed a flat VA range from 0.00 to -1.50 D (-0.05-0.02 logMAR). At near distance from -2.0 to -2.5 D a dip of DCVA was observed (0.14-0.28 logMAR). CS under photopic and mesopic conditions without and with glare was 1.56 logCS, 0.86 logCS, 0.78 logCS, respectively. Spectacle independence was achieved in 100% for distance and intermediate vision and 71% of patients at near distance.ConclusionThis EDOF IOL provides excellent intermediate and far (<0.1 logMAR) and acceptable near UCVA (<0.3 logMAR). Defocus curve testing confirmed an EDOF between far and intermediate distance and showed a dip at near distance. It provides good QoV, CS, high spectacle independence, perception of few optical phenomena, and particularly good night-driving capabilities.

Cases of replacing diffractive bifocal intraocular lens with extended depth of focus intraocular lens due to waxy vision.

Project description:PurposeTo investigate the postoperative course of patients who explanted a diffractive bifocal intraocular lens (IOL) due to waxy vision and implanted with an extended depth of focus IOL.MethodsThis study evaluated 29 eyes of 25 patients who underwent diffractive bifocal IOL explantation followed by TECNIS Symfony® implantation because of dissatisfaction due to waxy vision at the Takabatake West Eye Clinic between January 2018 and November 2019. The indication criteria for this surgery were patients with uncorrected distance visual acuity of 0.05 logMAR or better, without eye diseases that may affect visual function, and no dissatisfactions about photic phenomena. We investigated patient demographics, uncorrected and corrected visual acuity, manifest refraction, contrast sensitivity, subjective symptoms, time to IOL explantation, explanted IOL type, and spectacle independence.ResultsThe time to the IOL exchange after the initial IOL implantation was 55.3 ± 50.4 days (range: 14-196 days). The logMAR corrected distance visual acuity before and after IOL exchange were -0.13 ± 0.06 and -0.14 ± 0.06, respectively (p = 0.273). After IOL exchange surgery, the area under log contrast sensitivity function increased significantly from 1.07 ± 0.12 to 1.21 ± 0.12 (p < 0.001), and the waxy vision symptoms improved. The spectacle independence rate at the last visit was 88.0%.ConclusionFor patients who complain of waxy vision despite good visual acuity after diffractive bifocal IOL implantation, exchange to extended depth of focus IOL was considered one of the useful surgical options.
