In silico approaches for drug repurposing in oncology: Protocol for a scoping review of existing evidence.

ABSTRACT: Drug repurposing has been applied in the biomedical field to optimize the use of existing drugs, leading to a more efficient allocation of research resources. In oncology, this approach is particularly interesting, considering the high cost related to the discovery of new drugs with therapeutic potential. Computational methods have been applied to predict associations between drugs and their targets. However, drug repurposing has not always been promising and its efficiency has yet to be proven. Therefore, the present scoping review protocol was developed to screen the literature on how in silico strategies can be implemented in drug repurposing in oncology. The scoping review will be conducted according to the Arksey and O'Malley framework (2005) and the Joanna Briggs Institute recommendations. We will search the PubMed/MEDLINE, Embase, Scopus, and Web of Science databases, as well as the grey literature. We will include peer-reviewed research articles involving in silico strategies applied to drug repurposing in oncology, published between January 1, 2003, and December 31, 2021. Data will be charted and findings described according to review questions. We will report the scoping review using the Preferred Reporting Items for Systematic Reviews and Meta-Analyses Extension for Scoping Review guidelines (PRISMA-ScR).

Project description:IntroductionThe peripheral vascular catheter (PVC) is the most common intravenous medical device used in hospitals, given it is crucial for the delivery of intravenous therapies. Despite its widespread use, PVC complications such as occlusion, infiltration, extravasation, phlebitis, thrombophlebitis, catheter-related thrombosis and catheter-related bloodstream infection significantly affect patient safety, leading to increased morbidity and healthcare costs. One approach to maintain PVC patency and therefore device longevity is to flush the PVC. This usually involves the use of 5 to 10 mL syringes with normal saline. The evidence on PVC flushing practices is dispersed, making it difficult to draw clear conclusions about current practices and clinical implications. A scoping review is needed to explore and synthesise this evidence systematically and to systematically map the existing research on PVC flushing by assessing the scope, methodologies and outcomes of existing studies.Methods and analysisThe review will adhere to the Joanna Briggs Institute (JBI) methodology, using a three-phase search strategy as recommended by JBI and covering the full-time range available in each database. Systematic searches will be conducted across Embase, Web of Science, PubMed, Scopus, and CINAHL; clinical trial registries; and grey literature sources to identify potentially eligible studies published up to May 2024. Following duplicate removal, 3021 records will remain for screening. Two independent reviewers will screen the records and extract data based on the review questions. The reporting will follow the Preferred Reporting Items for Systematic Reviews and Meta-Analyses extension for scoping reviews, with the results and discussion structured according to the Patterns, Advances, Gaps, Evidence for practice and Research recommendations framework.Ethics and disseminationEthics approval is not required for this study. The findings will be shared through professional networks, presented at conferences and published in a scientific journal.

Project description:IntroductionDespite the availability of clinical practice guidelines for cancer symptom management, cancer care providers do not consistently use them in practice. Oncology nurses in outpatient settings are well positioned to use established guidelines to inform symptom assessment and management; however, issues concerning inconsistent implementation persist. This scoping review aims to (1) identify reported barriers and facilitators influencing symptom management guideline adoption, implementation and sustainability among specialised and advanced oncology nurses in cancer-specific outpatient settings and (2) identify and describe the components of strategies that have been used to enhance the implementation of symptom management guidelines.Methods and analysisThis scoping review will follow Joanna Briggs Institute methodology. Electronic databases CINAHL, Embase, Emcare and MEDLINE(R) and grey literature sources will be searched for studies published in English from January 2000 to March 2022. Primary studies and grey literature reports of any design that include specialised or advanced oncology nurses practicing in cancer-specific outpatient settings will be eligible. Sources describing factors influencing the adoption, implementation and sustainability of cancer symptom management guidelines and/or strategies to enhance guideline implementation will be included. Two reviewers will independently screen for eligibility and extract data. Data extraction of factors influencing implementation will be guided by the Consolidated Framework for Implementation Research (CFIR), and the seven dimensions of implementation strategies (ie, actors, actions, targets, temporality, dose, justifications and outcomes) will be used to extract implementation strategy components. Factors influencing implementation will be analysed descriptively, synthesised according to CFIR constructs and linked to the Expert Recommendations for Implementating Change strategies. Results will be presented through tabular/diagrammatic formats and narrative summary.Ethics and disseminationEthics approval is not required for this scoping review. Planned knowledge translation activities include a national conference presentation, peer-reviewed publication, academic social media channels and dissemination within local oncology nursing and patient networks.
