Variation in the Content of Bioactive Compounds in Infusions Prepared from Different Parts of Wild Polish Stinging Nettle (Urtica dioica L.).

ABSTRACT: Nettle is a common plant that offers many health benefits and is grown all over the world. The content of active compounds in roots, stems, and leaves was determined based on the extraction procedure optimized using the Central Composite Design. Flavonols, phenolic acids, trigonelline, nicotinamide, nicotinic acids, and short-chain organic acids were determined with the use of LC-MS/MS and capillary isotachophoresis. Trigonelline, which was not previously reported in the roots and stems of nettle, was found in all parts of the plant and considerable variations in its content were observed (2.8-108 µg g^-1). Furthermore, the Principal Component Analysis taking into account more variables demonstrated differences in the content of bioactive components between roots and aerial parts of nettle.

Insights into Lignan Composition and Biosynthesis in Stinging Nettle (Urtica dioica L.).

Project description:Stinging nettle (Urtica dioica L.) has been used as herbal medicine to treat various ailments since ancient times. The biological activity of nettle is chiefly attributed to a large group of phenylpropanoid dimers, namely lignans. Despite the pharmacological importance of nettle lignans, there are no studies addressing lignan biosynthesis in this plant. We herein identified 14 genes encoding dirigent proteins (UdDIRs) and 3 pinoresinol-lariciresinol reductase genes (UdPLRs) in nettle, which are two gene families known to be associated with lignan biosynthesis. Expression profiling of these genes on different organs/tissues revealed a specific expression pattern. Particularly, UdDIR7, 12 and 13 displayed a remarkable high expression in the top internode, fibre tissues of bottom internodes and roots, respectively. The relatively high expression of UdPLR1 and UdPLR2 in the young internodes, core tissue of bottom internode and roots is consistent with the high accumulation of lariciresinol and secoisolariciresinol in these tissues. Lignan quantification showed a high abundance of pinoresinol in roots and pinoresinol diglucosides in young internodes and leaves. This study sheds light on lignan composition and biosynthesis in nettle, providing a good basis for further functional analysis of DIRs and PLRs and, ultimately, engineering lignan metabolism in planta and in cell cultures.

An assessment of the evidence for antibacterial activity of stinging nettle (Urtica dioica) extracts.

Project description:Stinging nettles (Urtica spp.) have been used in a diverse range of traditional and historical medicines from around the world for the treatment of skin diseases, wounds, urinary disorders, respiratory diseases, bone and joint pain, anaemia and other circulatory problems, as well as in cosmetic preparations for skin and haircare. As part of an interdisciplinary exploration of nettle-based remedies, we performed a systematic review of published evidence for the antimicrobial activity of Urtica spp. extracts against bacteria and fungi that commonly cause skin, soft tissue and respiratory infections. We focussed on studies in which minimum inhibitory concentration (MIC) assays of U. dioica were conducted on the common bacterial opportunistic pathogens Escherichia coli , Pseudomonas aeruginosa , Klebsiella pneumoniae and Staphylococcus aureus . No studies used fresh leaves (all were dried prior to use), and no studies prepared nettles in weak acid (corresponding to vinegar) or in fats/oils, which are common combinations in historical and traditional preparations. We addressed this gap by conducting new antibacterial tests of extracts of fresh U. dioica leaves prepared in vinegar, butter or olive oil against P. aeruginosa and S. aureus . Our systematic review and additional experimental data leads us to conclude that there is no strong evidence for nettles containing molecules with clinically useful antimicrobial activity. It seems most likely that the utility of nettles in traditional topical preparations for wounds may simply be as a 'safe' absorbent medium for keeping antibacterial (vinegar) or emollient (oils) ingredients at the treatment site.

Stinging Nettle (Urtica dioica) Roots: The Power Underground-A Review.

Project description:Stinging nettle (Urtica dioica) is an herbaceous perennial plant native to Eurasia, wildly distributed throughout the temperate parts of the world. Although generally considered as a weed due to its fast growth and invasive capacity, stinging nettle is well suited to cultivation and is currently experiencing a revival as a beneficial crop due to its numerous potential applications. This interest reflects in an increasing number of scientific articles related to nettle in the last years. However, reports mostly focus on the aerial parts of the plant. Roots are rich in numerous phytochemicals such as phytosterols, lignans, coumarins, sugars, and lectins. By compiling the most relevant publications, the aim of this review is to gather the current knowledge about nettle roots, such as root system functioning, biochemical composition, and related functional activities. A special emphasis is placed on lectins (or UDA for Urtica dioica agglutinin) due to their functional activities. This review highlights the potential of nettle root as a source of biomolecules. Gaps of knowledge and possible future directions for nettle root research, production, and uses are discussed.

Wild Stinging Nettle (Urtica dioica L.) Leaves and Roots Chemical Composition and Phenols Extraction.

Project description:Stinging nettle (Urtica dioica L.) is an herbaceous plant that grows all over the world and is widely used as an edible and medicinal plant. Overall research results reveal that the chemical content and antioxidant activity of aerial parts and roots of stinging nettle depends on the growing region, soil, meteorological conditions (especially sunshine), collecting time, etc. The chemical composition of stinging nettle growing in Lithuania and the solid-liquid extraction efficiency of leaves and roots using different solvents were analysed. Additionally, we determined leaves phenols extraction efficiency using 96% methanol at different extraction conditions. Research results showed that a higher amount of crude fats, non-nitrogen extractives, and total carotenoids were in leaves, but the amount of crude proteins and ash did not differ significantly compared with roots. A higher amount of polyunsaturated fatty acids (PUFAs) and monounsaturated fatty acids (MUFAs) were detected in roots instead of leaves while saturated fatty acids (SFAs) were in leaves. The extraction results showed that the most effective solvent for total phenols and flavonoids in leaves was 96% methanol, for total phenols in roots was 50% methanol and 50% ethanol for total flavonoids in roots. The most effective temperature for the Urtica dioica L. leaves phenols extraction was 70 °C, while time does not have a significant influence. The present study's findings suggested that concentrated and binary solvents had different effects on the phenol's extraction efficiency from different stinging nettle parts and extraction temperature performed a key role instead of extraction time.

Urtica dioica (Stinging Nettle): A Neglected Plant With Emerging Growth Promoter/Immunostimulant Properties for Farmed Fish.

Project description:Urtica dioica (stinging nettle), is a perennial plant belonging to the family of Urticaceae, genus Urtica. Despite the use of nettle in folk veterinary medicine is well documented, U. dioica is today an underestimated and frequently neglected plant, considered by the contemporary agriculture as a weed to be eliminated. This mini review focus on very recent studies on dietary administration of U. dioica, both as a single herb or in combination with other herbs, to enhance growth and stimulate farmed fish immunity, thus enabling the fish to be more resistant against bacterial infections. Such an emerging feature, together with cost-effectiveness, adequate availability, and easy processing of nettle, could make this herb an excellent, inexpensive and widely used dietary supplement on intensive fish farms.

Aqueous extracts of Urtica dioica (stinging nettle) leaf contain a P2-purinoceptor antagonist-Implications for male fertility.

Project description:Stinging nettle root and leaf extracts were tested for their effect on prostatic smooth muscle contractility. Root extract did not affect electrical field stimulation induced-nerve mediated contractions of isolated rat prostates. On the other hand, leaf extract attenuated electrical field stimulation-induced contractions at all frequencies. Similarly, contractions elicited by exogenous administration of ATP and αβ-methylene ATP were inhibited by leaf extract, whereas contractions elicited by exogenous administration of noradrenaline or acetylcholine were unaffected. The active component was present within the aqueous phase of the leaf extract. In mouse mating studies, stinging nettle leaf extract (50 mg p.o. daily) reduced male fertility by 53% compared to vehicle-treated male mice. Cardiovascular parameters were unaffected by administration of stinging nettle leaf extract (p ≥ 0.057). Treated mice exhibited normal mating behaviour. Bladder and testes weighed less in stinging nettle leaf extract treated mice. All other organs and total body weight were unaffected. It is concluded that stinging nettle leaf extract reduces contractility of genitourinary smooth muscle by acting as an antagonist at postjunctional P2X1-purinoceptors. These data indicates that blocking sperm transport through pharmacological blockade of P2X1-purinoceptors via oral administration is consistent with an effective and convenient biological strategy male contraception.

Digestate Improves Stinging Nettle (Urtica dioica) Growth and Fiber Production at a Chlor-Alkali Site.

Project description:Marginal lands have been proposed to produce non-food crop biomass for energy or green materials. For this purpose, the selection, implementation, and growth optimization of plant species on such lands are key elements to investigate to achieve relevant plant yields. Stinging nettle (Urtica dioica) is a herbaceous perennial that grows spontaneously on contaminated lands and was described as suitable to produce fibers for material applications. Two mercury-contaminated soils from industrial wastelands with different properties (grassland soil and sediment landfill) were used in this study to assess the potential growth of stinging nettle in a greenhouse mesocosm experiment. Two organic amendments were studied for their impact on nettle growth. The solid digestate from organic food wastes significantly doubled plant biomass whereas the compost from green wastes had a lower impact. The highest doses of organic amendments significantly increased the number of fibers, which doubled following digestate application, while reducing leaf Hg concentration. Both amendments significantly improved soil respiration and enzymatic activities linked to the microbial biomass in the soil from the sediment landfill by the end of the experiment. In the context of a phytomanagement scenario, solid digestate would be a preferred amendment resource to improve nettle production on industrial wastelands.

Stinging Nettle (Urtica dioica L.) Attenuates FFA Induced Ceramide Accumulation in 3T3-L1 Adipocytes in an Adiponectin Dependent Manner.

Project description:ObjectiveExcess dietary lipids result in the accumulation of lipid metabolites including ceramides that can attenuate insulin signaling. There is evidence that a botanical extract of Urtica dioica L. (stinging nettle) improves insulin action, yet the precise mechanism(s) are not known. Hence, we examined the effects of Urtica dioica L. (UT) on adipocytes.Research designWe investigated the effects of an ethanolic extract of UT on free fatty acid (palmitic acid) induced inhibition of insulin-stimulated Akt serine phosphorylation and modulation of ceramidase expression in 3T3-L1 adipocytes. Adipocytes were exposed to excess FFAs in the presence or absence of UT. Effects on adiponectin expression, ceramidase expression, ceramidase activity, ceramide accumulation and insulin signaling were determined.ResultsAs expected, FFAs reduced adiponectin expression and increased the expression of ceramidase enzymes but not their activity. FFA also induced the accumulation of ceramides and reduced insulin-stimulated phosphorylation of Akt in adipocytes. The effects of FFA were partially reversed by UT. UT enhanced adiponectin expression and ceramidase activity in the presence of excess FFAs. UT abated ceramide accumulation and increased insulin sensitivity via enhanced Akt phosphorylation. A siRNA knockdown of adiponectin expression prevented UT from exerting positive effects on ceramidase activity but not Akt phosphorylation.ConclusionsIn adipocytes, the ability of UT to antagonize the negative effects of FFA by modulating ceramidase activity and ceramide accumulation is dependent on the presence of adiponectin. However, the ability of UT to enhance Akt phosphorylation is independent of adiponectin expression. These studies demonstrate direct effects of UT on adipocytes and suggest this botanical extract is metabolically beneficial.

In Vitro Bioactivities of Food Grade Extracts from Yarrow (Achillea millefolium L.) and Stinging Nettle (Urtica dioica L.) Leaves.

Project description:Yarrow (Achillea millefolium L., AM) and nettle (Urtica dioica L., UD) are bioactive plants used commercially in functional food and supplement applications and traditionally to alleviate gastric disorders. In this work, the effects of food-grade optimized extracts of Finnish early-season AM and UD were tested on bacterial growth including potential beneficial and foodborne pathogens, as well as murine norovirus (MNV). The anti-inflammatory properties of the extracts were also tested in vitro by NF-κB reporter cells. The food-grade extraction was optimized with the response surface modelling in terms of total carotenoid, chlorophyll, and phenolic compounds contents and antioxidant capacities. The optimal food-grade extraction parameters were a 1-h extraction in 70% ethanol at 45 °C for AM, and at 49 °C for UD. There were no significant effects on the beneficial bacteria (Lacticaseibacillus and Bifidobacterium strains), and the extracts were more effective against gram-positive than gram-negative foodborne bacteria and potential pathogens. Listeria innocua was the most susceptible strain in the optimized extracts with a growth rate of 0.059 ± 0.004 for AM and 0.067 ± 0.006 for UD, p < 0.05 compared to control. The optimized extracts showed a logarithmic growth reduction of 0.67 compared to MNV. The hydroethanolic extracts were cytotoxic to both cell lines, whereas aqueous AM and UD extracts induced and reduced TLR4 signalling in a reporter cell line, respectively. The results provide novel food-grade extraction parameters and support the bioactive effects of AM and UD in functional food applications, but more research is needed to elucidate the precise biological activity in vivo for gastric health.

Choline acetyltransferase in the nettle Urtica dioica L.

Project description:Extracts of acetone-dried powders prepared from nettle leaves were shown to catalyse the synthesis of acetylcholine. The specific activity of the enzyme in these extracts is of the same order as that of extracts from mammalian sources, such as ox brain, and the effects of temperature and pH are similar to those reported for mammalian choline acetyltransferase. Synthesis is not restricted to the younger leaves but appears to be continuous up to senescence.
