Project description:Retrosynthesis consists of recursively breaking down a target molecule to produce a synthesis route composed of readily accessible building blocks. In recent years, computer-aided synthesis planning methods have allowed a greater exploration of potential synthesis routes, combining state-of-the-art machine-learning methods with chemical knowledge. However, these methods are generally developed to produce individual routes from a singular product to a set of proposed building blocks and are not designed to leverage potential shared paths between targets. These methods do not necessarily encompass real-world use cases in medicinal chemistry, where one seeks to synthesize sets of target compounds in a library mode, looking for maximal convergence into a shared retrosynthetic path going via advanced key intermediate compounds. Using a graph-based processing pipeline, we explore Johnson & Johnson Electronic Laboratory Notebooks (J&J ELN) and publicly available datasets to identify complex routes with multiple target molecules sharing common intermediates, producing convergent synthesis routes. We find that over 70% of all reactions are involved in convergent synthesis, covering over 80% of all projects in the case of J&J ELN data. Scientific contributionWe introduce a novel planning approach to develop convergent synthesis routes, which can search multiple products and intermediates simultaneously guided by state-of-the-art machine learning single-step retrosynthesis models, enhancing the overall efficiency and practical applicability of retrosynthetic planning. We evaluate the multi-step synthesis planning approach using the extracted convergent routes and observe that solvability is generally high across those routes, being able to identify a convergent route for over 80% of the test routes and showing an individual compound solvability of over 90%. We find that by using a convergent search approach, we can synthesize almost 30% more compounds simultaneously for J&J ELN as compared to using an individual search, while providing an increased use of common intermediates.

Project description:Waste to energy (WTE) technologies have emerged as an alternative solution to municipal solid waste management. WTE systems provide major environmental and economic benefits by converting waste into accessible energy, as part of an integrated solid waste management (ISWM) strategy. However, previous studies showed that establishing an ISWM strategy based on a single type of WTE systems does not necessarily realize maximum benefits. Hence, optimizing the selection of WTE systems as part of a hybrid waste management strategy can potentially achieve maximum benefits and minimize negative impacts. However, such task is challenging due to the various alternatives and objectives, particularly those related to the material and energy recovery systems. This article presents the methods used to develop a systematic optimization framework that identifies the most beneficial set of ISWM systems through mathematical modelling. The methods include the procedures of the established framework, including base model computations, as well as the comprehensive modelling and optimization methods.•The energy recovery, carbon footprint, and financial profitability are computed for selected WTE facilities.•The multi-objective mathematical programming is solved using the weighted comprehensive criterion method (WCCM).•The model is implemented in CPLEX software using mathematical programming language (OPL).

Project description:High-quality reconstruction under a low sampling rate is very important for ghost imaging. How to obtain perfect imaging results from the low sampling rate has become a research hotspot in ghost imaging. In this paper, inspired by matrix optimization in compressed sensing, an optimization scheme of speckle patterns via measurement-driven framework is introduced to improve the reconstruction quality of ghost imaging. According to this framework, the sampling matrix and sparse basis are optimized alternately using the sparse coefficient matrix obtained from the low-dimension pseudo-measurement process and the corresponding solution is obtained analytically, respectively. The optimized sampling matrix is then dealt with non-negative constraint and binary quantization. Compared to the developed optimization schemes of speckle patterns, simulation results show that the proposed scheme can achieve better reconstruction quality with the low sampling rate in terms of peak signal-to-noise ratio (PSNR) and mean structural similarity index (MSSIM). In particular, the lowest sampling rate we use to achieve a good performance is about 6.5%. At this sampling rate, the MSSIM and PSNR of the proposed scheme can reach 0.787 and 17.078 dB, respectively.

Project description:Land managers increasingly rely upon landscape assessments to understand the status of natural resources and identify conservation priorities. Many of these landscape planning efforts rely on geospatial models that characterize the ecological integrity of the landscape. These general models utilize measures of habitat disturbance and human activity to map indices of ecological integrity. We built upon these modeling frameworks by developing a Landscape Integrity Index (LII) model using geospatial datasets of the human footprint, as well as incorporation of other indicators of ecological integrity such as biodiversity and vegetation departure. Our LII model serves as a general indicator of ecological integrity in a regional context of human activity, biodiversity, and change in habitat composition. We also discuss the application of the LII framework in two related coarse-filter landscape conservation approaches to expand the size and connectedness of protected areas as regional mitigation for anticipated land-use changes.

Project description:BackgroundRecent transformative changes in science education require new learning opportunities for teachers-opportunities that include rich images of classroom enactment of the reform vision. One fruitful way for doing that is to use video clips of instruction.Teachers do not, however, learn how to improve their instructional practice from simply watching and reflecting on classroom videos. The videos need to be carefully selected and embedded in professional development in ways that-through facilitator-led, participant-centered discussion-can help teachers to notice and reason about important aspects of instruction and learning that occur in the video. Consistent with the recent efforts to identify planning and facilitation approaches that guide effective professional development (PD) programs, in this paper, we adapted the Five Practices Framework for orchestrating productive classroom discussions to describe how PD facilitators plan for and enact professional learning tasks to help science teachers learn within a video-based PD program. These practices include anticipating, sequencing, monitoring, selecting, connecting and two additional practices that set the stage for the five practices (i.e., setting goals and selecting tasks).ResultsOur analyses of the video-based discussions in the PD provide insights into how the facilitators engaged teachers in video-based conversations by using the practices of monitoring, selecting, and connecting. The monitoring moves, such as clarifying, countering, and redirecting, were used by the facilitator in nearly all the PD sessions. Similarly, selecting moves were used and were consistent with the goals of the PD. Finally, analysis of facilitators' and participants' connecting comments indicated their increased capacity to make connections to the bigger ideas of teaching science by maintaining the cognitive demand on students' thinking.ConclusionsThis paper provides elaborated descriptions of the five practices for planning and facilitating video-based PD and the ways in which they were enacted in a video-based PD program in science. In so doing, it proposes five practices as a guiding framework to support teachers' learning from videos. Overall, the study's results endorse the promise of a goal-driven, theory-informed design that foregrounds careful attention to teachers' thinking in ways that support their understanding of complex classroom interactions.

Project description:By coupling a robot to a smart environment, the robot can sense state beyond the perception range of its onboard sensors and gain greater actuation capabilities. Nevertheless, incorporating the states and actions of Internet of Things (IoT) devices into the robot's onboard planner increases the computational load, and thus can delay the execution of a task. Moreover, tasks may be frequently replanned due to the unanticipated actions of humans. Our framework aims to mitigate these inadequacies. In this paper, we propose a continual planning framework, which incorporates the sensing and actuation capabilities of IoT devices into a robot's state estimation, task planing and task execution. The robot's onboard task planner queries a cloud-based framework for actuators, capable of the actions the robot cannot execute. Once generated, the plan is sent to the cloud back-end, which will inform the robot if any IoT device reports a state change affecting its plan. Moreover, a Hierarchical Continual Planning in the Now approach was developed in which tasks are split-up into subtasks. To delay the planning of actions that will not be promptly executed, and thus to reduce the frequency of replanning, the first subtask is planned and executed before the subsequent subtask is. Only information relevant to the current (sub)task is provided to the task planner. We apply our framework to a smart home and office scenario in which the robot is tasked with carrying out a human's requests. A prototype implementation in a smart home, and simulator-based evaluation results, are presented to demonstrate the effectiveness of our framework.

Project description:IntroductionConstantly evolving technology and techniques within radiation therapy require practitioners to maintain a continuous approach to professional development and training. Systems of performance appraisal and adoption of regular feedback mechanisms are vital to support this development yet frequently lack structure and rely on informal peer support.MethodsA Radiation Therapy Performance Appraisal Framework (RT-PAF) for radiation therapists in planning and simulation was developed to define expectations of practice and promote a supportive and objective culture of performance and skills appraisal. Evaluation of the framework was conducted via an anonymous online survey tool. Nine peer reviewers and fourteen recipients provided feedback on its effectiveness and the challenges and limitations of the approach.ResultsFindings from the evaluation were positive and suggested that both groups gained benefit from and expressed a strong interest in embedding the approach more routinely. Respondents identified common challenges related to the limited ability to implement suggested development strategies; this was strongly associated with time and rostering issues.ConclusionsThis framework successfully defined expectations for practice and provided a fair and objective feedback process that focussed on skills development. It empowered staff to maintain their skills and reach their professional potential. Management support, particularly in regard to provision of protected time was highlighted as critical to the framework's ongoing success. The demonstrated benefits arising in terms of staff satisfaction and development highlight the importance of this commitment to the modern radiation therapy workforce.

Project description:Policy Points: Strengthening accountability through better measurement and reporting is vital to ensure progress in improving quality primary health care (PHC) systems and achieving universal health coverage (UHC). The Primary Health Care Performance Initiative (PHCPI) provides national decision makers and global stakeholders with opportunities to benchmark and accelerate performance improvement through better performance measurement. Results from the initial PHC performance assessments in low- and middle-income countries (LMICs) are helping guide PHC reforms and investments and improve the PHCPI's instruments and indicators. Findings from future assessment activities will further amplify cross-country comparisons and peer learning to improve PHC. New indicators and sources of data are needed to better understand PHC system performance in LMICs.ContextThe Primary Health Care Performance Initiative (PHCPI), a collaboration between the Bill and Melinda Gates Foundation, The World Bank, and the World Health Organization, in partnership with Ariadne Labs and Results for Development, was launched in 2015 with the aim of catalyzing improvements in primary health care (PHC) systems in 135 low- and middle-income countries (LMICs), in order to accelerate progress toward universal health coverage. Through more comprehensive and actionable measurement of quality PHC, the PHCPI stimulates peer learning among LMICs and informs decision makers to guide PHC investments and reforms. Instruments for performance assessment and improvement are in development; to date, a conceptual framework and 2 sets of performance indicators have been released.MethodsThe PHCPI team developed the conceptual framework through literature reviews and consultations with an advisory committee of international experts. We generated 2 sets of performance indicators selected from a literature review of relevant indicators, cross-referenced against indicators available from international sources, and evaluated through 2 separate modified Delphi processes, consisting of online surveys and in-person facilitated discussions with experts.FindingsThe PHCPI conceptual framework builds on the current understanding of PHC system performance through an expanded emphasis on the role of service delivery. The first set of performance indicators, 36 Vital Signs, facilitates comparisons across countries and over time. The second set, 56 Diagnostic Indicators, elucidates underlying drivers of performance. Key challenges include a lack of available data for several indicators and a lack of validated indicators for important dimensions of quality PHC.ConclusionsThe availability of data is critical to assessing PHC performance, particularly patient experience and quality of care. The PHCPI will continue to develop and test additional performance assessment instruments, including composite indices and national performance dashboards. Through country engagement, the PHCPI will further refine its instruments and engage with governments to better design and finance primary health care reforms.

Project description:Due to their large surface area and pore volume, three-dimensional covalent organic frameworks (3D COFs) have emerged as competitive porous materials. However, structural dynamic behavior, often observed in imine-linked 3D COFs, could potentially unlock their potential application in gas storage. Herein, we showed how a pre-locked linker strategy introduces breaking dynamic behavior in 3D COFs. A predesigned planar linker-based 3,8-diamino-6-phenylphenanthridine (DPP) was prepared to produce non-dynamic 3D JUC-595, as the benzylideneamine moiety in DPP locked the linker flexibility and restricted the molecular bond rotation of the imine linkages. Upon solvent inclusion and release, the PXRD profile of JUC-595 remained intake, while JUC-594 with a flexible benzidine linker experienced crystal transformation due to framework contraction-expansion. As a result, the activated JUC-595 achieved higher surface areas (754 m2 g-1) than that of JUC-594 (548 m2 g-1). Furthermore, improved CO2 and CH4 storages were also seen in JUC-595 compared with JUC-594. Impressively, JUC-595 recorded a high normalized H2 storage capacity that surpassed other reported high-surface area 3D COFs. This works shows important insights on manipulating the structural properties of 3D COF to tune gas storage performance.

Project description:Retrosynthetic route planning can be considered a rule-based reasoning procedure. The possibilities for each transformation are generated based on collected reaction rules, and then potential reaction routes are recommended by various optimization algorithms. Although there has been much progress in computer-assisted retrosynthetic route planning and reaction prediction, fully data-driven automatic retrosynthetic route planning remains challenging. Here we present a template-free approach that is independent of reaction templates, rules, or atom mapping, to implement automatic retrosynthetic route planning. We treated each reaction prediction task as a data-driven sequence-to-sequence problem using the multi-head attention-based Transformer architecture, which has demonstrated power in machine translation tasks. Using reactions from the United States patent literature, our end-to-end models naturally incorporate the global chemical environments of molecules and achieve remarkable performance in top-1 predictive accuracy (63.0%, with the reaction class provided) and top-1 molecular validity (99.6%) in one-step retrosynthetic tasks. Inspired by the success rate of the one-step reaction prediction, we further carried out iterative, multi-step retrosynthetic route planning for four case products, which was successful. We then constructed an automatic data-driven end-to-end retrosynthetic route planning system (AutoSynRoute) using Monte Carlo tree search with a heuristic scoring function. AutoSynRoute successfully reproduced published synthesis routes for the four case products. The end-to-end model for reaction task prediction can be easily extended to larger or customer-requested reaction databases. Our study presents an important step in realizing automatic retrosynthetic route planning.
