Methionine oxidation of proteins analyzed by affinity capillary electrophoresis in presence of silver(I) and gold(III) ions.

ABSTRACT: Oxidative damage of biopharmaceuticals during manufacturing and storage is a key concern throughout pharmaceutical development. However, few simple and robust analytical methods are available for the determination of oxidation sites. Here, the potential of affinity capillary electrophoresis (ACE) in the separation of proteins with oxidized methionine (Met) residues is shown. Silver(I) and gold(I) ions have the attribute to selectively form complexes with thioethers over sulfoxides. The addition of these ions to the BGE leads to a selective complexation of Met residues and, thus, to a change of charge allowing separation of species according to the different oxidation states of Met. The mechanisms of these interactions are discussed and binding constants for peptides containing Met with silver(I) are calculated. Additionally, the proposed method can be used as an indicator of oxidative stress in large proteins. The presented technique is easily accessible, economical, and has rapid analysis times, adding new approaches to the analytical toolbox of Met sulfoxide detection.

Project description:Surface oxidation improves the dispersion of carbon nanotubes in aqueous solutions and plays a key role in the development of biosensors, electrochemical detectors and polymer composites. Accurate characterization of the carbon nanotube surface is important because the development of these nano-based applications depends on the degree of functionalization, in particular the amount of carboxylation. Affinity capillary electrophoresis is used to characterize the oxidation of multi-walled carbon nanotubes. A polytryptophan peptide that contains a single arginine residue (WRWWWW) serves as a receptor in affinity capillary electrophoresis to assess the degree of carboxylation. The formation of peptide-nanotube receptor-ligand complex was detected with a UV absorbance detector. Apparent dissociation constants (KD) are obtained by observing the migration shift of the WRWWWW peptide through background electrolyte at increasing concentrations of multi-walled carbon nanotubes. A 20% relative standard deviation in method reproducibility and repeatability is determined with triplicate analysis within a single sample preparation and across multiple sample preparations for a commercially available carbon nanotube. Affinity capillary electrophoresis is applied to assess differences in degree of carboxylation across two manufacturers and to analyze acid treated carbon nanotubes. The results of these studies are compared to X-ray photoelectron spectroscopy and zeta potential. Affinity capillary electrophoresis comparisons of carbon nanotube samples prepared by varying acid treatment time from 30 min to 3 h yielded significant differences in degree of carboxylation. X-ray photoelectron spectroscopy analysis was inconclusive due to potential acid contamination, while zeta potential showed no change based on surface charge. This work is significant to research involving carbon nanotube-based applications because it provides a new metric to rapidly characterize carbon nanotubes obtained from different vendors, or synthesized in laboratories using different procedures.

Project description:Covalently functionalized gold nanoparticles influence capillary electrophoresis separations of neurotransmitters in a concentration- and surface-chemistry-dependent manner. Gold nanoparticles with either primarily covalently functionalized carboxylic acid (Au@COOH) or amine (Au@NH(2)) surface groups are characterized using extinction spectroscopy, transmission electron microscopy, and zeta potential measurements. The impact of the presence of nanoparticles and their surface chemistry is investigated, and at least three nanoparticle-specific mechanisms are found to effect separations. First, the degree of nanoparticle-nanoparticle interactions is quantified using a new parameter termed the critical nanoparticle concentration (CNC). CNC is defined as the lowest concentration of nanoparticles that induces predominant nanoparticle aggregation under specific buffer conditions and is determined using dual-wavelength photodiode array detection. Once the CNC has been exceeded, reproducible separations are no longer observed. Second, nanoparticle-analyte interactions are dictated by electrostatic interactions which depend on the pK(a) of the analyte and surface charge of the nanoparticle. Finally, nanoparticle-capillary interactions occur in a surface-chemistry-dependent manner. Run buffer viscosity is influenced by the formation of a nanoparticle steady-state pseudostationary phase along the capillary wall. Despite differences in buffer viscosity leading to changes in neurotransmitter mobilities, no significant changes in electroosmotic flow were observed. As a result of these three nanoparticle-specific interactions, Au@NH(2) nanoparticles increase the mobility of the neurotransmitters while a smaller opposite effect is observed for Au@COOH nanoparticles. Understanding nanoparticle behavior in the presence of an electric field will have significant impacts in separation science where nanoparticles can serve to improve either the mobility or detection sensitivity of target molecules.
