Myelin like electrogenic filamentation and Liquid Microbial Fuel Cells Dataset.

ABSTRACT: Biofilm at water-oil interface of hypoxic water columns of microcosms, prepared from a lacustrine sample, that used diesel as a carbon source was found to show electrogenic properties. These microcosms named, Liquid Microbial Fuel Cells (L-MFCs) were electrically characterized using a custom electronic analyzer; accurate determination of voltage (V), power density (W/m 2), and current density (A/m2) for both charge and discharge phases was carried out. The instrument made it possible to carry out cell characterizations using resistive loads between 0 Ω (Ohm) and 10 kΩ. During the hypoxic and electrogenic phase, the synthesis of a system of "bacterial piping induction", produced filaments of hundreds of micrometers in which the microbial cells are hosted. Ultrastructural microscopy collected by scanning (SEM), transmission (TEM), immunofluorescence, Thunder Imager 3D, confocal laser scanning (CLSM) microscopy revealed a "myelin like" structure during filamentation processes; this "myelin like" structure exhibited cross-reactivity towards different epitopes of the myelin basic protein (MBP) and Claudin 11 (O4) of human oligodendrocytes. The disclosure of these filamentation processes could be helpful to describe further unconventional microbial structures in aquatic ecosystems and of the animal world. Project description:Microbial fuel cells (MFCs) incorporating air-breathing cathodes have emerged as a promising eco-friendly wastewater treatment technology capable of operating on an energy-free basis. However, the inevitable biofouling of these devices rapidly decreases cathodic catalytic activity and also reduces the stability of MFCs during long-term operation. The present work developed a novel microbial separator for use in air-breathing MFCs that protects cathodic catalytic activity. In these modified devices, microbes preferentially grow on the microbial separator rather than the cathodic surface such that biofouling is prevented. Trials showed that this concept provided low charge transfer and mass diffusion resistance values during the cathodic oxygen reduction reaction of 4.6 ± 1.3 and 17.3 ± 6.8 Ω, respectively, after prolonged operation. The maximum power density was found to be stable at 1.06 ± 0.07 W m−2 throughout a long-term test and the chemical oxygen demand removal efficiency was increased to 92% compared with a value of 83% for MFCs exhibiting serious biofouling. In addition, a cathode combined with a microbial separator demonstrated less cross-cathode diffusion of oxygen to the anolyte. This effect indirectly induced the growth of electroactive bacteria and produced higher currents in air-breathing MFCs. Most importantly, the present microbial separator concept enhances both the lifespan and economics of air-breathing MFCs by removing the need to replace or regenerate the cathode during long-term operation. These results indicate that the installation of a microbial separator is an effective means of stabilizing power generation and ensuring the cost-effective performance of air-breathing MFCs intended for future industrial applications. Project description:Microbial fuel cells (MFCs) are eco-friendly bio-electrochemical reactors that use exoelectrogens as biocatalyst for electricity harvest from organic biomass, which could also be used as biosensors for long-term environmental monitoring. Glucose and xylose, as the primary ingredients from cellulose hydrolyzates, is an appealing substrate for MFC. Nevertheless, neither xylose nor glucose can be utilized as carbon source by well-studied exoelectrogens such as Shewanella oneidensis. In this study, to harvest the electricity by rapidly harnessing xylose and glucose from corn stalk hydrolysate, we herein firstly designed glucose and xylose co-fed engineered Klebsiella pneumoniae-S. oneidensis microbial consortium, in which K. pneumoniae as the fermenter converted glucose and xylose into lactate to feed the exoelectrogens (S. oneidensis). To produce more lactate in K. pneumoniae, we eliminated the ethanol and acetate pathway via deleting pta (phosphotransacetylase gene) and adhE (alcohol dehydrogenase gene) and further constructed a synthesis and delivery system through expressing ldhD (lactate dehydrogenase gene) and lldP (lactate transporter gene). To facilitate extracellular electron transfer (EET) of S. oneidensis, a biosynthetic flavins pathway from Bacillus subtilis was expressed in a highly hydrophobic S. oneidensis CP-S1, which not only improved direct-contacted EET via enhancing S. oneidensis adhesion to the carbon electrode but also accelerated the flavins-mediated EET via increasing flavins synthesis. Furthermore, we optimized the ratio of glucose and xylose concentration to provide a stable carbon source supply in MFCs for higher power density. The glucose and xylose co-fed MFC inoculated with the recombinant consortium generated a maximum power density of 104.7 ± 10.0 mW/m2, which was 7.2-folds higher than that of the wild-type consortium (12.7 ± 8.0 mW/m2). Lastly, we used this synthetic microbial consortium in the corn straw hydrolyzates-fed MFC, obtaining a power density 23.5 ± 6.0 mW/m2.
