Population Genomics of New Zealand Pouched Lamprey (kanakana; piharau; Geotria australis).

ABSTRACT: Pouched lamprey (Geotria australis) or kanakana/piharau is a culturally and ecologically significant jawless fish that is distributed throughout Aotearoa New Zealand. Despite its importance, much remains unknown about historical relationships and gene flow between populations of this enigmatic species within New Zealand. To help inform management, we assembled a draft G. australis genome and completed the first comprehensive population genomics analysis of pouched lamprey within New Zealand using targeted gene sequencing (Cyt-b and COI) and restriction site-associated DNA sequencing (RADSeq) methods. Employing 16 000 genome-wide single nucleotide polymorphisms (SNPs) derived from RADSeq (n = 186) and sequence data from Cyt-b (766 bp, n = 94) and COI (589 bp, n = 20), we reveal low levels of structure across 10 sampling locations spanning the species range within New Zealand. F-statistics, outlier analyses, and STRUCTURE suggest a single panmictic population, and Mantel and EEMS tests reveal no significant isolation by distance. This implies either ongoing gene flow among populations or recent shared ancestry among New Zealand pouched lamprey. We can now use the information gained from these genetic tools to assist managers with monitoring effective population size, managing potential diseases, and conservation measures such as artificial propagation programs. We further demonstrate the general utility of these genetic tools for acquiring information about elusive species.

Project description:Lampreys are one of the two surviving groups of the agnathan (jawless) stages in vertebrate evolution and are thus ideal candidates for elucidating the evolution of visual systems. This study investigated the retinal amino acid neurochemistry of the southern hemisphere lamprey Geotria australis during the downstream migration of the young, recently-metamorphosed juveniles to the sea and during the upstream migration of the fully-grown and sexually-maturing adults to their spawning areas. Glutamate and taurine were distributed throughout the retina, whilst GABA and glycine were confined to neurons of the inner retina matching patterns seen in most other vertebrates. Glutamine and aspartate immunoreactivity was closely matched to Müller cell morphology. Between the migratory phases, few differences were observed in the distribution of major neurotransmitters i.e. glutamate, GABA and glycine, but changes in amino acids associated with retinal metabolism i.e. glutamine and aspartate, were evident. Taurine immunoreactivity was mostly conserved between migrant stages, consistent with its role in primary cell functions such as osmoregulation. Further investigation of glutamate signalling using the probe agmatine (AGB) to map cation channel permeability revealed entry of AGB into photoreceptors and horizontal cells followed by accumulation in inner retinal neurons. Similarities in AGB profiles between upstream and downstream migrant of G. australis confirmed the conservation of glutamate neurotransmission. Finally, calcium binding proteins, calbindin and calretinin were localized to the inner retina whilst recoverin was localized to photoreceptors. Overall, conservation of major amino acid neurotransmitters and calcium-associated proteins in the lamprey retina confirms these elements as essential features of the vertebrate visual system. On the other hand, metabolic elements of the retina such as neurotransmitter precursor amino acids and Müller cells are more sensitive to environmental changes associated with migration.

Project description:BACKGROUND:The Argentinian pouched lamprey, classified as Petromyzon macrostomus Burmeister, 1868 was first described in 1867 in De La Plata River, in Buenos Aires, Argentina, and subsequently recorded in several rivers from Patagonia. Since its original description, the validity of P. macrostomus was questioned by several ichthyologists and 36 years after its original discovery it was considered a junior synonym of Geotria australis Gray, 1851. For a long time, the taxonomic status of G. australis has been uncertain, largely due to the misinterpretations of the morphological alterations that occur during sexual maturation, including the arrangement of teeth, size and position of fins and cloaca, and the development of an exceptionally large gular pouch in males. In this study, the taxonomic status of Geotria from across the "species" range was evaluated using both molecular analysis and examination of morphological characteristics. METHODOLOGY/PRINCIPAL FINDINGS:Phylogenetic and species delimitation analyses based on mitochondrial DNA sequences of Cytochrome b (Cyt b) and Cytochrome C Oxidase Subunit 1 (COI) genes, along with morphological analysis of diagnostic characters reported in the original descriptions of the species were used to assess genetic and morphological variation within Geotria and to determine the specific status of the Argentinian lamprey. These analyses revealed that Geotria from Argentina constitutes a well differentiated lineage from Chilean and Australasian populations. The position of the cloaca and the distance between the second dorsal and caudal fins in sub-adult individuals, and at previous life stages, can be used to distinguish between the two species. In addition, the genetic distance between G. macrostoma and G. australis for the COI and Cyt b mitochondrial genes is higher than both intra- and inter-specific distances reported for other Petromyzontiformes. CONCLUSIONS/SIGNIFICANCE:Our results indicate that the Argentinian pouched lamprey, found along a broad latitudinal gradient on the south-west Atlantic coast of South America, should be named as Geotria macrostoma (Burmeister, 1868) and not as G. australis Gray 1851, returning to its earliest valid designation in Argentina. Geotria macrostoma can now be considered as the single lamprey species inhabiting Argentinian Patagonia, with distinct local adaptations and evolutionary potential. It is essential that this distinctiveness is recognized in order to guide future conservation and management actions against imminent threats posed by human actions in the major basins of Patagonia.

Project description:Tetrodotoxin (TTX) is a potent neurotoxin responsible for many human intoxications globally. Despite its potency and widespread occurrence in taxonomically diverse species, the primary source of TTX remains uncertain. Paphies australis, an endemic clam found in New Zealand, has been found to contain TTX in several locations. However, it is unknown if this represents endogenous production or accumulation from an external source. To address this question, the concentrations of TTX in whole P. australis and dissected organs (siphons, foot, digestive gland and the 'rest') from thirteen sites around New Zealand were determined using liquid chromatography-tandem quadrupole mass spectrometry analysis (LC-MS/MS). Depuration rate of TTX was also investigated by harvesting and measuring concentrations in P. australis maintained in captivity on a toxin-free diet every three to 15 days for 150 days. The LC-MS/MS analyses of the spatial samples showed that TTX was present in P. australis from all regions tested, with significantly (p < 0.001) higher concentrations (15-50 μg kg-1) observed at lower latitudes of the North Island compared with trace levels (0.5-3 μg kg-1) in the South Island of New Zealand. Tetrodotoxin was detected in all the dissected organs but the siphons contained the highest concentrations of TTX at all sites analysed. A linear model of the depuration data identified a significant (p < 0.001) decline in total TTX concentrations in P. australis over the study period. The siphons maintained the highest amount of TTX across the entire depuration study. The digestive glands contained low concentrations at the start of the experiment, but this depurated rapidly and only traces remained after 21 days. These results provide evidence to suggest that P. australis does not produce TTX endogenously but obtains the neurotoxin from an exogenous source (e.g., diet) with the source more prevalent in warmer northern waters. The association of higher TTX concentrations in shellfish with warmer environments raises concerns that this toxin's distribution and abundance could become an increasing human health issue with global warming.

Project description:This study aimed to elucidate the way in which larvae of the lamprey Geotria australis counteract the potential problems of the very high concentrations of non-haem iron they contain and thereby avoid the deleterious effects associated with iron overload in other vertebrates. Particular attention has been paid to ascertaining whether increasing concentrations of iron are accompanied by (i) change to a less readily available form of iron and (ii) an increase in the activity of those detoxifying enzymes responsible for minimizing the production of harmful hydroxyl radicals via the Haber-Weiss reaction. The mean concentrations of haemosiderin and ferritin in larval G. australis were each far higher in the nephric fold than in either the liver or intestine, but all these concentrations were much greater than those in rat liver. Since haemosiderin releases iron far more slowly than ferritin, the iron it contains is much less readily available to catalyse the Haber-Weiss reaction. It is thus relevant that (i) non-haem iron in the nephric fold occurred to a greater extent as large dense haemosiderin granules than as ferritin molecules and (ii) the proportion of iron in the form of haemosiderin rose with increasing concentration of total non-haem iron. A strong correlation was also recorded between the activity of superoxide dismutase in the nephric fold and the concentrations of total non-haem iron and its haemosiderin and ferritin components. This demonstrates that enzyme detoxification of O2.- rises with increasing amounts of iron. The exceptional iron concentrations in the nephric fold were not reflected by a greater measured activity of superoxide dismutase than that found in other tissues. However, the nephric fold was shown to contain an augmentation factor which is presumed to enhance the activity of this enzyme in vivo. The activity of catalase and glutathione peroxidase, which catalyse the breakdown of H2O2 to O2 and water, were each significantly correlated with the concentration of ferritin.

Project description:Southern right whales (Eubalaena australis) migrate between Austral-winter calving and socialising grounds to offshore mid- to high latitude Austral-summer feeding grounds. In Australasia, winter calving grounds used by southern right whales extend from Western Australia across southern Australia to the New Zealand sub-Antarctic Islands. During the Austral-summer these whales are thought to migrate away from coastal waters to feed, but the location of these feeding grounds is only inferred from historical whaling data. We present new information on the satellite derived offshore migratory movements of six southern right whales from Australasian wintering grounds. Two whales were tagged at the Auckland Islands, New Zealand, and the remaining four at Australian wintering grounds, one at Pirates Bay, Tasmania, and three at Head of Bight, South Australia. The six whales were tracked for an average of 78.5 days (range: 29 to 150) with average individual distance of 38 km per day (range: 20 to 61 km). The length of individually derived tracks ranged from 645-6,381 km. Three likely foraging grounds were identified: south-west Western Australia, the Subtropical Front, and Antarctic waters, with the Subtropical Front appearing to be a feeding ground for both New Zealand and Australian southern right whales. In contrast, the individual tagged in Tasmania, from a sub-population that is not showing evidence of post-whaling recovery, displayed a distinct movement pattern to much higher latitude waters, potentially reflecting a different foraging strategy. Variable population growth rates between wintering grounds in Australasia could reflect fidelity to different quality feeding grounds. Unlike some species of baleen whale populations that show movement along migratory corridors, the new satellite tracking data presented here indicate variability in the migratory pathways taken by southern right whales from Australia and New Zealand, as well as differences in potential Austral summer foraging grounds.
