In a recent article, we described the edxia framework, a user-friendly framework to analyse the microstructure of cementitious materials using SEM-EDS hypermaps. The manual approach presented was shown to be efficient to answer the relevant scientific questions. However, it is limited for batch analysis and (semi-)automated treatments. Project description:The scanning electron microscope/X-ray energy dispersive spectrometer (SEM/EDS) system is widely applied to rare earth minerals (REMs) to qualitatively describe their mineralogy and quantitatively determine their composition. The performance of multivariate statistical analysis on the EDS raw dataset can enhance the efficiency and the accuracy of phase identification. In this work, the principal component analysis (PCA) and the blind source separation (BSS) algorithms were performed on an EDS map of a REM sample, assisting to achieve an efficient phase map analysis. The PCA significantly denoised the phase map and was used as a preprocessing step for the following BSS. The BSS separated the mixed EDS signals into a set of physically interpretable components, bringing convenience to the phase separation and identification. Through the comparison between the independent component analysis (ICA) and the nonnegative matrix factorization (NMF) algorithms, the NMF was confirmed to be more suitable for the EDS mapping analysis.

Project description:The combination of scanning electron microscopy (SEM) images and energy-dispersive X-ray spectroscopy (EDS) maps (SEM-EDS analysis) enables the analysis of the relationship between the microstructures and elemental compositions of the surfaces of materials. However, conventional SEM-EDS analyses lack comprehensiveness and quantitativeness, resulting in potential inaccuracies in reflecting the properties of the entire sample and variations in the results depending on the analyst. Therefore, herein, we propose an objective SEM-EDS analytical process that addresses the aforementioned issues. Comprehensiveness was addressed by acquiring large volumes of SEM images through automated capturing, whereas quantitativeness was addressed through microstructural analysis of the SEM images based on image features, model-based dimension reduction and clustering methods, and similarity analysis of the elemental distribution in EDS maps based on statistical distances. The proposed method was used to analyze the degradation of lithium-ion battery electrodes, affording objective results that align with subjective insights into the changes in the morphology and composition of solid electrolyte interphase (SEI) films accompanying degradation.

Project description:An enhanced method to quantify energy dispersive spectra recorded in 3D with a scanning electron microscope (3D SEM-EDS) has been previously demonstrated. This paper presents an extension of this method using all the available X-ray lines generated by the beam. The extended method benefits from using high energy lines, that are more accurately quantified, and from using soft X-rays that are highly absorbed and thus more surface sensitive. The data used to assess the method are acquired with a dual beam FIB/SEM investigating a multi-element Ni-based superalloy. A high accelerating voltage, needed to excite the highest energy X-ray line, results in two available X-ray lines for several elements. The method shows an improved compositional quantification as well as an improved spatial resolution.

Project description:The oxidation states of colouring elements and the pigments in ancient rare glasses have been investigated in this study. Synchrotron X-ray, SEM-EDS, and Raman techniques revealed that Cu2+plays a major role in blue and green glasses. The lead stannate pigment gives glasses a yellow colour. Copper and lead stannate can cause the green colour in glasses, and iron gives rise to the colour of black glasses. Microcomputed tomography reveals the distribution of the heavy elements, pigments, and inclusions in the glasses. The Dvaravati glasses in Southeast Asia may have been imported or technologically transferred to domestic manufacturers during trading on the Silk Road that connected the East and the West.

Project description:In arsenopyrite bioleaching, the interfacial reaction between mineral and cells is one of the most important factors. The energy of the interface is influenced by the mineralogical and microbiological characteristics. In this paper, the interfacial energy was calculated, and the surface of arsenopyrite during the bioleaching process was characterized by 3D laser microscopy, scanning electron microscopy with energy-dispersive X-ray spectroscopy, and X-ray photoelectron spectroscopy, in order to assess the dissolution and oxidation behavior of arsenopyrite during bioleaching. The results showed that the contact angles of arsenopyrite were 22 ± 2° when covered with biofilms, but the reaction surface of arsenopyrite turned 103 ± 2°. However, the angle was 45-50° when covered by passive layer, which was half as that of arsenopyrite surface. The interfacial energy of arsenopyrite without biofilms increased from 45 to 62 mJ/m2, while it decreased to 5 ± 1 mJ/m2 when covered by biofilms during the leaching process. The surface was separated into fresh surface, oxidized surface, and (corrosion) pits. The interfacial energy was influenced by the fresh and oxidized surfaces. Surface roughness increased from 0.03 ± 0.01 to 5.89 ± 1.97 μm, and dissolution volume increased from 6.31 ± 0.47 × 104 to 2.72 ± 0.49 × 106 μm3. The dissolution kinetics of arsenopyrite followed the model of Kt = lnX, and the dissolution mechanisms were mixed controlled: surface reaction control and diffusion through sulfur layer. On the surface of arsenopyrite crystal, the oxidation steps of each element can be described as: for Fe, Fe(II)-(AsS)→Fe(III)-(AsS)→Fe(III)-OH or Fe(III)-SO; for S, As-S(-1) or Fe-S(-1)→polysulfide S→intermediate S-O→sulfate; and for As, As-1-S→As0→As+1-O→As+3-O→As+5-O.

Project description:A better understanding of automation disengagements can lead to improved safety and efficiency of automated systems. This study investigates the factors contributing to automation disengagements initiated by human operators and the automation itself by analyzing semi-structured interviews with 103 users of Tesla's Autopilot and FSD Beta. The factors leading to automation disengagements are represented by categories. In total, we identified five main categories, and thirty-five subcategories. The main categories include human operator states (5), human operator's perception of the automation (17), human operator's perception of other humans (3), the automation's perception of the human operator (3), and the automation incapability in the environment (7). Human operators disengaged the automation when they anticipated failure, observed unnatural or unwanted automation behavior (e.g., erratic steering, running red lights), or believed the automation is not capable to operate safely in certain environments (e.g., inclement weather, non-standard roads). Negative experiences of human operators, such as frustration, unsafe feelings, and distrust represent some of the adverse human operate states leading to automation disengagements initiated by human operators. The automation, in turn, monitored human operators and disengaged itself if it detected insufficient vigilance or speed rule violations by human operators. Moreover, human operators can be influenced by the reactions of passengers and other road users, leading them to disengage the automation if they sensed discomfort, anger, or embarrassment due to the automation's actions. The results of the analysis are synthesized into a conceptual framework for automation disengagements, borrowing ideas from the human factor's literature and control theory. This research offers insights into the factors contributing to automation disengagements, and highlights not only the concerns of human operators but also the social aspects of this phenomenon. The findings provide information on potential edge cases of automated vehicle technology, which may help to enhance the safety and efficiency of such systems.

Project description:Energy dispersive X-ray spectroscopy (EDS) carried out alongside scanning electron microscopy (SEM) is a common technique for elemental analysis. To investigate "wet" biological specimens, complex pre-treatments are required to stabilize them under the high vacuum conditions of high-resolution SEM. These often produce unwanted artifacts. We have previously reported that the polymerization of natural surface substances on organisms by the electron beam of the SEM setup or by plasma irradiation causes a nano-scale layer to form-called a "NanoSuit"-that can act as a barrier and keep organisms alive and hydrated in a field-emission SEM system. In the study reported herein, we examined the suitability of the NanoSuit method for elemental analyses of biological specimens by EDS. We compared experimental results for living Drosophila larvae and Aloe arborescens specimens prepared by the NanoSuit method and by conventional fixation. The NanoSuit method allowed accurate detection of the elemental compositions at high resolution. By contrast, specimens prepared by the conventional fixation method displayed additional EDS signals corresponding to the elements in the chemicals involved in the fixation process. Our results demonstrate that the NanoSuit method is useful for studying hydrous samples via EDS and SEM, particularly in biological sciences.

Project description:Overcoming the non-specificity of histological accentuated growth lines in hard tissues is an ongoing challenge. Identifying season at death and reproductive events has profound implications for evolutionary, ecological and conservation studies. Dental cementum is a mineralized tissue with yearly periodicity that continues deposition from tooth formation until death, maintaining a record spanning almost the entire life of an individual. Recent work has successfully employed elemental analysis of calcified incremental tissues to detect changes in extrinsic conditions such as diet and climate and to identify two important life-history milestones: weaning and sexual maturity. Here, we employ field-emission scanning electron microscopy and energy-dispersive X-ray analysis to measure the relative concentrations of calcium, phosphorous, oxygen, magnesium and sodium in the cementum of 34 teeth from seven male and female rhesus macaques with known medical and life-history information. We find that changes in relative magnesium concentrations correspond with reproductive events in females and breastfeeding in infants. Additionally, we observe seasonal calcium patterns in 77.3% of the samples.

Project description:Bacterial communities in water, soil, and humans play an essential role in environmental ecology and human health. PCR-based amplicon analysis, such as 16S rRNA sequencing, is a fundamental tool for quantifying and studying microbial composition, dynamics, and interactions. However, given the complexity of microbial communities, a substantial number of samples becomes necessary for analyses that parse the factors that determine microbial composition. A common bottleneck in performing these kinds of experiments is genomic DNA (gDNA) extraction, which is time-consuming, expensive, and often biased based on the types of species present. Direct PCR method is a potentially simpler and more accurate alternative to gDNA extraction methods that do not require the intervening purification step. In this study, we evaluated three variations of direct PCR methods using diverse heterogeneous bacterial cultures, including both Gram-positive and Gram-negative species, ZymoBIOMICS microbial community standards, and groundwater. By comparing direct PCR methods with DNeasy Blood and Tissue Kits for microbial isolates and DNeasy PowerSoil Kits for microbial communities, we found that a specific variant of the direct PCR method exhibits an overall efficiency comparable to that of the conventional DNeasy PowerSoil protocol in the circumstances we tested. We also found that the method showed higher efficiency for extracting gDNA from the Gram-negative strains compared to DNeasy Blood and Tissue protocol. This direct PCR method is 1,600 times less expensive ($0.34 for 96 samples) and 10 times simpler (15 min hands-on time for 96 samples) than the DNeasy PowerSoil protocol. The direct PCR method can also be fully automated and is compatible with small-volume samples, thereby permitting scaling of samples and replicates needed to support high-throughput large-scale bacterial community analysis. IMPORTANCE Understanding bacterial interactions and assembly in complex microbial communities using 16S rRNA sequencing normally requires a large experimental load. However, the current DNA extraction methods, including cell disruption and genomic DNA purification, are normally biased, costly, time-consuming, labor-intensive, and not amenable to miniaturization by droplets or 1,536-well plates due to the significant DNA loss during the purification step for tiny-volume and low-cell-density samples. A direct PCR method could potentially solve these problems. In this study, we developed a direct PCR method which exhibits similar efficiency as the widely used method, the DNeasy PowerSoil protocol, while being 1,600 times less expensive and 10 times faster to execute. This simple, cost-effective, and automation-friendly direct-PCR-based 16S rRNA sequencing method allows us to study the dynamics, microbial interaction, and assembly of various microbial communities in a high-throughput fashion.

Project description:In this work, the particulate matter (PM) from three different monitoring stations in the Monterrey Metropolitan Area in Mexico were investigated for their compositional, morphological, and optical properties. The main aim of the research was to decipher the different sources of the particles. The methodology involved the ex situ sequential analysis of individual particles by three analytical techniques: scanning electron microscopy-energy dispersive X-ray spectroscopy (SEM-EDS), polarized light microscopy (PLM), and micro-Raman spectroscopy (MRS). The microanalysis was performed on samples of total suspended particles. Different morphologies were observed for particles rich in the same element, including prismatic, spherical, spheroidal, and irregular morphologies. The sequential microanalysis by SEM-EDS/PLM/MRS revealed that Fe-rich particles with spherical and irregular morphologies were derived from anthopogenic sources, such as emissions from the metallurgical industry and the wear of automobile parts, respectively. In contrast, Fe-rich particles with prismatic morphologies were associated with natural sources. In relation to carbon (C), the methodology was able to distinguish between the C-rich particles that came from different anthopogenic sources-such as the burning of fossil fuels, biomass, or charcoal-and the metallurgical industry. The optical properties of the Si-rich particles depended, to a greater extent, on their chemical composition than on their morphology, which made it possible to quickly and accurately differentiate aluminosilicates from quartz. The methodology demonstrated in this study was useful for performing the speciation of the particles rich in different elements. This differentiation helped to assign their possible emission sources.
